Effects of Dietary Supplementation of Garlic by-products on Total Phenol Contents, DPPH Radical Scavenging Activity, and Physicochemical Properties of Chicken Meat (마늘 부산물을 사료에 첨가 급여한 계육의 총페놀함량, 전자공여능 및 이화학적 특성)

  • This study was conducted to investigate the effects of dietary supplementation of garlic by-products on TBARS, WHC (water holding capacity), shear force, pH, total phenol content, DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity, meat color, sensory evaluation, and fatty acid composition of chicken meat. Broiler chicks were fed for 5 wk with experimental diets of 0% garlic by-product (Control), 1% garlic by-product (T1), 2% garlic by-product (T2), and 5% garlic by-product (T3). TBARS and pH were significantly decreased by the supplementation of garlic by-products compared to the control (p<0.05). Compared to the control diet, the total phenol content and DPPH radical scavenging activity were significantly increased by the supplementation of garlic by-products (p<0.05). The total phenol content and DPPH radical scavenging activity of treatment groups were higher than the control; in particular, T3 was significantly (p<0.05) more effective in improving freshness compared to other treatment groups. CIE $a^*$ value of treatment groups (especially T3) showed significantly higher values compared to the control; however, no difference in the CIE $L^*$ and $b^*$ values were observed among treatments. In its fatty acid composition, amounts of linoleic acid and linolenic acid in chicken meat was increased by the supplementation of garlic by-products, but amounts palmitic acid were decreased. In conclusion, supplementation with garlic by-products was effective in decreasing TBARS, pH, and saturated fatty acids, and in increasing total phenol content, DPPH radical scavenging activity, and unsaturated fatty acids.

  • Objective: Human mesenchymal stromal cells (MSCs) exhibit variable differentiation potential and can be divided accordingly into distinct subpopulations whose ratios vary with donor age. However, it is unknown whether the same is true in pigs. This study investigated MSC subpopulations in miniature pig and compared their characteristics in young (2 to 3 months) and adult (27 to 35 months) pigs. Methods: Osteogenic, chondrogenic, and adipogenic capacity of isolated MSCs was evaluated by von Kossa, Alcian blue, and oil red O staining, respectively. Cell surface antigen expression was determined by flow cytometry. Proliferative capacity was assessed with the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. Expression of marker genes was detected by quantitative real-time polymerase chain reaction. Results: Porcine MSCs comprised cells with trilineage and bilineage differentiation potential (tMSCs and bMSCs, respectively) and non-differentiating stromal cells (NDSCs). The tMSC and bMSC fractions were smaller in adult than in young pigs (63.0% vs 71.2% and 11.6% vs 24.0%, respectively, p<0.05); NDSCs showed the opposite trend (25.4% vs 4.8%; p<0.05). Subpopulations showed no differences in morphology, cell surface antigen expression, or proliferative capacity, but octamer-binding transcription factor 4 (OCT4) expression was higher in tMSCs than in bMSCs and NDSCs (p<0.05), whereas sex determining region Y-box 2 (SOX2) expression was higher in tMSCs and bMSCs than in NDSCs (p<0.05). Aging had no effect on these trends. Conclusion: Porcine MSCs comprise distinct subpopulations that differ in their differentiation potential and OCT4 and SOX2 expression. Aging does not affect the characteristics of each subpopulation but alters their ratios.

Identification of Novel SNPs with Effect on Economic Traits in Uncoupling Protein Gene of Korean Native Chicken

  • The avian uncoupling protein (avUCP) is a member of the mitochondrial transporter superfamily that uncouples proton entry in the mitochondrial matrix from ATP synthesis. The sequencing analysis method was used to identify nucleotide polymorphisms within the avUCP gene in Korean native chicken (KNC). This study identified ten single nucleotide polymorphisms (SNPs) in the avUCP gene. We analyzed the SNPs of the avUCP gene to investigate whether polymorphism in the gene might be responsible for quantitative variations in economic traits in KNC. Three significant polymorphic sites for economic traits were avUCP C+282T (mean body weight, p<0.05), avUCP C+433T (daily percent lay, p<0.05), and avUCP T+1316C (daily percent lay, p<0.05). The frequency of each SNP was 0.125 (C+282T in avUCP gene exon 1 region), 0.150 (C+433T in avUCP gene intron 1 region), and 0.15 (T+1316C in avUCP gene exon 3 region), respectively. Among the identified SNPs, one pair of SNPs (genotype CC, C+282T and TT, avUCP C+433T) showed the highest daily percent lay (p<0.05) and mean body weight (p<0.05) and the frequency was 0.067. This study of the avUCP gene could be useful for genetic studies of this gene and selection on economic traits for KNC.

