The experiment was conducted to investigate accumulation o( phosphate in tobacco production area. The results are as follows: 1) The content of available P in burley production area was about 200ppm higher than that in flue-cured tobacco production area, the available P content of 20% tobacco fields surveyed was above 1000ppm P. 2) Tobacco fields having 200ppm of available P which were grouped as medium category for phosphate recommendation were more than 90% out of tobacco fields surveyed. 3) Total phosphate content was above 4000ppm in 42.5% burley tobacco fields before transplanting, and 2000~3000ppm in about 40% flue-cured tobacco fields. 4) Phosphate fractions in soil increased in order of Al-P > Fe-P > Ca-p, and their content was about 50% of total phosphate in tobacco fields surveyed. 5) pH value in most of tobacco fields surveyed ranged from 4.5 to 5.0 and, field soils with pH value below 5.5 were 64 %.
An experiment was conducted to investigate the effect of niacin supplementation on hepatic lipid metabolism in rabbits. Rex Rabbits (90 d, n = 32) were allocated to two equal treatment groups: Fed basal diet (control) or fed basal diet with additional 200 mg/kg niacin supplementation (niacin). The results show that niacin significantly increased the levels of plasma adiponectin, hepatic apoprotein B and hepatic leptin receptors mRNA (p<0.05), but significantly decreased the hepatic fatty acid synthase activity and adiponectin receptor 2, insulin receptor and acetyl-CoA carboxylase mRNA levels (p<0.05). Plasma insulin had a decreasing tendency in the niacin treatment group compared with control (p = 0.067). Plasma very low density lipoproteins, leptin levels and the hepatic adiponectin receptor 1 and carnitine palmitoyl transferase 1 genes expression were not significantly altered with niacin addition to the diet (p>0.05). However, niacin treatment significantly inhibited the hepatocytes lipid accumulation compared with the control group (p<0.05). In conclusion, niacin treatment can decrease hepatic fatty acids synthesis, but does not alter fatty acids oxidation and triacylglycerol export. And this whole process attenuates lipid accumulation in liver. Besides, the hormones of insulin, leptin and adiponectin are associated with the regulation of niacin in hepatic lipid metabolism in rabbits.
Garlic oil (GAR, Allium sativum L.) has been studied as a feed additive to improve animal production performance and decrease methane emission in ruminants. The present study was designed to determine the possible effect of GAR on fatty acid composition and accumulation in animal fat tissue using a cell model. 3T3-L1 preadipocytes at $2{\times}10^{4}\;mL^{-1}$ were seeded to 24-well plates and allowed to proliferate to reach confluence. The cells were then treated with media containing 0, 2.5, 5, 10, 20 and 40 $\mu{g}$$mL^{-1}$ of GAR during the differentiation period for 8 days. Media containing dexamethasone, methyl-isobutylxanthine and insulin was applied during the first 2 days of the early differentiation period. On day 8 sub-sets of the wells were stained with oil red-O and the remaining cells were harvested for determination of glycerol-3-phosphate dehydrogenase [EC 1.1.1.8] (GPDH) activity (n = 6) and cellular fatty acid concentration (n = 6). It was found that supplementation of GAR increased (p<0.05) the ratio of monounsaturated fatty acids/saturated fatty acids in the adipocytes and showed inhibitory effect (p<0.05) on the post-confluent proliferation. With relative low dosage, GAR (5-20 $\mu{g}$$mL^{-1}$) increased (p<0.05) the GPDH activity without affecting the cellular fatty acid concentration, while a high dosage (40 $\mu{g}$$mL^{-1}$) inhibited (p<0.05) fatty acid accumulation and decreased GPDH activity. Supplementation of GAR had an effect on cell post-confluent proliferation, differentiation and fatty acid accumulation. However, the effect may be diverse and depends on the dose applied.
Cultures of Saccharomyces cerevisiae were subjected to the effect of PEF (pulse electric field) and a source of selenium. The culture period after which yeast cells were subjected to PEF treatment was optimized, as was the duration of the exposure. Optimization of the nutrient medium composition in S. cerevisiae cultures resulted in an over 1.8-fold increase in selenium accumulation with relation to cultures on the initial substrate. Optimization of the pH value and of culture duration resulted in selenium accumulation increase by approximately 78%. A significant correlation was found between the accumulation of selenium in yeast cells and its concentration in the culture substrate. The highest accumulation of selenium in the biomass of yeast, approx. $240\;{\mu}g/g$ d.m., was obtained after 15-min exposure to PEF on a 20-h culture. An approx. 50% higher content of selenium in cells was recorded, as compared with the control culture without the application of PEF.
Heavy application of fertilizer and manure in excess from the optimum requirement for crop growth can increase phosphorus (P) accumulation and P release potential in soils. In this study, the relationship between soil test P and 0.01M $CaCl_2$ extractable P was analysed to evaluate the P release potential of agricultural soils under different land-use. The paddy, upland, plastic film house(PFH) soils were sampled from Tongyoung and Changnyeong, and Daegok areas in Gyeongnam province, respectively. With respect to the P accumulation, available P contents in upland and PFH soils were 619 and 796 mg $P_2O_5$/kg, respectively indicating that different land-use types can greatly impact soil P accumulation. As soil available P was increased in the paddy soil, the content of 0.01M $CaCl_2$ extractable P also linearly increased without change point. Comparatively, P threshold were detected at 520 mg $P_2O_5$/kg in both upland and PFH soils, indicating that P release potential were higher in these land-use systems. For reducing P release from agricultural soils, management of optimum P content is needed in soils possessing high P release potential. Further, the change point value, if it is to be used as an environmental indicator, requires more detailed investigation to cover a wide range of soil characteristics.
