Mapping quantitative trait loci controlling low-temperature germinability in rice

  • Low-temperature germination is one of the major determinants for stable stand establishment in the rice direct seeding method in temperate regions and at high altitude areas. Quantitative trait loci (QTL) controlling low-temperature germinability in rice were identified using 96 introgression lines (ILs) derived from a cross between Oryza rufipogon and the Korean japonica cultivar, 'Hwaseongbyeo'. The germination rate at $15^{\circ}C$ was measured to represent low-temperature germination and used for QTL analysis. The germination rate at $15^{\circ}C$ for 7 days of Oryza rufipogon and Hwaseongbyeo was 93.3 and 28.7%, respectively, and that of progenies ranged from 0 to 48%. A linkage map was constructed using 135 simple sequence repeat (SSR) markers. Five putative QTLs associated with low-temperature germination were detected on chromosomes 1, 3, 4, 10 and 11. The QTL, qltg10 on chromosome 10 accounted for 19.2% of the total phenotypic variation for low-temperature germinability. Four additional QTL, accounted for 10.4 - 15.1% of the total phenotypic variation. The O. rufipogon alleles in all detected QTLs loci increased the low-temperature germination rate. No QTL associated with low temperature germinability has been detected near the qltg10 QTL in this study suggesting that qltg10 is a new QTL. The locus, qltg10 is of particular interest because of its independence from undesirable height and maturity effects. The DNA markers linked to the QTL for low temperature germinability would be useful in selecting lines with enhanced low temperature germinability in rice breeding program.

종간교잡 유래 중생 다수성 벼 품종 "화원 2호" (A New Rice Variety Developed from an Interspecific Cross, "Hwaweon 2")

  • "화원2호"는 종간교잡을 통해 화성벼의 근동질계통을 육성하고자 '97년 화성벼와 O. rufipogon을 교배하고, 계속적인 여교배와 MAS를 병행 실시하여 CR121-1-1 계통을 선발하였다. 생산력검정 시험 결과 조사된 형질 중 천립중, 출수기, 간장, 수당립수 등에서 화성벼와 차이를 보이는 화성벼 근동질계통으로, 품종보호원 출원 조건에 부합하여 "화원2호"로 명명하고 품종보호원을 출원하였다. 화원 2호는 중간모본 혹은 분자연구를 위한 재료로서도 유용할 것이다. 1. 출수기는 보통기재배에서 8월 16일로 "화성벼보다 6일 정도 늦은 중만생종 품종이다. 2. 천립중은 24.4 g으로 "화성벼"보다 1.9 g 정도 증가하였으며 간장, 수당립수 등에서도 화성벼와 차이를 보였다. 3. 흰잎마름병에는 이병성이며, 줄무늬잎마름병에는 저항성이다. 5. 도정 특성, 아밀로스 함량은 화성벼와 비슷한 수준이다. 6. "화원 2호"의 정조수량은 '05~'06년 2개년간 실시한 생산력검정 시험에서 평균 7.68 MT/ha로 "화성벼" 대비 114% 수준이었다.

  • "화원1호"는 종간교잡을 통해 화성벼의 근동질계통을 육성하고자 '97년 화성벼와 O. rufipogon을 교배하고, 계속적인 여교배와 MAS(Marker aided selection)를 병행 실시하여 CR101-1-1 계통을 선발하였다. 생산력검정 시험 결과 조사된 형질 중 천립중을 제외한 기타 형질에서는 화성벼와 유사한 근동질계통이었으며, 품종보호원 출원 조건에 부합하여 "화원1호"로 명명하고 품종보호원을 출원하였다. 1. 출수기는 보통기재배에서 8월 10일로 "화성벼와 동일한 중생종 품종이다. 2. 천립중은 23.4 g으로 "화성벼"보다 무겁다. 3. 흰잎마름병에는 이병성이며, 줄무늬잎마름병에는 저항성이다. 5. 도정 특성, 아밀로스 함량은 화성벼와 비슷한 수준이다. 6. "화원 1호"의 정조수량은 '05~'06년 2개년간 실시한 생산력검정 시험에서 평균 7.43 MT/ha로 "화성벼" 대비 110% 수준이었다.

