Modulation of P-glycoprotein Activity by Flavonoids and Organic Isothiocyanates in Human Uterine Cells.

  • One of the possible mechanisms of multi-drug resistance found in cancer cells is the over-expression of P-glycoprotein (P-gp). Studies have shown that compounds found in plants including vegetables and fruits not only have anticancer activities but may also modulate P-gp activity. The effect of flavonoids and organic isothiocyanates on P-gp activity was studied in human uterine sarcoma cell lines, MES-Sa (sensitive) and MES-SD/DX5 (resistant). The accumulation of daunomycin (DNM), a P-gp substrate, was approximately 10 times greater in the sensitive cell as compared to the resistant cells over the entire time course (up to 2 hrs).   • The growth of baceriophage T4 is inhibited by the presence of the tin gene product o bacteriophage P2. The interaction between purified Tin and gp32 proteins was observed using coimmunoprecipitation experiments. The in vivo interaction was confirmed by yeast two-hybrid experiments. A deletion analysis showed that the Asp 163 region of gp32 to DNA substrates was not affected by the presence of Tin, Thus, it would appear that the inhibition of 4 growth by Tin was due to a protein-protein interaction rather than affecting the DNA-binding ability of gp32.

  • A ginseng preparation (GP) consisting of ginseng ex., lycium fructus ex., four kinds of vitamines and caffein was evaluated for acute toxicity and general pharmacology. Average lethal doses $(LD_{50})$ of GP in male mice were 2,988mg/kg (i.p.) and more than 3g/kg (p.o.). In dosage of 300 and 900mg/kg (p.o.) showed no analgesic activity in both tests of the writhing method induced by acetic acid and of tail pressure method and no effect on the pentetrazole-induced convulsion. However, it appeared to have a hypothermic action only in dose of 900mg/kg. The duration of hypnosis induced by hexobarbital sodium in mice was shortened by GP, being probably due to caffein. GP Produced no marked contraction of isolated ileum and uterus in high concentrations of up to $1{\pm}10^{-3}g/ml$. These results suggested that GP did not show any considerable central nervous depressant activity and exhibited very weak actue toxicity in mice.

Effects of Dietary Garlic Powder and Copper on Cholesterol Content and Quality Characteristics of Chicken Eggs

  • This experiment was conducted to investigate the dietary effect of garlic powder (GP) and copper (Cu) on laying performances and the cholesterol content and quality characteristics of eggs during storage in laying hens. A total of one hundred and eighty, 50-wk-old, Hy-Line Brown layers were divided into 6 groups with 3 replicates per group (10 layers per replicate) and fed one of six diets containing GP 0%, GP 1%, GP 3%, GP 5%, Cu 200 ppm, or GP 3%-Cu 200 ppm for 5 wks. There were no differences in the laying performances and feed intakes bertween treatments. Eggshell strength, eggshell thickness and yolk color were also not affected by feeding of GP and Cu. With increasing dietary GP, Haugh unit was linearly increased after 2 wk of storage (p<0.05). The levels of serum total cholesterol in hens fed diets containing GP or Cu were lower than that of the control (p<0.05), but high density lipoprotein-cholesterol was not influenced by dietary GP or Cu. The content of egg yolk cholesterol from hens fed diets containing GP or Cu was significantly decreased from that of the control, except for the GP 1% group. Based on the results of this experiment, the decrease of Haugh unit during storage was alleviated by feeding of GP. The feeding of GP or Cu alone and in combination altered the cholesterol fractions in serum and reduced the content of egg yolk cholesterol in laying hens.

