Effects of Feeding Charcoal Powder and Vitamin A on Growth Performance, Serum Profile and Carcass Characteristics of Fattening Hanwoo Steers (활성탄과 비타민 A의 급여가 거세 한우의 성장, 혈액성상 및 도체 특성에 미치는 영향)

  • Effects of supplemental charcoal powder and vitamin A on growth performance, serum profile and carcass characteristics with 24 Hanwoo steers were inves$\ell$ated. The Hanwoo steers were randomly assigned to one of the three dietary treatments: 1) Control(commercial feed), 2) Tl(commercial feed supplemented with 2% charcoal powder and 0.2 % vitamin A), 3) T2(commercial feed with 2 % charcoal powder). In the age 26 month, body weight of control Tl and T2 were 633.8 kg, 621.0 kg, 619.4 kg, respectively, with not significantly different(P> 0.05). The daily weight gain of control was the highest followed by Tl and T2. The contents of vitamin A(retinol) and total cholesterol in the blood of Tl was higher Tl(56.32 $\ell$ / dt, 163.83 mg/ dz) than the other treatment groups(P < 0.05). The average of WBC value in the blood was 6.86-" 9.83 k / ${\mu}\ell$, RBC value was 7.98 - 11.26 k / ${\mu}\ell$ and blood platelet was 236.7 - 287.0 k / ${\mu}\ell$. But The serum profile in blood was not significantly different between control and treatment groups. The carcass grade and characteristics of all treatments in Hanwoo steers were not significantly different except the texture.

  • This study is an attempt to investigate the effects of the water extracts of the whole plants of Prunella vulgaris (Labiatae) and Thesii chinense (Santalaceae) on the acute toxicity, the activities on central nervous system and the diuretic action. The acute toxicities shown by $LD_{50}$ were estimated to be more than 3000 mg/kg p.o. and 1,000 mg/kg s.c. in the extracts of Prunellae Herba and Thesii Herba, respectively, in mice. The extracts at a dose of 2,000 mg/kg p.o. did not show any activities on central nervous system, i.e, sedative, analgesic, hypothermic and anticonvulsant actions. The urination in rats was increased by 45.5% and 57.6% when 100mg/kg of each of the extracts were given orally. The results obtained revealed that the water extracts possessed weak diuretic actions without any of central nervous system activities. Furthermore, it is considered that the potassium in the extract may play a role in the diuretic action.

  • Piezoelectric and dielectric properties as functions of x and y mole ratio in yPb(ZrxTil-x)O$_3$(1-y)Pb(Mn$\_$1/3/Nb/2/3/)O$_3$, ceramics, PZT-PMN, are investigated for large displacement piezoelectric devices. From the experimental results, when y is 0.95 and x is 0.505, the piezoelectric and dielectric properties are maximum, that is, electromechanical coupling coefficient(kp), piezoelectric strain constant(d$\_$33/), permittivity($\varepsilon$$\_$33/$\^$T//$\varepsilon$$\_$0/), and Curie temperature are 58 %, 272 pC/N, 1520 and about 350$^{\circ}C$, respectively. Also, when y is 0.90 and x is 0.50, their properties are 56 %, 242 pC/N, 1220, and 290$^{\circ}C$, respectively. As MgO dopant is added from 0 wt% to 1 wt%, kp increases to 63 % and Qm decreases to 500 at the MgO dopant of 0.1 wt%, and then kp decreases to 57 % as MgO is added.

  • The sucrose, MSG, fresh oyster and salted-fermented anchovy and shirmp were added into Kimchi in the concentration range of $1{\sim}3%$ and studied for their effects on the changes in chemicl properties and fermentation rate. The control Kimchi was prepared with addition of 2% red pepper, garlic, Welsh onion and 1% ginger. It was found that the fermentation rate was increased as the concentration of all of ingradients tested increased. Among them M.S.G affected most significantly. However, ascorbic acid formed was reduced for those Kimchi added with salted-fermented anchovy and shrimp while the changes in pH and titrable acidity was affected a little.

