Complete genome sequence of Salmonella enterica strain K_SA184, multidrug resistance bacterium isolated from lamb (Ovis aries)

  • Salmonella enterica is a representative foodborne pathogen in the world. The S. enterica strain K_SA184 was isolated from the lamb (Ovis aries), which was collected from a local traditional market in South Korea. In this study, the S. enterica strain K_SA184 was sequenced using PacBio RS II and Illumina NextSeq 500 platforms. The final complete genome of the S. enterica strain K_SA184 consist of one circular chromosome (4,725,087 bp) with 52.3% of guanine + cytosine (G + C) content, 4,363 of coding sequence (CDS), 85 of tRNA, and 22 of rRNA genes. The S. enterica strain K_SA184 genome includes encoding virulence genes, such as Type III secretion systems and multidrug resistance related genes.

The First Case of Diarrhoea in Tibetan Sheep, Ovis aries, Caused by Balantidium coli in the Qinghai Tibetan Plateau Area, China

  • This study was carried out to determine the pathogen-causing diarrhoea in sheep Ovis aries in the Qinghai Tibetan Plateau Area, China. A trophozoite was identified as species of ciliate alveolates infecting the sheep based on morphological characteristics examined by microscope. It was mostly spherical, colourless and transparent, with many vesicles. Macronucleus and contractile vacuoles could not be distinguished. Size of the trophozoite was $80-180{\times}70-150{\mu}m$ and its surface was covered with cilia. Molecular analysis based on sequences of 18S rRNA and ITS genes confirmed the ciliate species as Balantidium coli. According to the literature, there have been many epidemiological investigations of B. coli infection in pigs, monkeys and humans. To our knowledge, this was the first report of B. coli infections in sheep in the Qinghai Tibetan Plateau Area of China, or eleswhere around the world. Importantly, the sheep case was rare but raised our concern that B. coli may spread across species and expand its host range.

Complete Mitochondrial Genome Sequences of Chinese Indigenous Sheep with Different Tail Types and an Analysis of Phylogenetic Evolution in Domestic Sheep

  • China has a long history of sheep (Ovis aries [O. aries]) breeding and an abundance of sheep genetic resources. Knowledge of the complete O. aries mitogenome should facilitate the study of the evolutionary history of the species. Therefore, the complete mitogenome of O. aries was sequenced and annotated. In order to characterize the mitogenomes of 3 Chinese sheep breeds (Altay sheep [AL], Shandong large-tailed sheep [SD], and small-tailed Hulun Buir sheep [sHL]), 19 sets of primers were employed to amplify contiguous, overlapping segments of the complete mitochondrial DNA (mtDNA) sequence of each breed. The sizes of the complete mitochondrial genomes of the sHL, AL, and SD breeds were 16,617 bp, 16,613 bp, and 16,613 bp, respectively. The mitochondrial genomes were deposited in the GenBank database with accession numbers KP702285 (AL sheep), KP981378 (SD sheep), and KP981380 (sHL sheep) respectively. The organization of the 3 analyzed sheep mitochondrial genomes was similar, with each consisting of 22 tRNA genes, 2 rRNA genes (12S rRNA and 16S rRNA), 13 protein-coding genes, and 1 control region (D-loop). The NADH dehydrogenase subunit 6 (ND6) and 8 tRNA genes were encoded on the light strand, whereas the rest of the mitochondrial genes were encoded on the heavy strand. The nucleotide skewness of the coding strands of the 3 analyzed mitogenomes was biased toward A and T. We constructed a phylogenetic tree using the complete mitogenomes of each type of sheep to allow us to understand the genetic relationships between Chinese breeds of O. aries and those developed and utilized in other countries. Our findings provide important information regarding the O. aries mitogenome and the evolutionary history of O. aries inside and outside China. In addition, our results provide a foundation for further exploration of the taxonomic status of O. aries.

Comparison of Female Reproductive Systems in Himalayan Tahr (Hemitragus jemlahicus) and Corriedale Sheep (Ovis aries)

  • A necropsy of a primiparous, 4-year-old, Himalayan tahr (Hemitragus jemlahicus) was performed in September 8, 2009. Typical appearances of ovaries, oviducts, uterine horns, cervix and caruncles were well shown. Five cervical folds were present in the cervix. Cervical opening can be reached in the length of less than 15 cm from the entrance of vagina. Development of two follicles was found in the left ovary even though breeding season of this species naturally starts in late November. The reproductive organs of a primiparous, 3-year-old, Corriedale sheep showed that differences in the length and morphological appearance of cervix exist between two species. Comparative understanding of reproductive systems in Himalayan tahr and Corriedale sheep could help advancing assisted reproductive technologies in feral goats.

ASSOCIATION OF RED CELL X-PROTEIN, NADH-DIAPHORASE 1, MALIC ENZYME AND SERUM ARYLESTERASE PHENOTYPES WITH ECONOMIC TRAITS IN SHEEP

  • The red cell X-protein, NADH-diaphorase 1, malic enzyme and serum arylesterase phenotypes of 50 Thai Longtail and 53 Cameroon X Thai Longtail ($F_1$) crossbred sheep were determined by horizontal starch gel electrophoresis. None of the economic traits was influenced by DIA1, ME and EsA phenotypes. However, XP phenotypes showed a highly significant association with body weight, body height, heart girth and back girth, with mean values of XP+ve phenotype greater than XP-ve. The $XP^+$ allele was associated with greater body weight, body height, heart girth and back girth.

