국산 산소 농축기의 개발 및 동물실험 (Development and Animal Tests of Prototype Oxygen Concentrator)

  • 장기간 산소 호흡치료가 필요한 만성호흡부전증 환자에서 가정내 산소 농축기의 사용은 이미 보편화 되어있고 적응 및 사용빈도도 증가추세에 있다. 본 연구팀이 최근 개발한 국산 산소농축기의 성능검사와 보완점을 찾기위해 이미 시판되어 사용중인 외국산 산소농축기(FOR LIFE\ulcorner, Air Sep Corp. USA)와 비교 분석하여 효용성 및 안정성을 검증하고자 하였다. 기계적 성능 시험으로는 제작한 시제품과 외굿산 산소농축기를 4시간 연속 작동후 발생되는 산소농도를 측정하였으며, 동물실험은 잡견 16마리를 각 군당 8마리씩 A군, B군으로 나누어 A군은 외국산 산소농축기로, B군은 자체 개발한 산소농축기로 분당 5 리터의 산소를 공급하여 기본활력증후, 동맥혈 가스분석 및 혈액 화학 검사 등을 검사하였다. 4시간 연속 작동후 발생하는 산소 농도는 외산 98$\pm$3%, 국산 91$\pm$1 %로 외산이 우수하였다. 동물실험에서 산소 공급후 1, 2, 3, 4시간에 산소마스크 주입구에서 측정한 산소 농도는 A군에서 각각 70.6$\pm$2.5%, 67.1$\pm$2.9%, 68.2$\pm$2.6%, 64.9$\pm$3.9%, B군에서는 65.1$\pm$4.8%, 65.2$\pm$3.6%, 68.7$\pm$4.3%, 66.0$\pm$5.0%로 두 군간에 유의한 차이를 보이지 않았으며 (반복측정 분산분석 p=0.70), 산소투여 전 및 투여 후 1, 2, 3, 4 시간의 동맥혈 산소분압은 A군에서 각각 87.2$\pm$2.5 mmHg, 347.4$\pm$29.3 mmHg, 353.4$\pm$21.2 mmHg, 343.0$\pm$28.8 mmHg, 321.6$\pm$24.4 mmHg, B군에서 각각 102.5$\pm$9.6 mmHg, 300.3$\pm$17.1 mmHg, 321.6$\pm$23.7 mmHg, 303.4$\pm$27.4 mmHg, 273.5$\pm$25.9 mmHg로 비록 통계적 유의성은 없었으나(p=0.24) 외산 산소 농축기를 사용한 군에서 동맥혈 산소분압이 다소 높은 경향을 보였다. 이상으로부터 본연구팀이 현재 개발중인 국산 산소 농축기의 시제품의 성능을 시험한 결과 외산 산소 농축기와 비교하여 비교적 만족스러운 성능을 보였으나 소음, 내구성 등의 개선할 부분이 지적되었다.

The Impact of Abrupt Climate Change on the Marine Ecosystem in the East Sea

  • Environmental changes caused by the abrupt climatic change are one of the important issues in the scientific community. In the East Sea, abrupt climatic shift, called Younger Dryas, is identified. The age of the Younger Dryas cold episode occurred at 11.2 ka. Overall, changes in circulation and bottom water conditions occurred during the Younger Dryas cold episode in the study area. Especially, climatic transition from meltwater spike to the Younger Dryas cold episode is characterized by significant shifts of oxygen isotope values, the coiling ratios of Neogloboquadrina pachyderma, and the planktonic foraminifers abundances. The impact of abrupt climate change on the ecosystem is very significant. In the East Sea, the calcium carbonate secreting organism(foraminifers) is replaced by silicon dioxide secreting organisms(diatom, radiolarian) after the abrupt and severe cold climatic event. Based on the Doctrine of Uniformitarianism, at least climate change for the next 100 years would be severely influence on the marine ecosystem.