Inhibition of P-Glycoprotein by Natural Products in Human Breast Cancer Cells

  • Multidrug resistance (MDR) is one of the most significant obstacles in cancer chemotherapy. One of the mechanisms involved in the development of MDR is the over-expression of P-glycoprotein (P-gp). It is widely known that natural compounds found in vegetables, fruits, plant-derived beverages and herbal dietary supplements not only have anticancer properties, but may also modulate P-gp activity. Therefore, the purpose of this investigation was to examine the effects of naturally occurring products on P-gp function in human breast cancer cell lines, MCF-7 (sensitive) and MCF-7/ADR (resistant). The accumulation of daunomycin (DNM), a P-gp substrate, was greater in the sensitive cells compared to the resistant cells, while the efflux of DNM was higher in the resistant cells compared to the sensitive cells over a period of 2h. The $IC_{50}$ value of DNM in the resistant cells was about 22 times higher than that in the sensitive cells, indicating an over-expression of P-gp in the resistant cells, MCF-7/ADR. All of the compounds tested, with the exception of fisetin, significantly decreased the $IC_{50}$ value of DNM. Biochanin A showed the greatest increase in $[^3H]-DNM$ accumulation, increasing by $454.3{\pm}19.5%$ in the resistant cells, whereas verapamil, the positive control, increased the accumulation by $229.4{\pm}17.6%$. Also, the accumulation of $[^3H]-DNM$ was increased substantially by quercetin and silymarin while it was reduced by fisetin. Moreover, biochanin A, silymarin, and naringenin significantly decreased DNM efflux from MCF-7/ADR cells compared with the control. These results suggest that some flavonoids such as biochanin A and silymarin may reverse MDR by inhibiting the P-gp function.

Epileptogenic Properties of Balloon Cells in Cortical Tubers of Tuberous Sclerosis : Upregulation of Drug Resistance Proteins

  • Objective : Balloon cells and dysplastic neurons are histopathological hallmarks of the cortical tubers of tuberous sclerosis complex [TSC] and focal cortical dysplasia [FCD] of the Taylor type. They are believed to be the epileptogenic substrate and cause therapeutic drug resistant epilepsy in man. P-glycoprotein [P-gp] is the product of multidrug resistance gene [MDR1], and it maintains intracellular drug concentration at a relatively low level. The authors investigated expression of P-gp in balloon cells and dysplastic neurons of cortical tubers in patients with TSC. Methods : An immunohistochemical study using the primary antibody for P-gp, as an indicative of drug resistance, was performed in the cortical tuber tissues in two patients of surgical resection for epilepsy and six autopsy cases. Results : Balloon cells of each lesion showed different intensity and number in P-gp immunopositivity. P-gp immunopositivity in balloon cells were 28.2%, and dysplastic neurons were 22.7%. These immunoreactivities were more prominent in balloon cells distributed in the subpial region than deeper region of the cortical tubers. Capillary endothelial cells within the cortical tubers also showed P-gp immunopositivity. Conclusion : In this study, the drug resistance protein P-glycoprotein in balloon cells and dysplastic neurons might explain medically refractory epilepsy in TSC.

Overexpression and Purification of p24 and gp41 Proteins of Human Immunodeficiency Virus Type 1 in E. coli (대장균에서 인간면역결핍 바이러스 1형의 gag p24 및 env gp41 단백질의 과발현 및 정제)

  • Synthetic genes encoding the gag p24 and the part of the envelope protein gp41 of the human immunodeficiency virus (HIV-1) were cloned and overexpressed as fusion proteins in Escherichia coli, using an expression vector carrying T7 promoter and the poly-histidine leader, sequence. The overexpressed p24 fusion protein was purified by centrifugation, Ni-affinity chromatography and CM-sepharose chromatography. The overexpressed gp41 fusion protein was purified by centrifugation, $C_4$ chromatography and DEAE-sepharose chromatography. The purified fusion proteins showed a high level of purity and immunoreactivity in SDS-polyacrylamide gel electrophoresis and western blot analysis. These results suggest that this prokaryotic - expression purification method is suitable for obtaining a large amount of the viral antigen which may be useful for screening of antibodies to HIV-1 in human blood samples.

