Oxidative stress on anaerobes

  • A strict anaerobe, Prevotella melaninogenica is highly sensitive to oxidative stress. Oxidative stress such as exposure to oxygen or addition of hydrogen peroxide, increased 8-hydroxydeoxyguanosine (80HdG), a typical of oxidative DNA damage, and decreased the bacterial cell survival rate. We could detect the generation of reactive oxygen species in P. melaninogenica after exposure to oxygen. UVA irradiation also increased 80HdG in the bacterium. On the other hand, such oxidative stress did not increase 80HdG in a facultative anaerobe. These findings suggest that P. melaninogenica is a suitable material to study the biological effects of oxidative stress, to evaluate antioxidants, and to study the effects of oxygen or reactive oxygen species on molecular evolution.

Control of Singlet Oxygen-induced Oxidative Damage in Escherichia coli

  • Singlet oxygen ($^1O_2$) is highly reactive form of molecular oxygen that may harm living systems by oxidizing critical cellular macromolecules. The oxyR gene product regulates the expression of the enzymes and proteins that are needed for cellular protection against oxidative stress. In this study, the role of oxyR in cellular defense against a singlet oxygen was investigated using Escherichia coli oxyR mutant strains. Upon exposure to methylene blue and visible light, which generates singlet oxygen, the oxyR overexpression mutant was much more resistant to singlet oxygen-mediated cellular damage when compared to the oxyR deletion mutant in regard to growth kinetics, viability and protein oxidation. Induction and inactivation of major antioxidant enzymes, such as superoxide desmutase and catalase, were observed after their exposure to a singlet oxygen generating system in both oxyR strains. However, the oxyR overexpression mutant maintained significantly higher activities of anticxidant enzymes than did the oxyR deletion mutant. These results suggest that the oxyR regulon plays an important protective role in singlet oxygen-mediated cellular damage, presumably through the protection of antioxidant enzymes.

  • By the oxygen plasma exposure to the organic light-emitting diodes, the turn-on voltage decreased from 10.5 to 7 V and luminance increased from 470 to $852\;cd/m^2$. Synchrotron radiation photoelectron spectroscopy results showed that during oxygen plasma exposure, oxygen ions were diffused into organic layer and induced p-type doping effect.

산소 플라즈마 처리된 d-PMMA 박막의 표면특성 분석 (Surface Characterization of the d-PMMA Thin Films Treated by Oxygen Plasma)

  • d-PMMA(deuterated poly(methyl methacrylate)) 박막 표면의 친수성을 향상시키기 위해 산소 플라즈마에 노출시켰다. 이 때 모든 조건은 동일하며, 플라즈마에 대한 노출 시간만을 0초에서 180초까지 변화를 주어 노출 시간에 대한 영향을 접촉각과 X-ray 반사율 장치, 중성자 반사율 장치, XPS(X-ray photoelectron spectroscopy)를 이용해 조사하였다. 노출 시간이 증가할수록 물 접촉각은 작아지며, 산소의 조성은 커짐을 확인함으로써 산소의 조성이 친수성 향상에 큰 영향을 미치는 것을 확인할 수 있었다. 또한, X-ray 반사율 장치를 이용해 얻은 에칭률을 통해서 d-PMMA 박막에 대한 산소 플라즈마의 노출 시간에 따른 물리적 특성을 연구하였으며, X-ray 반사율과 중성자 반사율, 그리고 XPS 측정 결과로부터 산소와 탄소의 조성뿐만 아니라 수소의 조성까지도 얻음으로써 플라즈마 처리된 박막의 화학적 성질을 보다 자세히 연구할 수 있었다.

저산소에 노출된 넙치(Paralichthys olivaceus)의 호흡대사와 혈액의 화학적 변화 (Changes in Respiratory Metabolism and Blood Chemistry of Olive Flounder Paralichthys olivaceus Exposed to Hypoxia)

  • This experiment investigated changes in metabolic rate (MO2), critical oxygen saturation (Scrit), and blood chemistry of olive flounder Paralichthys olivaceus exposed to progressive hypoxia and returned to normoxic water at 20°C. The normoxic standard metabolic rate (SMR) and routine metabolic rate (RMR) were 69.5-83.9 and 70.2-156.4 mg O2 kg-1h-1, respectively based on fish weight. Scrit was 31.0% dissolved oxygen (DO) at 20°C. After returning the fish to 70% DO following exposure to hypoxia (20% DO), MO2 increased two-fold compared to the normoxic SMR and then decreased into the range of the RMR with time. Blood PO2 and plasma lactate decreased significantly after exposure to hypoxia (20% DO) and then increased as ambient oxygen saturation decreased. Cortisol levels increased as ambient oxygen saturation decreased, but the levels decreased rapidly in the range of the normoxic control when the fish were returned to ambient water with 70% DO. Plasma glucose levels increased when the fish were returned to normoxic water after exposure to a progressively more hypoxic condition.