Heavy metal-tolerant microorganisms, such as Pseudomonas putida, P. aeruginosa, P. chlororaphis and P. stutzeri which possessed the ability to accumulate cadmium, lead, zinc and copper, respectively, were isolated from industrial wastewaters and mine wasewaters polluted with various heavy metals. Metal binding sites in the cells were investigated by extracting the components of the cells through pretreatments with hot water, acid, alkli, chloroform-methanol or chloroform-methanol/concentrated alkali. The heavy metal accumulation was drastically decreased by pretreatment with alkali or chloroform-methanol/concentrated alkali, but the heavy metal accumulation was not changed by pretreatment with chloroform-methanol. The amount of heavy metal accumulation was remarkably decreased by decreasing crude protein remaining in the cell. These results suggested that proteins of cell components played an important role on the heavy metal accumulation.

Nitrite accumulation is essential for constructing an anammox process. As the pH in the reactor exerts a complicated and strong influence on the reaction rate, we investigated its effects upon treatment of an ammonic wastewater (2,000 mgN/L) through modeling and experiment. The modeling results indicated that the reaction stability is strongly affected by pH, which results in a severe reduction of the 'stable region' of operation under alkaline environments. On a coordinate of the total ammonia nitrogen (TAN) concentration vs. pH, the maximal stable reaction rates and the maximal nitrite accumulation potentials could be found on the 'stability ridge' that separates the stable region from the unstable region. We achieved a stable and high ammonia oxidation rate (${\sim}6kgN/m^3-d$) with a nitrite accumulation ratio of ~99% when operated near the 'stability ridge'. The optimum pH that can be observed in experiments varies with the TAN concentrations utilized, although the intrinsic optimum pH is fixed. The direction of change is that the optimum operational pH falls as the TAN concentration increases, which is in excellent accordance with the observations in the literature. The optimum operational pH for 95% nitritation was predicted to be ~8.0, whereas it was ~7.2 for 55% partial nitritation to produce an anammox feed in our experimental conditions.

Differential-pulse cathodic stripping voltammetry was applied to the Sn(II)-cupferron complex in 0.1 M acetate buffer solution (pH 4.20). Effects of solution pH, ligand concentration, accumulation potential, and accumulation time on the reduction peak current for the adsorptive complex of Sn(II)-cupferron were investigated. Interferences by other metal cations that affected on reduction peak current were also discussed. The detection limit was 3.1${\times}$10-9 M (0.37 ppb) of Sn(II) with 60 seconds accumulation time. The relative standard deviation (n=8) for 5${\times}$10-8 M Sn(II) was 3.0%.
The characteristics of absorption and accumulation of inorganic germanium in Panax ginseng C. A. Meyer were examined. In 4-year-old P. ginseng, the germanium content of the field soil increased with increased amounts and frequencies of inorganic germanium application, while chemical components of the soil, such as available phosphate and exchangeable calcium, potassium, and magnesium, decreased with the increased inorganic germanium application. In the 4-year-old P. ginseng, the germanium content was highest in the rhizome and increased in the order of stem, leaf, lateral root, and main root, suggesting that inorganic germanium was absorbed from the root and translocated to the stem and leaf via the rhizome. As for changes in ginsenosides in 4-year-old P. ginseng rhizomes, the contents of ginsenosides $Rb_1$, $Rb_2$, Re, and Rf decreased as the germanium content in soil increased. Ginsenosides $Rb_1$, $Rb_2$, Rc, Re, and Rf in the main root also decreased with increasing germanium content in the main root. The results suggest that inorganic germanium treatment may increase organic germanium in harvested P. ginseng, thus enhancing the medicinal effi cacy of ginseng products.
Metformin is the most widely used anti-diabetic drug that helps maintain normal blood glucose levels primarily by suppressing hepatic gluconeogenesis in type II diabetic patients. We previously found that metformin induces apoptotic death in H4IIE rat hepatocellular carcinoma cells. Despite its anti-diabetic roles, the effect of metformin on hepatic de novo lipogenesis (DNL) remains unclear. We investigated the effect of metformin on hepatic DNL and apoptotic cell death in H4IIE cells. Metformin treatment stimulated glucose consumption, lactate production, intracellular fat accumulation, and the expressions of lipogenic proteins. It also stimulated apoptosis but reduced autophagic responses. These metformin-induced changes were clearly reversed by compound C, an inhibitor of AMP-activated protein kinase (AMPK). Interestingly, metformin massively increased the production of reactive oxygen species (ROS), which was completely blocked by compound C. Metformin also stimulated the phosphorylation of p38 mitogen-activated protein kinase (p38MAPK). Finally, inhibition of p38MAPK mimicked the effects of compound C, and suppressed the metformin-induced fat accumulation and apoptosis. Taken together, metformin stimulates dysregulated glucose metabolism, intracellular fat accumulation, and apoptosis. Our findings suggest that metformin induces excessive glucose-induced DNL, oxidative stress by ROS generation, activation of AMPK and p38MAPK, suppression of autophagy, and ultimately apoptosis.
