백굴채가 마우스의 세포성 및 체액성 면역반응에 미치는 영향 (The Effects of Chelidonii Herba Extract on the Cell Mediate and Humoral Immune Responses in Mice)

  • The effects of Chelidonii Herba on the cell mediate and humoral immune responses were investigated in ICR mice. ICR mice received intraperitoneal injection of methanol extracts (ether fraction, buthanol fraction, water fraction) for 5 days before sensitization. The change of body weight (%), the organ weight ratio (%), the numbers of RBC and WBC, the ear swelling by dinitrofluorobenzene, the titers of hemagglutination (HA) and hemolysin (HY) to SRBC were determined. The results were as follows; 1. The change of body weight (%) showed a tendency of increasing generally. 2. The weight of spleen (%) revealed decreasing but that of thymus revealed increasing. 3. Ear swelling was maxium at 48 hours after challenge and was significantly decreased in the groups treated with Ether, Buthanol and Water. 4. The numbers of RBC and WBC were decreased generally. 5. HA and HY titers were decreased significantly.

  • The effects of Asiasarum Root on the cell mediated and humoral immune responses were investigated in ICR mice.   • Certain hexavalent chromium compounds when administered via inhalation have the potential to induce lung injury in human and experimental animals. In present study, the inhalation effect of hexavalent chromium on morphological change and weight change of rat organ were investigated. Rats were exposed to hexavalent chromium ($Na_2CrO_4{\cdot}4H_2O$) at concentration of $0.36mg/m^3$ (group 1), $1.8mg/m^3$ (group 2), ascorbic acid and $1.8mg/m^3$ (group 3) and filtered air (group 0, control group) for I week, 2 weeks and 3 weeks. The weight of lung and kidney in group 2 and group 3 significantly higher than in control group at same exposure period. The epithilial cells of bronchiole in group 1, 2, 3 were more flatten than group 0. In the lung, the number of macrophage was significantly increased and morphologically changed macrophages were observed in group 1, 2, 3. The morphological change of the lung did not significant between group 2 and group 3, however, in group 1 was milder than in group 2 and group 3. The severity of morphological change were depend on exposure period in the lung. The morphological changes by hexavalent chromium of the liver and kidney were also observed These results suggest that inhalation of hexavalent chromium effects on not only respiratory organ, but also the liver and the kidney via blood stream.

단치소요산(丹梔逍遙散)의 항(抗)스트레스 효과(效果)에 대한 실험적(實驗的) 연구(硏究) (The Experimental Studies on the Anti-Stress Effects of Danchisoyosan(丹梔逍遙散))

  • This study was aimed to evaluate the anti-stress effect of danchisoyosan on the rats stressed by immobilization. The experimental animals were immobilized in the stress box($5{\times}5{\times}20cm$) for 12 hours in a day during 3 days, and administered $500mg/5m{\ell}/g$ of Danchisoyosan extract for 14 days before stress. There were measured the change of body weight and organ weight under immobilized-stress. The norepinephrine, epinephrine, dopamine, serotonine contents were measured by HPLC method in rat brain. There were measured the GOT, GPT contents in serum and tissue lipid peroxidation in the brain, liver, spleen, adrenalgland, pancreas, testes, thymus, heart. The following results were obtained: 1. The change of organ weight was significantly lower in control than normal group. Sample group inhibited decreased weight from stress comparing to control group. 2. Lipid peroxidation in the liver was significantly higher in control than normal group. Sample group shows significant decrease comparing to control group. 3. Lipid peroxidation in the kidney was significantly higher in control than normal group. Sample group shows significant decrease comparing to control group. 4. GPT contents in serum was significantly higher in control than normal group. Sample up shows significant decrease comparing to control group. 5. Dopamine contents in the brain was significantly higher in control than normal group. Sample group shows significant decrease comparing to control group. 6. Serotonine contents in the brain was significantly higher in control than normal group. Sample group shows significant decrease comparing to control group.