  • A bacterial strain with phytase and glucose-1-phosphatase activity was isolated from seawater. The colony was identified as an Enterobacter cloacae strain and named E. cloacae B11. A gene, agpEnB11, coding for an intracellular acid glucose phosphatase was cloned from the strain and sequenced. It comprised 1,242 nucleotides and encoded a polypeptide of 413 amino acids. Recombinant glucose-1-phosphatase (AgpEn) was overexpressed in Escherichia coli and purified using Ni-NTA column under native conditions. Purified protein displayed a single band of 47 kDa on SDS-PAGE. AgpEn hydrolyzed a wide variety of phosphorylated compounds, with high activity for glucose-1-phosphate and glucose-6-phosphate. Optimum pH and temperature for enzyme activity were pH 5.0 and $50^{\circ}C$, respectively. Enzyme activity was stimulated by $Ca^{2+}$ and $Co^{2+}$, and inhibited by $Cu^{2+}$.

The Effect of Docosahexaenoic Acid on Brain Function and Acetylcholine Level in Cerebral Cortex of Electroconvulsive Shock Induced Mice (Docosahexaenoic acid가 전기충격성 뇌장애 마우스의 기억력 및 Acetylcholine량 변화에 미치는 영향)

  • Electroconvulsive shock (ECS) increases the activity of acetylchohnesterase and decreases in brain acetylcholine levels. A large amount of free fatty acids accumulated in the brain tissue affects cerebral blood flow, brain edema and inflammation and results in brain injury. The present study examined the effect of docosahexaenoic acid (DHA) and D,L-pyroglutamic acid (D,L-PCA) on the learning and memory deficit using the passive avoidance failure technique and on the change of acetylcholine and choline level in the cerebral cortex of ECS-induced mice. The application of ECS (25mA, 0.5sec) induced a significant decrease in memory function for 30 min. ECS-induced a significant decrease in cortical acetylcholine and choline levels 1 min following the ECS application, which were almost recovered to ECS control level after 30 min. DHA (20 mg/kg, i.p.). administered 24 hr before shock. prevented the ECS-induced passive avoidance failure and the decrease of acetylcholine level 1 min following the ECS application. DHA failed to elicit a change in cortical choline level. DHA did not affect memory function and the cortical Ach and choline level of normal mice. The administration of D,L-PCA (500 mg/kg, i.p.) increased the effect of DHA on memory function and the change of cortical acetylcholine level of ECS induced mice. These results suggest that DHA treatment may be contributed to the prevention against memory deficit, and to the activation of cholinergic system in the ECS induced mice.

HPLC Analysis of Some Flavonoids in Citrus Fruits (조생온주 밀감의 품종에 따른 Flavonoids 함량 분석에 관한 연구)

  • New HPLC method was developed for determination of some flavonoids such as naringin, hesperidin, neohesperidin, rutin, quercitrin, naringenin, hesperetin and apigenin and their contents in citrus juice and citrus peel from citrus varieties Brown in Cheju. Detection was at 280nm and reverse phase ${\mu}$-Bondapak C-18 column was used. Water/methanol/acetic acid as the mobile phase was better than water/acetonitrile/acetic acid. Flavonoids were more stable in 20% n,n-dimethylformamide in methanol(20% DMF) than methanol and pH 12 adjusted by 1N-sodium hydroxide solution. Standard flavonoid solutions were injected three times consecutively and the reproduciability was 0.236 to 3.550%, Correlation coefficient of the calibration curve was 0.9946 to 0.9999. The exiraction efficiency of hesperidin from citrus peel was evaluated with different extraction method such as reflux, ultra-sonicating method, using three solvents (aqueous solutions with pH12 adjusted by 1N-sodium hydroxide, methanol and 20% DMF), respectively. The reflux for 4 hour in 20% DMF was the most efficient of the tested methods and solvents, and recovery percentage were 78.0∼130.0%. Flavonoids were determined in citrus juice. Naringin was 68.2mg/100$m\ell$ in Natsudaidai, Hesperidin were 85.6mg/100$m\ell$ in Sankyool and Neohesperidin was 25.3mg/100$m\ell$ in Dangyooja. Flavonoids were determined in citrus peel. Naringin was 110mg/g in Dangyooja, Hesperidin was 242mg/g in Hungjin and Neohesperidin was 87.9mg/g in Dangyooja.

