The patulin producing capicity of Penicillium crustosum, an isolate from Korean apple, in various broth culture media, was investigated, and compared with patulin production by the standard strain P. griseofulvum(ATCC 46037). The maximal patulin production capacity of the P. griseofulvum ATCC 46037 was 2,029-2,829 ppm in 5-GYEP, SY and MEB broth media. The patulin-producing capacity of the isolated fungus(P. crustosum) attained 2,794 ppm in a 5-GYEP broth medium, but was only 324 and 11 ppm in SY and MEB media, respectively. There were no significant correlations between mycelial growth levels and patulin-producing ability in either P. crustosum or P. griseofulvum. The patulin production of P. griseofulvum was induced in the wide pH range of pH 3.0-11.0, while that of P. crustosum was induced in the acidic pH range pH 3.0-5.0. Patulin production levels were dependent on the carbon sources in the media and maximal patulin production by P. griseofulvum and P. crustosum was observed in media containing glycerol and fructose, respectively.

This study was carried out to identify the causal pathogens and investigated the characteristics of Penicillium spp. isolated from postharvest decay of pear. One hundred and ninety eight Penicillium spp. were isolated from infected pear fruits. The lesions were formed when the isolated pathogen were inoculated into the wounds and unwounds of pear fruit. Total isolates were classified into 15 groups by the size, color, pigment of colony and shape of conidia. These isolates were identified to be P.expansum, P.solitum, and P.crustosum according to the types of morphological, cultural and physiological characteristics. The pathogenicity was higher in wound inoculation at low temperature than unwound one. This result confirmed that wound promoted the disease appearance. P.expansum was appeared to have the most strong virulence, whereas P.solitum and P.crustosum were classified as weak virulent species by pathogenicity test on pear fruits.
Blue mold is the most important postharvest disease of apples in Korea. Apple fruits with blue mold symptoms were collected from storages in different locations in Korea and were investigated for their association with Penicillium species. A total of sixty five isolates of Penicillium were sampled from the collected apples. Based on DNA sequence analysis of ${\beta}$-tublin gene and ITS and lsu rDNA (ID region) and morphological characteristics, they were identified as P. crustosum, P. expansum, P. italicum, P. solitum and P. sp.. P. sp. which is closely related to P. hirsutum is a new species, not reported before. P. expansum (35%) was predominant species followed by P. crustosum. The phylogenetic tree inferred from combined ${\beta}$-tublin and ID region sequence showed good correlation with species that are defined by morphological characteristics. In pathogenicity test, apples were wound-inoculated with conidial suspension and incubated at $20-22^{\circ}C$. The most severe and destructive species was P. expansum. The species caused a decayed area 42-50mm in diameter after 8-10days. Decayed area caused by P. crustosum and P. sp. was 26-32mm and 20-26mm, respectively. This is the first record of P. crustosum, P. italicum and P. sp. from apple in Korea.
Patulin is a food mycotoxin which induces genotoxicity and acute intestinal disease in infants. Patulin mainly originates from fruit putrefactive moulds, especially in apples, which necessitates the maintenance of strong safety standards against patulin for fresh and processed apples. To investigate the patulin producing moulds in Korean apples, 16 morphological types of fungi were isolated from Korean apples and a patulin producing fungus was identified based on a sequence analysis of the region of internal transcribed spacers (ITS5-5.8S-ITS4 region, 505 base pair) and the 26 rRNA D1/D2 region (527 base pair). Morphological analyses were also performed. The isolated patulin producing fungus was found to a representative species of Penicillium crustosum. The maximal patulin production ability of the isolated fungus (P. crustosum) and the patulin producing standard strain (P. griseofulvum, ATCC 46037) in an SY broth medium were 0.32 and 2.46 mg/L, respectively.
