Differences in the Community Structures of Macrobenthic Polychaetes from Farming Grounds and Natural Habitats in Gamak Bay (가막만 양식장과 자연 서식지에서의 대형저서다모류 군집구조 차이)

  • This study was carried out to investigate the differences in sedimentary environments and benthic polychaete communities between farming grounds and natural habitats (non-farming ground) in Gamak Bay. Sampling stations of natural habitats were evenly distributed in the entire bay. And mussel farm, oyster farm and ark-shell farm were selected as farming grounds. Dominant sedimentary facies was mud in most sampling stations of farming grounds and natural habitats. However organic contents were higher in the farming grounds than natural habitats of the bay. The species number and mean density of polychaetous community in the natural habitats were greater than those from the farming grounds. Lumbrineris longifolia, known as potential organic enrichment indicator species, was first dominant species both in farming grounds and natural habitats of the bay. However, the next dominant species consisted of different species between two benthic habitats. As a result of community analysis using cluster analysis and nMDS, the natural habitats were divided into several station groups, but most of stations in farming grounds were clustered into one group. Pearson' correlation analysis and PCA showed high relationships between sedimentary environmental factors and benthic polychaetous community in natural habitats, but low or no relationships in farming grounds. That means benthic polychaetous community established in farming ground was under unusual condition such as high input of organic matter. Thus it is necessary to improve the benthic environmental quality of the farming grounds as well as the north-western inner part in Gamak Bay through long-term monitoring efforts.

Dietary Calcium and Non-phytin Phosphorus Interaction on Growth, Bone Mineralization and Mineral Retention in Broiler Starter Chicks

  • An experiment was conducted to study the requirement of calcium (Ca) and non-phytin phosphorus (NPP) in commercial broilers during starter phase. Seven hundred and twenty day-old Vencob male broiler chicks were randomly distributed into 144 stainless steel battery brooders, 5 birds in each. Four levels each of Ca (6, 7, 8, and 9 g/kg) and NPP (3, 3.5, 4, and 4.5 g/kg diet) were fed in a factorial design in a corn-soya basal diet. Levels of dicalcium phosphate and oyster shell grit were adjusted to obtain the desired levels of Ca and NPP. Each diet was fed ad libidum to chicks in 9 battery brooders from one d to 21 d of age. Body weight gain and feed intake were depressed (p<0.01) by increasing the dietary Ca level (8 and 9 g/kg) at lower levels of NPP (3 and 3.5 g/kg diet). The growth depression observed at lower NPP level was alleviated by reducing the Ca content to 6 g/kg diet. The tibia ash content and tibia breaking strength increased with increase in both Ca (>6 g/kg) and NPP (>3 g/kg) levels. The leg abnormality score decreased (p<0.01) with increase in NPP content in the diet at all levels of Ca tested. The serum Ca and inorganic P levels were increased with increase in the level of the respective mineral in the diet, but the serum concentration of Ca and P were inversely related to the level of NPP and Ca, respectively /kg diet. In general, the excretion of macro minerals (Ca, and P), and micro minerals {zinc (Zn), manganese (Mn), iron (Fe), and copper (Cu)} was significantly lower at lower levels of Ca and NPP tested (6 and 3 g/kg diet, respectively). The mineral excretion increased with increase in dietary Ca and NPP levels, more conspicuously at the disproportionate ratio of these minerals (>2:1, Ca and NPP). Similarly, the retention of Zn, Mn, and Fe in liver was significantly higher (p<0.01) at lower levels of Ca and NPP tested. Results from this study indicate that the commercial broilers do not require more than 3 g NPP and 6g Ca/kg diet during starter phase (up to 21 d of age) for optimum weight gain, feed efficiency and utilization of Ca, P, Zn, Mn, Fe and Cu. However, the requirements of these minerals for optimum bone mineralization were higher than the levels suggested above.

