메뚜기류유충의 혈구(血球)에 대한 미세구조(微細構造) (An Ultrastructural Study on Larval Hemocytes of Orthoptera)

  • 방아깨비, 사마귀, 섬서구메뚜기의 종령유충을 재료로 하여 전자현마경적 관찰을 하였다. Prohemocyte는 세포중 가장 작고 구성비도 낮으며 그 구조상 다른 혈구의 기원이 되는것 같다. Plasmatocyte는 나비류에서 와는 달리 그 형태가 대부분 난형이고 허족과 소기관을 가장 많이 가진다. Granular Cell은 종에 따라 그 형태가 뚜렷이 구분되며 어느 경우나 구성비는 높았다. 같은 세포의 과립이라 할지라도 그 미세구조가 몇 가지로 구분되고 이것은 아마도 축적단계에서 오는 결과인듯 하다. Adipohemocyte 는 방아깨비에서만 나타났고 구성비는 대단히 낮았다.

Effect of different diets on growth and development of the two-spotted cricket, Gryllus bimaculatus (Orthoptera: Gryllidae)

  • Many insects have gained increasing attention as an alternative protein for humans. Among those, the two-spotted cricket, Gryllus bimaculatus (Orthoptera: Gryllidae), was recently approved as a general food ingredient by the Korean Ministry of Food and Drug Safety. For industrial utilization of G. bimaculatus, mass rearing techniques and production system should be standardized first. In this study, we investigated the effects of five different feeds on the growth and development of G. bimaculatus. Feed is the one of the key factors that has considerable effects on rearing insects. With five different kinds of feed on $1^{st}$, $3^{rd}$, and $5^{th}$ instar nymphs, the change of survival rate, body weight were monitored up to eight wk after hatching. We concluded that 50% of soybean flour, 20% of corn powder, 10% of rice bran, 9% of milk serum, 10% of rice flour, 0.5% of microorganisms, and 0.5% of multivitamins and minerals (diet C) was the best mix for promoting growth and development of $3^{rd}$ instar nymphs compared to the control diet.

The fatty acid composition of edible grasshopper Ruspolia differens (Serville) (Orthoptera: Tettigoniidae) feeding on diversifying diets of host plants

  • Ruspolia differens (Serville) (Orthoptera: Tettigoniidae) is a highly valued edible grasshopper species in Africa. However, the effects of plant diets on lipid content and fatty acid composition of R. differens are not well understood. We tested the effects of four diets on the total lipid content and fatty acid composition of R. differens. Sixth instar nymphs of R. differens were reared on one, and mixtures of two, three, and six natural plant inflorescences. Individuals collected from the field constituted a control treatment. We extracted lipids and analyzed the fatty acid methyl esters using gas chromatography-mass spectrometry. We analyzed if the total lipid content, body weight, and fatty acid composition differed among diets and between the sexes using two-way ANOVAs and a PERMANOVA model, respectively. The total lipid content and weight of R. differens did not differ among the diets. The nine common fatty acids were palmitic (mean across treatments, 26%), oleic (22%), palmitoleic (18%), linoleic (13%), stearic (7%), myristic (6%), myristoleic (4%), ${\alpha}$-linolenic (2%) and arachidic acid (1%). The composition of fatty acids and the proportion of essential fatty acids significantly differed among the diets. The proportion of essential fatty acids was highest in the control treatment (21%) but low in less diversified (one to three feed) diets (12-13%). This study demonstrates that the fatty acid composition in R. differens can be influenced through diet. Thus, with dietary manipulations, using local plants in Africa, it is possible to produce R. differens with preferred high quality essential fatty acids for human consumption.

Developmental characteristics and genetic diversity of the two-spotted cricket Gryllus bimaculatus De Geer, 1773 (Orthoptera: Gryllidae) in South Korea

  • In this study, we investigated the developmental characteristics and genetic diversity of seven populations of two-spotted crickets (Gryllus bimaculatus De Geer, 1773 (Orthoptera: Gryllidae)) raised in South Korea. Regarding the developmental characteristics of the species, we observed no statistically significant difference in the weight of the nymphs in the six populations we tested. After molting, although weight differences were observed between the populations in each stage of the developmental period, the average weight for each developmental stage was constant. We also analyzed mitochondrial COI gene sequences (DNA barcoding region) of the reared crickets collected from five insect farms and two national insect rearing facilities and the resultant sequences were analyzed together with the 12 sequences from foreign countries specimens obtained from public data. We detected six haplotypes from 111 specimens, indicating a low intraspecific genetic distance (~1.8%). The most dominant haplotype was overwhelmingly haplotype 1, which was found in all South Korean specimens and four specimens from China, Indonesia, and Germany. These findings indicate that the low genetic diversity of South Korean specimens can be explained by the fact that the G. bimaculatus population imported for feed from Japan in the early 2000s became a maternal group that spread throughout cricket farms in South Korea. In order to breed healthy cricket strains, it is necessary to increase genetic diversity by importing them from other countries through appropriate quarantine procedures.

