To clarify the antioxidation effect of the various solvent fractionation of Tetragonia tetragoniodes which has been known to superior plants for the traditional prevention and treatment of stomach-related diseases, total polyphenol and flavonoid contents, vitamin E content, the elimination activity of a DPPH free radical and superoxide anion, inhibition activity of the superoxide generation, reducing power and metal chelating effects were estimated. The contents of the polyphenol compounds were highest in the DCM and EA fractionation, and the content of flavonoid was high in the order of HX and EA fractionation. The vitamin E content showed high in the order of the HX and DCM fractionation among solvent fractions. $IC_{50}$ for the elimination effect of a DPPH radical were estimated as 554.25 and $394.96{\mu}g/ml$ in the DCM and EA fractions, respectively. These values were higher than that of BHT $784.7{\mu}g/ml$ widely used in antioxidation effect. The inhibition activity of the superoxide generation using the T. tetragoniodes solvent fractions represented the effects similar to that of ${\alpha}$-tocopherol known to prevent the lipoprotein oxidation, but lower consequences than that of the phenol-resins, BHT and BHA, respectively. In the antioxidation activity derived by the reducing capability, the EA fractionation in a 1.5 mg/ml concentration showed higher than that of ${\alpha}$-tocopherol.
This study suggests that fermented rice bran extract contains natural antioxidants. The contents of bioactive materials (e.g., polyphenolic compounds and flavonoids), antioxidative properties (DPPH (α,α'- diphenyl-β-picrylhydrazyl) free radical scavenging activity, Fe reducing, Cu reducing power, peroxidation of linoleic acid and rat hepatocyte microsome) were tested by in vitro experimental models using fermented rice bran (FRB) extract. The concentrations of phenolic compound and flavonoid were 19.92 mg/g and 11.56 mg/g, respectively. In oxidation in vitro models using DPPH free radical scavenging activity, (free radical scavenging activity 69.8%) Fe reducing power and Cu reducing power (effect of dose-dependent manner), Fe2+/ascorbate induced linolenic acid peroxidation by ferric thiocyanate and thiobarbituric acid (TBA) methods (inhibition activity 81%), and autooxidation of rat hepatic microsomes membrane (lipid peroxidation inhibition activity 38%), antioxidative activities were stronger in FRB extract than FRS (Fermented Rice and Soybean, positive control) extract and, these effects were dose-dependent manner. From these results, FRB extract was shown to have the most potent antioxidative properties and contain the highest amounts of antioxidative compounds such as phenolic compounds and flavonoids. Overall, these results may provide the basic data to understand the antioxidative properties of fermented rice bran for development of functional foods.

Mechanism of calcium transport inhibition of cardiac sarcoplasmic reticulum (SR) by oxygen free radicals was examined. Effects of oxygen free radicals generated by xanthine/xanthine oxidase (X/XO) system on isolated porcine ventricle SR were studied with respect to its calcium binding, lipid peroxidation, SH-group content and alteration of membrane protein components. The results are as follows. 1) Calcium binding of isolated SR was markedly inhibited by X/XO. 2) During the incubation of sarcoplasmic reticulum with xanthine/xanthine oxidase, there were marked inclose in lipid peroxidation and reduction of SH-group content. 3) An antioxidant, p-phenylenediamine effectively prevented the lipid peroxidation but partially prevented the calcium binding inhibition of X/XO treated SR. 4) The reduction of SH-group content of SR treated with X/XO was partially prevented by p-phenylendiamine. 5) When modifying SH-group of SR by treatment with DTNB, the inhibition of calcium binding activity was partially prevented. 6) On gel-permeation chromatography of X/XO-treated sarcoplasmic reticulum, there was an increase of small molecular weight products, probably protein degradation products. 7) Semicarbazide, which prevents the cross-linking reaction of protein components, did not affect the calcium binding inhibition of X/XO-treated SR. From these results, it is suggested that the inhibition of calcium binding of SR by oxygen free radicals results from the consequence of multiple changes of SR components, which are lipid peroxidation, SH-group oxidation and degradation of protein components.
Acid mine drainage (AMD) is one of the most severe environmental problem that results from the oxidation of pyrite $(FeS_2)$ and various other metal sulfides. In this study, the influence of microorganism was tested on the process where AMD was released and the method to inhibit AMD generated by microorganisms at abandoned mine area. The activity and growth rate of Acidithiobacillus ferrooxidans and Acidithiobacillus thiooxidans, common microorganisms affecting AMD occurrence, were measured. Chlorine dioxide $(ClO_2)$, NaCl, or surfactant (ASOR-770) was used as an inhibitor for working on activity and growth of microorganism. Among the three inhibitors, 10ppm of chlorine dioxide was the most effective inhibitor for AMD control due to the reduced the activity and growth of microorganisms by 20%.
