손바닥선인장(제주도 기념물 35호) 추출물이 면역계세포의 활성화에 미치는 영향 (Effects of Opuntia ficus-indica extract on immune cell activation)

  • Opuntia ficus-indca(Op) extract has been claimed to have several therapeutic properties in oriental medicine including anti-inflammatory and anti-rheumatoid arthritis effects. Little is known of its effect on the activation of immune cells such as T cells and macrophages. To evaluate the functional effect of Op extract on immune cells, we examined whether Op extract stimulates the proliferation of T cells and the secretion of cytokines including IL-1 beta, IL-6 and tumor necrosis factor-alpha in THP-1 cell lines by RT-PCR. Op extract significantly enhanced the proliferation of T cell clone(D10S). Transcription of cytokines including IL-1 beta, IL-6, and TNF-alpha peaked 6 hrs after exposure to Op extract(100g/ml) in the THP-1 cell line and declined and declined thereafter. In an experiment to test the dose dependency of transcription of cytokines, transcription increased at a dose of 10 g/ml and the maximum expression was obtained at 100 g/ml, 6 hrs after exposure to Op extract. These findings suggest that Op extract is a potent stimulant of immune cells including T cells and macrophages, which acts by stimulating T cell proliferation and upregulating cytokines. These phenomena imply that some edible plants may be beneficial to living animals through the activation of immune functions.

Optimum Conditions for the Extraction of Effective Substances from the Stem of Opuntia fiscus-indica

  • The conditions for the extraction of effective substances from the stem of Opuntia ficus-indica were optimized by a response surface methodology. The total extract yield was maximized under the temperature of $97.77^{\circ}C$, at a time duration of 145.82 min and a water to sample ratio 16.59 mL/g. Moreover, the optimum conditions for the extraction of effective substances were as follows: $84.95^{\circ}C$, 156.50 min and a water/sample ratio of 7.46 mL/g for the phenolics content; and $97.11^{\circ}C$, 139.03 min and a water/sample ratio of 10.91 mL/g for the pectin content. The range of optimum extraction conditions in consideration of the physicochemical properties of the extracts were shown to be as $95-100^{\circ}C$ as the extraction temperature, 120-180 min as extraction time and a water to sample ratio of 5-18 mL/g.

Studies on the Pharmacological Actions of Cactus:Identification of Its anti-Inflammatory Effect

  • The ethanol extracts of Opuntia ficus-indica fructus (EEOF) and Opuntia ficus-indica stem (EEOS) were prepared and used to evaluate the pharmacological effects of cactus. Both the extracts inhibited the writhing syndrome induced by acetic acid, indicating that they contains analgesic effect. The oral administrations of EEOF and EEOS suppressed carrageenan-induced rat paw edema and also showed potent inhibition in the leukocyte migration of CMC-pouch model in rats. Moreover, the extracts suppressed the release of $\beta$-glucuronidase, a lysosomal enzyme in rat neutrophils. It was also noted that the extracts showed the protective effect on gastric mucosal layers. From the results it is suggested that the cactus extracts contain anti-inflammatory action having protective effect against gastric lesions.

채취시기별 보검선인장 줄기의 항산화, 항당뇨 및 항알츠하이머 활성평가 (Antioxidant and Anti-Diabetic, Anti-Alzheimer Activities of Stem from Opuntia ficus-indica var. saboten Cultivated in Jeju at Harvest Time)

  • In this study, we assessed antioxidant, anti-diabetic, and anti-Alzheimer activities of Opuntia ficus-indica var. saboten (OFI) at harvest time. OFIs were cultivated December 2015~November 2016 in Jeju island. The 70% ethanol extracts of OFI were used to investigate total polyphenol and flavonoid contents, antioxidant(DPPH and ABTS radical scavenging assay), anti-diabetic(yeast ${\alpha}$-glucosidase and rat ${\alpha}$-glucosidase inhibition assay), and anti-Alzheimer(Acetylcholinesterase and butyrylcholinesterase inhibition assay) activities. Total polyphenol and flavonoid contents of OFIs were $17.40{\sim}23.11{\mu}g$ garlic acid/mg Ex and 2.17~6.22 ug (+)-catechine/mg Ex, respectively. DPPH and ABTS radical scavenging activity of OFIs were 131.98~184.90 mg ascorbic acid(AA) eq/100 g and 63.60~101.83 mg AA eq/100 g, respectively. In the anti-diabetic and anti-Alzheimer activities, 70% ethanol extracts of OFI exhibited moderate inhibition activity, compared to control (acarbose and beberine). Total polyphenol and flavonoid contents, antioxidant, anti-diabetic, and anti-Alzheimer activities were no significant differences by season, respectively. Therefore, information on comparative biological evaluations of OFI may be a beneficial in exploring functional food and drug development.

