Evaluation of bacteriophages for prevention and treatment of diarrhea due to experimental enterotoxigenic Escherichia coli K88 infection of weaned piglets (이유자돈의 실험적 enterotoxigenic Escherichia coli K88 감염에 의한 설사증에 대한 박테리오파지의 예방과 치료에 대한 효능 평가)

  • Colibacillosis in pigs remain a major swine industry bruden worldwide. Although some progress has been made in treating collibacillosis in pigs by using biosecurity and antimicrobials, it still remain a considerable problem. The use of host-specific bateriophages as a biocontrol is one possible alternative. The purpose of this study was to evaluate the effect of bacteriophage against enterotoxigenic Escherichia coli (ETEC) K88 infection in piglets. Twenty-eight piglets were randomly divided into four groups and each group was allocated with 7 pigs. Group B, C and D were inoculated with 5 ml of ETEC K88 ($1{\times}10^8$ CFU/ml) per head of piglet via oral. Group C and D were fed with bacteriophages (Group C, $1.0{\times}10^6$ PFU/g; Group D, $1.0{\times}10^8$ PFU/g; CJ CheilJedang Corp., Korea) orally as treatment. In piglets administrated bacteriophages and challenged with ETEC K88 (Group C and D), Clinical signs and the growth performance were improved and antibody titers were maintained low level compared with piglets challenged with ETEC K88 (Group B, P<0.05). Group B were shown high pH in the alimentary tracts compared with other piglets (P<0.05). In quantitative analysis by real-time PCR, the results of Group C and D were lower than those Group B in faecal and intestinal samples (P<0.05). Severe villus atrophy and crypt hyperplasia were observed in Group B consequently V/C ratio increased, compared with other piglets. These results indicate that feeding with bacteriophage has effect to prevent ETEC K88 infection in piglets and suggest that use of bacteriophage can be considered a valid antibiotic alternative.

Non-nutritive Sucking Habits of Preschool Children in Daejeon (대전지역 미취학 어린이의 비수유성 빨기 습관에 대한 조사 연구)

  • The purpose of this study was to determine the prevalence of non-nutritive sucking in preschool children in Daejeon. This study assessed the relationship between contributing factors influencing the prevalence of non-nutritive sucking. Also, the effects of non-nutritive sucking on the primary dentition were investigated. The study included 841 children 18~65 months of age that visited a pediatric private dental clinic for oral examination. Both children and their guardians were surveyed. It was found that 32.8% of children exhibited non-nutritive sucking, and 37.7% of females displayed this behavior. The number of females showing this behavior was significantly greater than that of males. The prevalence decreased significantly with the breast-feeding period, and there was no association with either birth rank or maternal occupation. Finger sucking (62.6%) was more prevalent and of longer duration than pacifier sucking (37.4%). Malocclusion of the primary dentition was associated with the duration of this behavior.

A Survey of the Effect of Nursing Students on Community Health Knowledge (간호학생 실습이 지역주민의 보건지식 및 태도에 미치는 영향 -연세대학교 간호대학 보건간호학 실습을 중심으로-)

  • The purpose of this study was to determine whether or not the delivery of health services effects the health knowledge of the recipients of that service. 10 was hypothesized that the practical application of theory, though health services rendered by public health nursing students during their field experiences. would have a positive relationship with the quantity and quality of health knowledge of the female population of a given community. In April 1970, ninety five women(between an age 15-49) from Koyang-Gun were interviewed regarding their knowledge and attitudes in four areas of health: maternal health, child health. family planning and tuberculosis prevention. After 11 years(Nov. l971), during which time the women received home visits by student nurses, the women were again interviewed using the original questionnaire. Additionally, they were asked where they had received information about each question. During the 1$\frac{1}{2}$ year period the visits were made by a total of 112 students. They visited the community twice a week. The result were as follows: Maternal health There was marked improvement in the knowledge of specific aspects of maternal health such as the proper instruments needed for cutting the cord and nutritional precaution of pregnancy (p<0.05). In each case 42.0 percent of more of this knowledge was attributed to information gained from the nursing students. However even after the nursing students'visits, the knowledge of other general areas of maternal remained poor. Child health There were increases in the knowledge of the respondents for the time to supplement breast feeding and for time to wean infants (p<0.05). In each case 35.0 percent or more of this information was attributed to the student. The increase in knowledge regarding types of immunization was more often attributed to the clinic nurse than to the nursing students. Knowledge of the necessity of booster immunization was poor both before and after the visits by the nursing students. Family planing There were significant increases in correct information regarding the time of I.U.D. insertion (P<0.05) and method of taking the oral pill (P<0.05). More than 42.0 percent of the increase was attributed to the nursing students'visits. However there was a slight decrease in correct information about concept of family planning. Tuberculosis There was a general lack of improvement in all areas concerned with tuberculosis. Two thirds of the respondents felt tuberculosis was hereditary and did not know what B.C.G. was. From this study, it could be concluded that in specific area the health knowledge of the female population studied was positively effected by the visits of the nursing students. The study also, suggests, however, that there was a marked unevenness in the overall increase in health knowledge. This raises serious questions which require further research. Suggested areas for further research are (1) Adequacy of the course content preparing nurses for the field experience. (2) A public health nursing manual for student reference. (3) Where and when specific aspects of health can be taught most effectively.

