Potato dextrose broth was the best inoculation medium to produce in vitro synnemata of Paecilomyces tenuipes. The optimum temperature for synnemata formation of P. tenuipes was $20^{\circ}C$ under 500 lx of light intensity. Highest synnemata production was observed at 18 hr of light period per day. The medium containing 50 g of each silkworm pupae and brown rice produced highest number of synnemata. The optimum ratio of brown rice to distilled water was found as 1:1. Mycelial growth and synnemata production of P. tenuipes was faster and higher in medium containing grinded pupae as compared to whole pupae. The optimum inoculum amount per bottle of medium was 15ml. The highest synnemata production of P. tenuipes was obtained by incubating rice pupae medium at $24^{\circ}C$ until mycelium grows sufficiently after inoculation and then transferring it to $20^{\circ}C$ chamber till harvest.
The optimum concentrations of pH, sucrose and vitamins for the growth and tropane alkaloid production of hairy root clone DTLA9 (best growth line) were investigated. The optimum pH in growth and tropane alkaliod production of DTLA9 clone in SH (Schenk and Hildebrandt, 1972) basal medium without growth regulator were pH 6.3 and 6.5, respectively. Also, the optimum sucrose concentration in growth and tropane alkaliod production in the same medium were 3.0 and 2.8%, respectively. The optimum concentrations of ascorbic acid, D-pantothenate, nicotinic acid, pyridoxine, riboflavin, and thiamine on the growth of DTLA9 clone in SH basal medium without vitamins were 0.1 mM, 0.003 mM, 0.07 mM, 0.002 mM, 0.025 mM, and 0.01 mM, respectively. In particular, supplement of 0.1 mM ascorbic acid to SH basal medium without vitamins stimulated the tropane alkaloid production.
A new synthetic medium was developed for the submerged mycelial cultures of Phellinus linteus. The medium for maximum mycelial growth of Phellinus linteus (3 days incubation, 28$^{\circ}C$, pH 5) consisted of (per 1 L): glucose, 90 g peptone, 10 g soluble starch, 10 g yeast extract, 3 g KH2PO4, 1 g MgSO4.7H2O, 1 g and CaCl2, 0.1 g. The concentrations of glucose, peptone, yeast extract, KH2PO4, MgSO4.7H2O, and CaCl2 were examined in the ranges of 10~90 g/L, 0~10 g/L, 0~15 g/L, 0~2 g/L, 0~1 g/L, and 0~0.5 g/L, respectively. The dry weight of mycelium in 3 days increased to 16.79 mg/mL using the new synthetic medium. The optimum temperature for mycelial growth of Phellinus linteus was 28$^{\circ}C$. The concentrations of KH2OP4, CaCl2, and yeast extract, which gave the maximum mycelial growth of Phellinus linteus, existed in the concentration ranges examined in this study. But, in the cases of other compositions (MgSO4.7H2O, peptone, and glucose), the mycelial growth of Phellinus linteus increased with the concentration in the ranges.
This study was carried out to obtain the basic data for artificial culture of veiled lady mushroom (Dictyophora spp). The optimal conditions for the mycelial growth were $25^{\circ}C$ and pH 5.0 for all isolates except the optimal temperature of $30^{\circ}C$ for D. echinovolvata ASI 32002 and Phallus rugulosus. The optimal medium for Dictyophora spp. was PBA (potato bamboo sawdust extract agar) medium. The strain ASI 32002, D. echinovolvata, grew faster than. D. indusiata ASI 32003 and Phallus rugulosus ASI 25007 on the medium. Carbon sources such as glucose, maltose and inuline were favorable for stimulating a mycelial growth of the two strains of ASI 32002 and ASI 32003. Asparagine and glutamine appeared to be favorable to the strain ASI 32002 and ASI 32003, where as alanine, one of nitrogen source also favorable to the strain ASI 32002. The optimum C/N ratio of the two isolates of ASI 32002 and ASI 32003 was about 25 : 1 when 2% glucose as carbon source was mixed with the basal medium. While, in the case of 4% as carbon source, the optimum C/N ratio was about 30 : 1.
In the process of oyster mushroom production, loading work of culture medium needs the most intensive labor power. Therefore, development of culture medium machine causes to reduce the manpower and cost. The main objective of this study is to develop the culture medium loading machine and investigate the optimal operation conditions and to evaluate the economic value of the machine. The results are summarized as follows: 1. Optimum transporting velocity of the conveyor was 0.61 m/s 2. Optimum speed of blower was 3183 rpm at the transporting velocity of 0.61 m/s with the loading quantity of 3.41 t/hr 3. Recommendable opening area ratio of pressure controller was 1/2 at the blower speed of 3183 rpm and the transporting velocity of 0.61 m/s 4. The break even point resulted in $240\;m^2$ of cultivating area compared to the method of with portable workbench, and $350\;m^2$ of cultivating area compared to the method of with a tractor and a truck.
