The purpose of this study was to measure the bioactivity and antioxidant activity of Orostachys japonicus A. Berger. Orostachys japonicus A. Berger have been known to contain functional materials such as kaempferol, hydroquinone, methyl gallate, quercetin, gallic acid etc. To identify the main functional materials of Orostachys japonicus A. Berger, the contents of flavonoid and phenol were measured. We extracted Orostachys japonicus A. Berger powder from four solvents such as chloroform:methanol (CM, 2:1, v/v), distilled water (DW), 70% methanol, 70% ethanol. After that, this study determined tannin, total phenol, flavonoid content, DPPH radical scavenging activity, ABTS radical scavenging activity, ferric reducing antioxidant power and reducing power of Orostachys japonicus A. Berger extracts and as results of comparing each extract. respectively. From the above results shows that antioxidant activity and bioactivity of Orostachys japonicus A. Berger extracts was higher in the order of 70% ethanol, 70% methanol, DW and CM (p<0.05). The results showed that antioxidant activity of Orostachys japonicus A. Berger extracts supposed to affect by the total phenol and flavonoid contents.
The content of two steroids-campesterol (1) and ${\beta}$-sitosterol (2), and four triterpenes-taraxetrone (5), ${\beta}$-amyrin (6), (-)-friedelin (7), glutinol (8) in the Orostachys japonicus A. Berger cultivated under various conditions was estimated and compared with those in wild one. The present investigation disclosed that there are no significant difference in their contents between cultivated Orostachys japonicus A. Berger and wild one. from viewpoint of the content of the steroids 1 and 2, and the triterpenes 5-8, the quality of cultivated Orostachys japonicus A. Berger is not inferior to the wild one.
The content of four phonolic acids 1-4, and two flavonol aglycones 14 and 15 from Orostachysjaponicus A. Berger grown under night-break and day-length controlled experiments was estimated and compared with those in wild plants. The amount of the phenolic acids 1-4 and the flavonol aglycones 14 and 15 increased with increasing light irradiation under both the night-break and day-length control conditions. It was disclosed that the cultivation conditions such as the night-break and the day-length control were not adversely affect the production of phenolic acids and flavonols in Orostachys japonicus A. Berger extracts.
This study examined the antioxidant activity of methanol and water extracts from Orostachys japonicus A. Berger according to harvest times, by measuring electron donating ability(EDA), reducing power, superoxide dismutase(SOD)-like activity, thiobarbituric acid reactive substances(TBARS) and antioxidant activity within a linoleic acid emulsion. EDA increased proportionally with the extract concentration and the methanol extract had slightly stronger effects than the water extract. And reducing power and SOD-like activity were highest in the methanol extract. Overall, based on the data, the methanol extract of O. japonicus A. Berger harvested during $August{\sim}October$ presented the highest level of antioxidative activity and may be a good candidate as a natural antioxidant source.
The purpose of this study was to analyze the changes in physicochemical characteristics and NDMA formation of Orostachys japonicus A. Berger according to harvest times. The results showed moisture, carbohydrate, crude fiber and crude protein contents of <$48.5{\pm}1.4%$, $38.2{\pm}1.2%$, $l5.8{\pm}0.4%$ and $13.2{\pm}0.6%$ respectively. The total mineral content was 4,817.1 mg/100 g, where Ca was highest($2,577.0{\pm}3.2$ mg/100 g), followed by K, Mg, Na, P and Fe. The composition amino acid of O. japonicus A. Berger showed high levels of glutamic acid and aspartic acid. And among the five free sugars detected, galactose and glucose were most abundant at $32.2{\pm}0.02$ mg/100 g and $25.3{\pm}0.1O$ mg/100 g, respectively. Upon comparing O. japonicus A. Berger extracts that came from plants harvested during $August{\sim}October$, total phenolic compound($2,703.l{\pm}5.2l{\sim}2,428.0{\pm}3.52$ mg/100 g) and flavonoid($634.2{\pm}2.33{\sim}1,324.6{\pm}1.87$ mg/100 g) contents were higher in the methanol extract than in the water extract. Also, within a reaction system, nitrite scavenging ability and NDMA inhibition were most effective at pH 2.5, and increased in proportion to the extract concentration. Finally, the methanol extract of O. japonicus A Berger harvested during $August{\sim}October$ had the highest nitrite scavenging and NDMA inhibition effects.

The water extract of the aerial parts of Orostachys japonicus A. Berger showed the inhibitory activity against HIV-1 protease. From the same parts of O. Japanicus, 4-hydroxybenzoic acid, 3,4-dihydroxybenzoic acid, gallic acid and methyl gallate, together with flavonoids, kaempferol, quercetin, kaempferol $3-O-{\beta}-D-glucoside$, kaempferol $3-O-{\beta}-D-galactoside$ and quercetin $3-O-{\beta}-D-glucoside$ were isolated and characterized by spectral data.
