Sequestration of Orthophosphate by D(+)-Mannose Feeding Increases Nonphotochemical Quenchings in Chinese Cabbage Leaves (Mannose 처리된 배추 잎의 무기인산 감소에 따른 비광화학성 소산의 증가)

  • Limitation of photosynthesis in detached Chinese cabbage (Brassica campestris L.) leaves was induced by feeding of mannose (25 mM) for 12 h in the light, and changes in the basic thylakoid functions under this condition were investigated. The acid soluble phosphate contend and CO2 uptake rate was decreased by 66% and 67%, respectively. However, the starch content was increased by 24% compared to those of controls. From the fast induction curves of chlorophyll fluorescence, dark level fluorescence (Fo) slightly increased while intermediate plateau fluorescence level (FI) to peak level fluorescence (Fp) transient was significantly decreased with a slight decrease in the Fo-to-FI transient. This data means that reduction of secondary electron acceptor of PSII (QB) might be more severely inhibited than that of primary electron acceptor of PSII (QA) by decrease in phosphate level. The strong decline of (Fv)m//Fm ratio suggests that efficiency of excitation energy capture by PSII was decreased markedly. The quenching of Fo (qO), an indicator of state transition, was also occurred over the slow induction kinetics of chlorophyll fluorescence. From quenching analysis, fluorescence was dominantly quenched by nonphotochemical quenchings (qE+qT). These results showed that the capture and transfer efficiency of excitation energy to PSII reaction center in thylakoid was decreased with the decline of leaf phosphate level, and that the state transition was occurred during the induction of photosynthesis under these conditions.

  • Effects of $H_2O_2$ addition for fenton oxidation on iron coagulation for treatment of phosphorus species, such as orthophosphate, metaphosphate, pyrophosphate, organic phosphate, were investigated. The effects of coagulant dosage, hydrogen peroxide dosage and the combined sequence ferric coagulation and $H_2O_2$ addition for fenton oxidation and coagulation were studied. The characteristics of floc growth rate were monitored using the PDA. The removal efficiencies of phosphorus species by iron coagulation were increased as Fe/P molar ratio increased. However, the removal efficiencies of metaphosphate, pyrophosphate, organic phosphate by a ferric coagulation were not increased as Fe/P molar ratio increased. The removal efficiency of metaphosphate, pyrophosphate, organic phosphate was increased by using iron coagulation and $H_2O_2$ addition for fenton oxidation. The result indicated that non-reactive phosphorus after iron coagulation was changed to reactive phosphorus by $H_2O_2$ addition for fenton oxidation and the oxidized iron enhanced the coagulation efficiencies.

A Study on Phosphorus Removal Process Using Steel Industry By-Products(Slag) at Dynamic condition (동적(動的) 상태(狀態)에서 산업(産業) 폐기물(廢棄物)을 이용(利用)한 인(燐) 제거(除去)에 관한 연구(硏究))

  • Excessive phosphorus (P as orthophosphate) is one of the major pollutants in natural water that are responsible for algal blooms and eutrophication. P removal by slag is an attractive solution if the P sorption capacity of the slag is significant. To design an efficient land treatment facility, basic information on the behaviour of P in the media-water environment is required. In this study, detailed column experiments were conducted to study the P transport under dynamic condition, and mathematical models were developed to describe this process. The column experiments conducted with dust and cake waste products (slag) from BHP steel industry in Australia as adsorbing media indicated that they had higher sorption capacity of P than that of a sandy loam soil from North Sydney, Australia. P transport in the dust and cake columns exhibited characteristics S-shaped or curvilinear breakthrough curves. The simulated results from a dynamic physical non-equilibrium sorption model (DPNSM) and Freundlich isotherm constants satisfactorily matched the corresponding experimental breakthrough data. The mobility of P is restricted proportionally to the adsorbent's sorption capacity.