Immunomodulatory Effects of a Methanol Extract from Opuntia ficus indica on Murine Splenocytes

  • Multiple beneficial properties of Opuntia ficus indica (OPF) are well established. In the present study, we have investigated the immunological role of OPF extract (OPFE) on murine splenocytes. OPFE dose- and time-dependently enhanced the proliferation of splenocytes without cytotoxicity. Our results also showed that the number of $CD4^+$ helper T cells and CD45R/$B220^+$ pan B cells increased markedly, but not $CD8^+$ cytotoxic T cells or $CD11b^+$ granulocytes/macrophages. In addition, OPFE significantly decreased the production levels of T helper (Th) 1 type cytokines, interferon (IFN)-$\gamma$, and tumor necrosis factor (TNF)-$\alpha$, although had no significantly differences in those of interleukin (IL)-4, a Th2 type cytokine in concanavalin A (Con A)-stimulated blastogenic cells. Furthermore, OPFE alone strongly increased IL-4 production and decreased TNF-$\alpha$ production even in the absence of Con A. On the basis of these results, this study suggests that OPFE enhances immunity by regulating the pro- and anti-inflammatory response, indicating that this extract exerts a marked immunomodulatory effect, confirming its usefulness as therapy for immune-related diseases.

백년초 추출물의 항균 및 항산화성 (Antimicrobial and Antioxidative Activities of Opuntia ficus-indica var. Saboten Extracts)

  • 백년초(손바닥 선인장)는 제주도를 비롯한 우리나라의 남해안에서 많이 생산되는 선인장의 일종으로 오래전부터 그 임상효능이 알려져 여러 가지 기능적 특성을 갖는 것으로 생각된다. 따라서 본 연구에서는 추출용매 및 방법에 따른 백년초 추출물에 대한 항균 및 항산화성에 대하여 조사하였다. 백년초의 항균활성은 대체적으로 환류추출법이 상온에서의 진탕추출법보다 조금 강하게 나타났고, 각 용매간에는 큰 차이가 없었다. 또한 모든 시험균주에 대하여 항균활성이 강하게 나타났고, 특히 Pseudomonas aeruginosa균주에 대해서는 진탕추출법에 의한 추출물이, Escherichia coli O-157균주에는 환류추출법에 의한 추출물이 상대적으로 다른 시험균주에 비하여 강한 항균활성을 나타내었다. 백년초 물추출물을 Salmonella enteritidis에 처리하였을 때 균체의 표층구조가 허물어지는 심한 형태학적 변화를 나타내었다. 백년초 추출물의 첨가시 0.1% BHT의 첨가시 보다는 약하였지만 상당한 정도의 수소공여능을 나타내었으며, linoleic acid에 대한 항산화력의 조사에서도 0.1% BHT 첨가시와 같은 정도의 강한 효과를 나타내었다.

노팔천연복합물이 Streptozotocin으로 유발된 당뇨 쥐의 혈당 및 췌장조직에 미치는 영향 (Effects of Opuntia ficus-indica Complexes on Blood Glucose and Pancreatic Islets Histology in Streptozotocin-induced Diabetic Rats)

  • 손바닥선인장의 한 종류인 노팔(Opuntia ficus-indica (L.) Mill)을 주재료로 하여 제조한 복합물(OF)의 항당뇨 효과를 알아보기 위해 8주령 수컷 SD-rat에게 streptozotoxin을 주사하여 당뇨를 유발하고, 사료에 OF를 첨가하여 3주간 급여하였으며, 1주일 간격으로 공복 시 혈당을, 3주 후에는 당내성과 혈장 인슐린 농도를 측정하고 췌장 조직에 면역조직화학 염색을 실시하였다. 실험동물은 정상 대조군(NC), 당뇨 대조군(DC), 2% OF 급여군(OF-2), 5% OF 급여군(OF-5)으로 구분되었으며, NC와 DC는 기초식이를, OF-2와 OF-5는 기초식이에 각각 2%와 5%의 OF를 섞어서 급여하였다. 실험 개시 후 1주마다 12시간 절식시켜 꼬리정맥에서 혈액을 채취하여 공복 혈당을 측정하였다. 실험 3주 후 12시간을 절식시켜 glucose(50 mg/kg BW)를 복강주사한 다음, 30, 60, 90, 120분 경과 후에 혈당을 측정하여 당내성을 측정하였고, 심장에서 혈액을 채취하여 혈중 인슐린 함량을 분석하였다. 또한 췌장 조직에 대해 면역조직화학 염색을 실시하여 조직학적인 변화를 알아보았다. 3주간의 공복 시 혈당은 OF-5와 OF-2 모두 유의적으로 감소하였다(p<0.05). 당내성 측정 결과, OF 급여군은 DC와는 달리 혈당 농도의 변화 추이가 NC와 유사하였으며, 특히 OF-5는 OF-2에 비해서도 혈당 강하 효과가 높았던 것으로 드러났다. 췌장 조직의 면역염색에 의하면, OF의 혈당강하 기작은 췌장 Langerhans' Islet의 ${\beta}$-세포를 생성시키고, ${\beta}$-세포의 사멸을 억제시켜 인슐린의 분비를 정상화시키는 것이었으며, 이러한 결과는 혈장 인슐린 함량의 증가로 재확인할 수 있었다. 결론적으로 OF는 I형 당뇨에서 현저한 혈당 강하 효과 및 Langerhans' Islet의 ${\beta}$-세포수를 회복시켜줌으로써 I형 당뇨의 치료에 효과가 있을 것으로 사료된다.