Objectives : The present study compared Aralia continentalis Root and Angelica pubescens Root used in Korea and China respectively concerning their anti-oxidant effect Methods : We tested the anti-oxidant effect through in vitro experiment and in vivo experiment that induced oxidative stress using ethanol. Results : 1. DPPH scavenging activity was stronger in Aralia continentalis Root than in Angelica pubescens Root 2. Superoxide anion radical scavenging activity was similar between Aralia continentalis Root and Angelica pubescens Root 3. The linoleic acid peroxidation inhibition effect was stronger in Aralia continentalis Root than in Angelica pubescens Root. 4. The phenolic component was higher in Aralia continentalis Root than in Angelica pubescens Root. 5. Both Aralia continentalis Root and Angelica pubescens Root increased the concentration of GSH and decreased SOD activity in mice, in which oxidative stress was induced, and the effect was stronger in Aralia continentalis Root. 6. Aralia continentalis Root increased GSH peroxidase activity but Angelica pubescens Root did not have such an effect. 7. Neither Aralia continentalis Root nor Angelica pubescens Root had a significant effect on catalase, ADH and ALDH in mice, in which oxidative stress was induced. Conclusions : Aralia continentalis Root has a stronger anti-oxidant effect than Angelica pubescens Root. Thus, although Aralia continentalis Root is not an original plant recorded in botanical literature, it may be usable based on the data about its effects.
This study was carried out to investigate the influence of dietary xanthophylls(lutein and apocarotenoic acid ethyl ester) supplementation on the antioxidation of broiler meat. The broilers fed with 10 ppm or 20 ppm xanthophylls were raised for 6 weeks and then slaughtered. The broiler meats were stored at 3$^{\circ}C$ for 9 days and frozen at -18$^{\circ}C$ for 4 months until analysis, respectively. The pH of all treatments significantly(p<0.05) increased during the storage periods. The pH of the thigh was higher than that of the breast. TBARS (thiobarbituric acid reactive substance) and POV(peroxide value) were higher in thigh than breast. All meats from broiler fed with lutein and apocarotenoic acid ethyl ester(apo-ester) had greater antioxidant properties during the storage period than control meat(p<0.05). Antioxidant activity of dietary xanthophylls supplementation was more effective in thigh than breast, and in broiler meats during frozen storage than chilled storage. The higher concentration of xanthophylls in feed, the more inhibition of lipid oxidation in meat during storage. The meat from broiler fed with 20 ppm of lutein showed the highest antioxidant property during both refrigerated and frozen storage although there was no significant difference between lutein and apo-ester(p>0.05). Consequently, this results indicated that the antioxidant activity of dietary xanthophylls(lutein and apocarotenoic acid ethyl ester) supplementation was more effective.
This study was carried out to evaluate biological activities to different ethanol extracts from unripened and ripened fruit (Rubus coreanus Miq.). 25% to 75% ethanol extracts of unripened and ripened fruits were similar to extract yield respectively. Yield of ethanol extract of ripened fruit were approximately 3 times higher than that of unripened fruit. 75% ethanol extract of unripened fruit showed the highest contents of total polyphenol ($180.04{\pm}0.41$ mg/g) and total flavonoid ($50.43{\pm}0.81$ mg/g). Contents of total polyphenol and total flavonoid of unripened fruit were about 2 times higher than those of ripened fruit. $IC_{50}$ values of DPPH radical scavenging activity and linoleic acid peroxidation inhibition activity of BHA and ${\alpha}$-tocopherol showed $13.19{\pm}0.21$ and $18.16{\pm}0.23{\mu}g/m{\ell}$, $4.25{\pm}0.04{\mu}g/m{\ell}$ and $5.56{\pm}0.10{\mu}g/m{\ell}$ respectively, but 75% ethanol extract of unripened fruit showed the lowest $23.85{\pm}0.10{\mu}g/m{\ell}$ and $7.34{\pm}0.07{\mu}g/m{\ell}$ among other all extracts. $IC_{50}$ values of LDL (low density lipoprotein) oxidation inhibition activity and ${\alpha}$-glucosidase inhibitory activity of 75% ethanol extract of unripened fruit showed the lowest $1.04{\pm}0.04{\mu}g/m{\ell}$ and $7.21{\pm}0.13{\mu}g/m{\ell}$ among other all extracts respectively. Specifically, 75% ethanol extract of unripened fruit has relatively better biological activities than other ethanol extracts, it could be potentially used as bioactive source for health functional foods.