Antioxidant capacity of silkworm pupa according to extraction condition, variety, pupation time, and sex

  • Silkworm pupa has been used as an edible insect with the high quality of protein and unsaturated fatty acids. In this study, antioxidant activities of pupa according to variety, pupation day, sex, and extraction solvent were analyzed. The 30% ethanol extract showed highest radical scavenging activity compared with the DW, hexane, and 70-100% ethanol extracts. In the DPPH and ABTS radical scavenging assay according to the type of pupa, the antioxidant effect was increased in female with the early stage of pupation day. In cell-based assay, reactive oxygen species (ROS) level was decreased in pupa groups by -30 to -50% followed by tert-butyl hydroperoxide (t-BHP) treatment. The ROS levels were significantly reduced in 7th day in each variety. In conclusion, the free radical and ROS scavenging effects were increased in female pupa with the early pupation day. The result could be used for development of bioactive materials using silkworm pupa.

Morphological Difference of Rice Seedling Grown under Different Dissolved Oxygen Conditions

  • The response of dissolved oxygen (DO) concentrations caused significant change in root number, root length, coleoptile length, shoot length and leaf age of seedlings. The genotypic difference in the effect of DO also highly significant (P<0.01) for all of the seedling traits. The number and length of root were extremely inhibited at the condition of $0.39\pm0.09$ DO concentration. While the coleoptile elongated markedly in the lowest DO concentrations, the shoot did not develop. The root growth was improved slightly at the $1.39\pm0.27mg L^{-1}$, however, there were no difference among genotypes at these two low DO concentrations. As the DO concentration become higher, the growth of root and shoot was improved remarkably. Root number, root length and shoot length was significantly different between $20\;and\;30^{\circ}C$ in DO rich and normal conditions, the development of those traits were apparently accelerated in high water temperature, however those traits of seedlings in DO deficiency were not different between the two temperatures except for shoot length. On the other hand the coleoptile length was not affected by the stagnant water temperature; it was stimulated by the low DO concentration. The competition of DO was greater as the seedling density was increased in the stagnant water, therefore the seedlings grown under high density have long and white coleoptiles, and the growth of roots and shoots was retarded severely.

전씨백출산(錢氏白朮散)이 생쥐의 체액성(體液性) 면역반응(免疫反應)과 세포성(細胞性) 면역반응(免疫反應)에 미치는 효과(效果) (Effect of Junsibaekchulsan on the humoral and cell-mediated immune responses in mouse)

  • Even though appropriate immune response is necessary for the survival of the individual, excessive or insufficient immune response might cause autoimmune or allergic disease respectively. So the immune response must be controlled to the degree that is beneficial for the well being of the individual. This study was undertaken to know the effects of Junsibaekchulsan(JB) on the immune system od the mouse. For the evalulation of the cell-mediated immunity(CMI), delayed-type hypersensitivity against dinitrofluorobenzene(DNFB) were measured, and humoral immunity, hemagglutinin and hemolysin titers against SRBCs(sheep red blood cells) were measured, and rosette formation of spleen cells with SRBCs were measured. For the evaluation of innate immunity, phagocytic activity of macrophages, natural killer cell activity, and reactive nitrogen and oxygen intermediates were measured. The results are as follows: 1. The administration of JB depressed the antibody formation (hemagglutinin and hemolysin) against SRBCs. 2. The administration of JB did not affect the delayed-type hypersensitivity against DNFB. 3. The administration of JB did not affect the cytotoxic activity of natural killer cells. 4. The administration of JB increased the phagocytic activity of macrophages. 5. The administration of JB increased the rosette formating cells of the spleen cells. 6. The exposure of JB induced the secretion of reactive nitrogen intermediates but administration of JB deperssed the production of reactive oxygen intermediates. Administration of JB selectively depressed the humoral immune response without affecting CMI and innate immunity. These results of JB on the immune system might be useful for the treatment of such.

