Effect of small Black Soybean Fraction on the T cell-mediated Immune Responses in vivo and Proliferation of Leukemia Cells in vitro

  • We investigated effect of small black soybean fraction (SBSF) T cell-mediated responses for tumor surveillance and proliferation in leukemia cells in vitro. Each SBSF butanol fraction (SBSFBu) and SBSF chloroform fraction (SBSFCh) was administered p.o. once a day far 21 days in BALB/c mice and then levels of serum cytokines and subpopulation of lymphocytes were measured. Moreover, SBSF fraction was treated into the cultured various cell lines for proliferation in leukemia cell lines, NO production by RAW264.7 cells, and expression of p53 gene in U937 leukemia cells. These results showed that SBSFBu increased levels of serum IL-4but not IL-2 and IFN-${\gamma}$, and increased expression of CD4$^+$ T cells and CD8$^+$ T cells in splenocytes in vivo, while SBSFCh increased levels of serum IL-2 and IFN-${\gamma}$ but decreased IL-4, and increased CD8$^+$ T cells but not CD4$^+$ T cells. Moreover, both of SBSFBu and SBSFCh inhibited proliferation of HL60, U937, and L1210 leukemia cell lines in a dose-dependent manner, up-regulated NO production by RAW264.7 cells in a dose-dependent manner, and enhanced expression of p53 gene in U937 leukemia cells. Our findings indicate that SBSFBu and SBSFCh may enhance T cell-dependent immune responses, and that both of SBSFBu and SBSFCh may inhibit proliferation of leukemia cells by up-regulation of NO production and expression of p53 gene.

In vitro Evaluation of Phalaris minor Seeds as Livestock Feed

  • The nutritional worth of Phalaris minor seeds was assessed in comparison to conventional cereal grains like maize and wheat. P. minor seeds had higher total ash and cell wall constituents as compared to wheat and maize grains. The CP content of P. minor was comparable to wheat grains but higher than maize grains. The in vitro studies revealed that the net gas production and availability of ME from P. minor was comparable to that of maize but the digestibility of nutrients was significantly (p<0.05) lower than that of conventional cereal grains. The digestion kinetic parameters for DM and CP revealed that P. minor had the highest (p<0.05) soluble fraction (a) followed by wheat and maize. Reverse trend was observed for insoluble but potentially degradable fraction (b). The effective and true DM and CP degradability was significantly (p<0.05) higher in wheat grains followed by that in P. minor and maize grains. The digestibility of OM and NDF was not affected by replacing cereal grains in concentrate mixture with P. minor seeds up to 75 per cent level. But the availability of ME from concentrate mixtures was comparable to control only up to 50% level of replacement. Replacement of cereal grains with P. minor did not affect the rapidly soluble fraction and insoluble but potentially degradable fraction of concentrate mixture containing P. minor up to 75 per cent, but it was depressed significantly at 100% replacement level. The effective and true degradability of DM of concentrate mixtures containing P. minor from 50 to 100 per cent was comparable to that of conventional concentrate mixture (CCM). The wheat based concentrate mixtures showed higher net gas production (208 vs. 201 ml/g DM/24 h), digestibility of nutrients and ME availability (9.64 vs. 9.54 MJ/kg DM) as compared to maize based concentrate mixture. The wheat based concentrate mixture had significantly (p<0.05) higher rumen undegradable fraction and effective degradability. The data conclusively revealed that conventional cereal grains could be replaced with P.minor seeds up to 75 per cent without affecting the availability of nutrients.

각종 변이원에 대한 쇠비름 추출물의 돌연변이 억제 효과 (Inhibitory Effects of Poturaca oleracea L. Extract on the Mutagenicity of Various Mutagen)

  • This study was performed to determine the effects of antimutagenicity of Porturaca oleracea L. in Korea. In Ames test, the ethanol extract of Poturaca oleracea L. inhibited mutagenic activity of N-methyl-N'-nitro-N-nitrosoguanidine(MNNG), , 4-nitroquinoline-1-oxide(4NQO), benzo($\alpha$)pyrene (B($\alpha$)P) and 3-amino-1,4-dimethyl-5H-pyrido-(4,3-b)indole (Trp-P-1) in Salmonella typhimurium TA98 and TA100. But hot-water extract Poturaca oleracea L. only Inhibited mutagenic activity of MNNG in Salmonella typhimurium TA100, On 4NQO, the ethanol extract 100-1,600$\mu\textrm{g}$/plate of Porturaca oleracea L. showed a slight inhibitory effect of 13-48%, 4-47% in TA98 and TA100, respectively, but on MNNG, it showed higher inhibitory effect of 6-86% in TA100, And the treatment of 1,600$\mu\textrm{g}$/plate of ethanol extract of Porturacea L. had strong antimutagenicity with 74-87% inhibition against TA98 and TA100 induced by B(a)P and with 85-93% inhibition against TA98 and TA100 induced by Trp-P-1. The ethanol extract was fractionated with ether. chloroform, ethylacetate, butanol and water. Among them, most of the fraction except water fraction showed strong antimutagenicity effects against mutation induced by 4-NQO, MNNG, B(a)P and Trp-P-1. Chloroform fraction had strong antimutagenicity with 91% inhibition against TA100 induced by MNNG, diethyl etherfraction had strong antimutagenicity with 92%, 98% inhibition against TA98 and TA100 induced by 4NQO, Chloroform fraction had strong antimutagenicity with 97% inhibition against TA100 induced by B (a)P and diethyl etherfraction had strong antimutagenicity with 98% inhibition against both strain Induced by Trp-P-1, respectively.

