Protective Effects of EGCG on UVB-Induced Damage in Living Skin Equivalents

  • In this study, we evaluate the effects of (-)-epigallocatechin-3-gallate (EGCG) on ultraviolet B(UVB)-irradiated living skin equivalents (LSEs). Histologically, UVB irradiation induced thinning of the LSE epidermis, whereas EGCG treatment led to thickening of the epidermis. Moreover, EGCG treatment protected LSEs against damage and breakdown caused by UVB exposure. Immunohistochemically, UVB-exposed LSEs expressed p53, Fas, and 8-hydroxy-deoxyguanosine (8-OHdG), all of which are associated with apoptosis. However, EGCG treatment reduced the levels of UVB-induced apoptotic markers in the LSEs. In order to determine the signaling pathways induced by UVB, Western blot analysis was performed for both c-Jun $NH_2$-terminal kinase (JNK) and p38 mitogen-activated protein kinase (MAPK), which are associated with UVB-induced oxidative stress. UVB activated JNK in the epidermis and dermis of the LSEs, and EGCG treatment reduced the UVB-induced phosphorylation of JNK. In addition, p38 MAPK was also found to have increased in the UVB-exposed LSEs. Also, EGCG reduced levels of the phosphorylation of UVB-induced p38 MAPK. In conclusion, pretreatment with EGCG protects against UVB irradiation via the suppression of JNK and p38 MAPK activation. Our results suggest that EGCG may be useful in the prevention of UVB-induced human skin damage, and LSEs may constitute a potential substitute for animal and human studies.

Isorhamnetin Protects Human Keratinocytes against Ultraviolet B-Induced Cell Damage

  • Isorhamnetin (3-methylquercetin) is a flavonoid derived from the fruits of certain medicinal plants. This study investigated the photoprotective properties of isorhamnetin against cell damage and apoptosis resulting from excessive ultraviolet (UV) B exposure in human HaCaT keratinocytes. Isorhamnetin eliminated UVB-induced intracellular reactive oxygen species (ROS) and attenuated the oxidative modification of DNA, lipids, and proteins in response to UVB radiation. Moreover, isorhamnetin repressed UVB-facilitated programmed cell death in the keratinocytes, as evidenced by a reduction in apoptotic body formation, and nuclear fragmentation. Additionally, isorhamnetin suppressed the ability of UVB light to trigger mitochondrial dysfunction. Taken together, these results indicate that isorhamnetin has the potential to protect human keratinocytes against UVB-induced cell damage and death.

Tat-CIAPIN1 protein prevents against cytokine-induced cytotoxicity in pancreatic RINm5F β-cells

  • Cytokines activate inflammatory signals and are major mediators in progressive β-cell damage, which leads to type 1 diabetes mellitus. We recently showed that the cell-permeable Tat-CIAPIN1 fusion protein inhibits neuronal cell death induced by oxidative stress. However, how the Tat-CIAPIN1 protein affects cytokine-induced β-cell damage has not been investigated yet. Thus, we assessed whether the Tat-CIAPIN1 protein can protect RINm5F β-cells against cytokine-induced cytotoxicity. In cytokine-exposed RINm5F β-cells, the transduced Tat-CIAPIN1 protein elevated cell survivals and reduced reactive oxygen species (ROS) and DNA fragmentation levels. The Tat-CIAPIN1 protein reduced mitogen-activated protein kinases (MAPKs) and NF-κB activation levels and elevated Bcl-2 protein, whereas Bax and cleaved Caspase-3 proteins were decreased by this fusion protein. Thus, the protection of RINm5F β-cells by the Tat-CIAPIN1 protein against cytokine-induced cytotoxicity can suggest that the Tat-CIAPIN1 protein might be used as a therapeutic inhibitor against RINm5F β-cell damage.

The Promoting Effect of Rumex japonicas Houttuyn ethanol extract on Hair Growth

  • Objectives: This study was conducted to evaluate the hair growth-promoting effects by Rumex japonicas Houttuyn ethanol extract (RJHEE) in C57BL/6N mice and HaCaT cells. Methods: The hair growth effect was examined by topical application of RJHEE on the shaved dorsal skin of C57BL/6 mice. Six-week old mice were depilated and separated in 4 groups; CON (vehicle treatment), MXD (2% Minoxidil), and RJHEE (2% and 4%). The treatments were applied daily for 17 days. The hair growth was determined photographically and the hair density, thickness and length were identified by Folliscope. In dorsal skin tissue, the expression of hair growth-related protein was analyzed by Western blotting. In HaCaT cells, the cell proliferation and the protection against H2O2-induced cell damage by RJHEE were analyzed. Results: Our results indicate that RJHEE promote the hair growth, hair density, thickness and length. RHE activate the Wnt/𝛽-catenin signaling and induced the expression of cell survival-related proteins, such as pERK/ERK and Bcl-2/Bax. In HaCaT, RJHEE accelerated the cell proliferation and protected the H2O2-induced cell damage. Conclusions: Our results strongly suggest that RJHEE promotes hair growth by regulating the activation of Wnt/𝛽-catenin signaling and cell survival signaling and protects oxidative stress-induced hair damage. Therefore, RJHEE has a hair growth activity and can be useful for the treatment of alopecia.

