감마선과 Methyl Bromide 처리된 팥과 녹두의 수침에 따른 용출 특성 (Water-Soluble Components of Small Red Bean and Mung Bean Exposed to Gamma Irradiation and Methyl Bromide Fumigation)

  • 팥과 녹두의 가해해충 살충을 위한 감마선과 methyl bromide(MeBr) 훈증 처리가 두류의 수침 시 용출 성분에 미치는 영향을 비교 검토하였다. 팥과 녹두를 $20^{\circ}C$에서 16시간 침지시킨 후 용출액의 성분을 분석한 결과, 고형분과 환원당 함량은 감마선 조사선량에 따라 증가하였는데 녹두에 비해 팥이 두드러졌다. 그리하여 5 kGy 이상 조사구에서는 유의적으로 증가하였으나 살충선량인 2.5 kGy 범위에서는 변화가 크지 않았다. MeBr 처리된 팥의 고형분 함량은 7.5 kGy 조사구와 유사한 수준의 변화를 보였으나 전반적으로 대조구와 거의 유사한 함량을 보였다. 용출액의 pH는 처리구에 따라 일정한 변화를 보이지 않았다. 용출액의 색도는 감마선 조사선량의 증가에 따라 두 시료에서 명도(L)의 감소와 적색도(a) 및 황색도(b)의 증가현상이 점차 나타났으며, 훈증처리 시료에서는 이 같은 현상이 매우 심하여 전반적 색차(${\Delta}E$)가 팥은 10 kGy, 녹두는 30 kGy 이상 조사된 시료와 유사한 값을 보였다. 수침에 따른 유리아미노산의 용출은 팥의 경우 asparagine, glutamic acid, alanine, valine 등이, 녹두의 경우asparagine, glutamic acid, proline, valine 등이 용출 비율이 높았으며, 조사선량이 증가함에 따라 총 유리아미노산의 용출 함량은 높게 나타났다. 그러나 MeBr 훈증 처리구의 유리아미노산 함량은 대조구보다 낮은 값을 보였다. 팥과 녹두 용출액의 고형분과 환원당 함량은 감마선 조사선량과 매우 높은 정의 상관관계를 보여주었다.

남한 탄산수의 지구화학적 특성과 안정동위원소 조성 (Geochemistry and Stable Isotopes of Carbonated Waters in South Korea)

  • 본 연구에서는 남한에 분포하는 23개의 약수지역을 대상으로 탄산수의 수질 특성과 탄소의 기원을 연구하기 위하여 용존 이온 및 동위원소 분석연구를 수행하였다. 지질 특성별로 분류.비교된 국내 탄산수는 대체로 Ca-HC $O_3$형에 속하며 pH는 5.3~6.3의 범위이다. 탄산수내 대부분은 양이온과 음이온의 농도가 각각 $Ca^{2+}$>$Na^{+}$>M $g^{2+}$>S $i^{4+}$>F $e^{2+}$> $K^{+}$ 와 HC $O_3$$^{-}$>S $O_4$$^{2-}$ >C $l^{-}$의 순이다. 탄산수의 수질 유형은 대체로 Ca-HC $O_3$형에 해당되나 지역별로 다소 차이가 나타나 강원 지역의 선캄브리아기 변성암류 및 쥬라기 화강암지역(GI)의 탄산수는 Ca-HC $O_3$형이 우세하나 전형적인 Na-HC $O_3$형을 보여주는 경우도 있다. 경상 퇴적분지내의 중생대 퇴적암 및 화강암류 지역(GII)의 탄산수는 Ca-HC $O_3$형 내지 Ca(-Mg)-HC $O_3$형이 우세하다. 옥천 변성퇴적암류 및 화강암류 지역(GIII)의 탄산수는 Ca-HC $O_3$내지 Ca(-Na)-HC $O_3$형을 보여준다. 산소 및 수소 동위원소 분석 결과, 탄산수의 기원은 탄산수 지역 부근의 순환수 기원이며 지형 특성에 따라 동위원소 고도 효과 및 위도 효과를 반영하고 있다. 탄산수의 탄산이온의 탄소의 $\delta$$^{13}$C(PDB)값은 -6.2~0.0$\textperthousand$범위이며, 그 기원은 대수층인 지층내 탄산염암 또는 탄산염 광물의 용해에서 유래한 무기기원 탄소로 해석된다.다.

