척추 수술 전 노인환자에게 적용한 아로마 향흡입요법이 통증 및 불안에 미치는 효과 (The Effect of Aroma Inhalation on the Preoperative Pain and Anxiety of the Elderly Preparing to Undergo Spine Surgery)

  • 본 연구는 아로마 향흡입요법이 척추 수술 노인환자의 수술 전 통증과 불안에 미치는 효과를 검증하고자 시도한 비동등성 대조군 전후시차설계의 유사실험연구이다. 실험군(n=30)에는 프랑킨센스, 마조람, 만다린을 혼합한 향유를 수술 전일 오후 8~9시와 수술 당일 수술실 가기 1시간 전에 흡입시켰고, 대조군(n=30)에는 일반적인 수술 전 간호를 하였다. 두 군의 통증, 상태불안, 활력징후를 사전과 사후에 측정한 결과, 실험군과 대조군에서 통증(t=-1.223, p=.226), 수축기혈압(t=-0.211, p=.833), 이완기혈압(t=0.638, p=.526), 호흡(t=0.734, p=.466)은 통계적으로 유의한 차이가 없었으나, 상태불안(t=-3.202, p=.002)과 맥박(t=-0.213, p=.037)은 실험군에서 대조군에 비해 유의한 차이가 있었다. 본 연구를 통해 아로마 향흡입요법이 수술 전 환자의 불안 완화에 적용 가능한 독자적 간호중재로 적용될 수 있다.

Increase of Yeast Survival under Oxidative Stress by the Expression of the Laccase Gene from Coprinellus congregatus

  • Coprinellus congregatus secreted a laccase isozyme when the culture was transferred to an acidic liquid medium (pH 4.1). The laccase cDNA gene (clac2) was used as a probe for cloning of the genomic laccase gene (lac2) including the promoter (Plac2). The open reading frame (ORF) of lac2 had 526 deduced amino acids and four conserved copper binding domains as other fungal laccases. Recombinant plasmid (pRSlac2p-cDNA) of lac2 cDNA with its own promoter was transformed in Saccharomyces cerevisiae. Expression of the transformed lac2 gene was induced by oxidative stress ($H_2O_2$) in yeast and the survival rate of the transformed yeast strain was greatly increased when compared with that of the control strain transformed with pRS316 yeast vector.

  • This observation was carried out to investigate the inorganic constituents of dental calculus of varying locations and different ages. Supragingival calculus was obtained from 540 patients with the periodontal disease. The results were as follows : 1. The weight loss rate in ashing by aging was the highest in the 2nd decade (63.7%) and followed in order by 3rd decade (60.8%), seventh decade 959.8%), fourth decade (55.9%), sixth decade (52.6%) and fifth decade (43.2%). 2. The weight loss rate by ashing was more prominent in the buccal surfaces of the upper posterior teeth (62.0%) than in the lingual surface of the lower anterior teeth (59.7%). 3. The difference in contents of the inorganic constituents by sex was not remarkable. (male, Ca:373.0 P:333.9, female, Ca: 380.2, P: 339.6 ㎍/mg dry weight) 4. In the dry calulus, contents of the inorganic constituents were as follows: Ca:325.8 P:269.10 Mg:1.21 Na:8.44 K:1.32, Zn:0.67 ㎍/mg. 5. The Ca/P ratio was the lowest in the upper anterior region (1.11) and the highest in the lower posterior region (1.29) and the average was 1.20.

손바닥선인장 복합물이 Streptozotocin으로 유발된 당뇨 쥐의 지질대사에 미치는 효과 (Effects of Opuntia ficus-indica Complex on Lipid Metabolism in Streptozotocin-induced Diabetic Rats)

