Construction of a Genetic System for Streptomyces albulus PD-1 and Improving Poly(ε-ʟ-lysine) Production Through Expression of Vitreoscilla Hemoglobin

  • Poly(ε-ʟ-lysine) (ε-PL) is a novel bioactive polymer secreted by filamentous bacteria. Owing to lack of a genetic system for most ε-PL-producing strains, very little research on enhancing ε-PL biosynthesis by genetic manipulation has been reported. In this study, an effective genetic system was established via intergeneric conjugal transfer for Streptomyces albulus PD-1, a famous ε-PL-producing strain. Using the established genetic system, the Vitreoscilla hemoglobin (VHb) gene was integrated into the chromosome of S. albulus PD-1 to alleviate oxygen limitation and to enhance the biosynthesis of ε-PL in submerged fermentation. Ultimately, the production of ε-PL increased from 22.7 g/l to 34.2 g/l after fed-batch culture in a 5 L bioreactor. Determination of the oxygen uptake rate, transcriptional level of ε-PL synthetase gene, and ATP level unveiled that the expression of VHb in S. albulus PD-1 enhanced ε-PL biosynthesis by improving respiration and ATP supply. To the best of our knowledge, this is the first report on enhancing ε-PL production by chromosomal integration of the VHb gene in an ε-PL-producing strain, and it will open a new avenue for ε-PL production.

Mechanical Properties of Polypropylene/Talc Composites Prepared via Solid-State Extrusion (고상 압출된 폴리프로필렌/탈크 복합재료의 기계적 물성)

  • We investigated the specific gravity and mechanical property changes of solid-state extruded polypropylene (PP)/talc composites before and after orientation. The specific gravity of the composites increases with increasing the filler contents. The specific gravity of the oriented specimen containing filler in PP matrix is found to be much smaller than that of pre-specimen due to the formation of more micro-voids. It was found that the tensile properties of the composites are increased up to the talc content of 10 wt%, but after the contents exceeding 10 wt%, the tensile properties are decreased. For oriented specimens, the tensile strength of the composites showed monotonously decrease with increasing talc contents. When the contents of talc is 10 wt%, the theoretical values according to Halpin-Tsai equation are close to the experimental values but over 20 wt% of talc contents, the deviation of the experimental values from the theoretical prediction becomes higher. The maximum flexural strength and modulus were observed for PP/talc composites when the talc contents was 10 wt% for both pre-specimen and oriented specimen.

Barrier Property and Morphology of Biaxially Oriented PP/EVOH Blend Film (이축연신 PP/EVOH 블렌드 필름의 차단 성질 및 모폴로지)

  • In this study, biaxially oriented film process was used to improve barrier property of polypropylene (PP)/ethylene-vinyl alcohol copolymer (EVOH) blends by inducing a laminar morphology of the dispersed phase in the matrix phase. In order to examine the extent of deformation during melt extrusion process, the rheological properties of the resins were measured and the viscosity ratio of the dispersed phase to the continuous phase was determined. The effects of compatibilizer content, draw ratio, and draw temperature on the oxygen permeability and morphology of biaxially drawn blend films were studied. The laminar morphology of the EVOH phase with a larger area of thinner layer induced by biaxial orientation was found to result in a significant increase in oxygen barrier property of PP/EVOH (85/15) blends by about 10 times relative to the pure PP When both PP-g-MAH and ionomer were used as the compatibilizers, there existed an optimum level of compatibilizer content for obtaining improved barrier properties with a well developed laminar structure. In addition, higher draw ratio and draw temperature were found to be more favorable processing conditions in obtaining higher barrier blends.

Changes of Thin Film Coating on Polymer Lenses with Varying Temperature (온도에 의한 고분자 렌즈의 재질별 코팅 박막의 변화)

  • Purpose: To observe changes of coatings and lens materials with varying temperature to understand effect of temperature on plastic lens. Methods: In this study, three lenses of different refractive indices (2 of thiourethane oriented lenses, an allyl diglycol carbonate oriented lens) were exposed to high temperature (50, 80, and 100 degree) for 5 hours and changes of individual coating (anti-refractive coating, hard coating, and water repellent coating) were measured. Results: As a result, high-refractive index lenses did not exhibit significant variation of hardness. However, hardness of mid-refractive index lens were decreased when exposed to high temperature and destructions of hard coating layer was inferred. Surface contact angles of lens were decreased with increasing temperature and water repellent coating layer were damaged at higher than 80 degree. Conclusions: Multi including water repellent coatings on all three lenses with different refractive indices were damaged when exposed to at or higher than 80 degree. The degree of changes in mechanical and physical properties were depended on polymer material type.

Effects of E-beam treatment on the interfacial and mechanical properties of henequen/polypropylene composites

  • In the present study, chopped henequen natural fibers without and with surface modification by electron beam (E-beam) treatment were incorporated into a polypropylene matrix. Prior to composite fabrication, a bundle of raw henequen fibers were treated at various E-beam intensities from 10 kGy to 500 kGy. The effect of E-beam intensity on the interfacial, mechanical and thermal properties of randomly oriented henequen/polypropylene composites with the fiber contents of 40 vol% was investigated focusing on the interfacial shear strength, flexural and tensile properties, dynamic mechanical properties, thermal stability, and fracture behavior. Each characteristic of the material strongly depended on the E-beam intensity irradiated, showing an increasing or decreasing effect. The present study demonstrates that henequen fiber surfaces can be modified successfully with an appropriate dosage of electron beam and use of a low E-beam intensity of 10 kGy results in the improvement of the interfacial properties, flexural properties, tensile properties, dynamic mechanical properties and thermal stability of henequen/polypropylene composites.

