Enhancement of Lysine Production in Recombinant Corynebacterium glutamicum through Expression of Deinococcus radiodurans pprM and dr1558 Genes (Deinococcus radiodurans 유래 DR1558과 PprM에 의한 Corynebacterium glutamicum의 라이신 생산 향상 연구)

  • The expression of Deinococcus radiodurans dr1558 and pprM genes was examined for enhanced lysine production in recombinant Corynebacterium glutamicum. These genes are known to confer high tolerance to pH and osmotic shock in Escherichia coli. D. radiodurans dr1558 and pprM genes were expressed in C. glutamicum by using 6 synthetic promoters of different strengths, to evaluate the effect of expression efficiency on lysine production. Recombinant C. glutamicum expressing DR1558 under the L26 and I64 promoters showed higher lysine production than that expressing DR1558 under other promoters. Similarly, recombinant C. glutamicum expressing PprM under same promoters (L26 and I64) showed a higher increase in lysine production compared to that expressing PprM under other promoters. In the absence of $CaCO_3$ in the medium, the expression of DR1558 or PprM also increased lysine concentration in C. glutamicum depending on the promoter used. Together, these results suggest that genes involved in radiation tolerance in D. radiodurans can be used to enhance production of amino acids and their derivatives.

  • The effects of Korean Red Ginseng (KRG) in rats submitted to exhaustion exercise have been studied, by measuring different enzymatic and hematological parameters in plasma and muscle. KRG powder was daily administered to 15 male Wistar rats for a period of two weeks. Another group of 15 rats with the same characteristics were administered physiological saline. Both groups were divided as follows: 5 control. 5 exercised till exhaustion and 5 recovered for 48 h after exhaustion. The following results were obtained for the groups treated with KRG in rapport to those treated with saline: - Higher endurance to running, - Increase of the osmotic resistence of red blood cells and higher presence of reticulocytes. - Lower triglyceride levels in plasma. - Increase non statistically significant of urea levels in plasma, - Lower non statistically significant hypoglycemia after exhaustion exercise. - Decrease of liver glycogen after exercise and faster recovery of the resting - level. - Protective effect on tissular damage produced by exhaustion exercise - Lower LDH activity in all studied muscles. only statistically significant in the WG.

Clinical Effect Of Low Level Laser Therapy In The Treatment Of Dentin Hypersensitivity Following Periodontal Surgery (치주수술 후 상아질 지각과민증에 대한 저수준 레이저 처치의 임상적 효과)

  • Root surface exposure due to gingival recession after periodontal surgery, dentin exposure after root planing elicit pain response when exposed to mechanical, heat, chemical or osmotic stimulation. Especially, patients treated with periodontal surgery, show high frequency and there have been reports showing the 1 out of 7 patients have dentin hypersensitivity. There have been many studies on the clinical effects of various materials on the treatment of dentin hypersensitivity. but, none could provide absolute clinical efficacy. In this study, 45 teeth from 30 patients, who had had periodontal surgery and showed dentin hypersensitivity after surgery were chosen for the experimental group and they were illuminated with laser, 15teeth were chosen for the control group and they were not exposed to laser. After this dentin hypersensitivity was elicited by tactile, compressed air, cold water and then, the degree was evaluated using NRS(Numerical Rating Scale). And during LLLT(Low Level Laser Therapy) semiconductor laser using Gallium - Arsenide as a diode was illuminated for 180 seconds at a frequency of 7(500Hz). This therapy was done 10 times, and each time the changes in dentin hypersensitivity was evaluated using NRS. The results were as follows : 1. After treat with LLLT on dentin hypersensitivity due to periodontal surgery, 22.2% showed total loss of dentin hypersensitivity, 60.0% showed loss of tactile dentin hypersensitivity, 48.8% showed loss of compressed air dentin hypersensitivity, 22.2% showed loss of cold water dentin hypersensitivity. 2. As a result of clinical evaluation of dentin hypersensitivity using NRS, there was significant increase in improvement of dentin hypersensitivity in the experimental group compare to the control group(P<0.05). And there was almost no natural loss of dentin hypersensitivity in the control group. 3. In comparison of the stages of evaluation, there was significant difference in between experimental and control group. after the second visit(P<0.05), and the difference increased with each visit.