Changes in the Quality of Beef Jerky Containing Additional Red Wine (적포도주를 첨가한 우육포의 저장 중 품질 변화)

  • This study was carried out to investigate the effect of the addition of red wine on the color, water activity, hardness, pH, 2-thiobarbituric acid (TBARS) value, volatile basic nitrogen (VBN) content and total plate count of beef jerky during storage at room temperature for 4 weeks. Beef jerky was prepared using three variations: beef jerky containing 50 mL water (T0), beef jerky containing a combination of 25 mL water and 25 mL red wine (T1) and beef jerky containing 50 mL red wine (T2). The lightness ($L^*$) was the lowest at the 4-week storage period, and the $L^*$ value of T1 and T2 were lower than that of T0 (p<0.05). The redness ($a^*$) tended to decrease during storage, with the $a^*$ value of T1 and T2 being higher than that of the T0 (p<0.05). The yellowness ($b^*$) during storage was not changed, and the $b^*$ value was not significantly different among the different samples tested. The water activity decreased during storage and T0, T1 and T2 was 0.57, 0.57 and 0.60 after 4 weeks of storage, respectively(p<0.05). The hardness increased during storage and the hardness of T2 stored for 4 weeks was lower than that of T0 and T1 (p<0.05). The pH was decreased with an increase in storage time (p<0.05). The TBARS value increased during storage and the TBARS value of T1 and T2 stored for 4 weeks was lower than that of T0 (p<0.05). The VBN content increased during storage and T2 stored for 4 weeks showed the lowest (p<0.05). The total plate count tended to increase during storage, and T0 stored for 4 weeks showed the highest (p<0.05).

Effect of Jujube Methanol Extract on Benzo(a)pyrene Induced Hepatotoxicity (대추 메탄올 추출액이 Benzo(a)pyrene에 의해 유도된 간기능 장해에 미치는 영향)

  • The protective effect of jujube methanol extract on benzo(a)pyrene(B(a)P)-induced liver injury was studied in rats in vitro and in vivo. Jujube methanol extract significantly recovered the enzyme activities(GOT, GPT, LDH and ALP) and lipid contents(total cholesterol, triglyceride and HDL-cholesterol) changed by B(a)P to normal levels in vivo. in viro experiment jujube methanol extract didn't stimulate hepatocyte proliferation but significantly recovered the enzyme activities(GOT, GPT and LDH) in comparison to group Ⅱ administered B(a)P only. It was suggested that jujube methanol extract have a protective effect on liver injury by B(a)P.

  • p-(2,2,3,3-Tetracyanocyclopropyl)phenyl acrylate (3a) and p-(2,2,3,3-tetracyanocyclopropyl)phenyl methacrylate (3b) were prepared by the reactions of bromomalononitrile with p-acryloyloxybenzylidenemalononitrile (2a) or p-methacryloyloxy-benzylidenemalononitrile (2b), respectively. Compounds 3a and 3b were polymerized with free radical initiators to obtain the polymers with multicyano functionalities in the cyclopropane ring. The resulting polymer 4a was soluble in acetone but the polymer 4b was not soluble in common solvents. The inherent viscosities of polymers 4a were in the range of 0.10-0.15 dL/g in acetone and those of 4b were in the range of 0.20-0.30 dL/g in 98% sulfuric acid. Solution-cast films were cloudy and brittle, showing $T_g$ values in the range of 106-125$^{\circ}$C.

  • Benzo(a)pyrene(B(a)P), an extensively studied polycyclic aromatic hydrocarbon(PAH), is a common contaminant produced through the burning of fossil fuels, particularly coal, and from the exhaust products of internal combustion engines. It produces a wide range of toxicities, including carcinogenicity in experimental animals. B(a)P has been shown to suppress systemic immunity in experimental animals, which may contribute to the growth of the chemical-induced tumors. Using colorimetric MTT assay natural killer(NK) cell-mediated growth inhibition of tomor cell was measured in normal and B(a)P-exposed C57BL/6 mice. Non-adherent splenocytes of normal or B(a)P-exposed mice were cultured with Yac-1 cells at four different effector/target(E/T) cell ratios ranging from 200/1, 100/1, 50/1, and 25/1 in an assay volume of 0.1 ml. After the optical density of culture wells containing MTT solution was measured at a wavelength of 540 nm, the percentage of dead cells relative to the control target cell number was calculated. The NK activity of B(a)P-exposed mice was markedly lower than that of non-exposed mice group at all E/T ratios. These results indicated that suppression of NK cell activity may play a role in allowing for the growth of tumors.

