배추 GROWTH-REGULATING FACTOR 유전자 발현이 유채 기관크기에 미치는 영향 (Effects of Overexpression of Brassica rapa GROWTH-REGULATING FACTOR Genes on B. napus Organ Size)

  • GROWTH-REGULATING FACTOR (GRF) 유전자는 QLQ와 WRC 도메인을 갖고 있는 식물 특이적 전사조절인자로 식물기관들의 생장과 발달에 중요한 역할을 하고 있다. 배추로부터 분리된 BrGRF3-1과 BrGRF9 유전자를 각각 35S 프로모터에 결합시켜 두 종류의 식물 형질전환 벡터를 제작한 후 아그로박테리움을 이용하여 유채에 형질전환하였다. 선발된 형질전환체의 특성을 분석한 결과 세포 크기 보다는 세포 수 증가에 의해 자엽, 잎, 종자의 크기가 커지는 것으로 나타났다. RT-PCR로 유전자 발현을 분석한 결과 BrGRF3-1과 BrGRF9 유전자는 기관 생장 발달에 관여하는 지베렐린(GA), 옥신(auxin), 세포분열 관련 유전자들의 발현을 조절함으로써 유채 기관의 생장과 발달에 중요한 역할을 하는 것으로 추측된다. 따라서 본 연구결과로 BrGRF 유전자는 생명공학기술을 활용하여 작물의 농업적 형질을 개선하기 위한 유전자원으로 이용될 수 있을 것으로 사료된다.

  • 징거미새우의 생식소 성숙 제어에 대한 광주성발달과 안병 X-organ과의 관계를 밝히기 위하여 1) 생식소 성숙 제어에 미치는 수온과 광주기의 영향, 2)생식소 성숙에 대한 안병 절제 효과, 3) 안병에 있는 외수 X-organ (MEX-organ)조직의 활성에 대한 계절적인 변화 등을 조사하였다. 전난황형성기(12월${\sim}$다음해 3월)와 난황형성기(4월${\sim}$5월)에 있어서, 생식소 성숙은 주로 수온에 영향을 받으며, 광주조건(12L/12D, 15L/9D)에 관계없이 수온상승에 의하여 GSI가 증가하고 생식소도 성숙하였다. 반면, 산란기(6월${\sim}8$월)에 있어서, 생식소 성숙은 주로 광주조건에 영향을 받으며, 15L/9D와 같은 장일 조건에서는 생식소 성숙이 지속되고 높은 GSI값을 나타내는 반면, 12L/12D와 같이 단일화된 조건에서는 난소내에 있는 대부분의 난황형성기 난모세포들은 퇴화${\cdot}$흡수되어 휴지기 상태의 난모세포로 되었고, GSI는 감소되었다. 휴지기(9월${\sim}11$월)에 있어서, 수온($16^{\circ}C,\;22^{\circ}C,\;28^{\circ}C$)과 광주조건(12L/12D, 15L/9D)을 조합한 모든 실험구에서는 재성숙이 일어나지 않았다. 생식소 성숙에 대한 안병 절제 효과는 전난황형성기와 산란기때 보다 휴지기 때가 더 강하게 나타났다. 안병 MEX-organ에서 직경이 $20{\mu}m$이상인 신경분비세포 수는 생식주기에 따라 변동을 보였으며, 휴지기 때 그 수는 $77{\pm}12$로 증가하였고, 난황형성기때는 $55{\pm}7$로 감소하였다. 그러나 Bellonci 조직의 크기(volume)는 휴지기 때에 비해서 난환형성기때 증가하는 경향을 보였다.

FDG PET-CT in Non-small Cell Lung Cancer: Relationship between Primary Tumor FDG Uptake and Extensional or Metastatic Potential