갈변반응을 이용한 흑양파의 제조 및 물 추출물의 항산화 효과 (Black Onions Manufactured via the Browning Reaction and Antioxidant Effects of Their Water Extracts)

  • 양파의 매운 맛과 냄새를 감소시켜 섭취가 용이하도록 흑양파를 제조하고자 하였으며, 제조조건을 선발하기 위해서 숙성온도, 양파 크기, 그리고 양파 외부의 처리방법을 다르게 하여 흑양파를 제조하고, 각각의 흑양파 물 추출물에 대한 항산화 효과를 평가함으로서 흑양파를 새로운 식품 소재로 사용할 수 있는 흑양파 제조의 기초자료를 조사하였다. 온도에 따른 흑양파 제조효과에서 온도 증가에 따라 갈변효과가 증가하였다. 양파의 크기별은 100 g의 작은 양파와 뿌리와 줄기를 자른 양파의 갈변효과가 좋았다. 생양파보다 제조한 흑양파의 색도와 ${\Delta}E$ 값은 크게 증가하였으며 $90^{\circ}C$일 때 5.53 이었고, 총 페놀 함량은 온도의 증가에 따라 증가하는 경향을 보였는데 $90^{\circ}C$에서 9.19 mg/g이었다. $90^{\circ}C$에서 껍질 있는 작은 양파의 총 페놀함량은 11.62 mg/g으로 가장 높았다. 각 항산화 효과를 놓고 본 때 $90^{\circ}C$에서 껍질있는 작은 흑양파의 효과가 가장 좋았으며, DPPH 라디칼 소거능은 48%에 달하였고 아질산염 소거능도 타 처리구 보다 좋은 결과이었다. 흑양파 제조방법에 따른 기능성 성분을 분석한 결과 박피하지 않고 뿌리와 줄기를 자른 90-S-C 처리구가 갈변도, 총 페놀의 함량이 가장 높게 검출되었다. 이상의 결과를 종합하여 볼 때 $90^{\circ}C$에서 껍질로 싸여 있는 100 g 작은 양파의 갈변효과가 가장 우수하였다.

  • 8.3%의 carbamide peroxide를 함유한 펜형 코팅용 미백제인 BlancTis Forte (NIBEC, Seoul & JinCheon)를 실험군으로, 3% hydrogen peroxide를 함유한 Whitening Effect Pen (LG, Seoul) 제재를 대조군으로 각각 피험자 20명에게 2시간씩 1일 2회 제조사의 지시대로 치아표면에 4주간 적용하도록 지시하고 색조개선 효능과 안전성을 평가하였다. 미백 효과는 미백 전 및 미백 2주, 4주, 및 미백 종료 4주 후에 $Shadepilot^{TM}$을 이용하여 색 변화를 측정하였고. 매 내원시기마다 모든 환자는 치수생활력 검사와 치주 및 치태 검사를 통해 부작용 여부 (치아과민증 및 구강 내 연조직의 부작용)를 기록한 후 다음과 같은 결과를 얻었다. 1, 실험군 및 대조군의 색 변화량 (${\Delta}E$)은 2이상으로 인지할 수 있는 색 변화를 보였으며, 두 제품간에는 통계적으로 유의한 차이를 보이지 않아 (p > 0.05) 유사한 미백효능을 나타냄을 알 수 있었다. 2. 미백효과는 명도의 개선보다는 주로 a, b값의 변화에 의한 것으로 특히 실험군에서 b값의 변화, 즉 황색조의 개선효과가 대조군에 비해 유의성 있게 높은 것으로 나타났다 (p < 0.05). 3. 치아나 치은의 과민증이나 이상증상을 호소하는 피험자는 없어 두 제품 모두 안전성을 확인할 수 있었다.

