Hybridization between Marine Medaka Oryzias dancena and Javanese Medaka Oryzias javanicus (바다송사리 Oryzias dancena와 자바송사리 Oryzias javanicus 간 잡종 유도)

  • Inductions of hybrids and reciprocal hybrids between Oryzias dancena and O. javanicus (ODJ and OJD) were conducted and backcross hybrids between female O. dancena and male ODJ were also produced for biological and cytogenetic analysis. Embryonic development of ODJ and OJD were compared with those of their parents. Developmental time was fastest in O. dancena and ODJ, followed by O. javanicus and OJD. Oryzias dancena hatched 11 days (d) after fertilization, ODJ at 13 d, O. javanicus at 14 d and OJD at 15 d. The abnormality of external morphology rate in ODJ was 10.6%; however, OJD showed a high degree of abnormality, over 90%. The proportion of males was 90.0% and 31.3% for ODJ and OJD, respectively. Cytogenetic analysis was conducted to obtain basic information for genetic identification of O. dancena, O. javanicus and their hybrids. The karyotypes of all experimental groups showed 2n=48 chromosomes and the fundamental number (FN) was 48. The first pair carried secondary constrictions near the centromeric regions. Erythrocyte area and volume were $9.8\;{\pm}\;0.5\;{\mu}m^2$ and $18.2\;{\pm}\;1.0\;{\mu}m^3$, respectively, for O. dancena, $8.3\;{\pm}\;0.5\;{\mu}m^2$ and $15.8\;{\pm}\;1.5\;{\mu}m^3$ in O. javanicus, and $18.3\;{\pm}\;0.5\;{\mu}m^2$ and $15.7\;{\pm}\;1.3\;{\mu}m^3$ in ODJ. Erythrocyte area and volume in ODJ were similar to those of O. javanicus. In backcross hybrids between female O. dancena and male ODJ, all embryos failed to develop and died in the late gastrula stage.

Early Gonadogenesis and Sex Differentiation of a Marine Medaka, Oryzias dancena (Beloniformes; Teleostei) (해산 송사리 Oryzias dancena의 초기 생식소 형성 및 성분화)

  • Sex differentiation and gonad development were investigated in a marine medaka species, Oryzias dancena (Beloniformes; Teleostei). The average time to hatch was 11 days post-fertilization (dpf) at $25^{\circ}C$. Primordial germ cell (PGC) was first observed at 5 dpf and migrated to presumptive gonadal area between the gut and pronephric duct at 9 dpf. Male and female gonads were morphologically differentiated at 12 days post-hatching (dph). Early oocytes at perinucleolus stage as well as the formation of spermatid and efferent duct were observed at 28 dph. At 6 weeks of age, the ovary exhibited yolk granulation in many oocytes, while testis possessed a considerable number of spermatogonia and spermatids. The first ovulation was observed in 9-week-old females, and at the same age, males contained fully-matured spermatozoa. Data obtained in this study indicate that the gonad differentiation of O. dancena is the typical type of differentiated gonochorism.

Tolerance Capacity to Salinity Changes in Adult and Larva of Oryzias dancena, a Euryhaline Medaka (광염성 송사리 Oryzias dancena 성체 및 자어의 염분도 변화에 대한 내성)

  • Osmoregulatory capabilities of a euryhaline medaka, Oryzias dancena (Beloniformes; Teleostei), was examined with a particular emphasis on adult and larval viability during direct salinity changes. O. dancena adults were highly capable for hyper-osmoregulation as well as hypo-osmoregulation, as evidenced by no adverse effect on their viability during the direct transfer either from complete freshwater ($0^{\circ}/_{\circ\circ}$) to $40^{\circ}/_{\circ\circ}$ salinity, or from $70^{\circ}/_{\circ\circ}$ to $0^{\circ}/_{\circ\circ}$. Furthermore, the phased increase of external salinity with acclimation periods allowed them to survive at a salinity as high as $75^{\circ}/_{\circ\circ}$. However, tolerant capability to acute salinity increase in early larval stage was much less than in adult stage, based on the finding that the tolerance range of salinity increase was only $15^{\circ}/_{\circ\circ}$ from freshwater, indicating that the hyper-osmoregulation system might not be fully developed in the early larval stage. On the contrary, the hypoosmoregulation system could be more solidified in O. dancena larvae, as evidenced by their good survival even after direct transfer from $45^{\circ}/_{\circ\circ}$ to $0^{\circ}/_{\circ\circ}$. Knowledge achieved in this study could form the basis for a wide scope of researches including ecotoxicogenomics and geneexpression assay using this model species.