The Growth of Vibrio vulnificus in Meat Homogenates of Fish and Shellfish (어패육에서의 Vibrio vulnificus의 증식에 관한 연구)

  • The change of cell counts of Vibrio vulnificus in meat homogenates of fish and shellfish by the storage time and temperature was examined to get basic information for precautionary steps against septicemia from slices of raw fish (sashimi). Therefore, we inoculated raw and cooked meat homogenates of fish and shellfish with Vibrio vulificus M-8 (isolated from shellfish ) and stored them at $-20^{\circ}C,\;4^{\circ}C\;and\;30^{\circ}C$ for 72 hours. Vibrio vulnificus M-8 was not detected in 32 hours when it was frozen and stored at $-20^{\circ}C$ after inoculating them into phosphate buffer solution at concentration of $10^5\;cell/ml$, while the existance of Vibrio vulnificus was identified after 72 hours of storage at the same temperature in case of inoculation into the meat homogenate of yellow tail. The cell count of Vibrio vulnificus was decreased as about $20\%$ of initial count after 2 hours storage at $4^{\circ}C$ in phosphate buffer solution with fish and shellfish homgenates. From the experimental results it was recognized that Vibrio vulnificus was labile to the cold stress. In comparison to the growth of growth of Vibrio M-8 at $30^{\circ}C$ in the raw and cooked meat of the yellow tail(Seriola guingueradita), snapper(Chrysophrys major), ark shell(Anadra brouhgtonii), and oyster(Crassostrea gigas), the raw meat homogenates were more excellent than the cooked ones though all fish and shellfish meat homogenates were proves to be good for the growth of the microbe.

Material Characteristics and Conservation Treatment for Floral Wall in Lee Sang-beom's House and Atelier (이상범 가옥 및 화실 내 꽃담의 재료학적 특성과 보존처리)

  • The floral wall of Lee Sang-beom's House and Atelier, which is No. 171 of the Registrated Cultural Heritage was conserved. In addition, materials characteristics and manufacturing technique have revealed through the scientific analysis. As a result of the analysis, samples were divided into three sections; the support layer that is made from block bricks, the paint layer and the cement mortar layer on the paint layer for the reinforcement of the construction. The higher layer lies, the finer grains it has. Furthermore, a little it was generated a small quantity of pores and calcium carbonates ($CaCO_3$) generated due to aeration of cement mortar. The patterns of letters, animals and plants pattern were expressed in the paint layer by relief and openwork. The results of qualitative analysis of the pigments of the paint layer were detected components of carbon black (C), Fe oxide ($Fe_2O_3$) and oyster shell white ($CaCO_3$, or quicklime). On the other hands, as the conservation of the floral wall, stainless frames were set up for the structural stability, the cement mortar were removed from the surface and the partly damaged and cracked areas were filled with KSE Filler A, B.

  • A focused ion beam (FIB) system produces a beam of positive ions (usually gallium) which are heavier than electrons and can be focused by electrostatic lenses into a spot on the specimen. With its ability milling of the specimen material by 10 to 100 nm with each pass of the beam, FIB is widely adopted in materials science, semiconductor industry, and ceramics research. Recently, FIB has been increasingly employed in the field of biomedical sciences. Here we provide a brief introduction to FIB and its applications for a wide variety of biomedical research. The surface of specimen can be in situ processed and quasi-real time visualized by two beam combination of FIB and field emission scanning electron microscope (FESEM). Due to its milling process, internal structures can be exposed and analyzed: yeast cells, fungus-inoculated wheat leaf, mannitol particles in inhalation aerosols, and oyster shell. Serial blockface tomography with the system kindles 3-dimensional reconstruction researches in the realm of nervous system and life sciences. Two-beam system of FIB/FESEM is a versatile tool to be utilized in the biomedical sciences, especially in 3-dimensional reconstruction studies.

Effects of Silver lon Exchanged Water Treatment Agent upon E. Coli RB 797 and Bacillus sp. (수처리제 은이온이 E. Coli RB 797과 Bacillus sp. 에 미치는 영향)