Effects of [D-$Pen^2$, D-$Pen^5$]-enkephalin on the Neuronal Activity of Medial Vestibular Nuclear Neurons

  • This study was designed to investigate direct effects of [D-$Pen^2$, D-$Pen^5$]-enkephalin, a $\delta$-opioid receptor agonist on the neuronal activity of medial vestibular nuclear (MVN) neurons by whole-cell configuration patch clamp experiments. The spike frequency of MVN neuron was increased to $9.50{\pm}0.55$ (P<0.05) and $10.56{\pm}0.66$ (P<0.05) by 5 and $10{\mu}M$ [D-$Pen^2$, D-$Pen^5$]-enkephalin from the control level of $8.05{\pm}0.55$ spikes/sec, respectively (n=18). The resting membrane potential of the neurons was increased to $-37.86{\pm}0.92$ and $-36.97{\pm}0.97$ (P<0.05) from $-38.74{\pm}1.13\;mV$ by 5 and $10{\mu}M$ [D-$Pen^2$, D-$Pen^5$]-enkephalin, respectively. The amplitude of afterhyperpolarization was decreased to $23.78{\pm}0.65$ and $21.67{\pm}0.89$ (P<0.05) from $23.73{\pm}0.53\;mV$ by 5 and $10{\mu}M$ [D-$Pen^2$, D-$Pen^5$]-enkephalin, respectively. The spike width was changed to $2.22{\pm}0.08$ and $2.24{\pm}0.07$ from $2.20{\pm}0.08\;mV$ by 5 and $10{\mu}M$ [D-$Pen^2$, D-$Pen^5$]-enkephalin, respectively. After pretreatment of naltrindole, a highly selective 8-opioid receptor antagonist, [D-$Pen^2$, D-$Pen^5$]-enkephalin did not change firing rate, resting membrane potential, afterhyperpolarization amplitude, and spike width of MVN neurons. The above experimental results suggest that [D-$Pen^2$, D-$Pen^5$]-enkephalin increases the neuronal activity of MVN neurons via inhibition of calcium-dependent potassium currents underlying the afterhyperpolarization.

신장(身長) 및 체중(體重)의 실측치(實測値)와 상용치간(常用値間)의 오차(誤差)에 영향을 미치는 인자(因子) (Study of Factors Affecting to Discrepancy between Self-Reported and Body Weight and Height)

  • 구미공단내의 체용신체검사자 및 근로자 정기건강진단시 남자 1,060명과 여자 818명을 대상으로 피검자 본인이 인지하고 있는 신장 및 체중과 실측체중 및 실측신장과의 오차에 영향을 줄 수 있는 변수를 연령, 실측신장, 실측체중, 비체중, 교육수준, 취업상태 및 혼인상태로 구분하여 cross-tubulation, 회귀분석에 의한 상관계수 및 분산분석에 의한 검정을 하여 분석하였다. 신장은 남녀 공히 실측치보다 크게 표현하였으며 체중은 실측치보다 남자는 높게 여자는 낮게 표현하였지만 그 오차는 평균 1% 미만으로 적었다. 연령에 있어서 체중은 남자가 $20{\sim}29$세군과 여자 40세 이상군에서 가장 정확한 체중을 표현하였으며, 남자는 연령이 높아짐에 따라 점차 실측치보다 무겁게, 여자는 연령이 낮아짐에 따라 점차 가볍게 표현하였다. 실측체중이 남자는 $60{\sim}64kg$군에서, 여자는 49kg이하군에서 정확하며 남여 공히 체중이 무거울수록 실체중보다 가볍게 표현하였으며 남자는 75kg 이상군에서 4.25% 적게, 여자는 65kg 이상군에서 4.06% 적게 나타났다. 비체중이 $100{\pm}10%$군인 경우 남녀가 가장 정확한 체중을 알고 있으며 비만에 가까울수록 실체중보다 적게 표현하였다. 교육수준이 높을수록 정확한 체중을 표현하나 국졸군에서 가장 정확함을 보인 것은 좀더 정신보건면에서의 연구 관찰이 요망된다. 신장에 있어서 가장 정확한 군은 최상위 실측신장군에서 남녀 공히 나타났으며 신장이 적어질수록 실신장보다 점차 크게 표현하였다. 여성 체중에 영향을 주는 인자 중 유의하게 나타난 변수는 비체중, 실측체중, 연령, 교육수준 및 취업상태이며 여자 신장에 영향을 주는 인자 중 유의한 변수는 실측신장, 실측체중 및 교육수준으로 나타났으며 한국여자는 신장보다 체중에 관심이 있음을 나타내고 있다.