The embryogenesis stimulating activity(ESA) had been shown in co-culture of embryos with bovine oviduct epithelial cell(BOEC) and culture in BOEG-conditioned medium. The present study was undertaken to purify and quantify the embryotropic proteins and to determine the optimum concentration of the embryotropic protein for the proper development of embryos. In BOEC-conditioned medium, five major bands of proteins were detected(66, 53, 40, 32 and 24 kDa) by SDS-PAGE. From these proteins, 288pg of protein that had a 32kDa molecular weight was purified by gel filtration column and perfusion chromatography ion-exchange column. When purified protein was supplemented to the in vitro culture media at various concentrations in protein-free media, 2.5$\mu$g /ml supplement group showed significantly higher rates of embryo development into morula /blastocyst stages than other groups(p<0.05). In conclusion, we purified 32kDa protein from BOEC-conditioned medium and this protein showed optimum embryogenesis stimulating effect at 2.5$\mu$g /ml.
Metacordyceps yongmunensis is a newly reported species from Korea, which is very similar to Cordyceps species in morphological characters. It grows on large lepidopteran pupa, and numerous white stromata grow on a single host. Mycelial growth characteristics of M. yongmunensis isolates were studied in different media and at different temperatures. Also, different carbon sources, nitrogen sources, and mineral salts were tested for mycelial growth of M. yongmunensis. Schizophyllum (mushroom) genetics complete medium plus yeast extract, Schizophyllum (mushroom) genetics minimal medium, and Martin's peptone dextrose agar produced longer colony diameters and more compact mycelial density than other media. The optimum temperature for mycelial growth was $25^{\circ}C$. Carbon sources such as sucrose, soluble starch, dextrose, glucose, dextrin, maltose, and fructose showed better mycelial growth, whereas peptone, yeast extract and tryptone resulted in the best mycelial growth of all of the nitrogen sources tested. All of the mineral salts tested showed similar growth as the control, except $K_2HPO_4$ which showed longer colony diameter and more compact mycelial density. The compact colonies were white and cottony with a greenish margin. The results showed that M. yongmunensis is an easy fungus to growas it grew from 30 to more than 50 mm in 2 wk.
This study examined the dye-properties of natural fabrics dyed with Coptis chinensis and Curcuma longa root fermented with fungi. The optimum culture conditions for the fermentation of microorganisms, the relationship between natural dye color and fermentation conditions were investigated. Two different medical herbs (ground to 80-100 mesh in size) were used as a natural dyeing source. Phellinus linteus (P. linteus), which can grow in different media, such as Agarmedium (only agar containing medium), maltose extract agar (MA) and potato dextrose extract agar (PDA) culture media, were isolated from the medium. P. linteus was confirmed to be the optimum microorganism for the fermentation of Coptis chinensis and Curcuma longa, and the MA medium was confirmed to be the best for culturing. When using the microorganism as the fermenting agent, $32^{\circ}C$ was found to be the optimum fermenting temperature for both natural colorants. Regarding the dyeing property of the fermented natural dye, silk was dyed quite darkly in an appearance by naked eye estimation and the K/S value in the color strength of silk reached a high level of 16 after the fermenting process. The washing fastness of dyed silk after treatment washing was reduced from 4 to under4 and indicates that dyed silk with fermented plant was not unsubstantial. The light fastness was 1 to 2, showing intended to maintain due to the fermentation process.
Pongamia pinnata Pierre is a tree legume, having potential in production of raw material for biodiesel. A protocol for in wk propagation of this plant was standardized using seedling explants. Growth regulators (GR) including gibberellic acid $(GA_3),\;N^6-benzylaminopurine(BA)$, thidiazuron (TDZ), and Adenine sulphate (Ads) were tested for optimum germination of seeds. Removal of seed coat prior to germination, controlled fungal growth partially but enhanced bacterial growth. Antibiotic cefotaxime was ineffective in controlling bacterial contamination. Seedling derived nodal explants and cotyledon nodes with attached cotyledons were excised and cultured for induction of shoots. Optimum sprouting and multiplication of shoot buds were obtained in MS medium supplemented with $8.88{\mu}M$ BA. These buds differentiated and rooted on medium devoid of GR. Optimum growth of Pongamia seedling was obtained in cotton plugged culture vessels. Reculturing of the cotyledon node explants produced more shoots from the same site. This process of removing shoots and reculturing of cotyledon node was followed for eight passages yielding 4 to 8 shoots in each cycle. The shoots (75%) rooted on half strength MS basal medium supplemented with 0.22% charcoal. All plants survived on transfer to soil. This is the first report on in vitro regeneration of Pongamia pinnata. This report demonstrates the possibility of coupling more than one parameter in single experiment to hasten the process of standardization. The process of cycling the nodal explant repeatedly for production of large number of shoots from single meristem may find application in genetic transformation experiments wherein meristems are used for transformation.
In this study, the medium optimization for cell growth and metabolite formation of Haematococcus sp. under mixotrophic cultivation was investigated. As a result, modified MS medium was selected as the basal medium; glucose was selected as the carbon source, with an optimum concentration of 10 g/l, and potassium nitrate was chosen as the nitrogen source, with an optimum concentration of 1.9 g/l. Under optimum conditions, Haematococcus sp. demonstrated an increase in biomass concentration from 0.18 gDW/l to 5.58 gDW/l in 14 days, after which there was a 31-fold increase in its growth. At the same time, the concentrations of chlorophyll and carotenoids were 172.16 mg/l and 42.33 mg/l, respectively. This work will contribute to the basic data for mass cultivation of microalgae.
