Comparison of Photorhabdus luminescens and Vibrio fischeri lux Fusions to Study Gene Expression Patterns

  • A comparison of promoter fusions with the luxCDABE genes from Vibrio fischeri and Photorhabdus luminescens was made using promoters from several genes (katG, sodA, and pqi-5) of E. coli that are responsive to oxidative damage. The respective characteristics, such as the basal and maximum bioluminescence and the relative bioluminescence, were compared. E. coli strains carrying fusions of the promoters to P. luminescens lux showed higher basal and maximally induced bioluminescent levels than strains carrying the same promoter fused to the luxCDABE genes from V. fischeri. The sensitivities between the strains were similar, regardless of the luciferase used, but lower response ratios were seen from strains harboring the P. luminescens lux fusions. Furthermore, using the two katG::lux fusion strains, the bioluminescence from the P. luminescens lux fusion strain, DK1, was stable after reaching a maximum, while that of strain DPD2511 decreased very rapidly due to substrate limitation.

Growth Optimization of Photorhabdus luminescens Isolated from Entomopathogenic Nematode Heterorhabditis bacteriophora (병원성 선충 Heterorhabditis bacteriophora에서 분리된 공생 박테리아 Photorhabdus luminescens의 생장조건)

  • The yield of infective juveniles of Heterorhabditis bacteriophora (Tf strain) in vitro monoxenic liquid culture was improved significantly as the amount of symbiont biomass, Photorhabdus sp. strain Tf, increased. To investigate the influence of abiotic factors on the growth and biomass production of Photorhabdus sp. strain Tf, triplicate flask cu1tmes were performed. The optinal temperature and medium pH for the growth of Photorhahdus sp. strain Tf were 30$^{\circ}$C and between pH 5.5-7.3, respectively. Aeration also improved greatly growth and yield of biomass of Photorhabdus sp. strain Tf. Photorhabdus sp. strain Tf in batch fermentation showed growth-associated pattem in terms of pigment production, and the pH of culture medium rose steadily until growth stopped dUling the fermentation. Both pigment production and culture pH rise would be useful parameters indicating a reliable growth of Photorhabdus sp. strain Tf.

Comparison of Marine Luminescence Bacteria and Genetically Modified Luminescence E. coli, for Acute Toxicity of Heavy Metals (재조합 발광대장균과 해양 발광 미생물을 이용한 중금속 급성독성평가)

  • The responses of two luminescence-based biosensors were studied on various heavy metals in aqueous solutions. One was recombinant E. coli ($DH5{\alpha}$/pSB311), genetically modified luminescence-based bacteria, and the other was Vibrio fisheri used for the LumisTox system. The recombinant E. coli was marked with the lux CDABE gene from multicopy plasmid, pACYC184, originally isolated from Photorhabdus luminescens. The $DH5{\alpha}$/pSB311 had a characteristic of no organic substrate for its luminescence reaction. Among the tested heavy metals Zinc and cadmium were less toxic than copper and mercury. The recombinant E. coli was more sensitive to toxicity of heavy metals than the LumisTox. The order of toxicity of the heavy metals to the recombinant E. coli was $Hg^{2+}>Cu^{2+}>Zn^{2+}>Cd^{2+}$. In case of the LumisTox, the order of the toxicity of heavy metals was $Hg^{2+}>Cu^{2+}>Cd^{2+}>Zn^{2+}$. The genetically modified luminescence-based biosensor offers a range of sensitive, rapid, and easy to use methods for assessing the potential toxicity of heavy metals in aqueous samples.