정미기의 능률에 미치는 기계적 요인및 작동조건에 관한 연구 (Mechanical and Operational Factors Affecting the Efficiency of Rice Polishing Machines)

  • In analyzing the operational characteristics of a rice whitening machine, the internal radial pressure of the machine was measured using strain gage equipment. Changes in cylinder and feed screw configurations, screen type, cylinder speed and counter-pressure levels were examined to determine their impact on the quality and quantity of milled rice and the performance of the machine. The results are summarized as follows: 1. The internal radial pressure in the whitening chamber varied with the surface condition of the grain being processed. During the first or second pass through the machine, pressure was relatively low, reached a maximum after two to three passes with combinations I and II, three to six with combination III and then began to fall. 2. The pitch of the feed screw and the size of the feed gate opening which determine the rate of entry of grain into the whitening chamber, appeared to be the most important factor aff-::cting the degree of radial pressure, quality and quantity of milled rice and the efficiency of the machine. Using a feed screw with a wide pitch (4.8cm), radial pressure was relatively high and head rice recovery ratio \vere quite low. In this case capacity and machine effic?iency were much higher than obtained when using a feed screw with a narrow pitch (2.3cm). Very significant responses in radial pressure, head rice recovery rates and machine capacity were observed with changes in cylinder speed and counter-pressure levels when using the wide pitch feed screw. 3. The characteristics of the screen which surrounds the whitening chamber had an important effect on whitening efficiency. The existence of small protuberances on the original screen resulted in significant increases in both machine capacity and efficiency but without a significant decrease in head rice recovery or development of excessive radial pressure. Further work is required to determine the effects of screen surface conditions and the shape of the cylinderical steel roller on the rate of bran removal, machine efficiency and recovery rates. The size of the slotted perforations 0:1 the screen affects total milled rice recovery. The opening size on the original screen was fabricated to accommodate the round shape of Japonica rice varieties but was not suitable for the more slender Indica type. Milling Indica varieties with this screen resulted in a reduction in total milled rice recovery. 4. An increase in cylinder speed from 380 to 820 rpm produced a positive effect on head rice recovery for all machine combinations at every level of counter-pressure used in the tests. Head rice recovery was considerably lower at 380rpm using a wide screw pitch when compared to the results obtained at speeds from 600 to 820 r.p.m. The effects of cylinder speed On radial pressure, capacity and machine efficiency showed contrasting results, depending on the width of the feed screw pitch. With a narrow feed screw pitch (2.3cm), a direct proportional relationship was observed bet?ween cylinder speed and both radial pressure and machine efficiency. In contrast, using a 4.8 centimeter pitch feed roller produced a series of inverse relationships between the above variables. Based on the results of this study it is recommended when milling Indica type long grain rice varieties that the cylinder speed of the original machine be increased from 500-600 rmp up to a minimum of 800 rpm to obtain a greater abrasive effect between the grain and the screen. The pitch of the feed screw should be also reduced to decr?ease the level of internal radial pressure and to obtain higher machine efficiency and increased quality of milled rice with increased cylinder speeds. Further study on the interaction between cylinder speed and feed screw pitch is recommended. 5. An increase in the counter pressure level produced a negative effect On the head rice recovery with an increase in radial pressure, capacity, and machine efficiency over all combinations and at every level of cylinder speed. 6. Head rice recovery rates were conditioned primarily by the pressure inside the whitening chamber. According to the empirical cha racteristics curve developed in this study, the relationships of head rice recovery ($Y_h$) and machine capacity ($Y_c$/TEX>) to internal radial pressure ($X_p$) followed an inverse quadratic function and a linear function respectively: $$Y_h^\Delta=\frac{1}{{1.4383-0.2951X_p^\ast+0.1425X_p^{\ast\ast}}^2} , (R^2=0.98)$$$$Y_c^\Delta=-305.83+374.37X_p^{\ast\ast}, (R^2=0.88)$$The correlation between capacity and power consumption per unit of brown rice expressed in the following exponential function: $$Y_c^\Delta=1.63Y_c^{-0.7786^\{\ast\ast}, (R^2=0.94)$$These relationships indicate that when radial pressure increases above a certain range (1. 6 to 2.0 kg/$cm^2$ based On the results of the experiment) head ricerecovery decrea?ses in a quadratic relation with a inear increase in capacity but without any decrease in power consump tion per unit of brown rice. On the other hand, if radial pressure is below the range shown above, power consumption increases dramatically with a lin?ear decrease in capacity but without significant increases in head rice recovery. During the operation of a given whitening machine, the optimum radial pressure range or the correct capacity range should be selected by controlling the feed rate and/or counter-pressure keeping in mind the condition of the grain, particulary the hardness. It was observed that the total number of passes is related to radial pessure level, feed rate and counter-pressure level. The higher theradial pressure the fewer num?ber of pass required but with decreased head rice recovery. In particular, when using high feed rates, the total number of passes should be increased to more than three by reducing the counter-pressure level to avoid decreaseases in head rice recovery (less than 65 percent head rice recovery on the basis of brown rice) at every cylinder speed. 7. A rapid rise in grain temperature seemed to have a close relationship with the pressure generated inside the whitening chamber and, subsequently with head rice reco?very rates. The higher the rate of increase, the lower were the resulting head rice recoveries.