Detailed Investigation on Factors Governing Liquid Crystal Alignment on Rubbed Polystyrene Films

  • The molecular reorientations and surface morphologies of rubbed films formed from atactic polystyrene (PS) samples with various molecular weights were investigated in detail. Previously unknown surface topography features were newly discovered in rubbed films, depending on molecular weights: submicroscale groove-like meandering structures composed of fine-grooves like pebbles in tens nanometers are present, oriented perpendicular to the rubbing direction. The vinyl main chains, however, were preferentially reoriented along the rubbing direction and the planes of the phenyl side groups were preferentially reoriented perpendicular to the rubbing direction with para-directions that were positioned nearly normal to the film plane. Nematic liquid crystal (LC) molecules were found to always align on the rubbed PS surfaces along the orientation direction of the submicroscale grooves generated by rubbing.

Homopolymer Distribution in Polystyrene - Poly(methyl methacrylate) Diblock Copolymer (폴리스티렌-폴리(메틸 메타크릴레이트) 이종 블록 공중합체 내의 단일중합체 분포)

  • Homopolymer distribution in block copolymer/homopolymer blends was investigated as a function of homopolymer concentration and homopolymer molecular weight. The deuterated poly(methyl methacrylate) or polystyrene was blended with a deuterated polystyrene-poly(methyl methacrylate) diblock copolymer up to a concentration of 20 wt%. Samples were characterized by small-angle X-ray scattering (SAXS), neutron reflectivity and transmission electron microscopy. The block copolymer with a thin-film geometry formed alternating lamellar microdomains oriented parallel to the substrate surface. By adding the homopolymer, the microdomain structure was significantly disturbed. As a consequence, a poorly ordered morphology appeared when the homopolymer concentration exceeded 15 wt%. Increasing the homopolymer concentration and/or the homopolymer molecular weight caused the microdomains to swell less uniformly, resulting in segregation of the homopolymer toward the middle of the microdomains.

The Studies of the Water-Vapour Transmission Rate and It's Proofness on the Various Commercial Polymer Films (상업용 고분자 필름의 투습속도 및 방습도에 관한 연구)

  • The water-vapour transmission ratios (WVTR) of the various commercial polymer films have been investigated at the constant pressure and relative humidity (RH). Water proofnesses, the reciprocals of WVTR for the various samples, were determined using a cup device and maintaining the sample films at a constant temperature ($40{\pm}1^{\circ}C$) and a constant R. H ($90{\pm}2%$) for 24 hours. The following order of the relative proofness was observed; oriented polypropyrene (O.PP) > high density polyethylene (HDPE, Inflation) > high density polyethylene (HDPE. T-die) > casted polypropylene (C. PP) > nonoriented polyester (N. PET) > low density polyethylene (LDPE) > oriented polyester (O. PET) > rigid polyvinyl chloride (Rigid PVC) > semirigid polyvinyl chloride (Semirigid PVC) > nonrigid polyvinyl chloride (Nonrigid PVC) > oriented nylon (O. Nylon) > nonoriented nylon (N. Nylon). And water proofness order was also observed to decrease with the temperature rising; HDPE (T-die) > C. PP > O. PET > LDPE > O. Nylon. The activation energies of LDPE, HDPE (T-die), C. PP, O. PET and O.Nylon films were 12.0, 11.1, 11.4, 11.7, 14.1 kcal/mole, respectively. The WVTR's of the films were increased with the polarity of polymer and the addition of plasticizer in PVC, decreased with the increase of the film thickness and mechanical orientation. The WVTR's of the laminated films O. PP/LDPE, N.Nylon/LDPE, C.PP/LDPE were also more dependent on the film thickness than the WVTR's of the single films.

Supramolecular Nanodevice Based on Helical Encapsulation of Photo-/Electroactive Oligomers

  • Photo-/electroactive donor-acceptor (D-A) chromophores were synthesized and investigated for energy- and electron-transfer (ET/eT) properties, for which the chromophores are supramolecularly integrated by encapsulation with helical amylose, rendering the chromophores aggregation-free and rigidified along the helical axis and thus, a remarkably enhanced fluorescence intensity. Fluorescence quenching studies indicated that the helical encapsulation gives the ET/eT a clear D-A distance dependence unlike with the encapsulation-free counterparts, being reflected in their florescence decay profile. Another notable difference is that the helical supramolecule of the chromophores forms a perpendicularly oriented self-assembly. Transport behavior in the solid state will be also discussed.

Bending and Pressing Tolerance of Flexible Polyoxetane based Liquid Crystalline Polymer/Low Molecular Weight Liquid Crystal Device

  • We have studied the realigning behavior of liquid crystal molecules in liquid crystalline polymer/liquid crystal(LCP/LC) system when they are exposed to external stimulation such as bending and pressing. The birefringence of the LCP/LC in a flexible display device was measured as a function of bending or pressing deformation. The microscopic dynamic behavior of main chain, side chain, and the LC were characterized by FTIR and polarization optical microscopy. When the device is deformed in scattering memory state, liquid crystal(LC) director is found to align from randomly oriented domain state(scattering state) to homeotropic state.