휴ㆍ폐광산지역에서 폐재내 중금속의 존재형태 및 용출특성에 관한 연구 (A Study on Fractions and Leaching Potential of Heavy Metals in Abandoned Mine Wastes)

  • 연구의 목적은 광산활동에 의해 발생된 폐재에 의해 오염된 토양에 포함된 중금속의 존재형태와 용출특성에 대하여 비교 분석하는데 있다. 실험에 이용된 중금속 존재형태 분류는 다섯가지로 adsorbed, carbonate, reducible, organic과 residual 형태로 나누었다. 토양내 유기물의 함량 정도는 상동광산에서 발생된 광미가 석탄폐재보다 낮은 것으로 나타났다. 오염된 토양의 중금속 용출 실험에 의하면 석탄폐석과 광미내의 카드뮴의 경우 낮은 용출량을 보였으며, 대부분이 reduciblel fraction에서 용출된 것으로 관찰되었다 상동광산에서 발생된 광미에 포함된 니켈은 혐기성 상황이 되면 총량의 약 46.4%가 용출될 것으로 사료되며, 구리의 경우 약 39.4%가 용출될 것으로 판단된다. pH 3.0과 pH 5.6의 용출조건하에서 석탄 폐석과 광미의 용출랑의 점진적인 증가는 30분안에 이루어졌으며, 광미의 경우 니켈은 pH 3.0과 pH 5.6에서 용출 3시간까지 계속 증가되는 상태를 보였다.

마와 꿀풀 추출물에 의한 Streptococcus mutans의 산 생성 및 Glucosyltransferase 저해효과 (Glucosyltransferase Inhibition of Streptococcus mutans by Dioscorea batatas and Prunella vulgaris extract)

  • 마, 꿀풀 추출물의 용매 분획별 구강병인균에 대한 항균 및 항우식 효과를 알아본 결과 다음과 같은 결과를 얻었다. S. mutans에 대한 마, 꿀풀 헥산 추출물의 상대성장률과 산 생성 억제 및 GTase 활성 저해효과을 알아본 결과, 마 헥산 추출물의 0.25 mg/ml 농도에서 100%, 꿀풀헥산 추출물의 0.5 mg/ml 농도에서 100% 성장을 억제하였다. 따라서 농도가 증가함에 따라 성장이 억제되는 것을 볼 수 있었다. 2.0 mg/ml 농도에서 마가 pH 7.2, 꿀풀이 pH 7.0로 나타나 pH 5.6, pH 5.2인 대조군과 비교하여 볼 때 마, 꿀풀 헥산 추출물을 첨가물이 산 생성을 억제하여 중성으로 보여주고 있으며, GTase 활성 저해효과는 마와 꿀풀의 헥산 추출물의 2.0 mg/ml농도에서 35%, 25%로 각각 활성을 저해하였다.

염산-에탄올에 의해 유발된 흰쥐 위염에 대한 마치현의 억제 작용 (The Inhibitory Effects of Portulaca oleracea L. on HCl-ethanol Induced Gastritis in Rats)

  • Objectives : The objective of this study was to examine the effects of P. oleracea into the HCl-ethanol induced gastritis in rats, and to isolate and determine the chemical compounds from P. oleracea. Methods : The rats were orally administered with crude extract or fractions or isolated compounds of P. oleracea 30 mins before the induction of gastric lesion by oral administration of HCl-ethanol. The gastric lesional area was measured using pixel counting software. Then the chemical compounds from P. oleracea was isolated and determined by LC-MS and NMR. Results : The inhibition effect of oral administration of crude extract of P. oleracea at a dose of 500 mg/kg in HCl-ethanol induced gastritis was similar to cimetidine. Then, aqueous fraction at a dose of 240 mg/kg exhibited the effects similar to cimetidine. Then, the aqueous fraction was further separated by MPLC and yielded four sub fractions. Among those sub fractions, agent II at a dose of 40 mg/kg possessed the strongest effect in the HCl-ethanol induced gastritis. The water fraction yielded-Uridine, Adenosine, Guanosine, which were characterized by Mass, 1H-NMR, 13C-NMR. Conclusions : This study suggest that a P. oleracea and its compounds showed potent efficacy on the development of HCl-ethanol induced gastritis. Thus, P. olaracea can be a potential natural resource for the management of gastritis although the mechanism of action involved in the treatment remains to be explored.