  • Objective: To explore the relationships between primary tumor $^{18}F$-FDG uptake measured as the SUVmax and local extension, and nodal or distant organ metastasis in patients with NSCLC on pretreatment PET-CT. Methods: 93 patients with NSCLC who underwent $^{18}F$-FDG PET-CT scans before the treatment were included in the study. Primary tumor SUVmax was calculated; clinical stages, presence of local extension, nodal and distant organ metastases were recorded. The patients with SUVmax${\geq}2.5$ were divided into low and high SUVmax groups by using the median SUVmax. The low SUVmax group consisted of 45 patients with SUVmax<10.5, the high SUVmax group consisted of 46 patients with SUVmax${\geq}10.5$. Their data were compared statistically. Results: 91 cases with SUVmax${\geq}2.5$ were included for analysis. The mean SUVmax in patients without any metastasis was $7.42{\pm}2.91$ and this was significantly lower than that ($12.18{\pm}4.94$) in patients with nodal and/or distant organ metastasis (P=0.000). In the low SUV group, 19 patients had local extension, 22 had nodal metastasis, and 9 had distant organ metastasis. In the high SUV group, 31 patients had local extension, 37 had nodal metastasis, and 18 had distant organ metastases. There was a significant difference in local extension (P =0.016), distant organ metastasis (P =0.046), and most significant difference in nodal metastasis rate (P =0.002) between the two groups. In addition, there was a moderate correlation between SUVmax and tumor size (r = 0.642, P<0.001), tumor stage (r = 0.546, P<0.001), node stage (r = 0.388, P<0.001), and overall stage (r = 0.445, P= 0.000). Conclusion: Higher primary tumor SUVmax predicts higher extensional or metastatic potential in patients with NSCLC. Patients with higher SUVmax may need a close follow-up and more reasonable individual treatment because of their higher extensional and metastatic potential.

  • Bovine serum albumin microspheres containing cytarabine were prepared using cross-linking agent, formaldehyde. The shape and the size distribution of them were observed. The shape of them was spherical and the susrface was compact and smooth. The size distribution of them was affected by dispersion forces during emulsfication. The release of cytarabine from albumin microspheres was dependent upon cross-linking time, amount of cross-linking agent and drug/albumin ratio. However, the difference of drug release by the dispersion forces was not great. After release test, the shape of albumin microspheres was nonspherical and the albumin matrix seemed to be a little relaxed. The degradation tests of albumin microspheres by the proteolytic enzyme showed that albumin microspheres were progressively digested according to the cross-linking degree.

USE OF MOLD INHIBITOR FOR FEED STORAGE AND IMPROVED CHICK PERFORMANCE

  • Two experiments were conducted to evaluate the effect of mold inhibitor in the ration which had two different protein levels (18% and 12%) and two different particle sizes (80 or 40% of the particles in the ration less than 1.19 mm). The experimental diets with ave. 12.7% moisture which were treated at the level of 0.1% mold inhibitor were stored under 85% humidity and at $29{\pm}1^{\circ}C$ for 10 to 40 days. In experiment 1, after 40 days of storage the $CO_2$ production in the feed treated with mold inhibitor was higher (p < 0.01) than when 40% of the ration's panicle size was 1.19 mm. Aflatoxin production in the experimental diet with mold inhibitor was affected (p<0.05) by the levels of protein and the different particle size ranges after 40 days storage. The interaction of protein levels and particle size ranges on the anatoxin and $CO_2$ production was significant (p<0.05) at 40 days storage. In experiment 2, there was a decrease in total body weight gain and total feed intake observed in chicks fed the untreated diet of 18% protein with 40% of the particles in the ration less than 1.19 mm stored for 40 days. Feed conversion was depressed (p<0.05) in the chicks fed the untreated diets of both particle sizes. Particle size X types of feed interaction in feed conversion was significant (p<0.05).

  • Ventricular septal defect complicating myocardial infarction is rare but fatal condition which requires early surgical intervention before end-organ failure ensues from cardiogenic shock. Since the first successful repair by Cooley et al in 1956, surgical skills and strategies were developed and modified to a great extent, and we adopted the new repair technique in our case which stresses that minimal or no part of the infarcted septum and left ventricular wall be resected. This technique obviates the need to resect the infarcted part of the septum and prevents recurrence of an even larger VSD, and provides adequate size and shape of the left ventricle after of transinfarction left ventriculotomy.

Re-188이 표지된 황 교질(Sulfur Colloid) 제조와 생체내 분포 (Preparation and Biodistribution of Re-188 Sulfur Colloid)

