Interspecific Relationships within the Fungal Genus Pleurotus by Isozyme Analysis (동위효소 분석에 의한 느타리속의 종간 유연관계)

  • Thirty six strains of Pleurotus spp., from world-wide nations, were examined for interspecific isozyme variation. A comparison of isozymes in mycelial extracts of the fungal genus Pleurotus was made by polyacrylamide gel isoelectric focusing. A total of one hundred and sixty six bands was resolved from six isozymes. A cluster analysis was done based on the zymograms for esterase, glucosephosphate isomerase, leucine aminopeptidase, malate dehydrogenase, peroxidase, and phosphoglucomutase. From the isozyme analysis, esterase showed higher degree of variability, while it was observed less variability for the enzymes such as glucosephosphate isomerase, malate dehydrogenase, and phosphoglucomutase. The species P. ostreatus, whose taxon is controversial, was discriminated from P. pulmonarius, while P. florida was classified as a distinct taxon. The clustering of P. sapidus and P. spodoleucus strains appeared to be more difficult. It was found that some strains were included to another cluster based on electrophoretic banding patterns. These results show that this lack of congruence among data sets may help explain the taxonomic difficulty within the genus Pleurotus. A dendrogram of genetic similarities was presented, and applications of isozyme data to the systematics of these commercially important fungi was discussed.

Genetic Relationship of Pleurotus ferulae Strains (아위버섯(Pleurotus ferulae) 균주의 유전적 유연관계)

  • This study was carried out to investigate the genetic relationship of Pleurotus ferulae, an edible mushroom found on a medicinal plant, Ferula assa-foetida, in central China. The genetic relationships of 15 Pleurotus species strains, including five P. ferulae strains were analyzed. The strains were divided into seven groups at 80% genetic similarity level according to random amplified polymorphic DNA (RAPD) analysis. Four out of the seven groups consisted of two to four strains, while the other three groups consisted of three strains. In each of the three groups, the three strains were from each of three different Pleurotus species (P. cornucopiae, P. florida and P. sajorcaju). Other strains grouped together for genetic similarity were P. eryngii 26060 and P. fuscus var. ferulae 26065, three strains of P. ostreatus, and four P. ferulae strains (Bakdal, Awi, Cheonsan 1, and Yesan). However, Japanese Seolyi which belongs to P. ferulae and Heukpyung which belongs to P. ostreatus were together in a separate group.

  • So far as a solution of the given consistent linear system is concerned many numerical methods - both mathematically non-iterative as well as iterative - have been reported in the literature over the last couple of centuries. Most of these methods consider all the equations including linearly dependent ones in the system and obtain a solution whenever it exists. Since linearly dependent equations do not add any new information to a system concerning a solution we have proposed an algorithm that identifies them and prunes them in the process of solving the system. The pruning process does not involve row/column interchanges as in the case of Gauss reduction with partial/complete pivoting. We demonstrate here that the use of pruning as an inbuilt part of our solution process reduces computational and storage complexities and also computational error.

  • 4H-SiC Schottky rectifiers were exposed to pure Ar discharges in a planar coil Inductively Coupled Plasma system, as a function of source power, of chuck power and process pressure. The reverse breakdown voltage ($V_B$) decreased as a result of plasma exposure due to the creation of surface defects associated with the ion bombardment. The magnitude of the decrease was a function of both ion flux and ion energy. The forward turn-on voltage ($V_F$), on-state resistance ($R_{ON}$) and diode ideality factor (n) all increased after plasma exposure. The changes in all of the rectifier parameters were minimized at low power, high pressure plasma conditions.

  • The internal regions of nuclear small subunit rRNA from 6 plaeurotus species and 5 Pleurotus ostreatus strains were amplified by PCR and sequenced. The DNA sequences of 8 Pleurotus strains (P. ostreatus NFFA2, NFFA4501, NFFA4001, KFFA4001, KFCC11635, P florida, P. florida, P. sajor-cuju, P. pulmonarius, and P. spodoleucus) were idential, but P. cornucopiae differed from them in two bases out of 605 bases. However, p[hylogenetic analysis of the sequences by DNA-distance matrix and UPGMA methods showed that P. ostreatus NFFA2m1 and NFFA2m2, known as mutants of P. ostreatus NFFA2, belonged to anther group of Basidiomycotina, which is close to the genus Auricularia. The difference of the SSU rDNA sequences of P. cornucopiae from other Pleurotus species tested corresponds to the difference of mitochondrial plasmid type present in Pleurotus species as observed by Kim et al. (1993, Korean J. Microbiol. 31, 141-147).ishement of silencing at the HMR/hsp82 locus can occur in G1-arrested cells. Cell cycle arrest at G1 phase was achieved by treatment of early log a cell cultures with .alpha.-factor mating pheromone, which induces G1 arrest. The result suggests that passage through S phase (and therefore DNA replication) is nor required for re-establishing silencer-mediated repression at the HMNRa/HSP82 locus. Finally, to test whether de nono protein synthesis is required for re-establishment of silencer-mediated repression, cells were pretreated with cycloheximide (500 /.mu.g/ml) 120 min. It was apparent that inhibiting protein synthesis delays, but does not prevent, re-establishment of silencer-mediated repression. Altogether, these results indicate that re-establishment of silencer-mediated repression is not dependent on the DNA replication and has no requirement for protein synthesis.

