Subpopulations of miniature pig mesenchymal stromal cells with different differentiation potentials differ in the expression of octamer-binding transcription factor 4 and sex determining region Y-box 2

  • Objective: Human mesenchymal stromal cells (MSCs) exhibit variable differentiation potential and can be divided accordingly into distinct subpopulations whose ratios vary with donor age. However, it is unknown whether the same is true in pigs. This study investigated MSC subpopulations in miniature pig and compared their characteristics in young (2 to 3 months) and adult (27 to 35 months) pigs. Methods: Osteogenic, chondrogenic, and adipogenic capacity of isolated MSCs was evaluated by von Kossa, Alcian blue, and oil red O staining, respectively. Cell surface antigen expression was determined by flow cytometry. Proliferative capacity was assessed with the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. Expression of marker genes was detected by quantitative real-time polymerase chain reaction. Results: Porcine MSCs comprised cells with trilineage and bilineage differentiation potential (tMSCs and bMSCs, respectively) and non-differentiating stromal cells (NDSCs). The tMSC and bMSC fractions were smaller in adult than in young pigs (63.0% vs 71.2% and 11.6% vs 24.0%, respectively, p<0.05); NDSCs showed the opposite trend (25.4% vs 4.8%; p<0.05). Subpopulations showed no differences in morphology, cell surface antigen expression, or proliferative capacity, but octamer-binding transcription factor 4 (OCT4) expression was higher in tMSCs than in bMSCs and NDSCs (p<0.05), whereas sex determining region Y-box 2 (SOX2) expression was higher in tMSCs and bMSCs than in NDSCs (p<0.05). Aging had no effect on these trends. Conclusion: Porcine MSCs comprise distinct subpopulations that differ in their differentiation potential and OCT4 and SOX2 expression. Aging does not affect the characteristics of each subpopulation but alters their ratios.

  • Polo-like kinase 1 (Plk1) is crucial regulator of cell cycle progression during mitosis. It is known to highly overexpress in many different tumor types, and has been implicated as a potential antimitotic cancer target. The phosphopeptide, Pro-Leu-His-Ser-p-Thr (PLHSpT), was shown a high level of affinity and specificity for the polo-box domain (PBD) of Plk1. However, the peptide has the limitation of cell permeability. We designed the derivatives to enhance the limitation of PLHSpT using drug delivery system. In addition, we synthesized and evaluated its radiotracer for tumor diagnosis. This review discusses the derivative and radiotracer that are suitable for tumor treatment and diagnosis for PBD of Plk1.

  • Determination of sex origin of cattle meat by fast and reliable molecular methods is an important measure to ensure correct allocation of export refunds particularly in European countries and also female cattle (cow) slaughter is legally banned in India because of religious beliefs. Based on the DEAD box protein gene located on the X and Y chromosomes, 2 pair of primers were designed and the system of PCR was optimized. Upon PCR amplification, male tissue showed 2 bands, while female tissue resulted in only one band. The accuracy and specificity of the primers was assessed using DNA template extracted from cattle meat of known sex. The protocol was subjected to a blind test and showed 100% concordance, proving its accuracy and reliability.

  • To investigate how the dyad symmetry region (DSR) and the distance between vir box and -35 sequence of the virE promoter plays a role in virE gene expression, two mutants were constructed by base substitution and insertional mutagenesis. The base substitutional mutation, a AAlongrightarrowCG substitution at positions -39 and -40 on the DSR, showed the level of $\beta$-galactosidase activity approximately 91% of the wild type virE promoter activity. Therefore, the native structure of the DSR seems to be not essential for virE expression. The insertional mutation, constructed by inserting 8 bp ClaI linker between -49 and -50, displayed the $\beta$-galactosidase activity at 12% of the native virE promoter activity. However, this striking reduction appears to be not caused by destruction of the native DSR structure, but by shifting the vir box far from putative -35 sequence.

  • In literature for thin-walled sections with L, [, +, I, and ${\Box}$- shapes the approximate torsion equations for stiffness are used which were proposed by Bach (Hsu 1984), p.30. New formulae for torsional stiffness of bars with L, [, +, I, and ${\Box}$ cross section valid not only for thin-walled sections are presented in this paper. These formulae are obtained by appropriate polynomial approximation of stiffness results obtained by means of method of fundamental solutions. On the base of obtained results the validity of Bach's formulae are verified when cross section is not thin-walled.