Objective: This study investigated the effect of extraction solvents on antioxidant bio-active compounds as well as potential antioxidant and lipid peroxidation inhibition of Phyllanthus acidus (P. acidus) leaf extract in minced pork. Methods: The effect of various solvent systems of water, 25%, 50%, 75% (v/v) ethanol in water and absolute ethanol on the extraction crude yield, total phenolic content, total flavonoid content and in vitro antioxidant activities of P. acidus leaves was determined. In addition, antioxidant activities of the addition of crude extract from P. aciuds leaves at 2.5 and 5 g/kg in minced pork on 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging, 2,2′-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical cation decolorization, reducing power and inhibition of lipid peroxidation (thiobarbituric acid reactive substances; TBARS) were determined. Moreover, sensory evaluation of the samples was undertaken by using a 7-point hedonic scale. Results: The results showed that the highest crude yield (2.8 g/100 g dry weight) was obtained from water which also had the highest recovery yield for total phenolic content, total flavonoid content and the strongest antioxidant activity. The addition of crude water extract from P. acidus leaves was more effective in retarding lipid peroxidation and higher antioxidant activity than control and butylated hydroxytoluene in minced pork. In particular, the samples containing P. acidus extract had no significant effect on the sensory scores of overall appearance, color, odor, texture, flavor, and overall acceptability compared to the control. Conclusion: Water solvent was an optimally appropriate solvent for P. acidus leaf extraction because of its ability to yield the highest amount of bio-active compounds and in vitro antioxidant property. Particularly, P. acidus crude water extract also strongly expressed the capacity to retard lipid oxidation, radical scavenging, radical cation decolorization and reducing power in minced pork. The results of this study indicated that P. acidus leaf extract could be used as natural antioxidant in the pork industry.
Mitochondria in L. edodes were separated and purified by stepped sucrose density gradient centrifugation. In our previous work, we have found that the activation wavelengths of the mitochondrial ATPase and ATP synthase were 680 nm and 470 nm within the range of 400-700 nm, respectively. The activities of the above enzymes with wavelengths of 300-400 nm region were investigated. The mitochondrial ATPase and ATP synthase were stimulated at 380 nm and 330 nm, respectively, for 30 min illumination compared with dark control group. They, however, were inhibited at 330 nm and 350 nm, respectively. The presence of FAD resulted in inhibition of the activity of the ATPase and stimulation of the activity of the ATP synthase by the activation and inhibition wavelengths. However, the activities of these enzymes were not changed by NADH for the above wavelengths. In the spectral properties, the oxidation of $FADH_2$ into FAD occurs in the presence of the enzymes for illumination of the activation and inhibition wavelengths. Therefore, we can predict that the mitochondrial ATPase and ATP synthase may function as oxidant in the redox reaction by the light illumination and that the light-induced pigment of the mitochondrial ATP synthase should be an oxidized form of a flavoprotein.
Effect of inhibitions of three kinds of Ginkgo biloba extracts(Ginkgo biloba extract, Ginkgolide A, and Ginkgolide B) on induction of reactive oxygen species and release of inflammation mediator arachidonic acid were tested. Three kinds of Ginkgo biloba extracts could not inhibit the pyrogallol auto-oxidation, but they showed the hydrogen atom donating activity in DPPH assay. When 10 ${\mu}M$ hydrogen peroxide and 400 ${\mu}g/mL$ of three kinds of Ginkgo biloba extracts were added to U937 monocytic macrophage, the induction of lipid peroxidation was not observed. The Ginkgo biloba extract showed the most powerful inhibition among the extracts. And only Ginkgolide A was good for the inhibition of the protein degradation. The release of inflammation mediator arachidonic acid was induced by adding TPA and calcimycin to U937. In this assay, even 10 ${\mu}g/mL$ of three different Ginkgo biloba extracts excellently blocked the release of arachidonic acid. Particularly, the inhibition efficiency of Ginkgolide B was about 11 times higher than that of induction, and was about 4 times higher than that of the control of noninduction. This result suggests that the release of arachidonic acid is not inhibited by the antioxidant activity of Ginkgo biloba extracts, but a pre-step of the release of arachidoinc acid is inhibited by Ginkgo biloba extracts.
