식물세포의 느린 생장과 낮은 생산량의 이유로 지금까지 는 주로 미생물이나 동물세포에서 유전자 재조합 단백질을 생산하여 왔다. 그러나 저렴한 배지 가격, 동물 유래 바이러스 감염 위험성으로부터의 안정성, glycosylation 등의 post-translational modification이 가능하다는 장점들로 인하여 최근 들어 식물세포배양은 생물학적 활성을 가진 고부가가치의 단백질을 생산하는데 많이 이용되고 있다. 본 연구에서는 생장배지에 첨가했던 sucrose의 소비와 induction 배지로의 교환에서 오는 배지내의 삼투압을 조절하여 hCTLA4-Ig의 생산성을 높이고자 하였다. 다양한 삼투압 조절제 첨가 실험을 통해 sorbitol을 선별하고, 40 mM의 sorbitol 첨가에서 상대적으로 높은 생존도와 induction 후 7일째 대조구보다 1.7배 높은 생산성을 확인하였다. 또한, 저농도의 glucose 첨가를 통한 생산성 증대에 있어서는 8 mM glucose에서 induction 이후에도 높은 세포농도를 유지하면서 최대 37.3 mg/L까지 hCTLA4-Ig 생산량을 증가시켰다. 5-L bioreactor에서 회분식 배양과 induction시의 hCTLA4-Ig 생산량을 비교한 결과 induction시 배양 18일째 최고 45.3 mg/L까지 높일 수 있었으며, 회분식 배양에 비해 2.1배 증가됨을 확인하였다.
The diurnal changes of the stomatal conductance, transpiration and leaf water potential were measured in order to assess the water relations characteristics of Pueraria thunbergiana in August of 1995 and 1996. The results showed two different responses depending on the duration of rainless days. The microclimatic conditions were highly stressful on 2 August. Daily maximum temperature reached to $39.0{\circ}C$ and vapor pressure deficit was 3.55 KPa. During this time the leaf water potential decreased to -1.02 MPa and a marked reduction of stomatal conductance was shown. However, on 15 August the stomatal conductance increased with increment of photon flux density, and transpiration was highly maintained during the day time. Minimum leaf water potential was only -0.47 MPa in spite of high transpiration rate. Furthermore, on 15 August reduced leaf water potential during the day time was recovered rapidly with decrease of photon flux density, whereas recovery of leaf water potential on 2 August was delayed. However, reduced leaf water potential on 2 August was recovered untile the next dawn. Osmotic potential at turgor loss point of Pueraria thunbergiana on 2, 3 and 15 August was -1.79, -1.70 and -1.60 MPa, respectively. The vapor pressure deficit is more contributive to the regulation of stomatal conductance than leaf water potential.
Objective : Subarachnoid hemorrhage[SAH] is commonly associated with polyuria [solute diuresis or water diuresis]. The authors investigate the incidence and clinical characteristics of polyuria with special reference to the administration of osmotic diuretics. Methods : One hundred and forty eight patients with high urine output [>200ml/hr] after ruptured cerebral aneurysm operated early from Jan 1998 to Jun 2003 were selected. Water diuresis [diabetes insipidus, DI] was differentiated from solute diuresis by lower urine specific gravity [<1.005] and higher plasma osmolality. The incidence and mode of onset of polyuria were compared between two types of diuresis. Additionally, the relationships between development of polyuria and clinical features including aneurysm location, clinical grade, Fisher grade, and outcome were analyzed. Osmotic diuretics were not routinely used in patients with Hunt-Hess grade I-III since July 2001. Results : Annual incidence of polyuria decreased markedly since July 2001 : 45.2% in 1998, 34.5% in 2001, 11.9% in 2003. Postoperative DI occurred in $2.4{\sim}11.1%$. DI developed mainly from ruptured anterior communicating artery aneurysm. The mean interval between the last SAH and the onset of DI was 7.1 days [range $1{\sim}27$ days] and lasted mean 4.6 days. When compared with solute diuresis, the development of DI was significantly delayed. Other clinical features were not closely related to polyuria. Conclusion : Uncontrolled polyuria may lead to cerebral ischemia and electrolyte imbalance because SAH patients are already predisposed to hypovolemia, and will risk precipitating the opposite situation with overhydration. We can decrease the development of polyuria without routine use of osmotic diuretics, by avoiding the increased intracranial pressure such as the intraoperative ventriculostomy and gentle brain retraction in good grade patients.