SOS Chromotest에서 숙지황 물 추출물의 세포내 항돌연변이 효과 (Bio-antimutagenic effects of water extract from Rehmannia glutinosa Liboschitz in SOS Chromotest)

  • 숙지황 물 추출물의 항돌연변이 활성을 4-nitroquinoline 1-oxide (4-NQO), N-methyl-N'-nitro-N-nitrosoguanidine (MNNG), mitomycin C (MMC), $aflatoxin\;B_1\;(AFB_1)$, benzo(a)pyrene [B(a)P]의 변이원성에 대하여 SOS Chromotest로 조사하였다. 숙지황 물 추출물을 메탄올 가용성 부분과 불용성 부분으로 분리하여 시험한 결과 5종의 변이원의 활성에 대하여 메탄올 가용성 분획은 불용성 분획 보다 강한 억제효과를 나타내었다. 따라서 메탄올 가용성 분획을 에틸아세테이트, 부탄올, 물 분획 순으로 순차용매 분리하였고 그 분획물 중, 물 분획이 5종 변이원의 돌연변이원성에 대하여 가장 강한 억제효과$(4.5{\sim}29.5%)$를 나타내었고, 에틸아세테이트 분획은 $AFB_1$의 변이원성을 촉진하였다. Sephadex LH-20 column chromatography를 이용하여 물 분획을 정제하여 9 분획을 얻었으며, 이들 중 fraction III가 시험한 5종의 변이원으로 각각 유도된 변이원성에 대하여 용량반응의 억제효과와 함께 가장 강한 억제활성을 나타내었다. $400\;{\mu}g/assay$ 농도의 Fraction III는 4-NQO, MNNG, MMC, AFB1 및 B(a)P의 돌연변이원성에 대하여 각각 29, 35, 38, 25, 24% 억제효과를 나타내었다. Fraction III는 4-NQO, AFB1의 변이원성에 대하여 40% 이상의 강한 세포내 항돌연변이원성을 나타내었다.

  • pH7.0으로 조절한 2M의 AsA 용액을 $85^{\circ}C$에서 25시간 갈변반응시켜서 생성된 물질 중 항산화효과를 지닌 물질을 분리하고 성질을 조사하였다. 투석내액 및 투석외액 갈변반응물질 모두 항산화성이 존재하였고 투석외액 갈변반응물질의 항산화효과가 더 크게 나타났다. 투석외액 갈변반응물질을 gel 여과하여 분리된 3가지 획분(A, B, C)중 갈변도가 가장 높고 환원력이 가장 낮은 획분(C)의 항산화효과가 가장 크게 나타났다. 항산화성이 중간정도인 획분(B)은 환원력의 세기와 갈변도가 세획분중 중간정도에 속하였고 환원력은 가장 높고 갈변도가 가장 낮은 획분(A)은 항산화효과가 가장 낮았다. UV-Visible spectrum에서 A획분과 B획분은 각각 266.1 및 257.4nm에서 최대흡광 peak를 나타냈으나 C획분은 274.8nm에서 약한 peak를 나타내었다. IR-spectrum 조사결과 A, B 및 C획분 모두 hydroxy 및 carboxylic group이 존재하였고 항산화성이 큰 B와 C분획은 carboxylic ester이었다.

  • 혐기성 Clostrium therrnocellurn의 배양액으로부터 hydroxyapatite column chromatography를 통하여 cellulase complex중의 $C_{1}$ enriched fraction을 분리하였다. 다은 호기성 미생물과 마찬가지로 분리된 $C_{x}$, fraction과 다른 $C_{x}$ 성분과의 synergism에 의해 볼용성 성유소의 분해가 현저히 촉진되였다. $C_{x}$ 성분과는 달리 $C_{1}$, fraction은 공기중에서 산화에 의해 잘 실활되었으나 환원제, 특히 $\beta$-mercaptoethanol에 의해 효소 환성이 강하게 증가되는 것으로 보아 $C_{1}$ component는 다량의 sulfhydryl grou을 가지고 있는 것으로 판단되었다. 이 fraction은 열에 매우 안정하였으며 최적 온도와 pH는 각각 $60^{\circ}C$와 6.0 이었다.

사상자의 항염증작용 (Anti-inflammatory Activities of Torilis japonica Fruit)

  • The methanol extract of Torilis Fructus showed potent anti-edematous effect in carrageenan-induced paw edema in rats. Systematic fractionation of the extract with hexane, chloroform, ethyl acetate and butanol resulted in potent anti-edematous action only in hexane fraction at an oral dose of 237 mg/kg which is corresponding to 4.8 g of raw Fructus. This hexane fraction showed inhibitory effects at 77 mg/kg p.o on vascular permeability in mice, at 8mg/pouch on leucocyte emigration in rats, and at 120 mg/kg p.o. on adjuvant arthritis model in rats. These results demonstrate that hexane fraction of the extract possesses potent anti-inflammatory effect which may support its traditional uses.