Protective Effect of Fucoidan Extract from Ecklonia cava on Hydrogen Peroxide-Induced Neurotoxicity

  • We evaluated the antioxidant activity and neuronal cell-protective effect of fucoidan extract from Ecklonia cava (FEC) on hydrogen peroxide ($H_2O_2$)-induced cytotoxicity in PC-12 and MC-IXC cells to assess its protective effect against oxidative stress. Antioxidant activities were examined using the ABTS radical scavenging activity and malondialdehyde-inhibitory effect, and the results showed that FEC had significant antioxidant activity. Intracellular ROS contents and neuronal cell viability were investigated using the DCF-DA assay and MTT reduction assay. FEC also showed remarkable neuronal cell-protective effect compared with vitamin C as a positive control for both $H_2O_2$-treated PC-12 and MC-IXC cells. Based on the neuronal cell-protective effects, mitochondrial function was analyzed in PC-12 cells, and FEC significantly restored mitochondrial damage by increasing the mitochondrial membrane potential (${\Delta}{\Psi}m$) and ATP levels and regulating mitochondrial-mediated proteins (p-AMPK and BAX). Finally, the inhibitory effects against acetylcholinesterase (AChE), which is a critical hydrolyzing enzyme of the neurotransmitter acetylcholine in the cholinergic system, were investigated ($IC_{50}$ value = 1.3 mg/ml) and showed a mixed (competitive and noncompetitive) pattern of inhibition. Our findings suggest that FEC may be used as a potential material for alleviating oxidative stress-induced neuronal damage by regulating mitochondrial function and AChE inhibition.

한국산 일시 수확형 고추 후레이크의 특성 및 다양한 드레싱에의 품질에 미치는 영향 (Characteristics of Korean Single-harvested Pepper(Capsicum annuum, L.) Flakes and the Effects on the Quality of Various Dressings)

  • This study analyzed the characteristics of single-harvested pepper flakes and investigated the effects of the addition of red and green pepper flakes on the quality of various dressings. Moisture contents of red and green pepper flakes were $14.57{\pm}0.13%\;and\;11.86{\pm}0.12%$, capsaicinoids contents on a dry-weight basis were $207.91{\pm}6.68\;mg/100\;g\;and\;172.32{\pm}5.94\;mg/100\;g$, and total free sugars were $21.63{\pm}1.81%\;and\;12.49{\pm}0.56%$, respectively. ASTA color of red pepper flakes was $150.93{\pm}4.56$. Viable cell numbers of red and green pepper flakes were $3.9X10^4\;CFU/g\;and\;5.2X10^5\;CFU/g$, respectively. The pH values in French dressing and Italian dressing, with added red and green pepper flakes, were slightly increased, but not in American Caesar dressing. Acidity and viscosity weren't statistically different in the 3 kinds of dressings by the addition of red and green pepper flakes. The color difference $({\Delta}E^*_{ab})$ for the storage periods was calculated in each dressing and that of American Caesar dressing with added red pepper flakes was extremely high. Viable cell number in each dressing was maintained at 2 log CFU/g during the storage periods. The sensory characteristics during the storage periods were assessed by members of a trained panel, and showed that the color and pungency of red and green flakes increased the overall preference in the Italian dressing and American Caesar dressing.
