Effects of Low Incident Energy Levels of Infrared Laser Irradiation on the Proliferation of Candida Albicans Part II : A Short Term Study during the Cell Cycle

  • This study was performed to investigate the biostimulation effects of low level laser therapy (LLLT) on the fungus, Candida albicans, during the short term of cell cycle. Samples were divided into 6 groups which were P7, P9, P11, P15< CW and CO. All samples were irradiated for 1 minute with 2 hours of elapsed time during about 27 hours of the cell cycle of Candida albicans, and the optical density was assessed by spectrophotometry every 2 hours. It was found that there was no difference between the control and any other groups irradiated with 2 hours of short interval.

  • This study was performed to investigate the biostimulation effects of low level laser therapy (LLLT) on the fungus, Candida alvicans, according to the interval of irradiation during the cell cycle. Samples were divided into 5 groups which were P7-2Hr, P7-4Hr, P15-2Hr, P15-4Hr and Co. Sample was irradiated for 1 minute with 2 hours or 2 minutes with 4 hours of elapsed time during 28 hours of the cell cycle of Candida albicans, and the optical density was assessed by spectrophotometry every 2 hours. It was found that there was significant difference, in the cell growth, between 2 hour and 4 hour interval laser irradiation, although the total energy of each group using identical pulse during exam was equal.

  • CD133 is one of the most important stem cell markers in solid cancers and Ki-67 is a marker that reflects cell proliferation. The relationships between the expression of CD133 and Ki-67 and prognosis in gastric carcinoma are unknown and need exploring. We examined 50 gastric cancer patients retrospectively in the Radiation Oncology Department of the Faculty of Medicine, Gazi University. CD133 and Ki-67 expression was examined using immunohistochemical staining. The survival rate in patients with CD133 positive expression was significantly worse than that in the patients with negative expression (p=0.04). Expression of CD133 had a positive correlation with that of Ki-67 (r=0.350; p=0.014). Multivariate analysis revealed that the expression of CD133 was an independent prognostic factor in gastric cancer (p=0.02). Conclusion, expression of CD133 may be a useful prognostic marker in gastric cancer.

Salvia miltiorrhiza Inhibits Tumor Cell Growth in Association with Rb Dephosphorylation through Up-regulation of p21 Via a p53-dependent Pathway

  • Background: Salvia miltiorrhiza (SM), a traditional oriental medicine, has been reported to have anti-tumor properties, but its exact mechanism remains to be elucidated. In this study, we investigated several of the molecular events that occur in human breast carcinoma MCF-7 cells and human pulmonary adenocarcinoma A549 cells. Methods: For this purpose, we evaluated the growth-inhibitory effect of SM in association with the expressions of p53, p21, cyclin D1, and pRb, which are known to be involved in cell cycle arrest. The extent of thymidine incorporation was also examined to assess G1/S phase cell cycle arrest in both cells by $^3H$-thymidine incorporation. Results: Our results show that SM inhibits the growth and the proliferation of MCF-7 and A549 cells. Furthermore, we also observed increased expression of p21 via a p53-dependent pathway in both cell lines after treating with SM. In addition, treatment with SM for 24 hours caused the suppression of hyperphosphorylated retinoblastoma protein (pRb) expression and the dephosphorylation of pRb. Conclusion: These findings suggest that the growth inhibitory and the anti-proliferation effects of SM on MCF-7 cells and A549 cells are mediated via the decreased expression and dephosphorylation of pRB by p21 up-regulation in a p53-dependent manner. To the best of our knowledge, this study is the first to report upon the molecular mechanisms involved in SM-induced tumor cell growth inhibition.

Antiproliferative Effects of Native Plants on Prostate Cancer Cells

  • As part of the research for the natural products about prostate-related disease, this study screened 159 plant species from 46 families, which included a total of 213 different kinds of local native plants and these plants were tested for the ability to inhibit LNCaP proliferation, an androgen-sensitive prostate cancer cell line, and DU145 proliferation, which is a more aggressive androgen-insensitive prostate cancer cell line. The results indicated that nineteen of 213 types of plants exhibited antiproliferative activity (cell viability < 30%, $500{\mu}g/mL$) on the growth of androgen-sensitive LNCaP cell lines, and five of them exhibited DU145 cell antiproliferative activity (cell viability < 30%, $500{\mu}g/mL$). The methanol extracts of Eurya emarginata (stems), Gleditsia japonica var. koraiensis (leaves), Photinia glabra (leaves) and Elaeagnus macrophylla (leaves) showed antiproliferative activity on both the androgen-sensitive LNCaP cells (cell viability < 30%) and androgen-insensitive DU145 cells (cell viability > 100%). The study also found that the methanol extracts of Styrax japonica (fruits), Aralia continentalis (leaves), Fagus crenata var. multinervis (stems), Thuja orientalis (stems) and Poncirus trifoliate (branches) presented the strongest activity and demonstrated potent antiproliferative activity on both cell lines (LNCaP and DU145 cell viability < 30%).