육상풍력발전단지 조성에 따른 곤충군집 변화에 관한 연구 (Changes in insect diversity and composition after construction of an onshore wind farm)

  • 이번 연구는 육상풍력발전단지 내의 육상 곤충상을 파악하고, 발전시설과 관리 도로의 조성으로 발생하는 산림훼손 및 파편화에 따른 곤충 군집의 변화를 알고자 수행하였다. 2017년에 준공된 경상북도 영양과 영덕에 있는 육상풍력발전단지 내 식생이 훼손된 임도와 인근 산림을 조사지로 선정하였고, 2022년 8~9월에 다양한 곤충 군집을 조사하기 위해 맨눈 조사와 야간 조사를 병행하였다. 총 11목 50과 246종 1,076개체가 채집되었고, 나비목 곤충이 38.8%로 가장 큰 비중을 차지했으며, 벌목, 노린재목, 메뚜기목 순으로 확인되었다. 또한 산림과 비교하여 초지의 곤충 종 수가 유의하게 높았고 종 구성도 산림과 초지가 다르게 배열됨을 확인하였다. 비록 단기간의 조사였으나, 육상풍력발전단지의 조성으로 일부 교목의 훼손이 오히려 식생의 다양성이 생겨 육상 곤충의 종 구성과 다양성에 좋은 영향을 줌을 확인하였다. 이 결과를 바탕으로 육상풍력 발전단지 조성 때문에 생기는 생물상의 변화를 이해하는데 기초적인 자료로 활용되기를 기대해 본다.

Mitochondrial COI sequence-based population genetic analysis of the grasshopper, Patanga japonica Bolívar, 1898 (Acrididae: Orthoptera), which is a climate-sensitive indicator species in South Korea

  • Patanga japonica Bolívar, 1898 (Orthoptera: Acrididae) is listed as a climate-sensitive indicator species in South Korea and is called southern group of insects in that the main distributional range is southern region of South Korea and Asian continent. In South Korea, thus, the species was distributed mainly in southern region of South Korea including southward a remote Jeju Island, but recently the species has often been detected in mid to northern region of South Korea, implying northward range expansion in response to climate change. Understanding the characteristics of the changes in genetic diversity during range expansion in response to climate change could be a foundation for the understanding of future biodiversity. Thus, in this study, we attempted to understand the changing pattern of the genetic diversity of the P. japonica in newly expanded regions. For the purpose of study, we collected 125 individuals from seven localities throughout South Korea including two newly distributed regions (Pyeongtaek and Yeongwol at ~37° N). These were sequenced for a segment of mitochondrial cytochrome oxidase subunit I (COI) and analyzed for genetic diversity, haplotype frequency, and population genetic structure among populations. Interestingly, northward range expansion accompanied only haplotypes, which are most abundant in the core populations, providing a significant reduction in haplotype diversity, compared to other populations. Moreover, genetic diversity was still lower in the expanded regions, but no genetic isolation was detected. These results suggest that further longer time would take to reach to the comparable genetic diversity of preexisting populations in the expanded regions. Probably, availability of qualified habitats at the newly expanded region could be pivotal for successful northward range expansion in response to climate change.

  • Ultrastructure of the hemopoietic organ in the grasshopper, Euprepocnemis shirakii, was studied using light and electron microscopes. The hemopoietic organs located on the upper surface of the dorsal diaphragm between 1st and 8th segments were consisted of 3 kinds of cells; the reticular cells, stem cells and two types of granulocytes. The reticular cells had numerous cytoplasmic processes and forming a complex network. Characteristically, pinocytotic vesicles and rough endoplasmic reticulum were well developed in this cell. The stem cells give rise to differentiating hemocytes of the different cell lineages. Granulocytes within the hemopoietic organs were originated from the stern cells and differentiated into two types of hemocytes ; granulocyte( I ), granulocyte( II ).