방풍갈근탕(防風葛根湯)의 안전성에 관한 연구 (Study on Safety of Bangpung-galgeun-tang)

  • The purpose of this study was to examine to observe single and four weeks repeated toxicity in mice of Bangpung-galgeun-tang (BGT). We investigated to ascertain safety and toxicity of BGT, we divided into single and four weeks repeated administration test. In single test, three groups were administrated different dosages and routes (2 g/kg/i.p., 4 g/kg/i.p. and 15 g/kg/p.o.) of BGT, and in four weeks repeated test, 0.8 g/kg BGT was administrated. Control groups were administrated with only saline according to on Korean Food and Drug Administration, respectively. We observed attentively motality, abnormal clinical sign, body weight change, organ weight, AST and ALT of mice after BGT administration. During toxicity experiment period, there was no difference in body weight change, organ weight, AST and ALT among different dose groups. Death were found 3 mice from day 2 to day 3 in single test i.p. group. (2 g/kg, 4 g/kg). Several individuals of single test i.p. group were observed that decreased locomotor activity, exophthalmos, bloodshot eyes, loss of eyesight and so on in early period after administration. But there was no difference in clinical signs among p.o. group. These results indicate that BGT have inhibition effects on allergy and suggest that no observable effect level of the test orally administration was considered to be more than 2 g/kg in mice under the conditions employed in this study.

Single Oral Dose Toxicity Study of Pinelliae Rhizoma Aqueous Extract in ICR Mice

  • This study was conducted to obtain acute information of the oral dose toxicity of lyophilized water extract of Pinelliae Rhizoma, a dried tuber of Pinellia ternata (PR) in male and female mice. In order to calculated 50% lethal dose (LD$_{50}$) and approximate lethal dose (ALD), test material was once orally administered to male and female ICR mice at dose levels of 2000, 1000, 500, 250, 125 and 0 (vehicle control) ml/kg (body weight). The mortality and changes in body weight, clinical signs, gross observation, organ weight and histopathology of principle organs were monitored 14 days after treatment with PR extract. We could not find any mortalities, clinical signs, changes in the body and organ weights, gross and histopathological findings except for dose-dependent increases in the hepatic fatty change frequencies detected in PR extract 2000 and 1000mg/kg treated in both male and female mice. The results obtained in this study suggest that LD$_{50}$ and approximate LD in mice after single oral dose of PR extracts were considered over 2000 mg/kg in both and female male mice, but more than 1000mg/kg of PR extracts treatment could induce slight hepatotoxicity the fatty changes in mice.

일측폐장절제가 잔류폐, 간 및 신장의 대상성 증식과 혈청전기영동상에 미치는 영향 (Effect of Unilatromral Pneumonectomy on the Compensatory Growth of the Residual Lung, Liver, and Kidney, ana Serum Electrophoresis Pattern)

  • In order to clarify the effect of the unilateral pneumonectomy on the compensatory growth of the residual lung, liver and kidney, and serum electrophoresis pattern, right lung pneumonectomy was performed on rabbits under general anesthesia with pentobarbital sodium. On the fifth day after the surgery, the weight of the residual lung, liver and bilateral kidneys was measured and organ weight-body weight ratio was calculated. And in an attempt to know whether the cells in the liver and the kidney were proliferated by unilateral pneumonectomy, DNA content was determined. The quantity(g/100 ml) of serum protein was determined also and serum electrophoresis was performed on cellulose acetate membrane. The results obtained are summarized as following. The weight of the residual lung and lung weight-body weight ratio was significantly increased respectively. The weight of the liver and organ weight-body weight ratio were not changed but the DNA content of the liver and kidney tissue increased significantly, illustrating that unilateral pneumonectomy caused cellular hyperplasia in the liver and in the kidney as well as in the residual lung. The quantity(g/100 ml) of serum protein was significantly increased and in the analysis of the electrophoregram, there was significant difference between the normal and pneumonectomy group. Taken together, these results indicate that unilateral pneumonectomy caused the compensatory hyperplasia of the liver, the kidney and the residual lung as well as the change of electro-phoretic pattern. And it also suggests that a humoral factor, which proliferates the cells in the residual lung, the liver and the kidney, existed in the pneumonectomized rabbits.

