Bacteriocin-producing lactic acid bacteria, Lactococcus sp. HY 449, was isolated from dairy products. Using response surface methodology, the various concentrations of medium compo- nents (tryptone, glucose, yeast extract, tween 80, and initial pH) were tested to find the optimum conditions for maximum bacteriocin production and growth of Lactococcus sp. HY 449. Central composite design was used to control the concentrations of medium components in the experiment. Bacteriocin production and cell growth of Lactococcus sp. HY 449 were most affected by glucose and yeast extract. Estimated optimum growth conditions of Lactococcus sp. HY 449 were as follows; tryptone 1.08%, glucose 1.129%, yeast extract 0.674%, tween 80 0.11%, and initial pH 7.19. Also estimated optimum conditions for bacteriocin production were tryptone 0.937%, glucose 1.108%, yeast extract 0.163%, tween 80 0.09%, and initial pH 6.98.

This paper has presented an application of an optimum grinding conditions based on the fuzzy logic. Fuzzy logic can handle vague and uncertain knowledge, and presents a scheme for integrating data with various kinds of grinding data. Especially, this research is capable of determining the grinding conditions taking into account some fuzzy membership function represented for trapezoidal form such as hardness and surface roughness of workpiece, material tensile strength and elongation, and requirement of grinding method. Larsen's fuzzy production method utilizing the fuzzy production rule can be applied on the establishment of grinding conditions, and also the output value obtained by the center of gravity method can effectively utilize the optimum grinding conditions.
Optimum conditions for the production of acetoin and ammonia as the precursors of tetramethylpyrazine(TMP) were determined using Lactococcus lactis subsp. lactis biovar. diacetylactis FC1 in a modified Lactose-citrate broth containing galactose, citrate, and arginine. The cell growth and the productivity of acetoin and ammonia were remarkably increased in an aerobic culture with 10 $\mu M$ of hematin. For the optimum conditions of acetoin and ammonia production, the concentration of citrate and arginine were adjusted to 156 mM and 50 mM after 18 hr cultivation, and citrate and galactose to 156 mM and 50 mM after 36 hr cultivation, respectively. In these conditions, acetoin and ammonia were produced to the final concentration of 127 mM and 195 mM, which were the highest concentations, respectively. The optimum conditions of the TMP production were also determined as follows; 4 hours at 121, pH 8.3, and the maximal yield of TMP under these conditions was 0.81 g/l.
For waste recyle, we were investigated on Gardenia blue color production using Gardenia by-product by Bacillus subtilits. Optimum conditions for producing blue pigment were found to be 30$^{\circ}C$, initial pH 6.5, glucose as a carbon source 3% and yeast extract as a nitrogen source 0.5%, respectively. Optimum conditions for fermentor culture were agitation speed 400rpm, aeration 2 vvm and inoculum 5%. The optimum perculture time for inoculum was 20 hrs for blue pigment production.
Optimum conditions of the cellulose-hydrolysis reaction with mixed enzymes(cellulase extracted from Penicellium funiculosum mixed with $\beta$-glucosidase extracted from Almod) were investigated to increase the production of glucose from cellulose. Experimental result showed that optimum conditions fro pH, activity ratio of $\beta$-glucosidase to cellulase, concentration of mixed enzymes, concentration of cellulose as a substrate, and temperature range were 4.2, 0.4, 0.8, U/mL, 40 g/L, and 37$\pm$3$^\circ C$, respectively. In these conditions, quantities of glucose productions by using mixed enzymes were larger than those by using cellulase at optimum conditions.
Schizophyllum commune의 균체 생육 및 다당체 생성에 미치는 여러 가지 배지성분의 영향을 조사하였다. 탄소원으로는 산업적인 경제성과 높은 다당체 생성량을 나타낸 glucose를 선택하였고, 무기질소원은 유기질소원에 비해 현저히 낮은 다당체 생성량을 나타내었으며, 그 결과 균체량과 다당체 생성량에서 높은 효과를 나타낸 yeast extract를 질소원으로 선택하였다.
We examined optimum culture conditions of Rhodopseudomonas sphaeroides B5 for effective utilization of substrate and sunlight for hydrogen production. The optimum concentration range of DL-lactate as electron donor for hydrogen production by resting cells was from 5 to 50mM, and optimun CN ratio (lactate/glutamat) for maintenence of hydrogen production activity by growing cultures was from 5 to 6. Hydrogen production by the cultures of low cell density (0.36mg/ml dry cells) was saturated with 10 Klux light intensity. Under constant illumination of 50Klux which was set up as the average medium value of annual variation of sunlight intensity, hydrogen production with various cell densities in the culture resulted in highest production rate (132${\mu}$l/hr/mg dry cells) up to 0.64mg/ml dry cells. However, the amount of total hydrogen production was saturated with cell density of 2.1mg/ml dry cells. In addition to these, the optimum inner thickness pervious to light of the culture vessel for hydrogen production which was measured under sunlight was 5 cm.
For the efficient production of a new exo-polysaccharide from Ganoderma lucidum ASI 7004, the optimum conditions and methods in submerged cultivation were investigated with an airlift fermenter system. The optimum aeration rate was 2.5 Wm at the initial pH 5.0 and 28$^{\circ}C$. The increase of dissolved oxygen concentration by pure oxygen supply during cultivation did not improved the exo-polysaccaride production and the mycelial growth. The maximum exo-polysaccharide production and the mycelial growth under the optimum culture condition were obtained in media of glucose 60g/L, yeast extract 6g/L, (NH4)2HPO4 1g/L and KH2PO4 0.5g/L. Under these optimum medium and culture conditions, about 7.15g/L of exo-polysaccharide and 13.9g/L of mycelial growth were producted, respectively.
For Trametes sp. CJ-105, a kind of white-rot fungi which was collected from the mountain of Korea and was proven to be effective in decolorizing a wide range of structurally different synthetic dyes, the optimum conditions for mycelial growth and laccase(E.C. 1.10.3.2) production were investigated. Among various carbon sources, glucose showed the highest potential for the mycelial growth and laccase production, the optimum concentration being 2% glucose. For the nitrogen source, asparagine was good for the mycelial growth, while ammonium tartrate for laccase production(optimum concentration: 0.04%). The addition of thiamine and biotin increased both th emycelial growth and laccase production. When 2,5-xylidine was added as an inducer after the first day of culture, the production of alccase was seven-times higher than that in the absence of the inducer. The optimum pH and temperature conditions for laccase production by Trametes sp. CJ-105 were pH 5.0 and $25^{\circ}C$, respectively. In the 5L fermentation, the production of laccase reached a maximum of 340U/ml at the time when the ammonium ion was being rapidly depleted.
The optimum conditions of the production of exo-polymer by Ganoderma lucidum WK-003 and it's hepatoprotective effect was studied. Optimum conditions for the production of exo-polymer (3.18 g/$\ell$) by using shaken flask culture of G. lucidum WK-003 were pH 4.5, 30$^{\circ}C$, 120 rpm for 18 days cultivation. Also exo-polymer productio (7.15 g/$\ell$) was optimized by 5$\ell$ jar fermenter cultivation with condition of pH 4.5, 30$^{\circ}C$, 200 rpm, 1.0 vvm for 6 days cultivation. Glutamic pyruvic transaminase(GPT) activities in serum of intoxicated Sprague-Dawley rat were decreased from 704 IU/L to 330 IU/L by oral administration of the exo-polymer (20mg/kg$.$day)f or 4 consecutive days.
