Pesticide Residue Survey and Estimate Intake Amount of Vegetables in Noeun Wholesale Market, Daejeon (대전시 노은 도매시장 채소류의 농약 잔류 실태 및 식이섭취량 추정)

  • Pesticide residues in some vegetables collected at Noeun wholesale market in Daejeon were surveyed and assessed their risk. In 100 samples, the detection rate of pesticide was 46.0% and the rate exceed MRL was 6.0%. Commodities showing high detection rate were lettuce(85.0%), perilla leaf(80.0%) and cucumber(60.0%). Dicarboximide, organochlorine, and azole fungicides and organophosphorus and pyrethroid insecticides were detected. Detection frequency of pesticide was in the order of procymidone, chlorpyrifos, chlorothalonil, cypermethrin and EPN. When the estimated intake amount of the pesticides were compared with ADI to assess their risk, bitertanol, triflumizole and iprobenphos in perilla leaf were higher rate than the other vegetables. However the total amounts of intake of pesticides were estimated to less than 0.46% of ADI.

Inhibition of acetylcholinesterase activity by impurities in technical grades and purified flupyrazofos (Flupyrazofos 원제 및 정제품의 불순물 조성과 Acetylcholinesterase에 대한 저해 비교)

  • Flupyrazofos (O,O-diethyl O-1-phenyl-3-trifluoromethylpyrazo-5-yl phosphorothioate) is an organophosphorus insecticide with a pyrazole moiety which is newly developed and commercialized by SUNGBO chemical company and Korean Research Institute of Chemical Technology for effectively control against diamond back moth. This study was conducted to determine the composition and quantity of impurities in technical 1 (94.5%), technical 2 (97.6%) and purified (99.2%) flupyrazofos using GLC/MSD. Bimolecular inhibition rate constant($k_i$) with acethylcholinesterase (in vitro) and $I_{50}$ with mouse brain acetylcholinesterase (in vivo) were measured for comparing inhibitory patterns of two technicals and purified flupyrazofos. Impurities of flupyrazofos were identified as O,O,O-triethylthio-phosphoric acid (TEA), 1-phenyl-3-trifluoromethyl-5-ethoxy pyrazole(PTMEP), O,O-diethyl O-1-phenyl-3-trifluoromethylpyrazo-5-yl phosphoric acid ester(flupyrazofos oxen), O,S-diethyl O-1-phenyl-3-trifluoromethylpyrazo-5-yl phosphorothionate (S-ethyl flupyrazofos). In in vitro, technical 1 showed the fastest inhibition on AChE activity among them. And technical 1 and 2 showed 40% higher in vivo inhibition against mouse brian AChE than purified flupyrazofos did. These results could be caused by the impurities such as flupyrazofos oxen and S-methyl flupyrazofos contained in technical grades of flupyrazofos.

ELISA Development for the residue of the organophosphorus insecticide acephate (ELISA에 의한 유기인계 살충제 Acephate 잔류물 분석법 개발)

  • A competitive indirect enzyme-linked immunosorbent assay (ci ELISA) for the organophosphorus insecticide acephate, O,S-dimethyl acetylphosphoramidothioate, was developed using a polyclonal antibody. Three different haptens mimicking the analyze and containing hexanoic acid moiety as a linker were synthesized, and then conjugated with the carrier proteins bovine serum albumin and keyhole limpet hemocyanin by the N-hydroxysuccinimide active ester method. Polyclonal antibodies raised against hapten-KLH conjugates in rabbits and the hapten-BSA conjugates as coating antigens were screened and selected for the assay in the homologous and/or heterologous ELISA system. The effects of various assay conditions, including blocking reagents, detergent content, organic solvents, pH, and preincubation of tile mixture of the polyclonal antibody and the analyze on the sensitivity were evaluated. The $IC_{50}$ value of acephate of 110 ng/mL was obtained in an optimized heterologous system using hapten-3-BSA as a coating antigen and a polyclonal antibody 8377, showing the detection range of 10-1000 ng/mL and the lowest detection limit of 4 ng/mL. The cross-reactivities of the structurally related insecticides, including methamidophos were less than 0.02%. These results indicate that the ELISA could be a convenient and alternative tool for monitoring acephate residues in agricultural products and environmental samples.

