Biological Control of Lycariella magi(Diptera: Sciaridae), a Pest of Oyster Mushroom, Pleurotus ostreatus Using Entomopathogenic Nematodes (곤충병원성 선충을 이용한 느타리버섯해충, 긴수염버섯파리 (Lycoriella mali)의 생물적방제)

  • The potential of two entomopathogenic nematodes, Sreinernema carpocapsae Pocheon strain and Heterorhabditis bacteriophora Hamyang strain as biological control agents was evaluated against mushroom ny, Lycoriella mali in laboratory and field. Mortality of L. mali was significantly different according to nematode species, concentration, temperature, and developmental stage of fly S. carpocapsae was more effective than H. bacteriophora. Mortality of L. mali was higher at $25^{\circ}C$ than at $20^{\circ}C$. In addition, the 3rd instal and the 4th instar of L. mali were more susceptible than the 2nd instar. The lowest $LC^{50}$ value was represented by S. carpocapsae, 20.0 infective juveniles (Ijs) in the 3rd instar, 27.5 Ijs in the 4th instar at $25^{\circ}C$. S. carpocapsae infected all the developmental stages of L. mali except egg stage and the 1st instar of larva. The highest mortality was shown in adult female representing 74.0% at$20^{\circ}C$ and 80.0% at $25^{\circ}C$.L. mali female adult was influenced by S. carpocapsae in oviposition. The number of eggs by L. mali female infected by nematodes was much lower than uninfected females. S. carpocapsae was dispersed by infected L. mali adult with higher numbers by females than males. When S. carpocapsae was applied at the rate of $2.25{\times}10^{5}\;and\;4.5{\times}10^{5}\;Ijs/1.5\;\textrm{m}^2$ in the mushroom house, mortalities were 42.2% and 81.6%, respectively. The infective juveniles of nematodes survived for 14 days in the mushroom medium. However, nematodes did not affect mushroom growth.

  • The sexual incompatibility system was investigated in the new strain Pleurotus ostreatus 'Miso'. Matings among monosporous progeny collected from fruiting bodies revealed that the new oyster mushroom is heterothallic and has a tetrapolar mating system. Single basidiospore isolates that did not have clamp connections were determined to be monokaryotic. Compatible parings of monokaryons were distinguishable macroscopically by the rapid growth and gross morphology to some extent but they were distinguished obviously by the presence of clamp connections from incompatible pairings under microscopic examination. Rarely dikaryotization was unidirectional with clamp connections only at the margin of one side of mating. In tetrapolarity of matings, four kinds of patterns, A1B1, A1B2, A2B1, and A2B2, were decided depending on compatibility tests in a parental strain with a mating factor of A1A2B1B2. The mating tests with the tester strains were performed for the detection of mating types on the rest of selected monokaryons.

  • Pleurotus eryngii(King oyster) is one of the most promising mushrooms produced on the domestic farms. The quality as well as quantity of Eryngii is sensitively affected by micro climate factors such as temperature, relative humidity, $CO_2$ concentration, and light intensity. To safely produce high-quality Eryngii all the year round, it is required that the environmental factors be carefully controlled by well designed structures equipped with various facilities and control systems. At the commercial mushroom cultivation house(A,B), this study was carried out to find out reasonable range of each environmental factor and yield together with economic and safe structures influencing on the optimal productivity of Eryngii. this experiment was conducted for about two-month from Nov. 11, 2002 to Dec. 30, 2002 in Eryngii. cultivation house-A, B. Ambient temperature during the experiment period was not predominantly different from that of a normal year. The capacity of the hot water boiler and the piping systems were not enough. Maximum air temperature difference between the upper and lower growth stage during a heating time zone was about 2~3$^{\circ}C$. The max. and min. relative humidity were ranged approximately 60~100%, and average relative humidity was ranged approximately 80~100%. And $CO_2$concentration increased until maximum 1,600~1,800 ppm with the passing growing period. The illuminance in cultivation house was widely distributed from 20lx to 160 lx in accordance with position, and it was maintained lower than the recommended illuminance range 100~200 lx. The average yield per bottle was about 67~85g. But the optimal productivity will be evaluated by considering the quality and quantity of mushroom production, energy requirements, facility construction and management cost, etc.