  • In this study, the elimination of $Ni^{2+}$ using 13X sorbent due to an electrostatic interaction was reported. The significant factors including pH, time and 13X sorbent amount were investigated using Box-Behnken design (BBD). In the optimum experimental conditions, the linear rang and limit of detection of the proposed method were 0.1-20 and $0.102mg\;L^{-1}$, respectively. The precision as RSD% was 1.3% for concentration of $2mg\;L^{-1}$. Concerning the excellent recoveries in a short time with highly efficient sample clean-up and removal, this method may be a very powerful and innovative future sample removal technique. To the best of our knowledge, this is the first report on using BBD for optimizing the parameters affected the removal of $Ni^{2+}$ by 13X zeolite sorbent.

  • The Korea high-speed rail, based on the French design. It also implements new concept to increase the strength of bridge deck by adding an impact factor (dynamic intensity factor) in static load. In order to assure the dynamic stability, SYSTRA and Jeseph Penzien, a professor in CEC (the US) conducted a dynamic stability review on design phase. Analyzing the review results, they developed design criteria for dynamic behavior. This study deal with operating PSC box GIRDER equipped with measurement equipment or measured data of Seoul $\∼$ Taejeon, P.S.C BOX GIRDER bridge and steel comsition bridge equipped with measurement equipment based on structual knowledge about configuration of measuring sensor, response analysis of structure when train runs was performed by using measured data of PSC box girder to directly compare with design criteria. moreover, the dynamic stability with comparison of high-speed rail construction criteria was reviewed and analyzed based on historical records.

Box-Cox Power Transformation Using R

  • If normality of an observed data is not a viable assumption, we can carry out normal-theory analyses by suitable transforming data. Power transformation by Box and Cox, one of the transformation methods, is derived the power which maximized the likelihood function. But it doesn't induces the closed form in mathematical analysis. In this paper, we compose some R the syntax of which is easier than other statistical packages for deriving the power with using numerical methods. Also, by using R, we show the transformed data approximately distributed the normal through Q-Q plot in univariate and bivariate cases with some examples. Finally, we present the value of a goodness-of-fit statistic(AD) and its p-value for normal distribution. In the similar procedure, this method can be extended to more than bivariate case.

Changes of Characteristics in Salted Baechu(Chinese Cabbage) and Its Exudate during Long Term Storage (장기저장중 절임 배추와 그 삼출액의 특성변화)

  • Chemical, physical md microbial analysis of salted baechu(Chinese cabbage) and its exudate were conducted during storage in large plastic bags(LDPE, HDPE, PVC-box) at 0$^{\circ}C$. Salinity was slightly lowered for 2 weeks storage and maintained that level thereafter in salted haechu, but in its exudate increased rapidly for 2 weeks storage and decreased thereafter. In all treatment pH decreased to 5.0 in salted baechu and to 4.5 in its exudate for 6 weeks storage. Reducing sugar content of salted baechu was 1.5∼2.0 fold higher than that of its exudate, and decresed slowly in both. Total viable cells increased rapidly for 2 weeks but maintained that level thereafter and lactic acid bacteria increased continuously until 6 weeks. Lightness of exudate decreased rapidly in PVC-box, but decreased slowly in LDPE. Compression force of salted baechu increased continuously untill 8 weeks except for PVC-box.

Finite strip analysis of a box girder simulating the hull of a ship

  • In the present study, the finite strip analysis of a box girder to simulate a ship's hull model is carried out to investigate its inelastic post-buckling behavior and to predict its ultimate flexural strength. Residual stresses and initial geometrical imperfections are both considered in the combined material and geometrical nonlinear analysis. The von-Mises yield criterion and the Prandtl-Reuss flow theory of plasticity are applied in modeling the elasto-plastic behavior of material. The Newton-Raphson iterative process is also employed in the analysis to achieve convergence. The numerical results agree well with the experimental data. The effects of some material and geometrical parameters on the ultimate strength of the structure are also investigated.