Embryogenesis and Early Ontogenesis of a Marine Medaka, Oryzias dancena (바다 송사리 Oryzias dancena의 난발생 및 자치어의 형태 발달)

  • The egg development and morphological changes of larvae, juveniles and adults of Oryzias dancena were observed. Fertilized eggs were incubated at $25{\pm}1^{\circ}C$; the process of embryonic development was observed by light microscopy and based on diagnostic features of the developing embryos. The average time to hatch was 11 days after fertilization. The hatched larvae averaged $4.40{\pm}0.24mm$ in total length (TL). The yolk sacs of the larvae were almost absorbed at 3 days after hatching and $4.55{\pm}0.23mm$ TL. At 21 days after hatching, the larvae were $7.23{\pm}0.73mm$ TL and had reached the juvenile stage. First ovulation was about 9 weeks after hatching and at $22.58{\pm}2.73mm$ TL.

Morphometric Characteristics of Diploid and Triploid Marine Medaka, Oryzias dancena

  • The morphometric truss characteristics and classical dimensions of the marine medaka, Oryzias dancena, that might distinguish diploid and triploid fish were examined. Significant differences in all the classical and truss dimensions of the diploid and triploid fish were observed in both sexes (p<0.01). All the dimensions of the triploid fish were greater than those of the diploid fish. The triploid marine medaka shows sexual dimorphism in these characters, and the sexual dimorphism of the triploid marine medaka is similar to that of the diploid marine medaka. Thus, when their classical dimension and truss dimension was measured, the growth of triploid marine medaka is faster than that of the diploid fish, and it displays clear sexual dimorphism, with male fish having longer dorsal and anal fins than female fish.

Toxicity and Effects of the Herbicide Glufosinate-Ammonium (Basta) on the Marine Medaka Oryzias dancena

  • Glufosinate-ammonium, a component of the herbicide Basta, is one of the most extensively used pesticides worldwide. In this study, we assessed subchronic and chronic toxicities of Basta and its histopathological effects on the marine medaka Oryzias dancena. Marine medaka were exposed to 0, 2, 4, or 8 mg/L of Basta for 28 or 42 days. The lethal concentration ($LC_{50}$) of Basta for 96 h is 8.76 mg/L. Histological changes in the gills and liver were evaluated with histopathological indices, allowing quantification of the damage to fish exposed to Basta. Blood congestion, lamellar fusion, and epithelial lifting were observed in the gills, and hydropic degeneration, fibrosis, lipid degeneration, leukocyte infiltration, and necrosis were found in the liver. These responses could be useful indicators of Basta toxicity in this species.

Effects of the Developmental Stage of Extract Donor Embryos on the Culture of Marine Medaka Oryzias dancena Embryonic Stem Cell-like Cells (배아추출물 공여 배아의 발생단계가 바다송사리(Oryzias dancena) 배아 줄기세포 유사세포의 배양에 미치는 영향)

  • Optimizing the conditions for stem cell culture is an essential prerequisite for the efficient utilization of stem cells. In the culture of fish embryonic stem cells (ESCs) or ESC-like cells, embryo extracts are important for stable growth, but there is no rule for determining the developmental stage of the embryos used to obtain extracts. Therefore, this study investigated the effects of the developmental stage of extract donor embryos on the culture of Oryzias dancena ESC-like cells. O. dancena ESC-like cells were cultured in different media containing each of four types of embryo extract depending on the developmental stage of the extract donor embryos. Growth, morphology, colony-forming ability, alkaline phosphatase (AP) activity, and embryoid body (EB) formation of the cells were investigated. While the developmental stage of the extract donor embryos did not influence the growth, morphology, AP activity, or EB formation of ESC-like cells, colony-forming ability was affected and the pattern of the effects differed completely between the two ESC-like cells investigated. These results suggest that the developmental stage of extract donor embryos should be selected carefully for the culture of ESC-like cells, according to the research purpose and type of cell line.