Extraction Efficiencies of Organophosphorus Pesticides Spiked in Fish Tissues by Supercritical Carbon Dioxide (초임계 이산화탄소에 의한 어류조직 중 유기인계 농약의 첨가회수율)

  • Fish tissues were spiked with organophosphorus pesticides (OPPs), mixed with a celite as a drying agent, and dynamically extracted with pure $CO_2$ or modified $CO_2$ for 10 min at different extraction temperatures, pressures and $CO_2$ flow rates. Recoveries of OPPs spiked in jacopever increased with the decrease of extraction temperature and pressure, and decreased with the increase of $CO_2$ flow rates. Modified $CO_2$ extractions with 10% methylene chloride showed a slight increase in the recoveries over pure $CO_2$ extraction. Quantity of fish tissues had great effect on their extraction efficiencies. Recoveries of OPPs were $66.7{\sim}86.3%$ for jacopever, $56.2{\sim}79.2%$ for yellow tail, $57.6{\sim}77.8%$ for blanquillo, $84.2{\sim}96.3%$ for sardine, $74.6{\sim}83.6%$ for mackerel. Application of supercritical carbon dioxide extraction offers an attractive alternative to the use of organic solvents for extraction of pesticide residues from fish tissues.

A Study of Current Status on Pesticide Residues in Commercial Dried Agricultural Products (서울시 유통 건조농산물 중의 농약잔류 실태 연구)

  • The purpose of this study was to investigate 253 kinds of pesticide residues in 58 commercial dried agricultural products in Seoul. The determinations of the pesticide residues were performed using multiresidue methods and were carried out by a gas chromatography-nitrogen phosphorus detector (GC-NPD), an electron capture detector ($GC-{\mu}ECD$), a mass spectrometry detector (GC-MSD) and high performance liquid chromatography-ultraviolet detector (HPLC-UV), and a fluorescence detector (HPLC-FLD). The pesticide residue detection rate in the commercial dried agricultural products was 24.1% (14 of 58 samples). Twelve pesticide residues without maximum residue limits (MRLs) were detected. In the vegetable groups, the frequency of pesticide residues was found to be in the increasing order of dried fruiting vegetables > dried leafy vegetables > dried stalk and stem vegetables. The pesticides used on dried red pepper in the dried fruiting vegetables were varied (7 kinds) and numerous (4 of 8 samples). The pesticide types detected in the commercial dried agricultural products were in the order of pyrethroid > organochloride > organophosphorus and insecticide > fungicide > herbicide ${\cdot}$ nematicide. The primary pyrethroid pesticide detected was cypermethrin. According to the producing areas of products, large numbers of pesticide residues were found in the order of Korea, China, North Korea, USA, and Vietnam.

Monitoring on Insecticide resistance of major insect pests in paddy field (주요 벼 해충에 대한 약제저항성 모니터링)

  • This study was carried out for looking into the status of susceptibility of pest insects to insecticides. Each insect Brown planthopper(BPH), Green leaf hopper(GLH), Smaller brown plant hopper(SBPH), Rice water weevil(RWW), were captured at various areas where the host crops were being cultivated and the susceptibility level of each pest insect was investigated. The susceptibility of each pest insect varied by insect species and areas where they were caught. BPHs kept higher level of susceptibility comparing to susceptible reference strain except to most of tested insecticides except imidacloprid (Resistance ratio was 68). The susceptibilities of GLH and SBPH to most of insecticides for their control did not developed markedly since 1976 except fipronil and imidacloprid which is widely used for WRR control. The insecticides used for control of WRR were very effective even at the concentration of one fourth of recommending concentration, but in 2000 suwon strain of WRR showed markedly reduction of susceptibility to carbofuran.

