We have screened the cytoprotective effect against $H_2O_2$ and $\gamma-ray$ radiation induced oxidative stress from 32 Korean plants. Betula ermani var.saitoana (caulis, leaves), Rosa wichuraiana (caulis), Sorbus commixta (caulis), Weigela florida (leaves), Cirsium rhinoceros (whole plant), and Viburnum erosum (caulis) were found to scavenge 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical and intracellular reactive oxygen species (ROS). As a result, extracts of six plants reduced cell death of Chinese hamster lung fibroblast (V79-4) cells induced by $H_2O_2$ treatment. In addition, these extracts protected cell death of V79-4 cells damaged by $\gamma-ray$ radiation. In addition, these extracts scavenged ROS generated by radiation. Taken together, the results suggest that Betula ermani var. saitoana, Rosa wichuraiana, Sorbus commixta, Weigela florida, Cirsium rhinoceros, and Vibumum erosum protect V79-4 cells against oxidative damage by radiation through scavenging ROS.
Objectives: SimJeok-Hwan(CP, Cardiotonic Pills) was made to treat patients with coronary arteriosclerosis, angina pectoris and hyperlipidemia. This study was designed to investigate the effects of CP on Proliferation rates neuroglia cells and protective effect of CP against oxidative stress, and also investigate the effects on regional Cerebral Blood Flow(rCBF) in normal rats. Methods: In this experiment, effects of CP on proliferation rates of neuroglia cells were measured using modified MTT methods. Oxidative stress was induced by treatment with 200 mM of hydrogen peroxide for 2 hr. rCBF and MABP were measured using Laser doppler flowmeter. Results: Treatment with CP elevated proliferation rates in C6 cells. In addition, CP protected cell death of C6 cells induced by oxidative stress. In results, rCBF was elevated by treatment with CP in normal rats. But, Mean Arterial Blood Pressure(MABP) did not affected by CP. In addition, the elevation of rCBF was blacked by pre-treatment with 1 mg/kg of indomethacin effectively. On the other hand, pre-treatment with 0.01 mg/kg of methylene blue did not affect rCBF level in normal rats. Conclusions: In conclusion, these results suggest that CP can act as anti-oxidant to protect neuroglia cells and also suggest that CP can elevate rCBF, which are involved in cyclooxygenase pathway.
The purpose of this study was to investigate the effect of vitamin C and vitamin E supplementation on the iron contents and oxidative stress of the rats. Rats were fed 18g ascorbic acid and 300IU $\alpha$-tocopherol/kg diet, respectively. Rats were sacrificed at 1, 3, 5 and 7 month of age. The blood, liver and brain were selected for the quantitation of iron and malondialdehyde(MDA) contents, glutathione peroxidase(GSHPx), superoxided dismutase(SOD) and catalase(CAT) activity. Iron and MDA contents and GSHPx activities were increased with aging. Vitamin C and Vitamin E supplementation increased iron contents of the plasma. Vitamin C raised iron contents, but vitamin E decreased iron contents of the liver. In the brain vitamin C and vitamin E did not affect the iron level. MDA levels were decreased with vitamin C and vitamin E supplementation in the erythrocyte and liver, and vitamin C supplementation elevated MDA levels in the brain. GSHPx activity was increased with vitamin C and vitamin E supplementation. SOD activities of erythroucyte and brain were not affected with age, but in the liver, SOD activity was raised with age and vitamin C supplementation. Vitamin C and vitamin E supplementation promoted CAT activity of erythroucyte and liver, and CAT activity of brain was eleveated with vitamin addition but was decreaed with vitamin E addition. Vitamin C and vitamin E decreased iron contents of blood plasma, MDA contents of plasma and liver, and CAT activity of erythrocyte. Above results indicated that iron contents and biomarkers of oxidative stress were more affected by age than antioxidant action of vitamin C and vitamin E.

Objectives : ROS are involved in a wide spectrum of diseases including chronic inflammation and cancer. S.chinensis Baill, a perennial herb commonly called Chinese lizard's tail or Sam-baek-cho in Korea, is used for the treatment of edema and inflammatory diseases in the Oriental folk medicine. In this study, we investigated the antioxidant activities and anti-inflammatory effects of the two extracts, water(WE) and ethyl acetate(EAE) from S.chinensis Baill. Methods : Anti-oxidant activity was evaluated using Fe2+ chelating and hydroxyl radical scavenging assay. DNA cleavage assay, and western blot and immunostaining for phospho-p65 were performed to evaluate anti-oxidative effect. Anti-inflammatory effect was performed using NO generation assay and western blot in LPS-stimulated RAW264.7 cell. Results : In Fe2+ chelating activity and hydroxyl radical scavenging activity, WE showed more strong scavenging activity for hydroxyl radical than EAE. WE scavenged hydroxyl radical by 12% at 3.2 ${\mu}g/ml$, 21% at 16 ${\mu}g/ml$, 32% at 80 ${\mu}g/ml$, 66% at 400 ${\mu}g/ml$ and 82% at 2000 ${\mu}g/ml$, respectively. In addition, WE showed more strong chelating activity than EAE. WE chelated Fe2+ ion by 1.1% at 3.2 ${\mu}g/ml$, 8.2% at 16 ${\mu}g/ml$, 26.3% at 80 ${\mu}g/ml$, 72% at 400 ${\mu}g/ml$ and 89% at 2000 ${\mu}g/ml$, respectively. Also, WE inhibited oxidative damage via its anti-oxidant activity. In anti-inflammatory effect, EAE inhibited NO production and iNOS expression. In addition EAE suppressed the NF-${\kappa}B$ and MAPK signaling pathway in LPS-stimulated RAW 264.7 cells. Conclusions : Together, these data indicate that S. chinensis Baill, shows anti-oxidant activity and anti-inflammatory effect.
