반추위에서 분리한 곰팡이의 Conjugated Linoleic Acid 생산과 ITS-1 영역의 염기서열 해석 (Isolation and Identification of Rumen Fungus and Its Produced Conjugated Linoleic Acid)

  • 반추위에서 분리한 곰팡이(IS-13)의 conjugated linoleic acid 생산 과정을 조사하고conjugated linoleic acid를 생산하는 곰팡이를 동정하기 위하여 본 연구를 실시하였다. IS-13 곰팡이가 함유된 배양액에 linoleic acid를 첨가한 결과 linoleic acid는 배양 12시간 이내에 conjugated linoleic acid와 trans-11 vaccenic acid로 biohydrogenation되었다. IS-13 곰팡이의 동정은 internal transcribed spacer 1 영역(ITS1)을 sequence하여 GenBank 유래의 관련 23속(종)의 반추위 곰팡이와 비교하였다. IS-13 곰팡이의 ITS1의 length는 218 bp인 것으로 확인되었으며, 염기서열 분석 결과 Orpinomyces species와 98% 일치하여 반추위내 Orpinomyces species는 conjugated linoleic acid 생산에 깊이 관여하는 것으로 확인되었다. 연구결과 본 균주는 Orpinomyces 속에 속하는 균으로 동정되었다.

Biohydrogenation Pathways for Linoleic and Linolenic Acids by Orpinomyces Rumen Fungus

  • The objective of this study was to identify biohydrogenation pathways for linoleic, linolenic, oleic and stearic acids by Orpinomyces species of rumen fungus during in vitro culture. Biohydrogenation of linoleic acid produced conjugated linoleic acid (cis-9, trans-11 C18:2), which was then converted to vaccenic acid (trans-11 C18:1) as the end product of biohydrogenation. Biohydrogenation of linolenic acid produced cis-9, trans-11, cis-15 C18:3 and trans-11, cis-15 C18:2 as intermediates and vaccenic acid as the end product of biohydrogenation. Oleic acid and stearic acid were not converted to any other fatty acid. It is concluded that pathways for biohydrogenation of linoleic and linolenic acids by Orpinomyces are the same as those for group A rumen bacteria.

Effect of Grass Lipids and Long Chain Fatty Acids on Cellulose Digestion by Pure Cultures of Rumen Anaerobic Fungi, Piromyces rhizinflata B157 and Orpinomyces joyonii SG4

  • The effects of grass lipids and long chain fatty acids (LCFA; palmitic, stearic and oleic acids), at low concentrations (0.001~0.02%), on the growth and enzyme activity of two strains of anaerobic fungi, monocentric strain Piromyces rhizinflata B157 and polycentric strain Orpinomyces joyonii SG4, were investigated. The addition of grass lipids to the medium significantly (p<0.05) decreased filter paper (FP) cellulose digestion, cellulase activity and fungal growth compared to control treatment. However, LCFA did not have any significant inhibitory effects on fungal growth and enzyme activity, which, however, were significantly (p<0.05) stimulated by the addition of oleic acid as have been observed in rumen bacteria and protozoa. This is the first report to our knowledge on the effects of LCFA on the rumen anaerobic fungi. Continued work is needed to identify the mode of action of LCFA in different fungal strains and to verify whether these microorganisms have ability to hydrogenate unsaturated fatty acids to saturated fatty acids.

Influence of an Anaerobic Fungal Culture (Orpinomyces sp.) Administration on Growth Rate, Ruminal Fermentation and Nutrient Digestion in Calves

  • The study was to see the effect of administration of ruminal fungi on feed intake, growth rate, rumen fermentation and nutrient digestion of calves (Tharparkar$\times$Holstein-Friesian, average age: 10 months, average body weight: 130 kg). The 6 calves in first group were fed a mixture consisted of 50% wheat straw and 50% concentrate (Maize 62%, Groundnut cake 35%, Mineral mix. 2% and Common salt 1%) along with 1 kg green oats $animal^{-1}$ $day^{-1}$ while second group calves were fed the above-mentioned diet in addition to a dose of 160 ml ($10^{6}$ CFU/ml) fungal culture $calf^{-1}$ $week^{-1}$. The average dry matter intake per day was slightly lowered in fungal fed calves yet feed conversion ratio was higher. The average daily weight gain was significantly higher (15.37%) in fungal administered group as compared to control. The nutrient digestibility was increased for crude fibre, NDF and ADF with fungal administration. Digestible energy value of straw-based diet in terms of percent TDN also increased. The pH and $NH_{3}$-N were lower whereas TVFA, total-N, TCA-N and number of zoospores were higher in rumen liquor in fungal administered group.