  • The cell wall is essential for the survival and osmotic integrity of fungal cells. It is the framework to which biologically active proteins such as cell adhesion molecules and hydrolytic enzymes are attached or within which they act. Recently it was shown that mutations in ${\alpha}-COP$, a subunit of COPI vesicle, is responsible for the thermo-sensitive osmo-fragile phenotype of fungi, such as Saccharomyces cerevisiae and Aspergillus nidulans, and suggested that ${\alpha}-COP$ may play a crucial role in translocation of protein(s) of the ${\beta}-1,3-gulcan$ synthase complex and cell wall proteins, thus may contribute to the maintenance of cell wall integrity. In this review, we summarized the relationship between the intra-cellular protein translocation machinery, especially the ${\alpha}-COP$ of COPI vesicle, and cell wall biogenesis in fungi. We also discussed potential use of secretory mutants in basic and applied research of the fungal cell walls.

  • Increasing harmful effects of climate change, such as its effect on water scarcity, has led to a focus on developing effective water purification methods to obtain pure water. Additionally, rising levels of water pollution is increasing levels of environmental degradation, calling for sources of water treatment to remove contaminants. To purify water, osmotic processes across a semipermeable membrane can take place, and recent studies are showing that incorporating nanoparticles, including carbon quantum dots (CQDs), graphene carbon dots (GQDs), and graphene oxide quantum dots (GOQDs) are making thin film composite (TFC) membranes more effective by increasing water flux while maintaining similar levels of salt rejection, increasing the hydrophilicity of the membrane surface, showing bactericidal properties, exhibiting antifouling properties to prevent accumulation of bacteria or other microorganisms from reducing the effectiveness of the membrane, and more. In the review, the synthesis process, applications, functionality, properties, and the role of several types of quantum dots are discussed in the composite membrane for water purification.

Effect of Membrane Module and Feed Flow Configuration on Performance in Pressure Retarded Osmosis (압력지연삼투(PRO) 공정에서 막 모듈 배치와 유입원수의 유입 흐름방식이 성능에 미치는 영향)

  • Recently, reverse osmosis (RO) is the most common process for seawater desalination. A common problem in both RO and thermal processes is the high energy requirements for seawater desalination. The one energy saving method when utilizing the osmotic power is utilizing pressure retarded osmosis (PRO) process. The PRO process can be used to operate hydro turbines for electrical power production or can be used directly to supplement the energy required for RO desalination system. This study was carried out to evaluate the performance of both single-stage PRO process and two-stage PRO process using RO concentrate for a draw solution and RO permeate for a feed solution. The major results, were found that increase of the draw and feed solution flowrate lead to increase of the production of power density and water permeate. Also, comparison between CDCF and CDDF configuration showed that the CDDF was better than CDCF for stable operation of PRO process. In addition, power density of two-stage PRO was lower than the one of single-stage. However, net power of two-stage PRO was higher than the one of single-stage PRO.

Effect of Pre-soaking in Salt and Sugar Solutions before Air Drying on Quality Characteristics of Dried Apples (염 및 당 침지 처리가 건조 사과의 품질특성에 미치는 영향)

  • In this study, we investigated the effects of pre-soaking in salt and sugar solution prior to air drying at $50^{\circ}C$ on the characteristics of dried apples. Pre-soaking solutions included single solutions of salt 2% and sugar 2%; and combined solutions of salt 2%+sugar 2% and salt 2%+sugar 5%, respectively. The effects of pre-soaking condition and drying were evaluated in terms of moisture content (MC), water activity, color, antioxidant activity determined by DPPH radical scavenging activity, shear force, microbial contents, and sensory evaluation of apple slices. The control sample without pre-soaking showed the most rapid drying rate; in addition, the single solutions showed higher MC and water activity after drying time of 120 min, as compared to combined solutions. In all samples, MC and water activity showed high correlation coefficients of 0.91 to 0.97; whereas, shear force was negatively correlated with MC and water activity. The single solution of salt 2% showed decrease in change of color, including L, a, and b values, and the number of aerobic bacteria during drying. In addition, highest antioxidant activity and values of sensory preferences were observed in the dried apple pre-soaked in single solution of salt 2%.