Distinct Bacterial and Fungal Communities Colonizing Waste Plastic Films Buried for More Than 20 Years in Four Landfill Sites in Korea

  • Plastic pollution has been recognized as a serious environmental problem, and microbial degradation of plastics is a potential, environmentally friendly solution to this. Here, we analyzed and compared microbial communities on waste plastic films (WPFs) buried for long periods at four landfill sites with those in nearby soils to identify microbes with the potential to degrade plastics. Fourier-transform infrared spectroscopy spectra of these WPFs showed that most were polyethylene and had signs of oxidation, such as carbon-carbon double bonds, carbon-oxygen single bonds, or hydrogen-oxygen single bonds, but the presence of carbonyl groups was rare. The species richness and diversity of the bacterial and fungal communities on the films were generally lower than those in nearby soils. Principal coordinate analysis of the bacterial and fungal communities showed that their overall structures were determined by their geographical locations; however, the microbial communities on the films were generally different from those in the soils. For the pulled data from the four landfill sites, the relative abundances of Bradyrhizobiaceae, Pseudarthrobacter, Myxococcales, Sphingomonas, and Spartobacteria were higher on films than in soils at the bacterial genus level. At the species level, operational taxonomic units classified as Bradyrhizobiaceae and Pseudarthrobacter in bacteria and Mortierella in fungi were enriched on the films. PICRUSt analysis showed that the predicted functions related to amino acid and carbohydrate metabolism and xenobiotic degradation were more abundant on films than in soils. These results suggest that specific microbial groups were enriched on the WPFs and may be involved in plastic degradation.

뇌기능 영상을 이용한 외부 산소 공급에 따른 공간 지각 능력 변화에 관한 연구 (A Study on Visuospatial Cognitive Performance Following Oxygen Administration using fMRI)

  • 본 연구에서는 일반 공기 중의 산소 농도 (21%) 환경에 비해 외부에서 고 농도 (30%)의 산소 공급이 공간 지각 능력에 어떠한 변화를 유발하는지 관찰하고자 한다. 8명의 남자 대학생 (평균 23.5세)을 뇌기능 연구의 실험 참여자로 선정하였고. 21%와 30% 산소 농도를 각각 8L/min의 양으로 일정하게 공급할 수 있는 장치를 개발하였다. 공간 지각 능력 측정을 위해 20 문항을 포함하는 두 개의 문제지를 제작하였고, 과제 수행 결과로부터 정답률을 산출하였다. 실험 패러다임은 30%의 산소 농도에서 과제를 수행하는 회기와 21%의 산소 농도에서 과제를 수행하는 회기로 각각 이루어져 있다. 각 회기는 네개의 black으로 구성되고, 각 block은 통제 과제 8문항. 공간지각 과제 5문항을 포함한다. 3T MRI기기를 사용하였고, single-shot EPI 방법으로 뇌기능 영상을 획득하였다. 산소 농도 21%에 비해 30% 상태에서 양측 후두엽 영역, 양측 상두정엽, 양측 하두정엽, 양측 쐐기앞소엽, 양측 중심후이랑 등의 두정엽 영역, 그리고 양측 중간전두이랑, 양측 하전두이랑, 양측 내측전두이랑 양측 상전두이랑, 양측 대상회 등의 전두엽 영역의 활성화가 증가하였고, 과제 수행 결과에서도 산소 농도 21%에 비해 30% 상패에서 정답률이 증가였다. 본 연구의 결과로부터 외부에서의 고농도의 산소 공급이 공간 지각 능력 증가에 긍정적인 영향을 미친다는 결론을 도출할 수 있다.

Effects of Restricted Oxygen, Nitric oxide, and Mercuric Chloride on the Seed Germination and Early Elongation Growth of Rice