Methanol extracts of Orostachys japonicus A. Berger (OAB) were found to exhibit apoptosis induction of HL60 human acute promyelocytic leukemia cells. Treatment of OAB exerted strong cytotoxicity against HL60 cells. OAB induced DNA fragmentation of HL60 cells in a dose dependent manner. Nitric oxide production were also increased in OAB-treated RAW264.7 macrophage cell lines. Treatment of OAB increased the expression of p53 and iNOS gene and the expression of p53, $NF-{\kappa}B$ and iNOS protein in cultured HL60 and RAW264.7 cells. These results suggest that OAB are effective on strong anti-cancer properties and can be useful as a chemo-preventive agents.
본 연구에서 사용된 와송(Orostachys japonica A. Berger)은 암 치료를 위한 민간요법으로 오랫동안 사용되어 온 약초이다. 본 연구에서는 와송(Orostachys japonica A. Berger) 에틸아세테이트 분획물의 항산화 효능 활성에 대하여 실험한 결과를 보고한다. 와송(Orostachys japonica A. Berger) 에틸아세테이트 분획물의 항산화 효능은 0.10 mg/mL 농도에서 78.54%의 DPPH 라디칼 소거활성을 보였고, 73.48%의 ABTS+ 라디칼 소거활성을 나타내어 항산화 효과가 있는 것을 확인하였다. 또한 대체실험동물 모델인 제브라피쉬를 이용한 독성실험 결과에서는 모든 농도에서 100%의 배아 생존율을 보이고 응고 또는 부화지연을 관찰할 수 없어 독성이 없는 것으로 나타났으며, UV-B조사에 의해 유도된 ROS생성은 와송 에틸아세테이트 분획물을 처리한 모든 larvae에서 전체적으로 형광강도가 감소하였다. 특히 3 ㎍/mL 농도에서 양성대조군 대비 35.7%의 감소율을 보여 효과적으로 ROS 생성을 억제하였음을 확인하였다. 위와 같은 결과로 미루어보아, 와송 에틸아세테이트 분획물은 천연 항산화제로서 화장품 분야에서 응용 가능성이 있을 것으로 기대한다.
본 연구에서 와송 에틸아세테이트 분획물을 항균활성 및 화장품 소재로 활용하기 위해 연구를 진행하였다. 와송 에틸아세테이트 분획물의 항균활성을 측정한 결과 Paper disc method에서 S. aureus, S. epidermidis, P. aeruginosa 균의 생육을 저해하였으며, 특히 S. aureus에 대한 생육저해환은 0.5 g/mL농도에서 18.35 ± 1.5 mm로 대조군인 methyl paraben(16.83 ± 1.0 mm)보다 우수한 항균활성을 나타내었다. MIC 측정을 통한 최소저해농도를 평가에서는 Candida. A를 제외한 나머지 S. aureus, S. epidermidis, E. coli P. aeruginosa 균주에서 17.5 mg/mL의 최소저해농도를 보였으며 화장품 에멀션 제형에 적용하여 Challenge test를 통해 방부 효능을 평가한 결과, S. aureus에 대하여 와송 에틸아세테이트 분획물을 0.3% 첨가한 에멀션에서 7일 후 100.0%균의 생장을 사멸시켜 화장품에 적용 시 우수한 방부효능을 나타내었다.
The hypolipidemic and hypoglycemic effects of two dietary dosages (0.1% and 0.5%) of water and 80% ethanol extracts from hot-air dried Orostachys japonicus A. Berger were evaluated in the serum and organ tissues of streptozotocin-induced diabetic rats. The STZ-induced diabetic groups supplemented with the O. japonicus extracts showed significantly higher body weight compared to a diabetic control group at the end of experiment. The extracts exhibited substantial hypoglycemic effects by significant reductions of fasting blood glucose levels at all time points tested compared to the initial stage before treatment of the extracts. Declines of serum and hepatic triglyceride levels were greater than declines of total cholesterol in the groups treated with the 0.5% O. japonicus extract (DBW2 and DBE2) when compared to the DBC group. Hepatic glycogen content was higher in the groups treated with O. japonicus extract, while lipid peroxide content was decreased in these treated groups compared to the DBC group. Hepatic antioxidant activity was significantly increased in the groups supplemented with the O. japonicus ethanol extract The hypolipidemic and hypoglycemic effects of the O. japonicus ethanol extract were significantly greater than the effects of the water extract. Based on this study, it seems that O. japonicus ethanol extract, due to its higher phenolic and flavonoid components than the water extract, may control blood glucose and alleviate hyperlipidemia in diabetes.