  • This study was conducted to examine the effects of Opuntia ficus-indica complex (OF) on the lipid metabolism, bile acid in feces, alanine aminotransferase (ALT) activity, aspatate aminotransferase (AST) activity, composition of urine and expression of cholesterol related mRNA in streptozotocin (STZ)-induced diabetic rats. Thirty two male Sprague-Dawley rats were randomly divided into non-diabetic control (NC), diabetic control (DC), diabetic OF of 2% (OF-2) and diabetic OF of 5% (OF-5), then each group was fed for 3 weeks. Plasma total cholesterol, non-esterified fatty acids (NEFA), total cholesterol, low density lipoprotein (LDL), very low density lipoprotein (VLDL) were decreased significantly (p<0.05) in OF-5 group compared to DC, but high density lipoprotein (HDL) was not changed. AST and ALT were also reduced and bile acid excretion was improved. Composition of urine in OF-5 was almost same in NC. The expression of cholesterol $7{\alpha}$-hydroxylase (CYP7A1), 3-hydroxy-3-methylglutaryl-coenzyme A reductase (HMG-CoA-R), Low density lipoprotein receptor (LDL-R) mRNA indicated that feeding OF have the effects of cholesterol decreation in plasma by synthesis of bile acid from cholesterol. These results provide experimental evidence about improved lipid metabolism of the OF feeding in the STZ-induced diabetic rats.

Genetic Analysis of a Structural Motif Within the Conserved 530 Stem-Loop of Escherichia coli 16S rRNA

  • The 530 stem-loop is a 46 nucleotide stem-loop structure found in all small-subunit ribosomal RNAs. Phylogenetic and mutational studies by others suggest the requirement for Watson-Crick interactions between the nucleotides 505-507 and 524-526 (530 pseudoknot), which are highly conserved. To examine the nature and functional significance of these interactions, a random mutagenesis experiment was conducted in which the nucleotides in the proposed pseudoknot were simultaneously mutated and functional mutants were selected and analyzed. Genetic analysis revealed that the particular nucleotide present at each position except 524 was not exclusively critical to the selection of functional mutants. It also indicated that basepairing interactions between the positions 505-507 and 524-526 were required for ribosomal function, and much weaker base-pairing interactions than those of the wild-type also allowed high ribosomal function. Our results support the hypothesis that the 530 pseudoknot structure may undergo a 'conformational switch' between folded and unfolded states during certain stages of the protein synthesis process by interacting with other ligands present in its environment.

영지버섯 백색 변이주의 형태적 특성 및 최적 배양조건 (Morphological Characterization and Culture Conditions of A White Mutant of Ganoderma Iucidum)

  • A morphologically different form of Ganoderma lucidum was isolated from a cultivator's farm, and its optimum growth conditions were determined. A major difference in their morphology was color of fruit bodies. Fruit bodies of the mutant were white wherase those of normal Ganoderma lucidum were red. Spores of the mutant were global and mycelia were thin. Mycelial growth of this white mutant was favorable on potato sucrose agar medium, and optimum pH of the medium was 5.5.

461-nm LED조사와 산의 병행처리가 Staphylococcus aureus와 Escherichia coli 생육에 미치는 영향 (Effects of the 461-nm LED Light and Combination with Acid Stress Treatment on Staphylococcus aureus and Escherichia coli)

  • 이 연구는 $15^{\circ}C$에서 10시간동안 pH 5 환경과 461-nm LED를 조사하였을 때 S. aurues (MRSA 포함), E. coli (E.coli O157:H7 포함)의 살균 효과를 측정하고자 수행하게 되었다. 461-nm LED만을 10시간동안 조사하였을 때. S. aureus (MRSA 포함)은 4 log CFU/mL 감소하였다. 그리고 E. coli (E. coli O157:H7 포함)는 9시간동안 조사하였을 때 불활성화되었다. 그러나 pH 5와 461-nm LED를 함께 조사한 결과, 모든 균주들은 7시간동안 조사하였을 때 불활성화되었다. 이 가시광선 불활성화 결과는 외인성의 광감각제없이 이루어 졌다. 461-nm LED만을 조사하였을 때, 가장 높은 D값은 S. aureus (ATCC 27664)으로 5.05시간이었으며 가장 낮은 D값은 E. coli O157: H7 (ATCC 35150)으로 1.39시간이었다. pH 5와 461-nm LED를 함께 조사한 결과, 가장 높은 D값은 S. aureus (ATCC 19095)의 1.58시간이었으며, 가장 낮은 D값은 S. aureus (ATCC 27664)의 0.83시간이었다. 본 연구 결과는 pH 5와 461-nm LED를 함께 조사한 것이 461 nm LED 한가지 환경보다 살균효과가 증가되었다는 것을 보여준다.