MECHANICAL PROPERTIES OF REUSED LITHIUM DISILICATE GLASS-CERAMIC OF IPS EMPRESS 2 SYSTEM

  • This investigation was designed to estimate the biaxial flexure strength and fracture toughness of lithium disilicate glass-ceramics of IPS Empress 2 system pressed with as-received ingots and their sprue buttons. Two groups of the lithium disilicate glass-ceramics were prepared as follows: group 1 is ingot-pressed group; group 2 is sprue button-pressed group. A ball-on-three-ball test was used to determine biaxial flexure strength (BFS) of disks in wet environment. Scanning electron microscopy(SEM) analysis was conducted to observe the microstructure of the ceramics. Unpaired t-test showed that there were no differences in the mean biaxial Hem strength (BFS) and KIC values between group 1 and 2 (p > 0.05). Two groups showed similar values in the KIC and the strength at 5% failure probability. The SEM micrographs of the IPS Empress 2 glass-ceramic showed a closely packed, multi-directionally interlocking pattern of numerous lithium disilicate crystals protruding from the glass matrix. The lithium orthophosphate crystals could not be observed on the fracture surface etched. There was no a marked difference of the microstructure between group 1 and 2. Although there were no tests including color stability, casting accuracy, etc., the results of this study implied that we could reuse the sprue button of the pressed lithium disilicate glass-ceramic of IPS Empress 2 system.

  • Nitrogen (N) is an important regulator of the expression of genes involved in carbon and N assimilation pathways in plants by selectively altering the levels of proteins and/or mRNAs. These in C4 plants include genes for such as phosphoenolpyruvate carboxylase, carbonic anhydrase, and pyruvate-Pi dikinase. The C4 genes are regulated in mesophyll cells by N availability both transcriptionally and posttranscriptionally through cytokinins and glutamine as signals. The level of both the signals is up-regulated by N availability: cytokinins in roots and glutamine in leaves. The level of glutamine is controlled by the differential expression by N of glutamine synthetase and ferrdoxin-dependent glutamate synthase genes which locate in the mesophyll cells of C4 plants. The results is discussed as molecular mechanism for the greater N use efficiency of the plants as well as N partitioning is the photosynthetic cells.

Characterization of C-P Lyase gene cluster by in vivo $^{31}$ P-NMR spectroscopy

  • $\^$31/ P-NMR experiment was performed to detect phophonates (Pn) utilization and degradation in the several different C-P lyase mutants of E. coli and in E. aerogenes and the recombinants. The relative peak intensity (RPI) for the standard samples of 0.5 mM methylphosphonate (MPn) and 1.0 mM aminoethylphosphonate in glucose-MOPS medium showed 0.5 : 1.0 ratio. In the case of BW14329 (.DELTA.phnC-P, .delta.phoA), RPI did not change significantly after 24 hrs culturing, which means it nearly could not utilize Pn. In vivo $\^$31/ P-NMR spectrum of E. aerogens (BWKL 16627) during 3 hrs starvation showed two intense peaks at 0-2 ppm and at near-10 ppm which indicate intracellular orthophosphate (Pi) and pyrophosphate (PPi), respectively. Both of them might be released by degradation of inorganic polyphosphate pool. When MPn is supplied to the medium as an unique P source, Pi content in the cell has the constant, but PPi seems to be slightly decreased. Recombinants (BWKL 16954) grew slower than E. aerogenes in the glucose-MOPS media with various P sources. In vivo $\^$31/ P-NMR spectrum of recombinant did not show any intense signal in the cell. Surprisingly, under the cultivation adding with MPn, a few intense peaks in the region of Pi AND phospate monoester were detected.

Effects of Different Hydraulic Retention Times on Contaminant Removal Efficiency Using Aerobic Granular Sludge (HRT 변경에 따른 호기성 그래뉼 슬러지의 오염원 제거효율에 미치는 영향)

  • The purpose of this study was to evaluate the effects of different Hydraulic Retention Times (HRTs) on the contaminant removal efficiency using Aerobic Granular Sludge (AGS). A laboratory-scale experiment was performed using a sequencing batch reactor, and the Chemical Oxygen Demand (COD), nitrogen, orthophosphate removal efficiency, AGS/MLSS ratio, and precipitability in accordance with the HRT were evaluated. As a result, the COD removal efficiency was not significantly different with the reduction in HRT, and at a HRT of 6 h, the removal rate was slightly increased owing to the increase in organic loading rate. The nitrogen removal efficiency was improved by injection of influent division at a HRT of 6 h. As the HRT decreased, the MLSS and AGS tended to increase, and the sludge volume index finally decreased to 50 mL/g. In addition, the size of the AGS gradually increased to about 1.0 mm. Therefore, the control of HRT provides favorable conditions for the stable formation of AGS, and is expected to improve the contaminant removal efficiency with the selection of a proper operation strategy.