Study on the characteristics of fruit body growth according to incubation temperatures and period for oyster mushroom (느타리버섯 병재배 배양온도 및 배양기간에 따른 생육반응 연구)

  • This study was carried out to determine the proper pin-heading induction time(spawn running time) when different incubation temperature were applied to Pleurotus ostreatus(Chunchu 2ho, Suhan 1ho, Heukpyung). Incubation period was 17 days at 23 and 21 days at 17 for Chunchu 2ho, 17 days at 23 and 26 for Heukpyung, and 22~23 days at 17~23 for Suhan 1ho. Incubation period for Suhan 1ho was not significantly affected by incubation temperature. The time required for initial pin-heading was 4~5 days at 17, 20, 23 and 26 for Chunchu 2ho as well as Heukpyung, and was 3 days at 17, 20, 23 and 5~6 days at 26 for Suhan 1ho. As the incubation period became longer, the available fruit-bodies at Chunchu 2ho were made more but they were short. The yield of Chunchu 2ho and Heukpyung increased when incubated for 22~27 days at 20~23 and that of Suhan 1ho also increased when incubated for 22~23 days at 17~23.

Thermophile mushroom cultivation in Cambodia: Spawn production and development of a new substrate, acacia tree sawdust

  • To minimize cultivation costs, prevent insect-pest infestation, and improve the production efficiency of thermophilic mushrooms, plant substrates obtained from local areas in Cambodia were used for production of both spawn and mushrooms. In this experiment, different sawdusts different organic wastes and grain ingredients and analyzed for improvement of spawn-production efficiency. Four thermophilic mushroom species, Pleurotus sajor-caju (oyster mushroom, Sambok), Ganoderma lucidum (deer horn shaped), Auricularia auricula (ear mushroom), and Lentinula edodes (shiitake), were used to identify efficient new substrates for spawn and mushroom production. Although the mycelia in the rubber tree sawdust medium showed a slightly slower growth rate (10.9 cm/15 days) than mycelia grown in grains (11.2 cm/15 days in rice seeds), rubber tree sawdust appeared to be an adequate replacement for grain spawn substrates. Th findings indicate that rubber tree sawdust, sugarcane bagasse, and acacia tree sawdust supplemented with rice bran and calcium carbonate could be new alternative the substrates for. Although sugarcane bagasse and rubber tree sawdust showed similarly high biological efficiencies (BE) of 60% and 60.8%, respectively, acacia tree sawdust exhibited relatively a low biological efficiency of 22.4%. However, it is expected that acacia sawdust has potential for the mushroom cultivation when supplemented with currently used sawdust substrates in Cambodia, because of its relatively low price. The price of the sawdust (20 kg sawdust= 6500 Riel or 1.6 USD) currently used was 6.5 times higher than the price of acacia sawdust (201000 Riel or 0.25 USD). Therefore, utilization for acacia sawdust for mushroom cultivation could become feasible as it would reduce by producing costs of mushrooms in rural areas of Cambodia.

Effects of Fly Ash,Gypsum,and Shell on the Chemical Properties of Soil and Growth of Chinese Cabbage in Plastic Film Housed Paddy (시설재배논에 석탄회,석고,패각시용이 토양화학성과 배추의 생육에 미치는 영향)

