제거된 ePTFE 막의 주사전자현미경적 소견과 치주임상상태의 비교연구 (The Comparative Study On Scanning Electron Microscopic Findings Of Retrived ePTFE Membrane With Clinical Conditions)

  • Ten intrabony defects in 10 patients were treated by flap surgery including root surface debridement and placement of an expanded polytetrafluoroethylene(ePTFE) membrane. The membranes were removed after 4-6 weeks. This study was performed to examine the retrived ePTFE membrane by scanning electron microscopy(SEM) for bacterial contamination and adherent connective tissue elements, and to compare it with clinical conditions. The cervical portion of the membrane, which in most cases had become partially exposed to the oral cavity, had a bacterial deposit. Small bacterial colonies and a scatter of single cells in some instances extended into the apical portion of the membrane. Fibroblast-like cells, erythrocytes and fibrous structures were seen in the apical portion of the membrane. Outer surface of membrane tends to more bacterial contamination than inner surface(p<0.01), and upper portions more than lower portions(P<0.01). Comparison of ultrastructural findings and clinical conditions revealed that extent of bacterial contamination of the membrane correlated with gingival inflammation and extent of membrane exposure, but it was not significant statistically. The results suggested that gingival inflammation and membrane exposure affect periodontal regeneration by the use of ePTFE membrane.

덴탈 바이오필름에서 분리한 세균의 주사전자현미경적 관찰 및 동정 (Identification and morphology of scanning electron microscopy(sem) of bacteria isolated from dental biofilm)

  • In this study, specimens such as tongue, supragingival and subgingival biofilm were taken from total 20 scaling subjects who visited the oral prophylaxis practice lab at department of dental hygienics, J Health College in order to observe bacterial distributions and morphology using scanning electron microscopy(sem). as a result, this study came to the following conclusions: 1. According to observation of tongue, supragingival and subgingival biofilm through sem, it is found that there are round colonies of gram-positive cocci and gram-negative bacilli on blood agar medium. 2. The observation of bacterial morphology on dental biofilm through sem, cocci in chain cocci in cluster and bacillus(rod) respectively. 3. For tongue biofilm, it is found that a variety of bacterial species are detected, such as Granulicatolla adiacens(1), Gemella morbillorum(3), Streptococcus mitis(2), Streptococcus sanguinis(1), Aerococcus viridans (2), Streptococcus equinus(1), Leuconostoc spp.(1), Gemella haemolysans (1) and Lactococcus lactis spp.(1) respectively. 4. For supragingival biofilm, it is found that a variety of bacterial species detected, such as Aerococcus viridans(1), Gemella haemolysans(2), Leuconostoc spp.(2), Gemella morbillorum(1) and Pseudomonas fluoescens (1) respectively. 5. For subgingival biofilm, it is found that a variety of bacterial species detected, such as Leuconostoc spp.(1), Staphylococcus lugdunensis(1) and Streptococcus salivarius(1) respectively.

구내 방사선 필름의 표면소독효과에 관한 연구 (SURFACE DISINFECTION OF INTRAORAL FILMS)

  • The purpose of the study was to determine whether Sodium hypochlorite and Glutaraldehyde would be effective for the surface disinfection of contaminated radiographic film pockets with saliva The following results were as obtained 1. Proper times for surface disinfection of 2.0% Glutaraldehyde and 3.5% Sodium hypochlorite were 60 seconds. 2. When films were immerged in 2% Glutaraldehyde solution for 1 minute, baterial colonies were present in 24 cases(80%). 3. When films were immerged in 3.5% Sodium hypochlorite solution for 1 minute, bacterial colony was absent in 25 cases(83.3%). 4. Differences of effectiveness on surface disinfection between 2% Glutaraldehyde and 3.5% Sodium hypochlorite were statistically significant.

Postantibiotic Effects and Postantibiotic Sub-MIC Effects of Antibiotics on Treponema Denticola

  • Postantibiotic effect (PAE) is defined as the length of time that bacterial growth is suppressed following brief exposure to an antibiotic. In this study, the in vitro PAE, postantibiotic sub-MIC effect (PA SME) and sub-MIC effect (SME) of antibiotics on Treponema denticola ATCC 35405 were investigated. The PAE of doxycycline and metronidazole were 20.3 h and 25.0 h, respectively. The PA SMEs examined by addition of 0.1, 0.2 and 0.3X MICs during the postantibiotic phase of the bacteria for metronidazole were longer than those for doxycycline. In contrast, the SMEs for doxycycline were longer than those for metronidazole. The PA-SME and SME values increased as the concentration of antibiotics increased. The present study illustrates the existence of PAE, PA-SME and SME for several antibiotics against T. denticola, thereby extending the pharmacodynamic advantages of these antibiotics.