An inventory of alien mammals for ecological risk assessment in South Korea (생태계위해성평가 기반 마련을 위한 한국 도입 외래 포유류 목록 구축)

  • This study investigated the exotic mammals introduced to Korea to present the basic data necessary for ecological risk assessment and the establishment of an alien species management database. For this purpose, a list of alien mammals introduced to Korea was established by investigating alien mammals introduced for zoo exhibits purposes, alien mammals in breeding environments traded between individuals, and alien mammals introduced into nature. A total of 163 taxa were identified as alien mammals introduced to Korea. There were 139 taxonomic groups of ornamental alien mammals, including 11 orders, 44 families, 129 species and 10 subspecies; 28 taxonomic groups of alien mammals traded between individuals, including five orders and 19 families; and eight taxonomic groups of alien mammals introduced into nature, including four orders, seven families, seven species, and one subspecies. For the effective management of alien mammals introduced to Korea, it is important to collect basic information, such as a list of all species introduced, their populations, and introduced areas. It is also necessary to apply proactive management policies according to the degree of potential risk by preemptively evaluating the ecological risks that may arise from the introduction of alien species into natural ecosystems. For the eight taxonomic groups of Capra hircus, Cervus nippon taiouanus, Felis catus, Myocastor coypus, Oryctolagus cuniculus, Ondatra zibethicus, Ovis aries, and Rattus norvegicus identified to be introduced into the natural ecosystem of Korea, the provision of proliferation control measures at the national level, continuous monitoring of changes in diffusion trends, and immediate response based on the degree of change are required.

Cloning and Sequence Analysis of Wild Argali ISG15 cDNA

  • The complete coding sequence of Wild Argali ISG15 cDNA was generated by rapid amplification of cDNA ends. The ISG15 cDNA was 642 bp with an open reading frame of 474 bp, which encoded a 17.47 kDa protein composed of 157 amino acids. Its amino acid sequence shared 97.9%, 80.8%, 91.4%, 94.3%, 78.3% identity with those of ISG15cDNA from Ovis aries (accession no. NM001009735.1), Capra hircus (accession no. HQ329186.1), Bos taurus (accession no. BC102318.1), Bubalus bubalis (accession no. HM543269.1), and Sus scrofa (accession no. EU647216.1), respectively. The entire coding sequence was inserted into the pET-28a vector and expressed in E. coli. The recombinant protein corresponded to the expected molecular mass of 25 kDa as judged by SDS-PAGE, and it was detected in the bacterial inclusion bodies. The expressed protein could be purified by $Ni^{2+}$ chelate affinity chromatography and the results from the lymphocyte proliferation test showed that the product could stimulate lymphocyte proliferation very well (p<0.05), which further confirmed its biological activity.

Some Motifs Were Important for Myostatin Transcriptional Regulation in Sheep (Ovis aries)

  • Many motifs along the 1.2 kb myostatin promoter (MSTNpro) in sheep have been found by the MatInspecter program in our recent study. To further verify the role of the motifs and better understand the transcriptional regulation mechanism of the myostatin gene in sheep, the reporter gene EGFP (enhanced green fluorescent protein) was selected and the wild-type (W) vector MSTNPro$^W$-EGFP or motif-mutational (M) vector MSTNPro$^M$-EGFP were constructed. The transcriptional regulation activities were analyzed by detecting the fluorescence strength of EGFP in C2C12 myoblasts transfected with the vectors. The results showed that E-box (E) 3, E4, E5 and E7, particularly E3, E5 and E7, had important effects on the activity of the 1.2 kb sheep myostatin promoter. In addition, we also detected several other important motifs such as MTBF (muscle-specific Mt binding factor), MEF2 (myocyte enhancer factor 2), GRE (glucocorticoid response elements) and PRE (progesterone response elements) along the sheep myostatin promoter by the mutational analysis.

Molecular Characterisation of Nilagiri Sheep (Ovis aries) of South India Based on Microsatellites

  • Genetic variation in Nilagiri sheep, the only apparel wool breed in South India was studied using 25 FAO recommended ovine-specific microsatellite markers. The number of observed alleles ranged from 3 to 8 with a mean of 5 across all loci. The size of alleles ranged from 72 to 228 bp. The frequency of alleles ranged from 0.0104 to 0.5781. In total, 125 alleles were observed at the 25 loci studied. The effective number of alleles ranged from 2.18 to 6.49. The mean number of effective alleles was 3.84 across all loci. All the 25 loci were found to be highly polymorphic. The PIC values ranged from 0.4587 to 0.8277 with a mean of 0.6485. Of 25 microsatellites studied, 17 were in Hardy-Weinberg Equilibrium proportions. The observed heterozygosity ranged from 0.4222 to 1.000 with a mean value of 0.7610 whereas the expected heterozygosity ranged from 0.5415 to 0.8459 with a mean value of 0.7213. Except six loci, the other loci revealed negative within-population inbreeding estimates (FIS) indicating excess of heterozygotes in the population of Nilagiri sheep.