  • In order to evaluate the utility of bituminous coal fly ash, gypsum, oyster shell as soil amendments, aadic clayloam paddy soils with low calaum content were amended in the upper 15㎝ with amendments, and then Chinese cabbage was cultivated under plastic film house. Amendments treated were, in metric tons per hectare, i) none(Check) ; ii) 80 fly ash(FA) ; iii) 4 shell(SH) ; iv) 56 fly ash+24 gypsum (FG) ; v) 40 fly ash+24 gypsum+0.8 shell(FGS). On the whole, soil chemical properties were improved by amendments treatments. Amongst treatments, FA prominently neutralized soil pH and increased contents of Av. $P_2O_5$, Ex. K, and Av. B in soils. Besides, it showed the highest ratio in bacteria/fungi and (bacteria+actinomycetes)/fungi. FGS also affected the neutralization of soil pH and the increment of Ex. Mg. Amendments plants appeared alkaline damages only at early growing stage, but showed positive responses in fresh weight yields : 23% for FGS : 21% for FG : 19 18% for SH. At harvesting, leaves both of FA and FGS plants had higher values in contents of N, P, K, Ca, Mg, Fe, Mn, Zn, B, reduang-sugar, and vitamin-C than of others. In especial, Check plants appeared the heart rot symptoms owing to calaum defiaency differently from amendments plants. Taken together, FGS was an effective combination enable to maximize the utility of fly ash, gypsum, shell as soil amendments, espeaally in cabbage yield and quality.

Annual Effective Dose of 210Po from Sea Food Origin (Oysters and Mussels) in Korea

  • Background: Ingestion of $^{210}Po$ laden seafood accounts for a substantial amount of the effective dose of $^{210}Po$. Among seafood items, mollusks, especially domestically produced oysters and mussels, are highly enriched in $^{210}Po$ and are consumed in large quantities in Korea. Materials and Methods: Oysters and mussels around the Korean coasts were collected from major farm areas in November 2013. Samples were spiked with an aliquot of $^{209}Po$ as a yield tracer, and they were digested with $6mol{\cdot}L^{-1}$ $HNO_3$ and $H_2O_2$. The $^{210}Po$ and $^{209}Po$ were spontaneously deposited onto a silver disc in an acidic solution of $0.5mol{\cdot}L^{-1}$ HCl and measured using an alpha spectrometer. The activity concentrations of $^{210}Pb$ and $^{210}Po$ were decay corrected to the sampling date, accounting for the possible in-growth and decay of $^{210}Po$. Results and Discussion: $^{210}Po$ activity concentrations in oysters were in a range from 41.3 to $206Bq{\cdot}(kg-ww)^{-1}$ and mussels in a range from 42.9 to $46.7Bq{\cdot}(kg-ww)^{-1}$. The $^{210}Po$ activity concentration of oysters in the turbid Western coast was higher than the Southern coast. The $^{210}Po$ activity concentration of the oysters was positively correlated ($R^2=0.89$) with those of the suspended particulate matter in the surface water. The calculated annual effective dose of $^{210}Po$ from oysters and mussels consumed by the Korean population was 21-104 and $5.01-5.46{\mu}Sv{\cdot}y^{-1}$. The combined effective dose due to the consumption of oysters and mussels appears to account for about $35{\pm}19%$ of that arising from seafood consumption in the Korean population. Conclusion: The annual effective dose of $^{210}Po$ for oysters in the Korean population was found to be higher than other countries. The total annual effective dose of $^{210}Po$ due to consumption of oysters and mussels consumed in Korea was found to be $76{\pm}42{\mu}Sv{\cdot}y^{-1}$, accounting for $28{\pm}16%$ of the total effective dose of $^{210}Po$ from food in Korea.

Some Properties and Microbial Community Changes of Gul (Oyster) Jeotgal during Fermentation