  • The metabolites of agrichemicals, such as organophosphorus pesticides, are known to be more hazardous than their parent pesticides. 3,5,6-trichloro-2-pyridinol (TCP) is a major degradation product of chlorpyrifos, one of the organophosphate insecticides widely used in agriculture. In vivo or in vitro exposure to chlorpyrifos has been known to interfere with male reproductive functions, leading to reduced fertility in mammals. Therefore, this study was performed to examine the changes in the fertilization competence of boar spermatozoa exposed to TCP. Sperm samples were subjected to varying concentrations of TCP (10, 50, 100, 200 µM) and different periods of incubation. Sperm motility, motion kinematics, viability, acrosome integrity, intracellular reactive oxygen species (ROS) production, and gene expression levels (ODf2, ZPBP2, AKAP3 and AKAP4) were evaluated after exposure of the sperm to TCP. A significant dose-dependent reduction in motility was observed in sperm samples incubated with TCP compared to the controls after both incubation periods. Sperm viability was significantly decreased in samples incubated with 50, 100, and 200 µM TCP in both incubation periods. A significantly lower percentage of normal acrosomes and gene expression levels were observed in sperm samples exposed to 50, 100, and 200 µM TCP after both incubation periods, compared to the controls. There was a significant increase in the ROS production in spermatozoa incubated with 100 - 200 µM TCP after both incubation periods. Consequently, the direct exposure of boar spermatozoa to TCP interferes with sperm functions and leads to decreased fertilization. In order to identify and address the various causes of reproductive decline, the impact of chemical metabolites needs to be discussed in depth.

Establishment of Simultaneous Analysis Method for the Detection of Multi-Pesticide Residue Used in Golf Courses (골프장 농약 검사를 위한 다성분 동시분석방법 확립에 관한 연구)

  • The possibility of multiresidue analysis of 24 pesticides out of 30 residual pesticides which are subjected to test in the golf courses was examined. The utility of multiresidue method for pesticide residue test was evaluated by recovery test through a standard addition method of pesticides in water, soil, and lawn grass. The experimental results of the recovery test for individual pesticides are as follows : The number of pesticide of which average recovery rate was over 70% regardless of media was 16 pesticides. These pesticides were composed of 8 organophosphorus pesticides (chlorpyrifos, chlorpyrifosmethyl, diazinon, EPN, fenitrothion, phenthoate, phosalone, and toclofos-methyl). 4-organochlorinated pesticides (daconil, captan, endosulfan, and tetradifon), 2-pyrethroid pesticides(fenpropathrin, lambda-cyhalothrin) and 2 other pesticides (bromopropylate, pendimethalin). On the other hand, in case of dicofol, average recovery rate was over 70% for water and lawn grass but only 53.3% for soil. Therefore, the multiresidue method applied in this experiment is not appropriate for analysis of dicofol in soil. Furthermore, among 7 pesticides, 2 pesticides(amitraz and pyraclofos) showed that theirs average recovery rate deviated from criteria($70{\sim}130%$) in almost ail media, while 5 pesticides(bensulide, deltamethrin, iprodione, phosphamidon and tralomethlin) were not detected from all media by GC/NPD or GC/ECD.

Effect of Parathion on Hematological Parameters in the Serum of a Male Bagrid Catfish (Pseudobagrus fulvidraco )

  • To evaluate the impact of parathion on aquatic organisms, a freshwater bagrid catfish (Pseudobagrus fulvidraco) was exposed to sublethal concentrations $(63,\;95,\;190\;and\;380\;{\mu}g/L)$ of parathion, organophosphorus pesticide for 30 days. Glucose level in the serum of the bagrid catfish was significantly increased than that of control groups in the $190\;{\mu}g/L$ concentration at 30 days and in the $380\;{\mu}g/L$ concentration after 10 days. Bilirubin level was significantly increased in the $190\;{\mu}g/L$ concentration at the end of the experiment. After 10 days, a significant differences of lactate dehydrogenase (LDH) and alkaline phosphatase (ALP) activity increased in the ${\geq}190\;{\mu}g/L$ and $380\;{\mu}g/L$ groups. Though cholesterol concentration was stable, aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities in serum were significantly reduced in the $190\;{\mu}g/L$ at 10 days and in the $380\;{\mu}g/L$ concentration at the end of the experiment. In conclusion, if P. fulvidraco was chronically exposed to ${\geq}190\;{\mu}g/L$ concentration of parathion, the hematological changes may be induced due to the stress response.