We investigated the anti-oxidative effect of the blueberry duke (Vaccinium corymbosum L., Ericaceae) ethanol extract in Caenorhabditis elegans model. The ethanol extract of blueberry duke showed relatively significant DPPH radical scavenging and superoxide quenching activities. To prove antioxidant activity of the extract, we checked the activities of superoxide dismutase (SOD), catalase, intracellular ROS, and oxidative stress tolerance in C. elegans. In addition, to verify if the increased stress tolerance of C. elegans by treating with the extract was due to regulation of stress-response genes, we checked SOD-3 expression using a transgenic strain. As a consequence, the blueberry duke ethanol extract increased SOD and catalase activities of C. elegans, and reduced intracellular ROS accumulation in a dose-dependent manner. Besides, blueberry duke ethanol extract-treated CF1553 worms showed higher SOD-3::GFP intensity.
Four naphthalene-degrading bacteria (Pseudomonas sp. strains O1, W1, As1, and G1) were isolated feom pollutant-contaminated sites. Examination of their substrate utilization and analyses of key naphthalene-catabolic regulatory genes revealed that the pathway and regulation of naphthalene-degradation in all four strains resemble those of NAH7 from P. putida G7. Superoxide anion production, superoxide dismutase activity, and catalase activity during their growth on naphthalene-amended medium increased significantly, compared with those with glucose-amended medium. Addition of ascorbate, an antioxidant, or ferrous iron ($Fe^{2+}$) increased the growth rates of all tested microorganisms on naphthalene. Northern blot and HPLC analyses showed that both nahA gene expression and naphthalene degradation increased under those conditions. Our data suggest that naphthalene degradation can impose severe oxidative stress, and defenses against oxidative stress would play an important role in the metabolism of naphthalene.
Endothelial cell dysfunction is considered to be a major cause of vascular complications in diabetes. In the present study, we investigated the protective effect of a baechu kimchi added Ecklonia cava extract (BKE) against high glucose induced oxidative damage in human umbilical vein endothelial cells (HUVECs). Treatment with a high concentration of glucose (30 mM) induced cytotoxicity, whereas treatment with BKE protected HUVECs from high glucose induced damage; by restoring cell viability. In addition, BKE reduced lipid peroxidation, intracellular reactive oxygen species and nitric oxide levels in a dose dependent manner. Treatment with high glucose concentrations also induced the overexpression of inducible nitric oxide synthase, cyclooxygenase-2 and NF-${\kappa}B$ proteins in HUVECs, but BKE treatment significantly reduced the overexpression of these proteins. These findings indicate that BKE may be a valuable treatment against high glucose-induced oxidative stress HUVECs.
Houttuynia cordata root on non-lipid oxidative damage. The antioxidative efects of methanolic (MeOH) extract of Houttuynia cordata rooton non-lipid, including liposome oxidation, oxidation of deoxyribose, protein oxidation, chelating, scavenging,and 2'-deoxyguanosine (2'dG) oxidation were investigated. Houttuynia cordata root exhibited highantioxidative effect in a liposome model system. The inhibitory effect of MeOH extract on deoxyribosedamage exhibited antioxidative effect and it afforded considerable protection against damage to deoxyribose.In addition, MeOH extract at over 300extracts exhibited metal binding ability for hydrogen peroxide. Furthermore, the oxidation of 2'dG to 8-hydroxy-2-deoxyguanosine was inhibited by MeOH extracts, and scavenging activity for hydroxyl radicalexhibited a remarkable effect. The present results on biological model systems showed that MeOH extractswas effective in the protection of non-lipids against various oxidative model systems.
Several lines of evidence indicate that the oxidative modification of protein and the subsequent accumulation of the modified proteins have been found in cells during aging, oxidative stress, and in various pathological states including premature diseases, muscular dystrophy, rheumatoid arthritis, and atherosclerosis. The important agents that give rise to the modification of a protein may be represented by reactive aldehydic intermediates, such as ketoaldehydes, 2-alkenals and 4-hydroxy-2-alkenals. These reactive aldehydes are considered important mediators of cell damage due to their ability to covalently modify biomolecules, which can disrupt important cellular functions and can cause mutations. Furthermore, the adduction of aldehydes to apolipoprotein B in low-density lipoproteins (LDL) has been strongly implicated in the mechanism by which LDL is converted to an atherogenic form that is taken up by macrophages, leading to the formation of foam cells. During the search for an endogenous inducer of cyclooxygenase-2 (COX-2), an inducible isoform responsible for high levels of prostaglandin production during inflammation and immune responses, 4-hydroxy-2-noennal (HNE), one of the most representative lipid peroxidation product, has been identified as the potential inducer of COX-2. In addition, the following study on the molecular mechanism of the COX-2 induction by HNE has unequivocally established that a serum component, which is eventually identified to be denatured LDL, is essential for COX-2 induction. Here I review current understanding of the mechanisms by which HNE in cooperation with the serum component activates gene expression of COX-2.

Anti-oxidative effect of Phellinus linteus (P. linteus) and red ginseng extracts on DNA damage induced by reactive oxygen species (ROS) were investigated in this study. P. linteus (PLE) and red ginseng extracts (RGE) inhibited the breaking of E. coli ColE1 plasmid DNA strands as well as nuclear DNA of rat hepatocytes damaged by oxidative stress. In addition, a reaction mixture of PLE and RGE showed synergistic inhibitory effect against DNA damage. These results suggest that PLE and RGE have a cellular defensive effect against DNA damage induced by ROS.
