A Study on the Application in Site of the Concrete Using Fly Ash Produced in Combined Heat Power Plant (열병합발전소 플라이애쉬를 사용한 콘크리트의 현장적용에 관한 연구)

  • This study dealt with the applicability and quality control of the concrete using fly ash produced in combined heat power in a construction site. Firstly, chemical and physical characteristics of the fly ash produced in combined heat power plant re analysed. Also, after investigating the properties of flesh and hardened concrete through various experiments, the fly ash concrete was placed in depositing construction in Ulsan Petrochemical Service Co. This field application was focussed on the quality control system in the site as production, placing and curing of concrete. As the result of this study, the quantity of CaO in the fly ash is relatively high based on the chemical analysis. The fly ash concrete showed slumping maintenance and high viscosity properties in the optimal mixing conditions (W/B:44~45%, S/a:$45\pm$2%, W:185kg/m). And, quality control and assurance of the fly ash concrete in actual site were verified by various testing methods.

  • In order to suggest the proper optimal conditions of the CNC milling machining for the Thin-wall surface, some experiments were carried out. The process was applied in the aerospace industry for the machining of light alloys, notably aluminium. In recent year, however, the mold and die industry has begun to use the technology for the production of components, including those manufactured from hardened tool steels. And the end mill is an important tool in the milling process. A typical example for the end mill is the milling of pocket and slot in which a lot of material is removed from the workpiece. Therefore the proper selection of cutting parameter for end milling is one of the important factors affecting the cutting cost. In this paper, we choose the optimal parameters(cutting forces) to cut thin-walled Al part by experiment.

  • The quality control literature contains a substantial number of articles concerned with how to optimally choose control limits in order to minimize production cost. The purpose of the this study is to determine the economic setting for the process mean of an industrial process. In this study it is assumed that the lower control limit is set by government regulations and the u, pp.r limit and process mean are chosen based on economic considerations. Much research has been conducted on this problem under the condition of the fixed rpm(rate per minute). However a variance can be increased in proportion to the level of rpm and the increase of the variance can change the optimal process mean. Therefore, it is desirable to determine both the process mean and the level of rpm simultaneously. In this paper, a mathematical model is presented which considers the u, pp.r limit and the rpm as variables.

Isolation and Characterization of Protease Producing B. amyloliquefaciens JH-35 from Food Waste (음식물 쓰레기로부터 Protease를 생산하는 B. amyloliquefaciens JH-35의 분리 및 특성)

  • BACKGROUND: Recent studies have described the importance of microbes and enzymes that can compost food waste. This study was carried out to improve production of protease of isolated microbes from food waste. METHODS AND RESULTS: Seven bacteria isolated from various sources were screened for protease production by adding skim milk into the agar medium. About 7 microbes producing protease were tested, and strain JH-35 showed the highest protease activity among them. The strain was identified as Bacillus amyloliquefaciens JH-35 based on morphological, cultural, physiological characteristics and 16S rRNA. In the fermentation experiment, the assay B. amyloliquefaciens JH-35 showed the highest protease activity in the condition of 1% glucose, 1.5% yeast extract and 0.2%$ K_2HPO_4$. The optimal condition of culture with temperature $35^{\circ}C$, initial pH of 7 and shaking speed of 200 rpm and 24 hr. CONCLUSION: The protease of the B. amyloliquefaciens JH-35 had its activity at pH 7 and the optimal culture time was 24 hr. Also, B. amyloliquefaciens JH-35 was high salt tolerance. Our results suggest that B. amyloliquefaciens JH-35 from food waste may have the potential to degrade protein and carbohydrate in food waste.

Study on Oil Production from Pyrolysis of Mixed Plastic Waste Using Multidimensional Chromatography (Multidimensional Chromatography/Mass Spectrometry를 이용한 혼합 폐플라스틱의 열분해 오일 특성 평가에 관한 연구)

  • The total hydrocarbon distribution of oil products obtained from the pyrolysis of four kinds of mixtures of polyethylene-polystyrene waste has been studied by multidimensional chromatography(high performance liquid chromatography followed by capillary gas chromatography)/mass spectrometry. Saturated, unsaturated and aromatic hydrocarbons in oil products were selectively pre-separated according to structural groups by HPLC and the weight fraction of each group was estimated by analysis of each component using GC-FID response factors. The hydrocarbon distribution of aliphatic fraction consists of $C_{5}$ to $C_{25}$ saturated and unsaturated hydrocarbons. And that of aromatics fraction consists of benzene, toluene, xylene, styrene, propenyl benzene, naphthalene, and some of derivatives. Pyrolysis temperature did not affect the ratio of total weight fraction of aliphatic over aromatic hydrocarbon distribution in case of PS only and PE-PS mixtures (1:1 and 1:4 wt. ratio) as a feed while affected the ratio of total wt. fraction in case of PE only. The optimal temperature for the maximum oil production was $600^{\circ}C$ for pyrolysis of PS and 1:1 and 1:4 mixtures of PE and PS. The optimal condition for aromatic recovery was $600^{\circ}C$ with 1:1 mixture of PE and PS. In this condition, aromatic was produced up to 90% of total oil product. The maximum yield of toluene, xylene, styrene, and propenyl benzene were 8.6, 8.9, 51.0 and 7.4% of feed for pyrolysis PS at $700^{\circ}C$, respectively. However, only 1.3% naphthalene was recovered at $700^{\circ}C$ with 1:1 PE:PS(by wt.).