State of Knowledge of Apple Marssonina Blotch (AMB) Disease among Gunwi Farmers

  • Purpose: Fuji apples are one of the top selling exports for South Korea bringing in over $233.4 million in 2013. However, during the last few decades, about half of the Fuji apple orchards have been infected by Apple Marssonina Blotch disease (AMB), a fungal disease caused by Diplocarpon mali., which takes about 40 days to exhibit obvious visible symptoms. Infected leaves turn yellow and begin growing brown lesions. AMB promotes early defoliation and reduces the quality and quantity of apples an infected tree can produce. Currently, there is no prediction model for AMB on the market. Methods: The Precision Agriculture Laboratory (PAL) at the University of Florida (UF) has been working with the National Academy of Agricultural Science, Rural Development Administration, South Korea to investigate the use of hyperspectral data in creating an early detection method for AMB. The RDA has been researching hyperspectral techniques for disease detection at their Apple Research Station in Gunwi since 2012 and disseminates its findings to the local farmers. These farmers were surveyed to assess the state of knowledge of AMB in the area. Out of a population of about 750 growers, 111 surveys were completed (confidence interval of +/- 8.59%, confidence level of 95%, p-value of 0.05). Results: The survey revealed 32% of the farmers did not know what AMB was, but 45% of farmers have had their orchards infected by AMB. Twenty-five percent could not distinguish AMB from other symptoms. Overwhelmingly, 80% of farmers strongly believed an early detection method for AMB was necessary. Conclusions: The results of the survey will help to evaluate the outreach programs of the RDA so they can more effectively educate farmers on the identifying, treating, and mediating AMB.

Management of Powdery Mildew in Squash by Plant and Alga Extract Biopesticides

  • Although many fungicides are registered for use to control powdery mildew on cucurbits, management of resistance to fungicides in pathogen populations still remains a major challenge. Two biopesticides Regalia SC and HMO 736 were evaluated in the greenhouse and field for their efficacy against powdery mildew in squash. In greenhouses, Regalia SC alone significantly (P < 0.05) reduced powdery mildew compared to the nontreated control, and was as effective as the chemical standard Procure 480SC (triflumizole). In alternation with Procure 480SC, Regalia SC demonstrated greater or equivalent effects on reducing the disease. HMO 736 alone showed varying levels of disease control, but alternating with Procure 480SC significantly improved control efficacy. In addition, application of Regalia SC or HMO 736 each in alternation with Procure 480SC significantly increased the chlorophyll content in leaves and the total fresh weight of squash plants, when compared with the water control, Regalia SC and HMO 736 alone. In field trials, application of Regalia SC and HMO 736 each alone significantly reduced disease severity in one of two field trials during the early stage of disease development, but not during later stages when disease pressure became high. Both Regalia SC and HMO 736 each applied in alternation with Procure 480SC significantly improved the control efficacy compared to Procure 480SC alone. Results from this study demonstrated that an integrated management program can be developed for powdery mildew in squash by integrating the biopesticides Regalia SC, HMO 736 with the chemical fungicide Procure 480SC.

  • Pleurotus florida was transformed by complementation of auxotrophic mutant using chimeric plasmid containing Flammulina velutipes leu 2 gene and pBR 322 replicon. Mycelial morphology of transformants was grown and compared on mushroom complete and minimal medium. Transformants were mated with monokaryon and their genetic recombination was investigated for the morphology of fruitbody and spore analysis. $Leu^+$ transformant showed same mating type of $A_1B_1$ as to the untransformed mutant. The transformant and the untransformed mutant were mated with monokaryon of which mating type is $A_2B_1$, respectively. Although fruitbody of the untransformed mutant was not produced, $leu^+$ transformant produced fruitbody. Spore analysis showed that leucine requiring spores from fruitbody of $leu^+$ transformant were diminished when compared with those of untransformed mutant.

Transfer of Isolated Nuclei from Pleurotus florida into Protoplasts of Pleurotus ostreatus (느타리버섯 원형질체내(原形質體內)에 사철느타리버섯 핵(核)의 전이(轉移))

  • The transfer of the isolated nuclei from P. florida into protoplasts of P. ostreatus was induced with polyethylene glycol and $CaCl_2$. Three types of transfer products of nuclei were obtained when transferred to MMM. Type 1 colonies were more vigrously growing mycelium and stable on MCM. One of the type 1 colonies, appeared segregation on MCM plus benomyl. The mycelium did not form clamp connection. These results suggest that type 1 colonies were nuclear hybrids or allodiploids. Type 2 was main products of nuclear transfer. The mycelium formed clamp connection and fertile on sawdust media. Type 3 was very slow growing or non-viable colonies after debris of nuclei or chromosomes transfer into recipient protoplasts. Isozyme pattern of esterase in type 1 produced a new band. Type 2 and type 3 could be characterized by parental bands.