Effect of Sub-Minimal Inhibitory Concentration Antibiotics on Morphology of Periodontal Pathogens

  • Minimal inhibitory concentration (MIC) is the lowest concentration of antibiotics that inhibits the visible growth of a microorganism. It has been reported that sub-MIC of antibiotics may result in morphological alterations along with biochemical and physiological changes in bacteria. The purpose of this study was to examine morphological changes of periodontal pathogens after treatment with sub-MIC antibiotics. Aggregatibacter actinomycetemcomitans, Fusobacterium nucleatum, and Porphyromonas gingivalis were used in this study. The MIC for amoxicillin, doxycycline, metronidazole, penicillin, and tetracycline were determined by broth dilution method. The bacterial morphology was observed with bright field microscope after incubating with sub-MIC antibiotics. The length of A. actinomycetemcomitans and F. nucleatum were increased after incubation with metronidazole; penicillin and amoxicillin. P. gingivalis were increased after incubating with metronidazole and penicillin. However, F. nucleatum showed decreased length after incubation with doxycycline and tetracycline. In this study, we observed that sub-MIC antibiotics can affect the morphology of periodontal pathogens.

S-PRG filler를 포함한 치면열구전색제의 Streptococcus mutans에 대한 항미생물 특성에 관한 연구 (Antibacterial Properties of Pit and Fissure Sealant Containing S-PRG filler on Streptococcus mutans)

  • 본 연구는 Planktonic growth inhibition test와 CDC Biofilm Reactor를 사용한 Biofilm assay를 통해 S-PRG filler를 함유하는 치면열구전색제의 S. mutans에 대한 항미생물 효과를 기존의 치면열구전색제와 비교하고자 하였다. S-PRG 필러를 함유하는 치면열구전색제인 BeautiSealant, 불소를 방출하는 치면열구전색제인 Clinpro$^{TM}$ sealant, 불소 미방출 치면열구전색제인 Concise$^{TM}$ sealant를 실험군으로 선정하였다. 성장억제평가를 위해 치면열구전색제를 사용하지 않은 군을 음성 대조군으로 설정하였으며, 3개의 실험군 모두 대조군보다 유의할 정도로 낮은 집락 형성 단위를 보였고, Clinpro$^{TM}$ sealant가 BeautiSealant와 Concise$^{TM}$ sealant보다 유의할 정도로 낮은 집락 형성 단위를 보였다. BeautiSealant와 Concise$^{TM}$ sealant 군간에는 유의한 차이가 관찰되지 않았다. 바이오 필름 평가에서도 Clinpro$^{TM}$ sealant군이 BeautiSealant와 Concise$^{TM}$ sealant군들에 비해 유의할 정도로 낮은 집락 형성을 보였으며, BeautiSealant와 Concise$^{TM}$ sealant 군간에 유의한 차이는 관찰되지 않았다. 본 연구 결과 S-PRG filler를 포함하는 치면열구전색제인 BeautiSealant는 기존의 불소방출 치면열구전색제에 비하여 낮은 항미생물 효과와 높은 바이오 필름 형성능을 보였다.

  • 지대주의 재료와 표면에 따라 연조직반응이 다르고 임플란트 주위염의 원인이 될 수 있다. 티타늄과 지르코니아는 우수한 생체적합성을 가져 안정적인 연조직반응을 보이고 금합금은 표면상태에 따라 연조직반응이 다르게 나타난다. 같은 재료라도 표면특성에 따라 연조직반응이 영향을 받는다. 표면거칠기 증가는 박테리아 부착을 유발하므로 연조직과 접촉하는 부분은 평활한 표면을 가져야 한다. 추가적인 표면처리를 통해 박테리아의 부착은 증가시키지 않으면서 세포반응을 촉진시킬 수 있다. 지대주의 형태 및 반복착탈도 연조직반응에 영향을 줄 수 있다. 초음파 세척과 아르곤 플라즈마 처리는 비교적 효과적으로 맞춤형 지대주의 잔류 미세입자를 제거할 수 있는 방법이다.