Degradation of Rice Straw by Rumen Fungi and Cellulolytic Bacteria through Mono-, Co- or Sequential- Cultures

  • Two strains of rumen fungi (Piromyces rhizinflata B157, Orpinomyces joyonii SG4) and three strains of rumen cellulolytic bacteria (Ruminococcus albus B199, Ruminococcus flavefaciens FD1 and Fibrobacter succinogenes S85) were used as mono-cultures or combinationally arranged as co- and sequential-cultures to assess the relative contributions and interactions between rumen fungi and cellulolytic bacteria on rice straw degradation. The rates of dry matter degradation of co-cultures were similar to those of corresponding bacterial mono-cultures. Compared to corresponding sequential-cultures, the degradation of rice straw was reduced in all co-cultures (P<0.01). Regardless of the microbial species, the cellulolytic bacteria seemed to inhibit the degradation of rice straw by rumen fungi. The high efficiency of fungal cellulolysis seems to affect bacterial degradation rates.

한우 및 산양의 장내 섬유소 분해 혐기 곰팡이의 분리 및 특성 구명 (Isolation and Characterization of Cellulolytic Anaerobic Fungi from the Guts of the Hanwoo Cattle and the Korean Native Goat)

  • 본 연구는 국내의 재래 반추동물인 재래산양과 한우의 장내에 서식하며 강력한 섬유소를 분해하는 혐기 곰팡이를 탐색하고 분리하여 섬유소 분해 특성을 구명하고자 실시되었다. 산양의 반추위로부터 16종과 한우의 십이지장 소화물로부터 5종의 혐기 곰팡이를 분리하여 총 21종의 혐기성 곰팡이가 분리되었다. 섬유소 분해효소의 활력을 측정하여 그 중 섬유소 분해력이 높은 4종의 곰팡이에 대하여 광학현미경에 의한 형태학적 관찰을 기초로 동정 작업을 수행하였다. NLRI-M003은 monocentric 성장형태, 구형의 포자낭, filamentous rhizoid 및 유주자의 flagella가 다수인 Neocallimastix sp., NLRI-M014는 monocentric 성장형태, 방추형의 포자낭, filamentous rhizoid 및 유주자의 flagella가 단수인 Piromyces sp.로, NLRI-T004는 monocentric 성장형태, 난형의 포자낭, filamentous rhizoid 및 유주자의 fagella 수가 다수인 Neocallimastix sp.로 각각 확인되었다. NLRI-M001은 Orpinomyces sp. 와 유사한 것으로 추측되나 지금까지 밝혀진 곰팡이 이외에 다른 밝혀지지 않은 곰팡이가 존재할 가능성이 있을 것으로 평가되어 더욱 더 세부적인 조사가 필요하다고 사료되었다. 혐기 곰팡이의 섬유소 분해 특성을 조사하기 위해 산양의 반추위로부터 분리된 NLRI-M003 혐기 곰팡이 배양액을 2% 첨가하여 혼합 반추위 미생물의 in vitro 건물 분해율을 볏짚과 filter paper를 기질로 하여 조사하였다. 모든 처리구에서 혐기 곰팡이 배양액을 첨가한 첨가구가 무첨가구에 비하여 볏짚의 경우 약 4%이상(p〈0.05) 그리고 filtre paper를 기질로 사용시 11% 이상(p〈0.001)의 분해율이 증가하였다. 또한 CMCase와 xylanase 효소의 활력도 첨가구에서 증가하였으며 특히 반추위 곰팡이는 강력한 xylanase 효소활력이 높음을 보여주었다.

반추위 곰팡이 다양성 조사 : 메타분석 (Diversity Census of Fungi in the Ruminal Microbiome: A meta-analysis)

  • 본 연구의 목적은 곰팡이 28S rDNA 염기서열의 메타분석을 통하여 반추위 곰팡이의 다양성을 조사하는데 있다. 'rumen'과 'ruminal'이 반추위 곰팡이 유래 염기서열들을 회수하기 위한 검색어로 사용되었다. 2016년 9월부로 모든 28S rDNA 염기서열이 보관되어 있는 Ribosomal Database Project(RDP, http://rdp.cme.msu.edu) 데이터베이스에서 반추위 곰팡이 유래 28S rDNA 유전자 염기서열(n=165)을 획득하였다. 총 165개의 염기서열은 분류학상의 '문(phylum)'인 Ascomycota, Neocallimastigomycota 및 Basidiomycota로 분류되었고, 165개의 염기서열 중에서 각각 109개, 48개, 8개의 염기서열을 차지하였다. Ascomycota 염기서열은 식물병원성곰팡이나 마이코톡신을 생성하는 곰팡이를 포함하고 있는 '속(genus)' Pseudonectria, Magnaporthe, Alternaria, Cochliobolus, Cladosporium 및 Davidiella로 분류되었다. 또한, Basidiomycota 염기서열은 식물병원성곰팡이를 포함하고 있는 '속(genus)' Thanatephorus와 Cryptococcus로 분류되었다. 뿐만 아니라, Neocallimastigomycota 염기서열의 경우 섭취된 조사료의 주요 구조탄수화물을 분해하는 '속(genus)' Cyllamyces, Neocallimastix, Anaeromyces, Caecomyces, Orpinomyces, Piromyces로 분류되었다. 본 연구는 처음으로 28S rDNA 염기서열의 메타분석을 통해 반추위 곰팡이 다양성에 대한 정보를 통합적으로 제공하였다. 본 연구의 결과는 향후 반추위 곰팡이 연구에 대한 방향을 제공할 것이고, 새로운 분석도구 개발에 응용될 수 있을 것이다.