Moringa oleifera Prolongs Lifespan via DAF-16/FOXO Transcriptional Factor in Caenorhabditis elegans

  • Here in this study, we investigated the lifespan-extending effect and underlying mechanism of methanolic extract of Moringa olelifa leaves (MML) using Caenorhabditis elegans (C. elegans) model system. To define the longevity properties of MML we conducted lifespan assay and MML showed significant increase in lifespan under normal culture condition. In addition, MML elevated stress tolerance of C. elegans to endure against thermal, oxidative and osmotic stress conditions. Our data also revealed that increased activities of antioxidant enzymes and expressions of stress resistance proteins were attributed to MML-mediated enhanced stress resistance. We further investigated the involvement of MML on the aging-related factors such as growth, food intake, fertility, and motility. Interestingly, MML significantly reduced growth and egg-laying, suggesting these factors were closely linked with MML-mediated longevity. We also observed the movement of aged worms to estimate the effects of MML on the health span. Herein, MML efficiently elevated motility of aged worms, indicating MML may affect health span as well as lifespan. Our genetic analysis using knockout mutants showed that lifespan-extension activity of MML was interconnected with several genes such as skn-1, sir-2.1, daf-2, age-1 and daf-16. Based on these results, we could conclude that MML prolongs the lifespan of worms via activation of SKN-1 and SIR-2.1 and inhibition of insulin/IGF pathway, followed by DAF-16 activation.

A Case of Ischemic Colitis Presenting as Bloody Diarrhea after Glycerin Enema in a Patient on Modified Fasting Therapy (절식요법 중 글리세린 관장 직후 혈성 설사로 발현한 허혈성 대장염 1예)

  • Case of ischemic colitis after enema for bowel cleansing have been rarely reported, but there has been no case report of a patient on modiefied fasting therapy. A 26-year old male patient with obesity admitted Korean medical hospital of Kyung Hee university for losing weight. He is on a special diet called modiefied fasting therapy, only took the fermented herbal drink. At 2nd day, he received an enema for bowel cleansing. A few hours after enema, he had a bloody diarrhea with lower abdominal pain. His colonoscopic and histologic findings presented ischemic colitis. He was advised to fast for two days and couldn't complete his diet program. We suggest 4 possible reasons : Increased intraluminal pressure by enema, vascular spasm caused by room-temperature glycerin solution colder than intraluminal temperature, predisposition to bleeding disturbances by taking selective serotonin reuptake inhibitor(SSRI) for depression history and mucosal injury by osmotic effect of glycerin solution itself. For reducing the risk of bowel cleansing, glycerin enema should be carefully prescribed and practiced concerning the condition of each patient.

  • Sodium chloride is present in our body fluids, and the blood contains approximately 0.9 wt% salt, which plays an important role in maintaining the osmotic pressure. However, the amount of salt intake has consistently increased, and an excessive intake can be the cause of high blood pressure, etc. In this study, it was investigated in vivo and in vitro whether biocompatible ionic polymers with K or Ca ions can be replaced by Na ions through an ion exchange process to be excreted. Among the polymers, Ca-polystyrene sulfonate, K-polystyrene sulfonate, Ca-carrageenan, and Ca-tamarind had an excellent Na exchange ability in the body temperature, simulated gastric fluid and also simulated intestinal fluid. The mechanism of Na removal by absorption and excretion without changing food taste in the mouth through the insolubility properties of these polymers is expected to be a solution for the current problems related with excess sodium intake.