Pressure retarded osmosis(PRO) has attracted much attention as potential technology to reduce the overall energy consumption for reverse osmosis(RO) desalination. The RO/PRO hybrid process is considered as the most logical next step for future desalination. The PRO process aims to harness the osmotic energy difference of two aqueous solutions separated by a semipermeable membrane. By using the concentrated water(RO brine) discharged from existing RO plants, the PRO process can effectively exploit a greater salinity gradient to reduce the energy cost of processing concentrated water. However, in order to use RO brine as the draw solution, PRO membrane must have high water flux and enough mechanical strength to withstand the high operational pressure. This study investigates the development of a thin film composite PRO membrane and spiral wound module for high power density. Also, the influence of membrane backing layer on the overall power density was studied using the characteristic factors of PRO membranes. Finally, the performance test of an 8-inch spiral wound module was carried out under various operating conditions(i.e. hydraulic pressure, flow rate, temperature). As the flow rate and temperature increased under the same hydraulic pressure, the PRO performance increased due to the growth of water permeability coefficient and osmotic pressure. For a high performance PRO system, in order to optimize the operating conditions, it is highly recommended that the flow pressure be minimized while the flow rate is maintained at a high level.
영양생리적 기능이 우수하며, 식품위생적인 면으로도 안전하여 소비자들의 기호에 맞는 반염건고등어를 제조한 다음, 이의 효율적 이용을 위해 저온저장 중 품질안정성에 대하여 검토하였다. 제조직 후 반염건고등어의 수분함량은 $59.4{\sim}62.4%$의 범위였으나 저온저장 중 전제품이 약간씩 감소하였다. 저장 중 생균수는 저장 5일 초기의 경우 탈수방법의 차이에 관계없이 거의 변화가 없거나 약간 감소하였으나 저장 중기 이후에는 계속 증가를 하였고, 그 증가폭에 있어서는 거의 차이가 없었다. Histamine 함량의 경우 전제품 모두 제조직 후 $2.5{\sim}3.6mg/100g$의 범위였고, 저장 15일째에도 $3.6{\sim}4.1mg/100g$을 수준으로 histamine의 식중독 한계선인 100mg/100g에는 훨씬 못미치는 함량이었다. 저장 중 반염건고등어의 과산화물값, 혼합지방산 조성 및 갈변도 변화의 결과로 미루어 탈수시트로 탈수하여 제조한 반염건고등어가 재래식 건조법에 의하여 제조한 반염건고등어보다 저장 중 지질산화가 억제되었다. 저장일수가 경과할수록 염용성질소의 용해도는 감소하였고, 그 감소폭은 탈수시트로 제조한 반염건고등어가 전통적인 방법으로 제조한 것보다 적었다. 텍스튜어의 경우 제조방법에 관계없이 저장일수가 경과할수록 경도는 감소하는 경향을, 응집력은 거의 변화가 없거나 증가하는 경향을 나타내었고, 이등의 값은 탈수시트로 제조한 반염건고등어가 재래식건조법에 의하여 제조한 반염건고등어보다 높았다. 이화학적 검사 및 관능검사의 결과로 미루어 볼 때 탈수시트로 제조한 반염건고등어가 재래식방법으로 제조한 것보다 품질유지기간이 긴 것으로 판명되었다.
본 연구에서는 품종 및 전처리방법에 따른 복숭아의 냉해동 특성을 비교하였다. 전처리 없이 냉해동한 대옥계와 황도는 변색과 maximum force 증가, 드립로스 발생 현상을 보였는데, 이러한 문제점들은 전처리 방법에 의해 개선할 수 있었다. 전처리 직후 냉동 전에 전처리 별로 수분함량을 비교했을 때 sucrose를 포함한 삼투건조 시 수분이 감소하였으며 이는 해동 후 drip loss 감소에 영향을 주었다. 변색과 성분 및 항산화능력의 감소는 SB 3분 이상, ascorbic acid를 포함한 삼투건조 시 크게 개선되었다. Maximum force는 steam blanching, water blanching 시간이 증가할수록 감소하였으며, ACS의 경우 다른 삼투건조 처리구에 비해 높은 값을 보였다. 두 품종 모두 sucrose를 포함한 삼투건조 시 Brix가 크게 증가하였으며 pH와 산도는 품종 별로 차이는 있었으나 전처리 방법에 따라서는 큰 차이가 없었다. 물성 및 색도, 드립로스 등으로 판단하였을 때 steam blanching 3분 처리구가 전처리 조건 중 가장 적절한 방법으로 생각된다.