Effect of Dietary Protein and Fiber on Ethanol-induced Hepatotoxicity in Rats (흰쥐의 에탄올성 간장해에 미치는 식이 단백질과 섬유소의 영향)

  • This study was conducted to investigate the effect of dietary protein and fiber levels on the activities of ethanol metabolizing enzymes of liver in ethanol-treated rats. Sprague-Dawley male rats were fed on diets containing two levels of protein(7, 20%/kg diet) and pectin(5, 10%/kg diet). In ethanol experiments, ethanol(25% v/v) was administered by oral intubation(5g/kg body weight) at the same time once a day Control animals received an isocaloric dose of sucrose. The rats were sacrificed after 5 weeks of feeding periods. Alcohol dehydrogenase and microsomal ethanol oxidizing system activities of hepatic tissue were increased more in ethanol-treated groups than in control groups. Increment of activities predominated in normal protein normal fiber group. Aldehyde dehydrogenase activity was decreased in ethanol-treated groups and significantly decreased in normal Protein normal fiber group. Cytochrome P-450 content was significantly increased in ethanol-treated groups and Predominated in normal protein groups. Xanthine oxidase activity was increased in ethanol-treated groups, but not significantly except normal protein normal fiber group. Glutathione content tended to increase in proportion to level of dietary protein and was higher in normal fiber groups than in high fiber groups, whereas it was decreased by ethanol treatment. Lipid Peroxide content was significantly increased in low Protein normal fiber groups.

Effect of low salinity probiotics on the growth and non-specific immunity of whiteleg shrimp Litopenaeus vannamei cultured under low salinity conditions (저염도에서 생육가능한 미생물 probiotics가 저염분 양식의 흰다리새우(Litopenaeus vannamei)의 성장 및 면역능에 미치는 영향)

  • The whiteleg shrimp Litopenaeus vannamei can survive in a wide range of salinity (1-40 psu). However, such variation, particularly at low salinity (1-5 psu), can affect various physiological changes such as survival rates, non-specific immunity and disease resistance. In this study, growth, non-specific immunity and disease resistance were measured following simultaneous oral feeding and addition of probiotic microbes into culture water for 73 day period. The salinity of the culture water was maintained at 3 psu by periodical salt additions. The result shows that survival rate increased significantly (5.6 vs. 15.4%) after 73 day rearing. Significant increases were identified in reactive oxygen species (ROS) production and phenol oxidase (PO) activity. However, superoxide dismutase (SOD) activity was not influenced. When the shrimp was artificially challenged with Vibrio alginolyticus, slight mortality reduction was observed in the probiotics-treated group (100 vs. 79%). In conclusion, the production of cultured whiteleg shrimp at low salinity might be increased by probiotics survivable at low salinity levels.

  • Although functional gastrointestinal disorders (FGIDs) are very common in pediatric patients, there is a scarcity of published epidemiologic data, characteristics, and management patterns from Saudi Arabia, which is the 2nd largest Arabic country in terms of area and the 6th largest Arabic country in terms of population, with 10% of its population aged <5 years. Functional constipation (FC) is an FGID that has shown a rising prevalence among Saudi infants and children in the last few years, which urges us to update our clinical practices. Nine pediatric consultants attended two advisory board meetings to discuss and address current challenges, provide solutions, and reach a Saudi national consensus for the management of pediatric constipation. The pediatric consultants agreed that pediatricians should pay attention to any alarming signs (red flags) found during history taking or physical examinations. They also agreed that the Rome IV criteria are the gold standard for the diagnosis of pediatric FC. Different therapeutic options are available for pediatric patients with FC. Dietary treatment is recommended for infants with constipation for up to six months of age. When non-pharmacological interventions fail to improve FC symptoms, pharmacological treatment with laxatives is indicated. First, the treatment is aimed at disimpaction to remove fecal masses. This is achieved by administering a high dose of oral polyethylene glycol (PEG) or lactulose for a few days. Subsequently, maintenance therapy with PEG should be initiated to prevent the re-accumulation of feces. In addition to PEG, several other options may be used, such as Mg-rich formulas or stimulant laxatives. However, rectal enemas and suppositories are usually reserved for cases that require acute pain relief. In contrast, infant formulas that contain prebiotics or probiotics have not been shown to be effective in infant constipation, while the use of partially hydrolyzed formula is inconclusive. These clinical practice recommendations are intended to be adopted by pediatricians and primary care physicians across Saudi Arabia.