Ingestion of Korean Red Ginseng after Noise Exposure Can Potentiate Rapid Recovery of Hearing in Mice

  • One mechanism of inner ear damage by noise-induced hearing loss (NIHL) is the production of reactive oxygen species (ROS). Because Korean red ginseng (KRG) has an anti-ROS effect in various tissues, KRG may have a role in preventing NIHL. A window period exists in which ROS formations continue after noise exposure, and further damage can be prevented by antioxidants. In this study, we aimed to investigate the effects of KRG after exposure to noise. KRG (200 mg/kg) was fed to mice for 3 days after noise exposure. The change in hearing level was analyzed by measuring the auditory brainstem response. To induce a temporary threshold shift (TTS) of hearing, mice were exposed to 110 dB white noise for 3 hours. Fast recovery of hearing was observed in mice fed KRG 1 hour and 1 day after noise exposure for 3 days. The expression of 8-oxoguanine was not observed in the inner ears of mice fed KRG 1 hour after noise exposure, but was evident in the stria vascularis of mice in the control group (noise exposure only). From this study, we conclude that KRG acted as an effective inhibitor of NIHL in TTS cases.

  • 텅스텐 표면의 (110)면에 흡착된 산소량을 18O(P,$\alpha$)15N 핵반응으로 직접 측정하여 미세분석 하였다. Van de Graaff 가속기로 양성자를 가속시켜 시료 표면 가까이에 있는 원 자와 충돌시켜 생성되는 $\alpha$ 입자를 측정함으로써 표면에 있는 미소량의 원소도 측정이 가능 했다. $\alpha$ 입자의 수율은 oxygen exposure에 비례하였는데 coverage ($ heta$)는 5 Langmuir에서 0.5가 되었고, 15 Langmuir에서 포화되었다.

호기중 에탄(ethane)측정을 통한 산소중독시 지질과산화평가에 관한 실험적 연구 (Lipid Peroxidation in Vivo Monitored as Ethane Exhalation in Hyperoxia)

  • In vivo ethane production in rats was used as an index of oxygen toxicity. The rats were allocated to four exposure conditions; hyperbaric oxygenation (HBO=5 ATA, 100% $O_2$), normobaric oxygenation (NBO=1 ATA,100% $O_2$), hyperbaric aeration (HBA=5 ATA, 21% $O_2$) and normobaric aeration (NBA=1 ATA, 21% $O_2$). After 120 minutes of exposure, the rats exposed to high concentration and/or high pressure oxygen exhaled significantly larger amounts of ethane than those exposed to NBA, and the differences in ethane production between any two groups were statistically significant (p<.01). This finding supports the hypothesis that hyperoxia increases oxygen free-radicals and the radicals produce ethane as a result of lipid peroxidation. It is notable that the ethane exhalation level of the HBA group was significantly higher than that of the NBO group. This difference could not be accounted for by the alveolar oxygen partial presure difference between the two groups.

산, 담즙산, 산소 노출에 대한 비피도박테리아의 생존에 관한 연구 (Viability of Bifidobacterial Strains against Acid, Bile Acid, and Oxygen Exposure)

  • 한국인 유아에서 분리한 13개 균주를 포함한 19개 bifidobacteria 균주를 대상으로 산, 담즙산 및 산소 노출시의 생균수의 변화를 측정하였다. 내산성은 pH 4.0, 3.0, 2.0에서 2시간까지 측정하였는데, 실험 균주마다 매우 상이한 결과를 나타내었으며, 실험 균주 중에서 B. bifidum B3, B. longum D6, B. adolescentis Fl이 우수한 생존력을 나타내었다. 실험 균주를 0.2% thioglycolic acid와 0.2% oxgall이 함유된 MRS 배지에서 생장을 측정한 결과, B. bififdum B3과 B. longum D6이 담즙산에 우수한 저항성을 나타냈으며, 균종간의 경향은 관찰되지 않았다. 호기 상태에서 48시간 노출시킨 후에 생균수의 변화를 측정한 결과, 실험 균주마다 상이한 결과를 나타내었으며 상대적으로 B. bifidum과 B. longum균주들의 생존력이 우수하였다. 산, 담즙, 산소 노출에 대한 생존력은 균종간의 경향을 나타내지 않고 균주마다 상이한 결과를 나타내었는데, 실험 균주 중에는 B. bifidum B3과 B. longum D6 균주가 모두 우수한 생존율을 나타내었다. 이들 균주는 현재 시판되는 상업용 종균과 비교해서도 동등한 성적을 나타내어 상업용 종균으로서의 사용 가능성을 시사하였다.

Deterioration in the fertilization capability of boar spermatozoa upon exposure to mancozeb

  • Although pesticides are recognized as necessary substances to improve agricultural production, exposure to pesticides is known to have a direct or indirect adverse effect on the reproductive function of mammals. The present study examines the effects of mancozeb, a well-known fungicide, on the fertility capacity of spermatozoa. Boar spermatozoa exposed to varying concentrations of mancozeb (0.01 - 0.5 µM) were evaluated for motility, motion kinetic parameters, viability, acrosome integrity and the generation of intracellular reactive oxygen species (ROS) after 30 min or 2 hrs of incubation. A significant reduction in the motility of spermatozoa was observed upon exposure to mancozeb. Similarly, there was a significant reduction of the motion kinematics of sperm treated with mancozeb as compared to untreated controls (p < 0.05). The sperm viability percentage and acrosome integrity also showed dose-dependent decreases upon exposure to mancozeb. High concentrations of mancozeb (0.2 - 0.5 µM) induced higher levels of intracellular ROS production, which resulted in the loss of the sperm membrane and decreased sperm motility due to oxidative stress. Taken together, the results here indicate that direct exposure to mancozeb affects the sperm fertility capacity. Hence, careful research that examines the interaction between reproduction and environmental toxins is crucial to prevent fertility disorders in animals.