A severe fruit rot of Persimmon (Diospyros kaki cv: Fuyu) was occurred during the storage and transport that infected with blue mold in Sweet Persimmon Experiment Station, Gyeongsangnam-do Agricultural Research and Extension Services, Korea. Fruit surfaces were infected with the fungus first and the colonized fungus formed mycelial mats. From the point of infection, fruits become collapsed and mostly ruptured. The pathogenic fungus from infected fruits was isolated and cultured on PDA. Colony color of the fungus was white at frist than became green on Malt Extract Agar and Czapek Yeast Extract Agar. Conidia were ellipsoid subglobose and 2.6${\sim}$3.8 ${\times}$ 2.4${\sim}$3.8 ${\mu}m$ in size. Stipes were 86${\sim}$320 ${\times}$ 2.8${\sim}$4.3 ${\mu}m$ in size. Rami were 7.5${\sim}$32.6 ${\times}$ 2.6${\sim}$4.2 ${\mu}m$ in size, Ramuli were 12.4${\sim}$14.8 ${\times}$ 3.2${\sim}$3.8 ${\mu}m$ in size, Metulae were 8.9${\sim}$13.6 ${\times}$ 2.8${\sim}$4.6 ${\mu}m$ in size. Phialides were ampulliform, 8.2${\sim}$12.4 ${\times}$ 2.3${\sim}$3.6 ${\mu}m$ in size. Based on the cultural and mycological characteristics and pathogenecity test on host plants, the fungus was identified as s, This is the first report on the blue mold of Persimmon (Diospyros kaki) caused by P. crustosum in Korea.

Fifteen isolates from pear, and sixteen isolates from KCTC, KCCM, and Chungnam Univ. of Penicillium spp. were investigated for the analysis of their relationships of cultural characteristics and RAPD genetic variation by RAPD. The cultural characteristics of Penicillium spp. were shown different growth rate, morphology, and color. In addition, the cultural characteristics and RAPD analysis were conducted for the pear rot pathogens and related isolates. RAPD patterns were applied to compare the taxonomic and genetic diversity of the Penicillium species between 15 groups isolated from pear fruits and 16 standard species. The genomic DNA were amplified from $0.1{\sim}2.0kb$ by five URP primer and 744 bands were detected. The cluster analysis showed four genomic DNA RAPD groups and its similarity was 47.7%. Intraspecific relationships were 87.4, 97.5 and 95.2%, in P. expansum, P. solitum, and P. crustosum, respectively. These results appeared to be that there were high similarities between isolates, and consistent with the results of cultural morphological characteristics analysis.

Penicillium species are commonly isolated from various outdoor and indoor environments, including marine environments such as sponges, algae and sand. Penicillium is especially important because numerous bioactive compounds have been isolated. Penicillium was the most common species in intertidal zone in Korea, however the diversity and ecological roles of Penicillium in intertidal zone are not clarified. We explored diversity and ecological roles of marine-derived Penicillium from tidal flat and sea sand in Korea. The diversity of marine-derived Penicillium from Korea was investigated using both culture-dependent and culture-independent approach by ${\beta}$-tubulin sequence. In addition, we evaluated optimal temperature, halo-tolerance, and enzyme activity of Penicillium strains, such as extracellular alginase, endoglucanase, ${\beta}$-glucosidase, and protease. For culture-dependent approach, a total of 182 strains of 62 Penicillium species were isolated, with 53 species being identified. The most common species was Penicillium oxalicum, followed by P. crustosum, P. brasilianum, P. koreense, and P. griseofulvum. Species richness and composition were not significantly different by season, substrates, and seaside. For culture-independent approach using Illumina sequencing, 73 OTUSs were detected. The most frequently observed species was P. antarcticum, followed by P. koreense, P. crustosum, and P. brevicompactum. Diversity of Penicillium was higher during winter season than during summer season and in western sea than in southern sea, respectively. Community structure was significantly different by season and sea side. 52 species were detected by both methods. Unique species were isolated from each of methods - 10 from culture methods and 21 from Illumina sequencing. Furthermore, salinity adaption of the Penicillium varied depending on species. Many Penicillium species showed endoglucanase, ${\beta}$-glucosidase, and protease activity. Some species including P. paneum and P. javanicum degraded the polycyclic aromatic hydrocarbons. Thus, our results demonstrate that intertidal zone in Korea harbors diverse Penicillium community and marine-derived Penicillium play important ecological roles as decomposers of organic material in marine environments.