흰쥐 초기배아 발생과정 중 착상부위 분화에 미치는 Progesterone의 영향 (The Effect of Progesterone on Differentiation of Implantation Sites during Early embrynic development of the Rat)

  • 포유류 배아의 착상기작을 규명하고자 본 연구에서는 착상시기에 수정된 배아가 착상부위를 어떻게 인식하는 지 그리고 착상부위의 분화유발요인이 무엇인지를 조사하였다. 착상시기(임신 제6일)와 착상후 초기배아 발생기간(임신 제9일)중 혈청내 estradiol (E2)과 Progesterone(P4)의 농도를 측정하였으며, 자궁내막 조직을 착상부위 (antimesome trium)와 비 착상부위(mesometrium)로 분리하여 P4의 수용체 농도를 측정하였다. 혈청내 E2의 농도는 임신 제6일군에서 E2처리군에서 가장 높았으며, 임신 제9일에서는 intact 대조군에서 가장 높고 제9일의 모든 실험군의 농도는 임신 제6일군보다 높았다. 혈청내 P4 농도는 임신 제6일과 9일에서 다같이 대조군에서 가장 높으며 처리군 중에서는 P4처리군에서 높아지고 있다. P4 농도 역시 임신 제9일의 모든 실험군에서 제6일의 실험군보다 높았다. P4수용체 농도는 착상부위가 비 착상부위보다 높으며, 대조군(P < 0.01)과 P처리 군(P < 0.05)에서는 유의한 차이로 비 착상부위보다 높았다. 자궁내막조직 착상을 위한 분화에는 P4의 영향이 크며 P4의 혈청내 농도와 핵의 수용체 농도는 모든 실험군에서 상응하는 관계를 나타내었다. 이 결과는 앞서 일차적으로 발표한 알카리성 phosphatase (ALPase) 활성과도 상응하는 것이다.

Large deformations of a flexural frame under nonlinear P-delta effects

  • In this paper, nonlinear P-delta effects are studied on the seismic performance, and the modal responses of a flexural frame, considering large deformations. Using multiple scales method, the nonlinear differential equations of motion are estimated, and the nonlinear interactions between the frame's degrees of freedom are outcropped. The results of time and frequency domain analyzes of a dynamic model are examined under internal resonance cases, and the linear and nonlinear responses are investigated in each modal cases. Also, changing the modal responses with respect to the amplitude and frequency of the harmonic forces is evaluated. It is shown that the dominant absorption of energy is in the first natural frequency of the frame, in the case of earthquake excitation, and when a harmonic force is applied to the frame, the peaks of the frequency domain responses depending on the frequency of harmonic force are in the first, and second or third natural frequency of the structure.

합천호 수질 및 영양단계에 대한 평가 (Estimation of Water Quality and Trophic State in the Hapcheon Lake)

  • 호수 유입 하천인 황강의 $1996{\sim}2001$년 년도별 평균 수질은 COD $3.1{\sim}4.2\;mg/L$, T-N $2.460{\sim}3.550\;mg/L$, T-P $0.111{\sim}0.201\;mg/L$ 이였다. 호수내의 평균 수질은 COD가 1996년 1.9 mg/L에서 2001년 2.7 mg/L로 매년 조금씩 증가하는 것으로 나타났다. 호수내의 $1996{\sim}2001$년 년도별 평균 T-N 농도는 $1.383{\sim}1.792\;mg/L$이었으며, T-P농도는 $0.018{\sim}0.023\;mg/L$이였다. 식물성 플랑크톤의 발생 정도를 나타내는 Chl-${\alpha}$와 T-N, T-P와의 상관계수는 각각 0.382, 0.372였으며, 또한 수온과 강우량에 대한 상관계수는 각각 0.589, 0.526이였다. 따라서 합천댐의 경우 식물성 플랑크톤의 발생이 T-N, T-P보다 수온 및 강수량과 더 밀접한 관계가 있는 것으로 나타났다. 그리고 호수의 영양단계는 중영양${\sim}$부영양단계에 있는 것으로 평가되었다.