  • The effects of the silver ion-exchanged water treatment agent (Ag-Os) upon E. coli RB 797 and Bacillus sp. have been discussed in this study. Silver ion causes a number of toxic effects with no known biological function. Silver ion-exchanged water treatment agent (Ag-Os) using oyster shell here showed antimicrobial activities. the soluble form of silver ion in water is more toxic to the growth of Bacillus sp. than that of E. Coli RB 797. The minium amount of Ag-Os needed for growth inhibition is 0.2 mg/ml for E. Coli RB 797 and 0.02 mg/ml for Bacillus sp., which is consistant with the data of the survival cell fraction. Binding studies suggested that binding of silver to the cell surface was a rapid, metabolic-independent process and different from active transport. Bacillus sp. showed more binding than E. Coli RB 797. Reducing substances of the cell cultures in the presence of Ag-Os was detected using Methylen blue as an indicator. From these results, we suggest that Ag-Os is effective as an antimicrobial agent on E. Coli RB 797 and Bacillus sp. and silver binds to the cells through rapid, metabolic-independent process and might complex to sulfur group in the cells for its toxicity.

Characteristics of Soil Chemical Properties in Abandoned Coal Mine Forest Rehabilitation Areas in Boryeong City, Chungcheongnam-do

  • The objectives of this study were to investigate soil chemical characteristics for forest rehabilitation and suggest management in abandoned coal mine areas in Boryeong City, Chungcheongnam-do. Total study sites were seven sites, and soil properties analyzed were soil pH, total organic carbon (TOC), total-N, C/N ratio, and available $P_2O_5$ (A.v. P). Average soil pH (range) was 5.9 (4.5~7.0). Three study sites (Samgwang, Shinsung1, and Shimwon1) showed lower soil pH than standard (pH 5.6~7.3) of Korea Industrial Standards (KS) for abandoned coal mine forest rehabilitation. Average contents of TOC, and total-N were 1.5% (0.1~4.7%), and 0.10% (0.03~0.23%), respectively. Five study sites where the collapsed time was less than 10 years (Wangjashingang, Wonpoong, Samgwang, Shinsung1, and Shinsung2) showed lower TOC level than standard of KS (more than 1.2%). Wangjashingang, Wonpoong, Samgwang, and Shinsung1 showed lower level of total-N than standard of KS (more than 0.09%). C/N ratio of six study sites except Shimwon1 was out of proper range (15:1~30:1). Average A.v. P (range) was $20.7mgkg^{-1}$ (4.8~63.1), less than other abandoned coal mine fores rehabilitation areas in Mungyong City, and Hwasun-gun. TOC, total N and A.v. P increased with elapsed time from forest rehabilitation, while other soil properties did not show distinct pattern. Betula platyphylla was planted in Samkwang and Sinsung where soil pH was less than KS standard. Because the growth of Betula platyphylla can be limited in acid soil, it is necessary to neutralize soil pH to proper level with some soil amendment such as lime or shell of oyster. Furthermore, TOC, total-N and A.v. P in early stage of forest rehabilitation showed lower level than proper to vegetation growth. Therefore it needs continuous monitoring of soil characteristics and fertilization for vegetation growth and influx from surrounding forest in early stage of rehabilitation.

Spawning Behavior and Early Life History of the Elegant Blenny, Omobranchus elegans (Pisces: Blenniidae) (앞동갈베도라치의 산란행동 유도 및 초기생활사)

  • This study was conducted to observe the spawning behavior and early life histroy of elegant blenny, Omobranchus elegans reared in the laboratory. The elegant blenny were caught at Dolsan lsland, Yeosu-si, Jeollanamdo from June in 2012. As a result of observeation, male fish attracted female continuously and guide lead to spawning site and clean the surface of inner pant in oyster shell or barnacle shell. Female left after spawning and male protected their eggs until they had hatched out. The fertilized eggs were spherical in shape (mean diameter: 1.06 mm; mean oil globule diameter: 0.24 mm) and transparent. Larvae hatched at 203 hrs 40 mins after fertilization at $25.5{\sim}28.5^{\circ}C$ (mean $27.0^{\circ}C$). The newly hatched larvae were 3.04~3.09mm(mean 3.06 mm, n=10) in total length and their mouth and anus were already opened. They began to eat rotifer and transformed to postarvae stage. 10 days after hatching postlarvae was measured 6.39~6.45mm(mean 6.42 mm, n=10) in total length. 60 days after hatching juvenile was measured 21.5~22.2 mm (mean 21.8 mm, n=10) in total length with dosal fin rays XI-22; ventral fin rays I-2; anal fin rays II-23; caudal fin rays 21; pectoral fin rays 13.

