Isolation and Identification of Adenosine and Phlomuroside from the Aerial Parts of Oryza sativa L. (벼(Oryza sativa L.)의 지상부로부터 adenosine과 phlomuroside의 분리 및 동정)

  • Fresh and chopped aerial parts of Oryza sativa were extracted in 80% aqueous mehthanol, and the concentrated extract was successively partitioned in n-hexane, ethyl acetate (EtOAc), n-butanol (n-BuOH), and $H_2O$ fractions. From the n-BuOH fraction, two compounds were isolated through repeated silica gel and ODS column chromatography (c.c.). Based on nuclear magnetic resonance (NMR), mass spectrometry and infrared spectroscopy spectroscopic data, the compounds were identified to be adenosine (1) and phlomuroside (2). Especially, the configuration of both the anomer hydroxyl groups was determined as ${\beta}$ from the coupling constants of the anomer protons (J =6.0 and 7.6 Hz) in the $^1H-NMR$ spectra. This is the first report for the isolation of these compounds from Oryza sativa L.

Changes in ROS-Scavenging Enzyme Activity in Rice (Oryza sativa L.) Exposed to High Salinity

  • We studied changes in the biochemical and physiological status and ROS-scavenging enzyme (superoxide dismutase, catalase and peroxidase) activity in leaves and roots of rice (Oryza sativa L.) plants exposed to high salinity. Under salt stress, the reduction in RWC (relative water content) in leaves was relatively severe in comparison with that of roots. The proline content was also significantly higher in leaves of rice plants following salt treatment. The activities of CAT and POX in roots increased with increasing NaCl concentration, but the activity of SOD decreased. These results suggest that the increase of endogenous proline is closely associated with the increase of CAT and POX activities, which may play important roles in salt tolerance. Therefore, we conclude that the alleviation of oxidative damage and increased resistance to salinity may result from the presence of efficient antioxidative systems.

RGISS: Rice (Oryza sativa L. ssp. japonica) Genome Information Service System

  • We have constructed the Rice Genome Information Service System (RGISS), which is an information service system of the Oryza sativa L. ssp. japonica (rice) genome, using the released version of rice Build 3.0 pseudomolecules based on the Ensembl architecture. The nonredundant library, composed of 3,360 clones of BACs, PACs, and fosmids, was used to construct supercontigs. RGISS contains 50,717 annotated genes from GenBank, 56,161 predicted genes from FgeneSH, and information on 9,587 markers, which includes STS, SSR, and EST-based RFLP. The 20,180 ESTs sequenced by the Korea National Institute of Agricultural Biotechnology (NIAB) were aligned and mapped into 168,792 exons. By gene ontology analysis, the classified protein numbers in the rice genome were 6158, 4531, and 12,364 proteins, which were mapped to molecular function, cellular component, and biological process, respectively.

  • The present work was dealt with the twelve species of orbatid mites from leaves, stems and stubbles of rice( Oryza sativa L). Among them six species, that is, Protoribates agricola, Scheloribates decarinatus, Scheloribates oryzae, Sacculozets filosus, Punctoribates manzanoensis, and Paralamellobates ceylanica are new ones in Korea. The other species are Epilohmannia palida pacifica, Podorbates cuspidatus, Tricogaulumna nipponica, Peloribate sp., Scheloribates sp., Galumna sp.

Morphological Characters of Panicle and Seed Mutants of Rice

  • Phenotypes of panicle, hull and seed of mutant rice (Oryza sativa L.) were characterized. Panicle mutants were classified in 4 groups with their internode length of main rachis, primary rachis, secondary rachis and pedicel. Hull and seed mutants were grouped into 12 based on their mutant characters in shape, size and color of seeds. These natural and spontaneous mutant collections showed distinct phenotypes to wild type rice. This might be useful for the identification of the functions of genetic factors in the Mendelian inheritance.

  • A positive selectable marker system was adapted for transformation of mature embryo-derived calli of Indica rice (Oryza sativa L.) utilizing the PMI gene encoding for phosphomannose-isomerase that converts mannose-6-phosphate to fructose-6-phosphate. The transformed cells grew on medium supplemented with 3% mannose as carbon source and calli were selected on media containing various concentrations of mannose. Molecular analyses showed that the transformed plants contained the PMI gene. The results indicate that the mannose selection system can be used for Agrobacterium-mediated transformation of mature embryo in rice to substitute the use of conventional selectable markers in genetic transformation.

Studies on the Leaf Photosynthesis of Salt-Stressed Rice Cultivars (염류처리에 따른 벼의 개엽광합성에 관한 연구)

  • The effects of NaCl salinity on the leaf photosynthesis and water relation of two cultivars of rice(Oryza sativa L.) , the salt-tolerant cultivar Seohae and the salt-senstive cultivar Iri-380 were exam-ined. Two cultivars of rice were grown for 14 days in nutrient solution at SOmM NaCl. Comparing theieaf Na content of two cultlvars, Seohae showed high accumulation of Na content in the leaf blade, while Iri-380 showed low. The Na content in leaf blade reduced the rate of leaf photosynthesis. Salt-tolerant cultivar Seohae was less decreased the rate of leaf photosynthesis than salt- sensitive cultivarIri-380. And Seohae showed larger decreased the osmotic potential in the leaves than Iri-380. This in-dicates that in the salt-tolerant cultivar, osmotic adjustment is developed under saliniEation.

New Stigmastane Steroids Constituents from Rice Hulls of Oryza sativa and Inhibitory Activity on Radish seed

  • Two new compounds 1'-(stigmast-11,20(21),25-trien 3$\alpha$,9$\alpha$-diolyl)-3'-(pimara-11,15-dien-3$\alpha$-olyl) glycerol (1) and stigmast-5-en-3$\alpha$,26-diol (2) along with known fatty acids n-hexacosanoic acid (3) and hexadecanoic acid (4), have been isolated from the methanol extract of rice hulls of Oryza sativa. The structures of the compounds were elucidated using 1D and 2D NMR spectral methods, viz; $^1H, ^{13}C, ^{13}C$-DEPT, $^1H-^1H$ COSY, $^1H-^{13}C$ HETCOR, HSQC and HMBC aided by IR, EIMS, FABMS and HRFABMS. Compound (1) showed inhibition to radish germination, growth of shoot and root length.