저출력 레이저 조사가 치은염증에 미치는 영향에 관한 연구 (Study on the Effect of Low Density Power Laser Radiation in Treating Gingival Inflammation - Clinical, Microbiological, Histological Study -)

  • 레이저의 의학적 이용은 고출력 레이저의 외과적 이용과 더불어 많은 진전을 하게 되었다. 근래에는 인체에 손상을 주는 고출력 레이저 외에도 조직의 파괴없이 인체조직에 유용한 효과를 줄 수 있는 저출력 레이저에 대한 연구가 적극적으로 시도되고 있다. 본 연구는 치은염을 가진 13명의 대상에 대해 Split-mouth design으로 대조군 및 실험군을 설정하여 실험군의 치은에 저출력 레이저(Biotherapy dental Laser 3DL)를 격일로 4회 조사한 후 plaque index, pocket depth, sulcus bleeding index, supragingival과 subgingival plaque의 bacterial morphotype의 비율 및 광학현미경을 통한 조직학적 염증 침윤상을 측정 평가하여 다음과 같은 결과를 얻었다. 1) 레이저 조사후 SBI(p<0.01)는 감소하였으나 PI는 별다른 변화가 없었다. 2) 레이저 조사에 따라 spirochetes(p<0.01)를 포함하여 motiles(p<0.05, p<0.01)은 감소된 반면 nommotiles(p<0.05, p<0.01) 증가하였다. 3) 레이저 조사 치은에서는 염증세포 침윤이 Dental(p<0.05) 및 oral(p<0.01) 양측 모두 감소되었다.

Cholera Toxin Disrupts Oral Tolerance via NF-κB-mediated Downregulation of Indoleamine 2,3-dioxygenase Expression

  • Cholera toxin (CT) is an ADP-ribosylating bacterial exotoxin that has been used as an adjuvant in animal studies of oral immunization. The mechanisms of mucosal immunogenicity and adjuvanticity of CT remain to be established. In this study, we investigated the role of indoleamine 2,3-dioxygenase (IDO), which participates in the induction of immune tolerance, in CT-mediated breakdown of oral tolerance. When IDO-deficient ($IDO^{-/-}$) mice and their littermates were given oral ovalbumin, significant changes in antibody responses, footpad swelling and $CD4^+$ T cell proliferation were not observed in $IDO^{-/-}$ mice. Feeding of CT decreased IDO expression in mesenteric lymph nodes (MLN) and Peyer's patch (PP). CT-induced downregulation of IDO expression was reversed by inhibitors of nuclear factor-kappa B (NF-${\kappa}B$), pyrrolidine dithiocarbamate and p50 small interfering RNA. IDO expression was downregulated by the NF-${\kappa}B$ inducers lipopolysaccharide and tumor necrosis factor-${\alpha}$. CT dampened IDO activity and mRNA expression in dendritic cells from MLN and PP. These data indicate that CT disrupts oral tolerance by activating NF-${\kappa}B$, which in turn downregulates IDO expression. This study betters the understanding of the molecular mechanism underlying CT-mediated abrogation of oral tolerance.

Antimicrobial Effect of Polyphenon 60 against Streptococcus mutans and Streptococcus sobrinus

  • Polyphenon 60 refers to the mixture of catechins present in green tea. The aim of this study was to investigate the antimicrobial activities of polyphenon 60 against 4 strains of Streptococcus mutans and 2 strains of Streptococcus sorbrinus, which are the major causative bacteria of dental caries. The minimum bactericidal concentration (MBC) values of polyphenon 60 for S. mutans and S. sobrinus were determined and the effect of biofilm formation inhibition of that was evaluated. The MBC value of polyphenon 60 against the bacterial strains was 2.5 mg/ml except for one particular strain, S. mutans KCOM 1128 for which the value was 1.25 mg/ml. The results of biofilm formation inhibition assay revealed that polyphenon 60 inhibited biofilm formation more than 90% at a concentration of 2.5 mg/ml. It was apparent that polyphenon exhibited biofilm formation inhibition activity along with bactericidal effect against S. mutans and S. sobrinus. Therefore, it is proposed that polyphenon 60 as one of the components of bactericidal agents could be useful in developing oral hygiene products, toothpaste or gargling solution.