Development studies of microalgae-based closed recirculating bivalves adults conditioning system: I. Induction of the gametogenesis (이매패류 어미관리를 위한 미세조류 기반 폐쇄-순환여과시스템 개발 연구: I. 성 성숙 유도)

  • Techniques were developed for holding and conditioning of Pacific oysters, Crassostrea gigas, in a closed recirculating system. Experimental adults were used 500 oysters ( x two system, total 1,000 oysters) which were collected in $20^{th}$ March 2016 from long-line aquaculture farm at the south coast of Korea. During conditioning periods concentrated live microalgae as Isochrysis sp. $15{\times}10^7cells/mL$, Tetraselmis sp. $2{\times}10^7cells/mL$ and Pheaodactylum sp. $18{\times}10^7cells/mL$ were added 5 L every day, respectively which micro algae were functioned as diets and biological filter. Over all experimental periods total water exchange rate was 21.3% (daily 0.5%). Over 42 days conditioning, female and male oysters were maturated 90.9% and 94.4%, respectively. Survival rate was 98.7%. Mean shell hight (8.3 mm), total wet weight (19.2 g), meat wet weight (5.0 g) and shell wet weight (13.6 g) were significantly increased (P < 0.05). Water quality parameters including the water temperature ($22.1{\pm}0.4^{\circ}C$), salinity ($24.9{\pm}04$), dissolved oxygen (5.1-7.9 mg/L) and pH ($7.93{\pm}0.15$) were kept stable. Concentration of dissolved inorganic nutrient as ammonia (1.96-0.35 mg/L), nitrite (0.03-0.16 mg/L), nitrate (1.34-0.47 mg/L), DIP (0.42-0.03 mg/L) and silicate (3.83-0.00 mg/L) were significantly decreased throughout experiment except nitrite which was increased (P < 0.05), but nitrogenous components stayed below toxic levels (ammonia 0.0-5.5 mg/L, nitrite 0.0-460.0 mg/L) which indicated that closed recirculation system with microalgae based bio-filter could supply sufficiently environment condition to holding and conditioning of oyster.

Epizootiology of Perkinsus sp. Found in the Manila Clam, Ruditapes philippinarum in Komsoe Bay, Korea (곰소만에 있어 바지락포자충, Perkinsus sp.의 출현에 관하여)

  • Mass mortality of the Manila clam, Ruditapes philippinarum has been reported all along the west and south coast of Korea for the past several years. As a pathogenic agent, Perkinsus sp., an endoparasitic protozoan has been identified in this study and believed to be responsible for the mass mortalities. Prevalence and infection intensity of Perkinsus sp. was investigated from a Manila clam population inhabiting at Komsoe Bay in the west coast where mass mortality of the clam has been reported. A total of 142 Manila clam, 50 oyster, Crassostrea gigas, 10 ark shell, Scapharca broughtonii, and 5 predatory gastropada, Rapana venosa were examined for the presence and the quantity of Perkinsus sp. Ray's fluid thioglycollate medium method (FTM method) with modified Mackin's infection intensity scale and Choi's quantitative method were used in detecting and quantifying the parasite. All individuals of R. philippinarum examined in this study were infected with Perkinsus sp., indicating $100\%$ prevalence while none of the oysters and the gastropods exhibited the parasite. Six to ten individual hypnospores of Perkinsus sp. were counted from the ark shells. The number of hypnospores in the clam tissues varied from 16,667 to 4,091,667, with a mean number of 1,077,628. Average infection intensity according to Mackin's was 2.87, indicating a moderate infection. A negative correlation was observed between the number of Perkinsus sp. in the tissue and the condition index, a ratio tissue wet weight to shell cavity volume. The clam size and the infection intensity in terms of total number of parasites were positively correlated; the bigger clam, the heavier infection. Such high number of Perkinsus sp. counted in the clams could be enough to cause physiological disturbance of clams, such as retarded growth and reproduction. It is also believed that such a high infection leads mortality of the clam via continuous draining of the energy by metabolic activities and reproduction of the parasites. Correlation between the condition index and the infection intensity observed in this study supports this hypothesis.