The toxicity of Aceporol 460 as a novel high loading capacity solubilizer of paclitaxel

  • Previously, we reported a novel polymeric micellar solubilizer, Aceporol 330, that showed relatively low toxic effects when it was compared with that of Cremophor EL which is currently being used for paclitaxel. In this study, we have developed a new micellar solubilizer, Aceporol 460, that has 3-4 times higher loding capacity for paclitaxel than Aceporol 330. The single-dose and the repeated-dose toxicity of Aceporol 460 were evaluated in ICR mice. For single dose toxicity test, male and female mice were randomly assigned to one of five study groups to receive, and injected intravenously with dosages of 0, 3, 4mL Cremophor EL/kgbody weight, and 3, 4mL Aceporol 460/kg body weight, respectively. In both male and female mice, LD50 for Aceporol 460 can not he determined even at the maximal administrable dosage, 4mL/kg due to the high viscosity of chemical and there was no significant change in body weight, hematological and serum biochemical analysis, organ weight, and histopathological examination compared with that of Cremophor EL. For the repeated dose toxicity test, male and female mice were given the dosage of 0, 1.6mL Cremophor EL/kgbody weight/day, and 1.6mL Aceporol 460/kg body weight/day for 2 weeks. Results of repeated dose toxicity tests for 2 weeks suggested that Aceporol 460 treated group show no significant toxicological findings with body weight, hematological and serum biochemical analysis, organ weight, urinalysis, and ophthalmoscopic and histopathological examination compared with that of Cremophor EL. These results indicate that Aceporol 460 have higher paclitaxeL-loading capacity than Aceporol 330 and less toxic effects than Cremophor EL in male and female mice.

산화네오디뮴(Nd2O3) 기도투여에 따른 흡입독성 (Pulmonary Toxicity in Rats by Intratracheal Instillation with the Rare-Earth Metal Neodymium Oxide)

  • Objectives: This study was performed to produce data on the pulmonary toxicity of neodymium oxide($Nd_2O_3$) by intratracheal instillation. Methods: Two groups of rats were exposed to neodymium oxide by intratracheal instillation with doses of 0.5 mg and 2.0 mg, respectively. At two days, four weeks and 12 weeks after exposure, body weight change, organ weight change and histopathological change were observed. At 12 weeks after exposure, lung function change was measured. Results: The body weight of rats in the high concentration group decreased after 12 weeks by 4-5% compared with the control group. At four weeks and 12 weeks after the administration of neodymium oxide, the absolute weight of the lungs of the high concentration group were significantly increased when compared with the control group(p<0.05). At 12 weeks after the injection of neodymium oxide, breath frequency and respiratory minute volume were increased, but inhalation time and expiratory time were decreased. Bronchiolar epithelial hyperplasia, alveolar type II cell hypertrophy/hyperplasia and foreign body granulomatous inflammation were observed in the high exposure group. Conclusions: Body weight decrease, lung absolute weight and breath frequency increase, and pathological lung change were all observed. We found that pulmonary toxicity of neodymium oxide nanoparticles by intratracheal instillation could be confirmed.

십이미관중탕(十二味寬中湯)이 Wister rat의 노화에 따른 변화에 미치는 실험적(實驗的) 고찰(考察) (Studies of SipIMiGwanJungTang that get weight, hematology, biochemistry change by Wistar rat's aging)

  • Administrating PalMoolGoonJaTang and SipIMiGwanJungTang to 40-weeks-(old Wistar rat for 10 weeks so, I researched into weight change, weight change of internal organ, and hemtological and serological changews. Then I got these conclusions. 1. Observed gain in weight than control group form of SipIMiGwanJungTang to aged Wistar rat. 2. Is thought to promote activation of living body action gaining intestine weight along with gain in weight. 3. Displayed decrease of MDA's content of serum than control group form of SipIMiGwan JungTang to aged Wistar rat. 4. Change that is Wistar rat's hematological value by aging according to 10, 30, 50 week WBC, RBC, Hgb, monocytes, eosinophil etc. increase, and HCT, PLT etc. showed tendency that decrease according to old-week, and observed improvement that is hematological value than control group form of SipIMiGwanJungTang 5. Change that is Wistar rat's biochemical value by aging was measured highest in 50 week because ALT, AST, BUN, CRN, T-bili., T-chol., TG, TP, ALB, A/G, P etc. increase according to 10, 30, 50 week, and observed improvement that is biochemical value than control group form of SipIMiGwanJungTang As those results, PalmoolGoonJaTang and SipIMiGwanJungTang are effective against dacline of physical function as aging. Moreover, considered to the serogical results, SipIMiGwanJungTang is much more effective than the other.