  • 목적 : Re-188 황 교질을 제조하여 관절염 활액막절제술, 복강내 암전이 치료제로서의 사용 가능성을 알아보았다. 대상 및 방법: Re-188 황 교질의 표지에 EDTA $Na_2.2H_2O$의 양, $KReO_4$의 존재유무, pH의 영향을 관찰하였다. Re-188 황 교질의 안정성을 실온에서 24 시간, 사람혈청과 활액에서 72 시간동안 관찰하였다. Re-188 황 교질의 입자 크기 분포는 여러 가지 크기의 필터를 사용하여 측정하였다. Re-188 황 교질을 실험 동물에 투여하여 생체내 분포와 체외 대사량을 관찰하였다. 결과: Re-188 황교질은 sodium thiosufate 40 mg, EDTA $Na_2.2H_2O$ 0.8 mg, $KReO_4$ 0.8 mg, pH 1의 조건에서 60분 동안 가열시 표지효율이 $64.5{\pm}5.8%$ (n=5)로 가장 높았고, 이를 정제하였을때 방사화학적 순도는 99%이상이었다. Re-188 황 교질은 실온에서 24 시간, 혈청과 활액에서 72시간 동안 모두 방사화학적 순도가 99%이상으로 매우 안정하였다. Re-188 황 교질의 입자크기 분포는 $1{\mu}m$ 이하가 0.3%, $1{\sim}5{\mu}m$ 11.2%, $5{\sim}10{\mu}m$ 35.8%, $10{\mu}m$ 이상은 52.8%으로 나타났다. 마우스의 꼬리정맥으로 Re-188 황 교질을 주사한 후 1 시간에 폐와 간의 섭취율($37.30{\pm}5.36$, $32.33{\pm}5.79%$ ID/organ)이 가장 높았으며, 근육의 섭취율은 $6.60{\pm}0.02 %$ID/organ였다. 마우스복강내에 Re-188 황 교질을 주사한 후 1, 24 시간에 복강외 장기의 섭취율은 $0.1{\pm}0.1$, $0.4{\pm}0.1%$ ID/organ으로 매우 낮았으며, 주사 후 70 시간까지 뇨와 대변으로 배설된 양은 $2.68{\pm}0.50%$$0.95{\pm}0.17%$였다. 토끼 무릎관절에 Re-188 황 교질을 주사한 후 24 시간에 무릎을 제외한 모든 장기에서의 섭취율은 0.07% ID/g 이하로 매우 낮았다. 결론: 이실험에서 표지한 Re-188 황 교질은 표지효율과 안정성이 높고 임상적으로 방사선 활액막 절제술 등에 사용할 수 있을 것으로 생각한다.

Overexpression of CXCL2 inhibits cell proliferation and promotes apoptosis in hepatocellular carcinoma

  • C-X-C motif chemokine ligand 2 (CXCL2) is a small secreted protein that exhibits a structure similar to the proangiogenic subgroup of the CXC chemokine family. Recently, accumulating evidence suggests that chemokines play a pivotal role in cancer progression and carcinogenesis. We examined the expression levels of 7 types of $ELR^+$ CXCLs messenger RNA (mRNA) in 264 clinical samples. We found that CXCL2 expression was stably down-regulated in 94% of hepatocellular carcinoma (HCC) specimens compared with paired adjacent normal liver tissues and some HCC cell lines. Moreover, CXCL2 overexpression profoundly attenuated HCC cell proliferation and growth and induced apoptosis in vitro. In animal studies, we found that overexpressing CXCL2 by lentivirus also apparently inhibited the size and weight of subcutaneous tumours in nude mice. Furthermore, we demonstrated that CXCL2 induced HCC cell apoptosis via both nuclear and mitochondrial apoptosis pathways. Our results indicate that CXCL2 negatively regulates the cell cycle in HCC cells via the ERK1/2 signalling pathway. These results provide new insights into HCC and may ultimately lead to the discovery of innovative therapeutic approaches of HCC.

  • An anatomically detailed elderly human body model is under development. Using the anthropometric database of domestic nation-wide size survey, SizeKorea, a standard size and shape of 50th %tile elderly was constructed. Through the local recruitment process, a male volunteer with 71 years of age, 163cm of height and 63kg of weight has been selected. The exterior (skin) and interior (skeleton and organ) geometries were acquired from whole body 3D laser scan and various medical images such as CT, X-ray, and Ultrasonic of the volunteer. A particular attention has been paid into the combining process of exterior and interior geometries especially for joint articulation positions since they were measured at different postures (sitting vs. supine). A whole ribcage of PMHS which possessed similar anthropometry and age of standard 50th %tile elderly was prepared and dissected for the precise gauge of cortical rib bone thickness distributions. After completing the morphological construction of elderly human body, the finite element modeling will be processed by meshing elements and assigning mechanical properties to various biological tissues which reflect the aging effect.