Demand for a rice cake, a popular traditional food in Korea, is rising, but its industrial-scale production is extremely difficult due to its short shelf-life caused by starch retrogradation and microbial spoilage. By means of the sourdough fermentation technique, we attempt to develop rice cakes with a longer shelf-life. Heterofermentative lactic acid bacteria (Weissella koreensis HO20, Weissella kimchii HO22) isolated from kimchi were used to ferment wet-milled rice flour for their abilities to produce exopolysaccharides and to inhibit the microbial spoilage of rice cakes. After 24 hr of fermentation at $25^{\circ}C$, viable cell counts in rice dough increased from $10^6$ CFU/g to $10^8$ CFU/g and total titratable acidity increased from 0.05% to 0.20%, whereas pH decreased from 6.5 to 5.1. Fermented rice flour showed significantly lower peak, trough, and final viscosities as well as breakdown and setback viscosities measured by rapid viscoanalyzer. Both lactic acid bacteria showed in vitro antifungal activity against Penicillium crustosum isolated from rice cakes. The antifungal activity remained constant after the treatments with heat, proteinase K and trypsin, but fell significantly by increase of pH. Rice cakes made of fermented rice flour were found to retard mycelial growth of P. crustosum. The degree of retrogradation as measured by the hardness of the rice cake was significantly reduced by the use of fermented rice flour. The results suggest that use of fermented rice flour has a beneficial role in retarding starch retrogradation and in preventing fungal growth, hence extending the shelf-life of rice cakes.
Marine macroalgae are important in coastal ecosystems and interact with marine microorganisms. In this study, we isolated fungi from seven types of marine macroalgae including Cladophora sp., Gloiopeltis furcate, Gracilariopsis chorda, Hydroclathrus clathratus, Prionitis crispata, Sargassum micracanthum, and Ulva lactuca collected in Korea. Morphological and phylogenetic analyses identified the isolates as four Aspergillus spp. (A. fumigatus, A. sydowii, A. tamarii, and A. terreus), three Penicillium spp. (P. crustosum, P. jejuense, and P. rubens), and Cladosporium tenuissimum. Among them, A. fumigatus TOP-U2, A. tamarii SH-Sw5, and A. terreus GJ-Gf2 strains showed the activities of all enzymes examined (amylase, chitinase, lipase, and protease). Based on the enzymatic index (EI) values in solid media, A. terreus GJ-Gf2 and C. tenuissimum UL-Pr1 exhibited the highest amylase and lipase activities, respectively. Chitinolytic activity was only observed in A. terreus GJ-Gf2, A. tamarii SH-Sw5, and A. fumigatus TOP-U2. Penicillium crustosum UL-Cl2 and C. tenuissimum UL-Pr1 showed the highest protease activities. To the best of our knowledge, this is the first report of lipolytic and proteolytic activities in a marine-derived C. tenuissimum strain. Overall, the fungal strains isolated from the marine macroalgae in this study actively produced industrially important enzymes.
A total of 65 isolates of Penicillium were isolated from decayed post-harvest fruits of apple pear citrus and grape. The Penicillium species isolated from the apple were idnetified as P. aurantiogriseum and P. expansum those from the pear were P. crustosum and P. expansum and those from the grape were P. aurantiogriseum and P. expansum, From decayed citrus fruits. P. digitatum and P. italicum were isolated. Citrinin and patulin from these species in the YES(yeast extract sucrose) broth were extracted with ethyl acetate and purified by thin-layer chromatography(TLC) and high performance liquid chromatography(HPLC) Among 51 isolates of Penicillium from apple pear and grape 7 isolates produced citrinin 13 isolates produces patulin and 12 isolates produced citrinin and patulin also. All 14 isolates of Penicillium from citrus produced only patulin.