Nucleotide and protein researches on anaerobic fungi during four decades

  • Anaerobic fungi habitat in the gastrointestinal tract of foregut fermenters or hindgut fermenters and degrade fibrous plant biomass through the hydrolysis reactions with a wide variety of cellulolytic enzymes and physical penetration through fiber matrix with their rhizoids. To date, seventeen genera have been described in family Neocallimasticaceae, class Neocallimastigomycetes, phylum Neocallimastigomycota and one genus has been described in phylum Neocallimastigomycota. In National Center for Biotechnology Information (NCBI) database (DB), 23,830 nucleotide sequences and 59,512 protein sequences have been deposited and most of them were originated from Piromyces, Neocallimastix and Anaeromyces. Most of protein sequences (44,025) were acquired with PacBio next generation sequencing system. The whole genome sequences of Anaeromyces robustus, Neocallimastix californiae, Pecoramyces ruminantium, Piromyces finnis and Piromyces sp. E2 are available in Joint Genome Institute (JGI) database. According to the results of protein prediction, average Isoelectric points (pIs) were ranged from 5.88 (Anaeromyces) to 6.57 (Piromyces) and average molecular weights were ranged from 38.7 kDa (Orpinomyces) to 56.6 kDa (Piromyces). In Carbohydrate-Active enZYmes (CAZY) database, glycoside hydrolases (36), carbohydrate binding module (11), carbohydrate esterases (8), glycosyltransferase (5) and polysaccharide lyases (3) from anaerobic fungi were registered. During four decades, 1,031 research articles about anaerobic fungi were published and 444 and 719 articles were available in PubMed (PM) and PubMed Central (PMC) DB.

Effects of Non-ionic Surfactants on Enzyme Distributions of Rumen Contents, Anaerobic Growth of Rumen Microbes, Rumen Fermentation Characteristics and Performances of Lactating Cows

  • A series of experiments was carried out to determine the possibility for the non-ionic surfactant (NIS) as a feed additive for ruminant animals. The effect of the NIS on (1) the enzyme distribution in the rumen fluids of Hereford bulls, (2) the growth of pure culture of rumen bacteria and (3) rumen anaerobic fungi, (4) the ruminal fermentation characteristics of Korean native cattle (Hanwoo), and (5) the performances of Holstein dairy cows were investigated. When NIS was added to rumen fluid at the level of 0.05 and 0.1% (v/v), the total and specific activities of cell-free enzymes were significantly (p<0.01) increased, but those of cell-bound enzymes were slightly decreased, but not statistically significant. The growth rates of ruminal noncellulolytic species (Ruminobacter amylophilus, Megasphaera elsdenii, Prevotella ruminicola and Selenomonas ruminantium) were significantly (p<0.01) increased by the addition of NIS at both concentrations tested. However, the growth rate of ruminal cellulolytic bacteria (Fibrobacter succinogenes, Ruminococcus albus, Ruminococcus flavefaciens and Butyrivibrio fibrisolvens) were slightly increased or not affected by the NIS. In general, NIS appears to effect Gram-negative bacteria more than Gram-positive bacteria; and non-cellulolytic bacteria more than cellulolytic bacteria. The growth rates of ruminal monocentric fungi (Neocallimastix patriciarum and Piromyces communis) and polycentric fungi (Orpinomyces joyonii and Anaeromyces mucronatus) were also significantly (p<0.01) increased by the addition of NIS at all concentrations tested. When NIS was administrated to the rumen of Hanwoo, Total VFA and ammonia-N concentrations, the microbial cell growth rate, CMCase and xylanase activities in the rumen increased with statistical difference (p<0.01), but NIS administration did not affect at the time of 0 and 9 h post-feeding. Addition of NIS to TMR resulted in increased TMR intake and increased milk production by Holstein cows and decreased body condition scores. The NEFA and corticoid concentrations in the blood were lowered by the addition of NIS. These results indicated that the addition of NIS may greatly stimulate the release of some kinds of enzymes from microbial cells, and stimulate the growth rates of a range of anaerobic ruminal microorganisms, and also stimulate the rumen fermentation characteristics and animal performances. Our data indicates potential uses of the NIS as a feed additive for ruminant animals.