Trait-based algal community assembly associated with Pectinatella magnifica (Bryozoa, Phylactolaemata)

  • Habitat-forming species increase spatial complexity and alter local environmental conditions, often facilitating the assembly of plants and animals. We conducted a trait-based approach to algal assemblages associated with the freshwater bryozoan, Pectinatella magnifica. Association with algae leads to the inner bodies of the bryozoans being colored green; this is frequently observed in the large rivers of South Korea. We collected the green-colored gelatinous matrices and phytoplankton from waterbodies of the two main rivers in South Korea. Algal assemblages within the colonies and in the waterbodies were compared using the three diversity indices (richness, diversity, and dominance), and the composition of functional groups (FGs) and morphologically based functional groups (MBFGs) between the colonies within and outside of P. magnifica colonies. The most dominant and common species within the colonies were Oscillatoria kawamurae and Pseudanabaena catenata, both of which were assigned to the same FG (codon S1). Of the algal assemblages within the colonies, the dominance was higher, while the richness and diversity were lower, than those in the waterbodies. There was variation in the compositions of FGs and MBFGs in the waterbodies outside the colonies. Total nitrogen and orthophosphate led to dominance, and were significant factors for the variation in FGs in the waterbodies, whereas there were no such significant factors within the colonies. This trait-based approach to the community structure of associated algae provides the status and habitat gradient of these communities, which are stable, isolated, and consistent with the overgrowth of shade-adapted tychoplanktonic cyanobacteria.

Flame Retardancy of Novel Phosphorus Flame Retardant for Polyurethane Foam (새로운 인계 난연제가 연질폴리우레탄 폼의 난연성에 미치는 영향)

  • Novel phosphrous flame retardant tetramethylene bis(orthophosphorylurea) (TBPU) was synthesized by the reaction of diphosphoric acid with 1,4-butanediol and urea, and charaterized by Fourier transform infrared spectroscopy (FT-IR). As the amount of TBPU added in polyurethane foam (PUF) was increased, the flame retardancy of PUF was increased and the mechanical properties were not decreased. Also, in the flame resistance test after the reduced pressure storage, the flame retardancy of TBPU added PUF was retained. We could find out that the thermal resistance of TBPU added PUF increased compared to that of pure PUF.

Biomass Production Potential of Chlorella vulgaris Under Different CO2 Concentrations and Light Intensities

  • The increasing demand of the crops (soybean and corn) for biofuel production has increased the focus of the animal nutritionists to look for alternative feeds, which are economic and environmental friendly. To identify microalgae as suitable candidate as an alternative feed, growth response of Chlorella vulgaris was studied under varying concentrations of carbon dioxide (0.07, 1.4, 3.0 and 5.0%) and photon densities (39.19, 72.97, 105.41, 116.22, 135.14, $175.68\;{\mu}mol/m^2/s$) by employing a photo-bioreactor. Swine wastewater was also investigated as nutritional source to economize the biomass production. Results showed that the higher biomass production was found to be at 3.0% $CO_2$ compared to other $CO_2$ concentrations. However, no difference in biomass production was found at $105.41\;{\mu}mol/m^2/s$ and above photon densities with 12 h of photoperiodicity. It was observed that C. vulgaris could easily grow in 200 times diluted swine wastewater and growth was found to be similar with that of artificial medium. Provided the conducive conditions for optimal growth, it has also the potentiality of depleting ammonia nitrogen ($NH_4$-N) and orthophosphate ($PO_4^{3-}$-P) completely from the wastewater after 3~4 days of cultivation. Thus, growing C. vulgaris would not only solve the problem of animal feed, but also help in biological $CO_2$ mitigation and wastewater treatment.