두충(杜沖)이 수컷 노화쥐의 우울증에 미치는 효과 (Effects of Eucomiae Cortex on the Depression of Male Aged Mice)

  • In order to investigate the effects of Eucomiae Cortex extracts on the depression caused by aging, histochemistry and immunohistochemistry were performed on the hippocampus of aged rats and the following results were obtained. Experimental animals were divided into three groups as follows: 8 week old ICR male mice, Aging-elicited group (AE group) and Eucomiae Cortex treatment group (EC group) 50 week old male ICR mice were used. The control group and AE group did not take any treatment and did not restrict diets and negatives. In the EC group, 0.51g/kg of Eucomiae Cortex extract was dissolved in distilled water once a day for 6 months. The Eucomiae Cortex extract reduced pyramidal neuronal damage in C3 hippocampus and dentate gyrus, increased DJ-1, SHH positive responses in aged mouse hippocampus, and 8-OHdG positivity was reduced, ${\beta}$-endorphin positivity was reduced in aged mouse substantia nigra. Therefore, based on the above results, Eucomiae Cortex extract reduces damage of pyramidal neurons in the hippocampus caused by aging, inhibits neuronal cell death, induces proliferation and differentiation of stem cells in the hippocampus, reduces DNA damage-induced oxidative stress, so improves the reduction of hippocampus volume. It is also thought to improves depression due to aging through ${\beta}$-endorphin which enhances mood through the inhibition of pain.

Ginsenoside compound K reduces the progression of Huntington's disease via the inhibition of oxidative stress and overactivation of the ATM/AMPK pathway

  • Background: Huntington's disease (HD) is a neurodegenerative disorder caused by the expansion of trinucleotide CAG repeat in the Huntingtin (Htt) gene. The major pathogenic pathways underlying HD involve the impairment of cellular energy homeostasis and DNA damage in the brain. The protein kinase ataxia-telangiectasia mutated (ATM) is an important regulator of the DNA damage response. ATM is involved in the phosphorylation of AMP-activated protein kinase (AMPK), suggesting that AMPK plays a critical role in response to DNA damage. Herein, we demonstrated that expression of polyQ-expanded mutant Htt (mHtt) enhanced the phosphorylation of ATM. Ginsenoside is the main and most effective component of Panax ginseng. However, the protective effect of a ginsenoside (compound K, CK) in HD remains unclear and warrants further investigation. Methods: This study used the R6/2 transgenic mouse model of HD and performed behavioral tests, survival rate, histological analyses, and immunoblot assays. Results: The systematic administration of CK into R6/2 mice suppressed the activation of ATM/AMPK and reduced neuronal toxicity and mHTT aggregation. Most importantly, CK increased neuronal density and lifespan and improved motor dysfunction in R6/2 mice. Conversely, CK enhanced the expression of Bcl2 protected striatal cells from the toxicity induced by the overactivation of mHtt and AMPK. Conclusions: Thus, the oral administration of CK reduced the disease progression and markedly enhanced lifespan in the transgenic mouse model (R6/2) of HD.

  • The recombinant bacteria strain DPD2540, containing a fabA::luxCDABE fusion, was used to detect the toxicity of various chemicals in this study. Membrane damaging agents such as phenol, ethanol, and cerulenin induced a rapid bioluminescent response from this strain. Other toxic agents, such as DNA-damaging or oxidative-damaging chemicals, showed a delayed bioluminescent response in which the maximum peak appeared over 150 min after induction. This strain was also tested for measurement of toxicity in field samples such as wastewater and river water effluents.

  • Living organisms have antioxidant enzymes, such as superoxide dismutase, catalase SE glutathione peroxidase, that protect themselves from the toxic effect of superoxide free radicals. Some report says that intracellular oxidation stress is involved in pathogenesis of chronic complications of diabetes mellitus. This study was performed to evaluate the effect of red ginseng on lipid peroxidation of red blood cell and antioxidant SOD activity of serum in NIDDM patients. As a result, there were trends for decrease of lipid peroxidases of RBC and Increase of SOD activity of serum in ginseng group but that were not statistically significant. Therefore, we suggest long term and large sized control study is necessary to confirm the protective effects of red ginseng on oxidative damage in NIDDM patients.

  • Fretting of fuel rod cladding material, Zircaloy-4 tube, in PWR nuclear power plants must be reduced and avoided. Nowadays the introduction of surface treatments or coatings is expected to be an ideal solution to fretting damage since fretting is closely related to wear. corrosion and fatigue. Therefore. in this study the fretting wear experiment was performed using TiAlN coated Zircaloy-4 tube as the fuel rod cladding and uncoated Zircaloy-4 as on of grids, especially concentrating on the sliding component. Fretting wear resistance of TiAlN coated Zircaloy-4 tubes was improved compared with that of TiN coated tubes and uncoated tubes and fretting wear mechanisms were brittle fracture and plastic flow at lower slip amplitude but severe oxidation and spallation of oxidative layer at higher ship amplitude.

Targeting Cellular Antioxidant Enzymes for Treating Atherosclerotic Vascular Disease

  • Atherosclerotic vascular dysfunction is a chronic inflammatory process that spreads from the fatty streak and foam cells through lesion progression. Therefore, its early diagnosis and prevention is unfeasible. Reactive oxygen species (ROS) play important roles in the pathogenesis of atherosclerotic vascular disease. Intracellular redox status is tightly regulated by oxidant and antioxidant systems. Imbalance in these systems causes oxidative or reductive stress which triggers cellular damage or aberrant signaling, and leads to dysregulation. Paradoxically, large clinical trials have shown that non-specific ROS scavenging by antioxidant vitamins is ineffective or sometimes harmful. ROS production can be locally regulated by cellular antioxidant enzymes, such as superoxide dismutases, catalase, glutathione peroxidases and peroxiredoxins. Therapeutic approach targeting these antioxidant enzymes might prove beneficial for prevention of ROS-related atherosclerotic vascular disease. Conversely, the development of specific antioxidant enzyme-mimetics could contribute to the clinical effectiveness.