DNA Sequencing and Expression of the Circumsporozoite Protein of Plasmodium vivax Korean Isolate in Escherichia coli

  • To obtain the recombinant circumsporozoite (CS) protein for the diagnosis of patients and seroepidemiology of Plasmodium vivax malaria which have been prevalent in northern part of Kyonggido, the CS protein gene was amplified by the polymerase chain reaction (PCR) from genomic DNA of the Korean vivax malaria patient. The gene consists of 1,123 nucleotides except signal peptide sequences and had an uninterrupted reading frame encoding a protein of 374 amino acids with a central region of 20 tandem repeats of the nonapeptide. The CS protein gene was expressed in Escherichia coli and purified, the molecular weight of recombinant CS protein was about 44 kDa (monomer) under denaturing purification and about 65 kDa (dimer) under native purification by SDS-PAGE. The purified recombinant CS protein which has antigenicity to malaria patients in Western blot analysis and Enzyme-linked immunosorbent assay, reacted only with the serum of P. vivax (PV210) infected malaria patients with no cross reaction to the P. falciparum malaria patient. The recombinant CS protein purified in this study will serve as a useful antigen to support the diagnosis of malaria patients and seroepidemiology.

Pharmacodynamic Interactions of Diazepam and Flumazenil on Cortical Eeg in Rats (흰쥐 대뇌피질의 뇌파에 대한 diazepam 및 flumazenil의 약력학적 상호작용)

  • Diazepam, a benzodiazepine (BDZ) agonist, produces sedation and flumazenil, a BDZ antagonist, blocks these actions. The aim of this study was to examine the effects of BDZs on cortical electroencephalogram (EEG) in rats. The recording electrodes were implanted over the frontal and parietal cortices bilaterally, and the reference and ground electrodes over cerebellum under ketamine anesthesia. To assess the effects of diazepam and flumazenil, rats were injected with diazepam (1 mgHg, i.p.) and/or flumazenil ( 1 mg/kg, i.p.), and the EEG was recorded before and after drugs. Normal awake had theta peak in the spectrum and low amplitude waves, while normal sleep showed large amplitude of slow waves. The powers of delta, theta and alpha bands were increased during sleep compared with during awake. Diazepam reduced the mobility of the rat and induced sleep with intermittent fast spindles and large amplitude of slow activity, and it produced broad peak over betaL band and increased the power of gamma band, which were different from EEG patterns in normal sleep. Saline injection awakened rats and abolished fast spindles for a short period about 2-5 min from EEG pattern during diazepam-induced sleep. Flumazenil blocked both diazepam-induced sleep and decreased the slow activities of delta, theta, alpha and betaL, but not of gamma activity for about 10 min or more. This study may indicate that decrease in power of betaL and betaH bands can be used as the measure of central action of benzodiazepines, and that the EEG parameters of benzodiazepines have to be measured without control over the behavioral state by experimenter.

  • This paper proposed an ESD protection device that improved the current driving ability through Self-bias than the existing ESD protection device. The new proposed structure is based on the LRSCR structure and adds an N+ diffusion region, and reduces resistance by connecting the gate and the P+ diffusion region. As a result, it was confirmed that the proposed ESD protection device exhibits a trigger voltage of 11.8V and a holding voltage of 5.9V. It can be used in 5V applications for low voltage and is expected to have excellent current driving capability.