  • Germination and early elongation of rice after germination were investigated in anoxic air treatment, nitric oxide gas treatment, and six concentrations of mercuric chloride solutions to determine the effects of limited oxygen environment, nitric oxide, and inhibited water flux through cell membrane in $17^{\circ}C$. Anoxic air treatment affected germination of tested six varieties very little. However root elongation rates were severely inhibited while shoot growth was affected less. Reductions in shoot and root elongations demonstrated genotypic variations. Nitric oxide delayed the germination of rice even though it didn't affect the final percent germination. Elongations of root and shoot were inhibited in nitric oxide treatment. The inhibitor effect of nitric oxide on the shoot elongation of rice was less severe, while nitric oxide completely inhibited the root emergence of rice. Concentrations of $HgCl_2$ greater than $300{\mu}M$ dramatically reduced the rate and percentage of germination when compared to distilled water treatment. The reduced percent germination showed the greatest variation among rice varieties in $500{\mu}M$ solution of mercuric chloride. Ansanbyeo, Jinheung, and Odaebyeo were affected less by $HgCl_2$, Nonganbyeo and Sangmibyeo were intermediate, and the germination of Andabyeo was greatly reduced by $HgCl_2$. Root elongation of germinated rice seedlings was more sensitive to oxygen deficits, nitric oxide, and $HgCl_2$ treatments than germination and shoot elongation. In conclusion, poor seedling establishment of rice sown in flooded paddy soils, in which the oxygen supply to the seeds is restricted, appears to the result of limited root elongation rate.

Methemoglobinemia caused by a low dose of prilocaine during general anesthesia

  • Methemoglobinemia is a blood disorder in which an abnormal amount of methemoglobin is produced, and prilocaine is one of the drugs that can cause this disorder. The maximum recommended dose of prilocaine is 8 mg/kg. We report a case of methemoglobinemia caused by the administration of 4.2 mg/kg of prilocaine without other methemoglobinemia-inducing drugs during general anesthesia. A 17-year-old girl with hyperthyroidism and anemia was scheduled to undergo maxillary sinus floor elevation and tooth extraction. The patient's peripheral oxygen saturation (SpO2) decreased from 100% at arrival to 95% after receiving prilocaine with felypressin following induction of general anesthesia. However, the fraction of inspired oxygen was 0.6. Blood gas analysis showed that the methemoglobin level was 3.8% (normal level, 1%-2%), fractional oxygen saturation was 93.9%, partial pressure of oxygen was 327 mmHg, and arterial oxygen saturation was 97.6%. After administration of 1 mg/kg of methylene blue, her SpO2 improved gradually to 99%, and the methemoglobin value decreased to 1.2%. When using prilocaine as a local anesthetic, it is important to be aware that methemoglobinemia may occur even at doses much lower than the maximum recommended dose.

김수육군전(金水六君煎)이 흡연(吸煙)한 백서(白鼠)의 폐손상(肺損傷)에 미치는 영향(影響) (Experimental Study of the Effects of Gyumsuyukgunjun on the Pulmonary Injury Caused by Exposure of Cigarette Smoke in Rats)

  • This experiment was performed in order to study the clinical effects of Gyumsuyukgunjun on the pulmonary injury caused by exposure of cigarette smoke in rats. Therefore, a writer reports on present experimental results having significant effects on the level of oxygen consumption, the lungs volume, the leukocyte, the hemoglobin level, and the $PaO_2$, $PaCO_2$ after administration of Gyumsuyukgunjun in order to study the clinical effects of Gyumsuyukgunjun on the pulmonary injury caused by exposure of cigarette smoke in rats. The results were obtained as follows: 1. In comparison with control group, The group of Gyumsuyukgunjun administration was revealed significant effects(P<0.005) of increase on the level of oxygen consumption. 2. In comparison with control group, The group of Gyumsuyukgunjun administration was revealed significant effects(P<0.005) of decrease on the lungs volume. 3. In comparison with control group, The group of Gyumsuyukgunjun administration was revealed significant effects(P<0.05) of decrease on the leukocyte counts. 4. In comparison with control group, The group of Gyumsuyukgunjun administration was revealed significant effects(P<0.05) of decrease on the hemoglobin level. 5. In comparison with control group, The group of Gyumsuyukgunjun administration was revealed significant effects(P<0.01) of increase on the $PaO_2$ level only of $PaO_2$, $PaCO_2$ level. It was concluded from the above results that Gyumsuyukgunjun has a significant effects on the pulmonary injury caused by cigarette smoke.