Optimal Condition for Citric Acid Production from Milk Factory Waste Water by Using the Immobilized Cells of Aspergillus niger (고정화 Aspergillus niger 세포를 이용한 우유공장 폐수로부터 구연산 생산의 최적 조건)

  • Immobilized cells of Aspergillus niger was employed to produce citric acid by fermentation of milk factory waste water. A. niger ATCC 9142 as a citric acid production strain was cultured for 3 days and was entrapped with Ca-alginate bead about 2.5∼3.5 mm. The optimal pH and temperature were estimated to be 3.0 and $30^{\circ}C$, respectively. Dilution rate for fermentation was calculated to be $0.025 h^{-1}$ . Maximum amount of citric acid was obtained at 4.5 g/$\ell$ with the optimized fermentation condition. The yield of citric acid produced by immobilized A. niger ATCC 9143 was 70.3%. The yield was increased by 20% with immobilized cell, compared to that of the shake flask culture. Hence, the milk factory waste water is worthy to be used for the substrate of citric acid fermentation.

Production of Inulase Using Jerusalem Artichoke Tuber Extract (돼지감자 추출물을 이용한 이눌라아제 생산)

  • To produce inulase from Kluyveromyces fragilis No.351 using Jerusalem artichoke tuber extract, optimization of the condition was conducted. As results, the optimal concentration of artichoke tuber extract was 3.5 % and bactocasitone showed better production than yeast extract. The optimal temperature and pH were $30^{\circ}C$ and 5.5, respectively. The addition of $NH_4H_2PO_4$ increased the enzyme production. Inulase synthesis was growth-associated and the enzyme production increased as concentration of dissolved oxygen increased. A higher quantity of industrial grade inulase was prepared by the combination of ultrafiltration and ethanol precipitation with 72% recovery.

  • In the present work, production of organophosphorus hydrolase (OPH) that is secreted in periplasmic space of recombinant Escherichia coli was performed for degradation of environmental toxic organophosphate compounds, paraoxon. The optimal conditions for enhancement of OPH production were 1.0 mM isopropyl-${\beta}$-D-thiogalactopytanoside (IPTG), 0.25 mM $Co^{2+}$, and 0.1 mM ethylenediamine tetraacetate (EDTA). Under these culture conditions, the maximum OPH production was $174Unit/L{\cdot}OD$. In addition, 1 mM of paraoxon was completely degraded by OPH. These results can be used as a bioremediation tool for removal of environmental toxic organophosphate compounds remaining in soil and aquatic environment.

Isolation of Pseudomonas putida BM01 Accumulating High Amount of $PHA_{MCL}$

  • A Pseudomonas putida strain able to accumulate high amount of polyesters of medium-chain-length 3-hydroxyalkanoic acids ($PHA_{MCL)$) was isolated from soil in a landfill site using an enrichment technique. Culture condition of the isolated strain for polyester production in a one-step culture was optimized in a mineral-salts medium against pH and concentrations of ammonium sulfate, carbon source(e.g., octanoate), and phosphate. The optimal values for maximal cell growth and PHA accumulation were: pH; 7$\sim$8, $(NH_4)_2SO_4$; 8 mM, octanoate; 40 mM. The optimum temperature was in the range of $20\sim30^{\circ}C$, which was rather broader than in other bacteria. Cell growth was strongly inhibited by the phosphate limitation to less than 1 mM. An increase of phosphate concentration above 1 mM showed little effect on cell growth and polyester accumulation. When the strain was grown on octanoate under this optimized condition it produced 3.4 g dry biomass per liter and yielded 1.7 g PHA per liter amounting to 53 wt% of dry cells. The monomer units composing the polyester synthesized from octanoate were 3-hydroxyoctanoate (3HO), 3-hydroxycaproate (3HC), and 3-hydroxybutyrate (3HB) (85:13:2, mole ratio). Other low linear $C_3\simC_{10}$ monocarboxylic acids were also tested for polyester production.

The Optimal Design of POF Optical Connector for Medical Image Transmission System (의료영상전송시스템을 위한 POF 광커넥터의 최적 설계)

  • For mass information transfer, the optical communication using optic fiber has been widely used. Especially, in the field of medical image, the large data is digitalized based on the standard image and it is used for telemedicine with this method. Therefore, to transfer the large amount of data fast and effectively POF (Plastic Optical Fiber) can be used and the development of optic connector for connection between POFs is very important. In this study, for stable optical coupling of POF optic fiber Ferrule and Sleeve were designed and produced by considering the bond stability and the insertion loss according to the physical contact and roughness profile was evaluated. As a result of examining the insertion loss by physical contact method of two optic fibers, it showed the loss was about 1.895dB. According to the results from studying the condition of grinding section for POF mass production, the mass production condition was established as POF profile roughness of 6nm and the loss of 0.2dB or lower by controlling the film size and time step by step.