본 연구에서는 M16 A계통 '부지화'를 강세대목인 대목 '스윙글 시트루멜로' 또는 보통대목인 '탱자'에 접목하여 무가온 하우스에서 재배한 잎과 과실 사양의 수분상태 변화 및 과실 성숙기의 과실 가용성 고형물과 산 함량변화를 조사했다. 강세대목인 '스윙글 시트 루멜로'와 보통대목인 '탱자'에 접목한 M16 A계통 '부지화'의 대목에 따라 과실의 품질은 영향을 받는 것으로 나타났는데, '스윙글 시트루멜로' 대목은 '탱자' 대목보다 과실 가용성 고형물과 산함량이 낮았다. 동트기 직전에 측정된 과실사양의 수분포텐셜 및 팽압은 '스윙글 시트루멜로' 대목이 '탱자' 대목보다 조금 높은 경향을 보였으나, 삼투 포텐셜의 값은 '탱자' 대목이 '스윙글 시트루멜로' 대목보다 낮았다. 과실사양과 같은 시간대에 측정된 엽수분포텐셜 값의 변화도 과실 사양의 삼투포텐셜 변화와 유사한 경향을 보여주었다. 이상의 결과로 보아, M16 A계통 '부지화'에 있어 두개의 대목에 따라 잎의 수분 상태와 과실사양의 삼투포텐셜 차이가 있었으며, 이러한 차이로 인하여 과실 가용성 고형물과 산 함량에 차이가 나타나는 것으로 판단되며, 대목에 따른 수분 상태와 과실 품질의 차이는 대목의 뿌리분포가 영향을 미쳤기 때문인 것으로 사료된다.
We examined changes in the physiological responses of gray mullet Mugil cephalus exposed to acidic seawater (pH 6.0, 6.5, 7.0) and normal seawater (pH 8.0, control) for 15 days. As pH decreased, survival rate and body weight also decreased. Levels of aminotransferase, total protein and triglycerides also differed significantly with changes in pH, presumably due to stress caused by exposure to acidic water. The level of osmotic pressure was significantly higher in the pH 6.0 group than in other groups. Superoxide dismutase was significantly higher in the pH 6.5 and 7.0 groups than in the pH 8.0 group, and glutathione level was lowest in the pH 6.0 group. We conclude that decreasing the pH level of seawater induces a stress response in fish, damaging their ability to control their hematological and osmotic pressure. Antioxidant enzymes are generally sensitive to osmotic stress; in this study, antioxidant activity significantly changed with pH level. These results indicate that physiological stress induced by exposure to acidification reduces survival rates and inhibits growth in M. cephalus.
Titrated Extract of Centella asiatica (TECA) is a poorly water-soluble extract from the Centella asiatica. Despite excellent wound preparation causes pain due to a low aqueous solubility and high hypertonicity and therefore, the patient's compliance of this drug is low. The objective of this study is to design a formulation of TECA with an improved therapeutic applicability via an adequate solubilization. A mixture of propylene glycol and ethoxylated hydrogenated caster oil achieved an acceptable solubilization of TECA (i.e. 10%) via a formulation of micelle. A preparation of extemporaneous TECA micelle was achieved by a dilution of the original micellar formulation with either water or saline. An estimated distribution of particle size was between 15.9 and 32.6 ㎜. The osmotic pressure of the formulation was found to be much lower than that found In a commercially available injectable (i.e. Madecassole). The erthrocytic hemolysis of micellar solution was lower than that with the conventional dosage form, consistent with the osmotic pressure data. Pain score after an injection of the micellar solution was assessed by the hind-paw writhing test using ICR mice and compared with that found with the conventional injectable. The data indicated that the injection of the micellar solution was a significantly less painful. These results indicated that a micellar solubilization, followed by an extemporaneous dilution, is a novel formulation of TECA with an improved therapeutic applicability.