Effects of Ad libitum and Restricted Feeding of Concentrates on Body Weight Gain, Feed Intake and Blood Metabolites of Hanwoo Steers at Various Growth Stages (배합사료의 자유 및 제한 급여가 거세한우의 성장단계별 증체, 사료섭취량 및 혈중 대사물질에 미치는 영향)

  • Two hundred fifty eight Hanwoo steers were used in a completely randomized design experiment to determine the effects of ad libitum or restricted feeding of concentrates on body weight(BW) gain, feed intake, blood metabolites and hematological parameters. Steers were assigned at 6 months of age to feeding groups of ad libitum(T1) or restricted(T2) by 18 months of age. Steers in both groups were fed ad libitum from 19 months of age. The restrictive feeding levels were 1.2-1.5% of BW for the growing period and 1.7-1.8% of BW for the early fattening period. Average daily gains were significantly higher in T1 than in T2 from 10 to 14 months of age, but were significantly higher in T2 than in T1 from 20 to 24 months of age(p<0.05). Total dry matter intake(DMI) was higher in T1 than in T2 at 10, 12 and 16 months of age(p<0.05). Total DMI of T2 was higher than that of T1 at 22 months of age(p<0.05). Feed conversions were significantly lower in T2 than in T1 from 20 to 30 months of age(p<0.05). Blood albumin concentrations were significantly higher in T2 than in T1 at 12, 14, 16 and 18 months of age. Blood triglyceride concentrations were significantly higher in T1 than in T2 at 14 and 16 months of age(p<0.05). Blood inorganic phosphorus concentrations were significantly higher in T2 compared with T1 at 8, 10, 16 and 22 months of age(p<0.05). Mean corpuscular volume and mean corpuscular hemoglobin were significantly lower in T2 than in T1 from 8 to 12 months of age(p<0.05), but those were significantly higher in T2 than T1 from 10 months to 12 months of age(p<0.05). Present results may indicate that the restricted feeding for the growing period does not show adverse effects on body weight gain with better feed conversion for the following late fattening period.

  • A study was made to find out a new method of calculating the survival rate of a fish population from length composition and growth equation. 1. Evaluation for the simultaneous Removal of Organic Matters and Nutrients by the RBC and tapered Aeration Processes with Bacillus sp. for the high Strength of Dairy Wastewater (바실러스 미생물을 이용한 고농도 유가공 폐수처리에 있어서 유기물질과 영양염류의 동시제거에 대한 평가)

  • The evaluation of organic matters and nutrients removal was investigated for the synthetic wastewater and the high strength of dairy wastewater. Two different systems were performed for this research. System A composing of a single RBC with tapered aeration was fed with the synthetic wastewater for 74 days with 173L/day of influent, 200% of internal return and 100% of sludge return for the period 1 and 2. The feed conditions were maintained 346L/day of influent, 50% of internal return and sludge return for the period 3. The dairy wastewater was introduced to evaluate treatment efficiency for system B composing of dual RBCs and tapered aeration tanks for 50 days of experimental run time, in which hydraulic rates were maintained at the constant ratios of 346L/day, 50% of internal return and 50% of sludge return. The spiral string media made of nylon fibre was attached by Bacillus sp. in RBC for both systems. The specific area of string media was $1.4m^2$/m and biomass was maintained at the concentrations of 23g/m. The synthetic wastewater was supplied by 1,800mg/L of glucose, 500mg/L of $NH_4Cl$, and 500mg/L of $KH_2PO_4$ to system A. The dairy wastewater was supplied to system B from dairy production plant. The average influent concentrations were 1,334mg/L of BOD, 2,014mg/L of CODcr, 160mg/L of T-N, and 12mg/L of T-P in system A. The average influent concentrations of parameters were 1,006 mg/L for BOD, 1,875mg/L for $COD_{cr}$, 51.6mg/L for T-N and 8.9mg/L for T-P in system B. Results indicated that removal efficiencies of BOD and $COD_{cr}$ were more than 90% however, the removal efficiency of T-N was 87%, and that of T-P was 82% for system A. Removal efficiencies were 98.5% of BOD, 91.3% of nitrogen and 89% of phosphorus for system B. The removal efficiencies of organic matters, T-N and T-P were higher in system B than in system A. The effluent quality issued by the stringent national legislations for the discharge of the high strength of dairy products wastewater can be improved using sequential RBCsand tapered aeration reactors rather than a single RBC and tapered aeration reactors with Bacillus sp.