Determination of garlic extract efficacy on growth, nutrient digestibility, and fecal score of growing pigs via diet supplementation

  • Garlic powder (GP) was supplemented in the diet to assess the growth efficiency, nutritional retention, and fecal score of growing pigs. In total, 80 growing pigs ([Yorkshire × Landrace] × Duroc) were arbitrarily allotted (22.04 ± 0.56 kg primary average body weight) to two different trial groups with eight repetitions (three males and two females) in each pen for the 42-day trial. The dietary treatment consisted of control (CON, basal diet), which included 0.2% GP (TRT1). Growing pigs fed the GP diet exhibited an increase (p < 0.05) in body weight on day 42 and a lower (p < 0.05) overall feed conversion ratio compared to the CON pigs; however, the average daily gain and feed intake showed no difference (p > 0.05). Additionally, nutrient utilization of dry matter was greater (p < 0.05) in the GP-supplemented diet group than in the CON group. The feeding methods did not adversely affect the nitrogen and energy utilization and the fecal score significantly (p > 0.05). In summary, growing pigs fed the GP diet showed increased growth and nutritional utilization, and no adverse impact on the fecal score; thus, it could be utilized as a stimulant for improved growth performance.

Storage characteristics of organic chicken stock containing plum extract and green tea powder

  • This study investigated quality characteristics of chicken stock with added plum extract (PE) and green tea powder (GP) stock during storage. Plum extract (0, 0.5, 1, 3%) and green tea powder (0, 0.5, 1, 3%) were added at three levels. Chicken stock was stored at room temperature for 14 days. The pH of the chicken stock decreased significantly as the content of PE and GP increased (p < 0.05). The group with 3% plum extract added showed significantly lower pH values (p < 0.05). Total numbers of microorganisms (TMC) showed significant differences according to the storage period (p < 0.05), where the groups with PE and GP added showed lower TMC values than the control group, This indicates that PE and GP could inhibit the growth of microorganisms. The addition of 3% PE or GP decreased lipid oxidation (TBARS) and protein deterioration (VBN) values. In sensory test, the results showed that adding PE or GP has a positive effect on storage characteristics of chicken stock. The addition of PE rather than GP is effective not only in improving sensory evaluation, but also in minimizing changes in quality by suppressing lipid oxidation and protein deterioration during storage. In conclusion, 3% PE addition was found to be the most optimal supplementation choice for increasing the storability of chicken stock.

Characterization of gp64 Gene of Bombyx mori Nucleopolyhedrovirus and Development of a Transient Expression Vector (누에 핵다각체병 바이러스 헤 gp64 유전자의 특성조사 및 transient 발현 벡터 개발)

  • Expression of the baculovirus major envelope glycoprotein gene(gp64) is regulated by transcription from botha early and late promoters. To develop a transient expression vector under the control of gp64 gene promoter, the gp64 gene of Bombyx mori nucleopolyhedrovirus-K1(BmNPV-K1) was characterized. The gp64 gene was local-ized at EcoR I-Pst I 7.38-kb fragment of the BmNPV-K1 genome. The EcorR 1-Pst I 7.38-kb fragment was cloned and the nucleotide sequence of 2,277 bases including the coding region of gp64 gene was determined. Based on these results, transient expression vector using gp64 gene promoter was constructed and named as pBm64. E.coli lacZ gene was introduced onto pBm64 as a reporter gene and expressed transiently in B. mori 5(Bm 5) cells. The expression vector transfected into the cells was maintained stably for 1 to 5 days. In order to confirm the expression of the reporter gene by gp64 promoter, recombinant virus was constructed. The recombinant virus has two independent transcription units in opposite orientations with two promoters; gp64 and polyhedrin gene promoters each initiating transcription of $\beta$-galactosidase and polyhedrin, respectively. Polyhedra formation and expression of $\beta$-galactosidase in Bm5 cells infected with the recombinant virus were observed with phase contrast microscope and in situ staining.