Effect of Feeding with Different Source of Carbohydrate and Fiber on Carbohydrate and Lipid Metabolism in Type 2 Diabetic Rats (탄수화물의 급원과 식이섬유의 종류를 달리한 식이가 제 2형 당뇨 쥐의 당대사 및 지질대사에 미치는 영향)

  • This study was designed to evaluate the effects of fructose(F) or sucrose(S) and guar gum intake on carbohydrate and lipid metabolism in 15-week-old male Goto-Kakizaki(GK) rats. Fifty rats were randomly assigned to 5 groups which were different in carbohydrate(25% of carbohydrate) and fiber(5% w/w) sources. The carbohydrate(CHO) sources of each group were comstarch(control group, 100% of CHO), fructose with cellulose(F), fructose with guar gum(FG), sucrose with cellulose(S), and sucrose with guar gum(SG). Each group was fed exterimental diet for 4 weeks. We measured food intake, body weight gain, adipose tissues weight and organs weight. We conducted oral glucose tolerance test(OGTT) and measured plasma insulin concentration to examine carbohydrate metabolism. To evaluate lipid metabolism, we measured the lipid profile of plasma, liver and feces. Food intake and weight gain of FG or SG groups tended to be less than those of F or S groups. Perirenal and epididymal fat pad weights of SG group were significantly lower than those of S group and those of FG group tended to be lower than those of F group. In OGTT, blood glucose values of F or S groups were significantly higher than those of C group, and FG or SG groups tended to be lower than those of F or S groups during the experimental time. The area under the curve(AUC) of C group was significantly highest among the groups, AUC and plasma insulin concentration of FG or SG groups tended to be lower than those of F or S groups. Plasma and hepatic triglyceride (TG) of FG and SG groups were significantly lower than those of F and S groups, plasma and hepatic total lipid(TL) and total cholesterol(TC) of FG and SG groups tended to be lower than those of F and S groups. Fecal TL, TG and TC of FG or SG groups tended to be higher than those of F and S groups. In conclusion, intake of guar gum should improve carbohydrate and lipid metabolism in partial substitution of fructose or sucrose for cornstarch in GK rats.

Surgical Outcomes of Cervical Esophageal Cancer: A Single-Center Experience

  • Background: Cervical esophageal cancer is a rare malignancy that requires specialized care. While definitive chemoradiation is the standard treatment approach, surgery remains a valuable option for certain patients. This study examined the surgical outcomes of patients with cervical esophageal cancer. Methods: The study involved a retrospective review and analysis of 24 patients with cervical esophageal cancer. These patients underwent surgical resection between September 1994 and December 2018. Results: The mean age of the patients was 61.0±10.2 years, and 22 (91.7%) of them were male. Furthermore, 21 patients (87.5%) had T3 or T4 tumors, and 11 (45.8%) exhibited lymph node metastasis. Gastric pull-up with esophagectomy was performed for 19 patients (79.2%), while 5 (20.8%) underwent free jejunal graft with cervical esophagectomy. The 30-day operative mortality rate was 8.3%. During the follow-up period, complications included leakage at the anastomotic site in 9 cases (37.5%) and graft necrosis of the gastric conduit in 1 case. Progression to oral feeding was achieved in 20 patients (83.3%). Fifteen patients (62.5%) displayed tumor recurrence. The median time from surgery to recurrence was 10.5 months, and the 1-year recurrence rate was 73.3%. The 1-year and 3-year survival rates were 75% and 33.3%, respectively, with a median survival period of 17 months. Conclusion: Patients with cervical esophageal cancer who underwent surgical resection faced unfavorable outcomes and relatively poor survival. The selection of cases and decision to proceed with surgery should be made cautiously, considering the risk of severe complications.

The effects of Galgŭnhaegit'ang and Won's-Galgŭnhaegit'ang for Taeŭmin on the immune response (태음인(太陰人) 갈근해기탕(葛根解肌湯)과 원특상(元特上)의 갈근해기탕(葛根解肌湯)이 면역반응(免疫反應)에 미치는 영향(影響))