Establishment and Characterization of Permanent Cell Lines from Oryzias dancena Embryos

  • The development of species-specific fish cell lines has become a valuable tool for biological research. In recent years, marine medaka Oryzias dancena has been recognized as a good experimental model fish but there are no reports of establishment of cell lines from this fish. In this study, two cell lines from O. dancena blastula embryos were established from 41 total trials (4.9%). The two cell lines displayed typical in vitro morphology and have been cultured for >121 passages, which corresponds to 293 days. The doubling times of the cell lines were 29.84 and 28.59 h, respectively, and both possessed the potential to expand in a clonal manner, albeit with significant differences between the two cell lines. The absence of any of the four main medium supplements; i.e., fish serum, fetal bovine serum, basic fibroblast growth factor, and medaka embryo extract, significantly inhibited growth. The proportion of cells possessing normal chromosome number was 45% and 46.7% of the cell lines, respectively. Taken together, two cell lines that proliferate continuously were established from marine medaka and these cell lines may provide a basic tool for characterizing the unique features of this fish species.

Spawning Performance, Embryonic Development and Early Viability under Different Salinity Conditions in a Euryhaline Medaka Species, Oryzias dancena (서로 다른 염분도 조건하에서 광염성 송사리 Oryzias dancena의 산란, 발생 및 초기 생존)

  • Effects of different salinity levels on spawning performance, embryonic development and early viability of a euryhaline medaka species, Oryzias dancena, were examined. O. dancena were able to spawn eggs in a wide range of salinity from 0 to 70$^{\circ}/_{\circ\circ}$, however, the spawning frequency was lowered in complete freshwater (0$^{\circ}/_{\circ\circ}$) and in highly salted water (70$^{\circ}/_{\circ\circ}$). Fertilization success was negatively affected when the environmental salinity was higher than the salt concentration found in normal seawater. Embryonic viability and hatching success were also inversely related with the salinity levels. Typical abnormality was observed in developing embryos incubated at high salinities (30, 45 and 60$^{\circ}/_{\circ\circ}$). In addition, the time to hatch was significantly delayed with increasing salinities: peak hatching occurred at 12~14 days post fertilization (dpf) in freshwater and at least at 17 to 18 dpf in 60$^{\circ}/_{\circ\circ}$. Mean survival rates of the hatched larvae up to 7 days post hatching (dph) were at least 97% in salinity levels ranging from 0 to 30$^{\circ}/_{\circ\circ}$. However, larvae reared in 45 and 60$^{\circ}/_{\circ\circ}$ experienced significant mortality, especially in the early phase, resulting in only 75% and 64% survival rates up to 7 dph, respectively.

Changing Temperature Affects Anesthetic Effects and Physiological Stress Responses in Marine Medaka, Oryzias dancena (해산송사리, Oryzias dancena의 수온 변화에 의한 마취 효과와 생리적 스트레스 반응)

  • 본 연구는 온도 변화에 따른 해산송사리, Oryzias dancena의 마취 효과 및 생리적 스트레스 반응을 평가하고, 고온 및 저온에서 최적의 마취 수온을 구명하였다. 해산송사리에서 고온 실험($36{\sim}42^{\circ}C$)과 저온 실험($4{\sim}10^{\circ}C$)을 통해 마취 효과를 조사하였으며, 각 마취수온에서 전어체의 cortisol과 glucose를 측정하였다. 각 수온 마취 실험 후, 전개체는 모두 생존하였으며, 해산송사리의 마취시간은 고온 실험에서 고온 일수록, 저온 실험에서는 저온 일수록 유의적으로 빨랐다(P<0.05). 회복 시간은 유의하게 고온 실험에서 수온이 감소할수록, 저온 실험에서는 수온이 증가할수록 유의적으로 느렸다(P<0.05). 아가미 운동수는, 고온 실험에서 수온이 증가함에 따라 빨랐으며, 저온 실험에서는 수온이 감소할때 빨랐다(P<0.05). 수온이 $38^{\circ}C$$8^{\circ}C$ 일때의 마취 조건에서 실험직후 전어체 cortisol이 최대치로, 실험후 6시간까지 점진적으로 감소한 반면, 전어체 glucose는 실험후 1시간에 최고치를 보이고 실험후 2시간까지는 감소하였다. 기존의 마취제를 쓰지 않은 본 연구로 인해, 해산송사리의 여타 연구에서 샘플 및 사용자가 보다 더 안전하고 용이한 샘플 취급이 가능할 것으로 사료된다.