  • In this study, the (p)ZnTe/(n)Si solar cell and (n)CdS-(p)ZnTe/(n)Si poly-junction thin film are fabricated by vaccum deposition method at the substrate temperature of $200{\pm}1^{\circ}C$ and then their electrical properties are investigated and compared each other. The test results from the (p)ZnTe/(n)Si solar cell the (n)CdS-(p)ZnTe/(n)Si poly-junction thin fiim under the irradiation of solar energy $100[mW/cm^2]$ are as follows; Short circuit current$[mA/cm^2]$ (p)ZnTe/(n)Si:28 (n)CdS-(p)ZnTe/(n)Si:6.5 Open circuit voltage[mV] (p)ZnTe/(n)Si:450 (n)CdS-(p)ZnTe/(n)Si:250 Fill factor (p)ZnTe/(n)Si:0.65 (n)CdS-(p)ZnTe/(n)Si:0.27 Efficiency[%] (p)ZnTe/(n)Si:8.19 (n)CdS-(p)ZnTe/(n)Si:2.3 The thin film characteristics can be improved by annealing. But the (p)ZnTe/(n)Si solar cell are deteriorated at temperatures above $470^{\circ}C$ for annealing time longer than 15[min] and the (n)CdS-(p)ZnTe/(n)Si thin film are deteriorated at temperature about $580^{\circ}C$ for longer than 15[min]. It is found that the sheet resistance decreases with the increase of annealing temperature.

Sodium Butyrate Alters Cell-Cell Interactions through Up-Regulation of E-Cadherin in Human Hepatocellular Carcinoma Cells (Sodium butyrate에 의한 E-cadherin의 발현증가와 세포간 상호작용의 변화)

  • Sodium butyrate (NaBt), a naturally occurring short chain fatty acid derived from carbohydrate metabolism in the gut, is known to exhibit strong anti-cancer potentials in various human cancer cells; however, its action mechanism is poorly understood. In the present study, we demonstrated that NaBt up-regulates levels of E-cadherin, a key cell adhesion molecule implicated as a tumor suppressor, in a cell type-specific manner. Although levels of p21, a potential activator for E-cadherin expression, were also up-regulated by treatment with NaBt in several types of cells, it does not seem to be associated with the activation of E-cadherin in the NaBt-treated cells. Instead, the data from promoter analysis suggest that NaBt up-regulates expression of E-cadherin at the transcription level by enhancing its promoter strength via a CCAAT-box. The elevated E-cadherin in the presence of NaBt was primarily localized at the cell-cell contacts, converting Hep3B cells into a more differentiated form.

Development of Somatic Cell Nuclear Transfer Embryos Following Donor Cell Type and Cell Treatment in Cattle (Donor 세포의 종류 및 세포처리에 따른 소 체세포 핵이식란의 체외발육)

  • This study was conducted to investigate the effects of donor cell type, individual, passage number and trypsinization time on the in vitro development of bovine somatic cell nuclear transfer embryos. Three cell types (skin, muscle and cumulus cells) and cells from 3 individuals were used for nuclear transfer. Cell were passaged by 5, 15 or 30 times, and cell were trypsinized for 1 or 3 min before injection. Nuclear transfer were performed by conventional fusion method. Development rates to the blastocyst stage were not significantly different among three cell types (16.5∼23.9%) and individuals (16.4∼19.5%). Blastocyst formation rate of cloned embryos reconstituted with cells at passage 30 (5.8%) was significantly lower than those of embryos reconstituted with 5- and 15-passaged cells (25.3 and 23.5%, respectively, P<0.05). The rate of embryos developed to the blastocyst stage was higher in embryos reconstituted with cells trypsinized for 1 min (30.7%) compared to embryos reconstituted with cells trypsinized for 3 min (P<0.05). The result of the present study indicates that different donor cell types and individuals used in this study did not affect the development of cloned bovine embryos. However, passage number and trypsinization time of donor cells affect the in vitro development of cloned bovine embryos.

Effect of Conditioned Medium of Human Endothelial Cell Line(tHUE-2 cell) on In Vitro Development of Mouse 1-cell Embryos In Vitro Fertilized (체내 수정된 Mouse 1-세포배의 체외발생에 미치는 혈관내피세포주(tHUE-2세포) 배양액의 영향)

  • Culture medium (ASF-301) of tHUE-2 cell, human endothelial cell line, and culture medium of these cells (conditioned medium : CM) which affect embryonic development of in vivo fertilized 1-cell embryos of mouse were examined. Two-cell stage block of mouse embryos was overicomed in ASF-301 and CM without EDTA, which usually added in basic medium (modified Whitten Medium: MWM, control) to overcome the 2-cell stage block. The developmental rates of embryos to the blastocyst stage were significantly increased in MWM containing 12.5% of growth factors added to ASF-301 (10mg/ $\ell$ transferrin, 1mg/$\ell$ insulin, 0.01mg/$\ell$ EGF) than those of 100% addition and control, 78.0% vs 20.8 and 52.3%, respectively (P<0.05), but the growth factors was not affected the hatching rate of blastocyst. Using ASF-301 or CM which was not treated, embryonic development into the blastocyst and hatched blastocyst stages were not affected. However, proportions of embryonic development into the blastocyst and hatched blastocyst stages were significantly higher in dilution (ASF-301 1:10; CM 1:3~1:6) than those in control (P,0.05). In ASF-301 dialyzed M.W.<10000 dialysis membrane, the developmental rate upto the hatched blastocyst stage was significantly increased, compared to ASF-301 which was not dialyzed (P<0.05), and hatching rate of blastocyst of these group was singnificantly increased than those in MWM (P<0.05). Compared to CM which was not dialyzed, however, in dialyzed CM was significantly decreased, compared to untreated CM (P<0.05), especially any hatched blastocyst was not appeared. As a result of these experiments indicated that a kind or porper treatment such as a dilution of complex synthetic cell culture medium and conditioned medium, and that a optimal concentration of growth factors are usuful for embryo cultrue in vitro.

  • N+-P/P+solar cells were fabricated by using the polycrystalline silline wafer with the resistivity of 3-6 ohm-cm. minority carrier lifetimes, measured by Nd: YAG laser, were from 100ns up to 150ns. Conversion efficiency measured under AM 1 irradiation, were about 4%.