Mapping QTL for ratooning ability in advanced backcross lines from an Oryza sativa × O. rufipogon cross

  • Ratooning ability is one of the major different traits from perennial to cultivated rice and annual type. We developed a set of 126 introgression lines derived from a cross between Hwayeong and W1944 (O. rufipogon) to gain an insight into the genetic factors underlying differences between common wild rice and cultivated rice. One IL, CR6 among the 126 ILs of $BC_3F_4$ showed a significant difference in rationing ability compared with Hwayeong. To further characterize the rationing ability, CR6 was selected and crossed to Hwayeong to produce three secondary populations, $BC_4F_2$, $BC_4F_3$ and $BC_5F_2$. In the Hwayeong background, the W1944 allele was associated with an increase in rationing ability. QTL analysis showed that the qRAT5 for rationing ability was linked to RM194 ($R^2$=6.6%, 19.6%, and 44.5% in the $BC_4F_2$, $BC_5F_2$, and $BC_5F_3$, respectively). The putative qRAT5 was also tightly linked to QTLs for spikelets per panicle and grain weight indicating that this region harbors a QTL cluster related to domestication. To our knowledge, this is the first report to map the major QTL for ratooning ability in rice. The SSR markers linked to qRAT5 would be useful in marker-assisted selection for breeding lines with enhanced ratooning ability.

청주 소로리 유적의 제4기 퇴적층과 볍씨 출토의 의미 재고찰 (Rethinking of Quaternary deposits and implication of rice seeds in Cheongju Sorori Site, Korea)

  • 금강 중류의 소로리 고기 하성퇴적층의 형성시기는 해면변동성 하안단구를 적용시켜 최종 간빙기(MIS 5)로 추정되며, 신기 하성퇴적층은 최종빙기최성기(MIS 2) 이후의 뵐링-알러뢰드 간빙기(12,700~14,700 cal-yrBP)를 포함하며, 이후 영드라이아스기(11,700~12,900 cal-yrBP)에 걸치는 것으로 해석된다. 소로리 유적의 신기 하성퇴적층 중에서 Oryza rufipogon 등 같은 야생벼가 출토되는 중부 토탄층 상부는 보정연대로 약 15,000년전 이후에 형성되었다. 소로리 유적에서는 화분대 OC-2 상부구간에서는 화분조성상 아고산성 침엽수림보다는 낙엽활엽수로의 천이가 뚜렷한 편이다. 특히 중부 토탄층의 상부에 해당하는 OC-2 화분대는 B/A아간빙기에 형성되었다. 이 기간은 청주소로리 볍씨가 온난기의 화분 및 배후습지성 곤충들과 공반 출토되었던 만빙기(Lastest Glacial) 기간 중으로 간주되며 상대적으로 현재와 유사한 온난한 기후가 나타났던 시기로 볼 수 있다.

The NILs from an interspecific cross show enhanced plant height and antioxidant activity

  • A high-resolution physical map targeting a cluster of yield-related QTLs on the long arm of rice chromosome 9 was constructed across a 35.5kb region containing the six predicted genes including the probable ascorbate peroxidase (OsApx). The $BC_3F_6$ near isogenic lines (NILs) were derived from a cross between the Oryza sativa Hwaseong and O. rufipogon. The plant height and length of internodes were compared between Hwaseong and NILs. There were significant differences in plant height between Hwaseong and NILs. The NILs internodes were longer than Hwaseong, showing dramatic elongation in the first and fourth internodes; thereby, leading to increased plant height. The antioxidant activity of Hwaseong and NILs was also analyzed by 3,3-diaminobenzidine (DAB) staining and 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay. In order to understand whether or not OsApx gene is important in scavenging $H_2O_2$ in rice, DAB staining was used. Intense dark-brown coloration was observed in Hwaseong than NILs. In addition, DPPH scavenging ability of Hwaseong showed lower value than NILs. These results indicated that the internode elongation and antioxidant activity might possibly be controlled by OsApx. To know the causative relationship of the gene and phenotype, we will further analyze the gene expression and use it for functional studies by complementation transgenic approach.