  • In order to compare the dffects of $Galg{\breve{u}}nhaegit'ang$(葛根解肌湯) of "Dongeuisusebowon(東醫壽世保元)and Won's(元)-$Galg{\breve{u}}nhaegit'ang$(葛根解肌湯) of "Dongeuisasansinpyun(東醫四象新編)" on the immune respone, Sprague-Dawley male rats were used and randomly divided into four groups. Normal group was under normal condition, Control group was injected i.v. with 2mg/kg Methotrexate(MTX) on the 9th day and 11th day after sensitization with SRBC on the 5th day, $Galg{\breve{u}}nhaegit'ang$ group was fed with 1ml of $Galg{\breve{u}}nhaegit'ang$ and Won's-$Galg{\breve{u}}nhaegit'ang$ group was fed with 1ml of Won's-$Galg{\breve{u}}nhaegit'ang$ by oral during eighteen days. In the 9th day and the 11th day after oral feeding with medication, MTX was injected in tail of rats in order to reduce immune function. Leukocyte count, lymphocyte ratio, lymphocyte count of spleen, lymphocyte count of bone marrow, contact hypersensitivity to DNFB, morphologic change of thymus cell, and electropherogram of serum protein were estimated and compared according to the group. The results are as follows : 1. Before and after MTX injection, leukocyte(WBC) count was increased signigicantly in Won's-$Galg{\breve{u}}nhaegit'ang$ group compared to control group. $Galg{\breve{u}}nhaegit'ang$ group had no significant difference compared to control group. 2. Before and after MTX injection, lymphocyte ratio was not significantly different in Won's-$Galg{\breve{u}}nhaegit'ang$ group and in $Galg{\breve{u}}nhaegit'ang$ group compared to control group. 3. The lymphocyte count of spleen was increased significantly in $Galg{\breve{u}}nhaegit'ang$ group compared to control group and Won's-$Galg{\breve{u}}nhaegit'ang$ group. Won's-$Galg{\breve{u}}nhaegit'ang$ group had no significant difference compared to control group. 4. The lymphocyte count of bone marrow was increased significantly in Won's-$Galg{\breve{u}}nhaegit'ang$ group compared to control group and $Galg{\breve{u}}nhaegit'ang$ group. $Galg{\breve{u}}nhaegit'ang$ group had no significant difference compared to control group. 5. Contact hypersensitivity was increased significantly in Won's-$Galg{\breve{u}}nhaegit'ang$ group compared to other group. $Galg{\breve{u}}nhaegit'ang$ group had no significant difference compared to control groups. 6. In the morphologic change of thymus cell, control group compared to normal group had a indistinct boundary between cortex and medulla and lymphocyte cell density of thymus was low. $Galg{\breve{u}}nhaegit'ang$ group and Won's-$Galg{\breve{u}}nhaegit'ang$ group compared to control group had a definite boundary between cortex and medulla and lymphocyte cell density of thymus was high. 7. In the SDS-PAGE electropherogram of serum protein, Won's-$Galg{\breve{u}}nhaegit'ang$ group had a wide band of nearby 25,000 Dalton, and which meant IgG generated more actively. Considering this results, $Galg{\breve{u}}nhaegit'ang$ group and Won's-$Galg{\breve{u}}nhaegit'ang$ group have an effect on the depression of immune function induced by MTX, and especially Won's-$Galg{\breve{u}}nhaegit'ang$ group has an significant effect than $Galg{\breve{u}}nhaegit'ang$ group.

Proteomic Assessment of the Relevant Factors Affecting Pork Meat Quality Associated with Longissimus dorsi Muscles in Duroc Pigs

  • Meat quality is a complex trait influenced by many factors, including genetics, nutrition, feeding environment, animal handling, and their interactions. To elucidate relevant factors affecting pork quality associated with oxidative stress and muscle development, we analyzed protein expression in high quality longissimus dorsi muscles (HQLD) and low quality longissimus dorsi muscles (LQLD) from Duroc pigs by liquid chromatography-tandem mass spectrometry (LC-MS/MS)-based proteomic analysis. Between HQLD (n = 20) and LQLD (n = 20) Duroc pigs, 24 differentially expressed proteins were identified by LC-MS/MS. A total of 10 and 14 proteins were highly expressed in HQLD and LQLD, respectively. The 24 proteins have putative functions in the following seven categories: catalytic activity (31%), ATPase activity (19%), oxidoreductase activity (13%), cytoskeletal protein binding (13%), actin binding (12%), calcium ion binding (6%), and structural constituent of muscle (6%). Silver-stained image analysis revealed significant differential expression of lactate dehydrogenase A (LDHA) between HQLD and LQLD Duroc pigs. LDHA was subjected to in vitro study of myogenesis under oxidative stress conditions and LDH activity assay to verification its role in oxidative stress. No significant difference of mRNA expression level of LDHA was found between normal and oxidative stress condition. However, LDH activity was significantly higher under oxidative stress condition than at normal condition using in vitro model of myogenesis. The highly expressed LDHA was positively correlated with LQLD. Moreover, LDHA activity increased by oxidative stress was reduced by antioxidant resveratrol. This paper emphasizes the importance of differential expression patterns of proteins and their interaction for the development of meat quality traits. Our proteome data provides valuable information on important factors which might aid in the regulation of muscle development and the improvement of meat quality in longissimus dorsi muscles of Duroc pigs under oxidative stress conditions.