  • Gul jeotgals (GJs) were prepared using solar salt aged for 3 years. One sample was fermented using starters, such as Bacillus subtilis JS2 and Tetragenococcus halophilus BS2-36 (each $10^6CFU/g$), and another sample was fermented without starters for 49 days at $10^{\circ}C$. Initial counts of bacilli and lactic acid bacteria (LAB) in non-starter GJ were found to be $3.20{\times}10^2$ and $7.67{\times}10^1CFU/g$ on day 0, and increased to $1.37{\times}10^3$ and $1.64{\times}10^6CFU/g$ on day 49. Those of starter GJ were found to be $2.10{\times}10^5$ and $3.30{\times}10^7CFU/g$ on day 49, indicating the growth of starters. The pH values of GJ were $5.93{\pm}0.01$ (non-starter) and $5.92{\pm}0.01$ (starter) on day 0 and decreased to $5.78{\pm}0.01$ (non-starter) and $5.75{\pm}0.01$ (starter) on day 49. Amino-type nitrogen (ANN) production increased continuously during fermentation, and $407.19{\pm}15.85$ (non-starter) and $398.04{\pm}13.73$ (starter) mg% on day 49. Clone libraries of 16S rRNA genes were constructed from total DNA extracted from non-starter GJ on days 7, 21, and 42. Nucleotide sequences of Escherichia coli transformants harboring recombinant pGEM-T easy plasmid containing 16S rRNA gene inserts from different bacterial species were analyzed using BLAST. Uncultured bacterium was the most dominant group and Gram - bacteria such as Acidovorax sp., Afipia sp., and Variovorax sp. were the second dominant group. Bacillus amyloliquefaciens (day 7), Bacillus velezensis (day 21 and 42), and Bacillus subtilis (day 42) were observed, but no lactic acid bacteria were detected. Acidovorax and Variovorax species might play some role in GJ fermentation. Further studies on these bacteria are necessary.

Comparison in the productivity of Pleurotus ostreatus sawdust spawn based on the medium composition (느타리 톱밥종균 배지조성에 따른 생산성 비교)

  • This study was performed to find a medium material that can replace Douglas fir sawdust and rice bran in spawn media for growing spawn directly in the oyster mushroom farm. The pH range, total nitrogen source, and total carbon source of the mixed spawn medium were 5.3~5.9, 0.65~1.11%, and 47.0~49.1%, respectively. The C/N ratio was high in the mixed medium of poplar sawdust, with a low total nitrogen content. The protein content was high in the medium containing fermented Douglas fir sawdust. The mycelium growth rate was higher in the medium containing wheat bran than that in the medium containing rice bran. The highest yield per bottle was observed with poplar sawdust, oak sawdust, and rice bran mixed at a ratio of 40:40:20 (v/v/v); however, there was no significant difference observed in terms of productivity with the other treatments. Thus, when growing sawdust spawns in farms, it is efficient to use poplar sawdust, oak sawdust, and wheat bran, which are also easily available, instead of Douglas fir sawdust and rice bran.

Effect of Chitosan-Ascorbate and Morea(roasted of oyster shell at $1300^{\circ}C$) on Growth of Contaminating Bacteria in Dombaeki(traditional shark dish) during Storage (돔배기의 저장 중 오염미생물의 생육에 미치는 키토산-아스코베이트 및 모려의 처리효과)

  • The effects of 0.01%(w/v) chitosan-ascorbate(CA) and 10 ppm morea on the number of total microbes, Escherichia coli levels, and growth of food poisoning bacteria in dombaeki during storage at $10^{\circ}C$ over 6 days were investigated. Total microbes in meat, cartilage, and skin of untreated samples increased by 4.24, 3.81, and 2.20 logs compared to the zero timepoint, respectively, but, in CA-treated samples, counts fell by 2.66, 2.37, and 1.24 logs. Total microbial levels in morea-treated meat, cartilage, and skin showed similar tendencies but the effects were slightly less than seen in CA-treated samples. E. coli numbers in CA-treated meat, cartilage, and skin stored for 6 days decreased by 1.69, 1.25, and 1.52 logs respectively, compared with control samples. Morea-treated samples showed similar falls, but the effects were again slightly less than seen after CA-treatment. Both Salmonella and Vibrio parahaemolyticus were detected in untreated meat stored for 3 or 6 days. Food poisoning bacteria were found in both untreated and morea-treated samples stored over 6 days. However, no such bacteria were detected in CA-treated samples. Also, CA-treated meat, cartilage, and skin showed low degrees of degeneration. Thus, CA treatment enhanced shelf-life and dombaeki quality by inhibiting microorganism growth and tissue breakdown during storage.
