The large process plant is currently implementing predictive maintenance technology to transition from the traditional Time-Based Maintenance (TBM) approach to the Condition-Based Maintenance (CBM) approach in order to improve equipment maintenance and productivity. The traditional techniques for predictive maintenance involved managing upper/lower thresholds (Set-Point) of equipment signals or identifying anomalies through control charts. Recently, with the development of techniques for big analysis, machine learning-based AAKR (Auto-Associative Kernel Regression) and deep learning-based VAE (Variation Auto-Encoder) techniques are being actively applied for predictive maintenance. However, this predictive maintenance techniques is only effective during steady-state operation of plant equipment, and it is difficult to apply them during start-up and shutdown periods when rises or falls. In addition, unlike processes such as nuclear and thermal power plants, which operate for hundreds of days after a single start-up, because the pumped power plant involves repeated start-ups and shutdowns 4-5 times a day, it is needed the prediction and alarm algorithm suitable for its characteristics. In this study, we aim to propose an approach to apply the optimal predictive alarm algorithm that is suitable for the characteristics of Pumped Storage Power Plant(PSPP) facilities to the system by analyzing the predictive maintenance techniques used in existing nuclear and coal power plants.
Objective: This study aimed to assess the impact of the dilution ratio of Tris diluent, storage at 0℃, and long-distance transportation on the spermatozoa of Simmental cattle. It also validated the feasibility of the regional distribution of fresh semen. Methods: In experiment 1, semen was diluted at four dilution ratios (1:6, 1:9, 1:12, and 1:15) to determine the optimal dilution ratio of Tris diluent. In experiment 2, we assessed sperm viability, progressive motility (objectively assessed by computer-assisted sperm analyzer), and acrosome intactness in Tris dilutions kept at constant 0℃ for 1, 3, 6, 9, and 12 days. We compared them to Tianshan livestock dilutions (Commercial diluent). In experiment 3, semen was diluted using Tris diluent, and sperm quality was measured before and after long-distance transport. Artificial insemination of 177 Simmental heifers compared to 156 using Tianshan Livestock dilution. Results: The outcomes demonstrated that 1:9 was the ideal Tris diluent dilution ratio. The sperm viability, Progressive Motility, and acrosome integrity of both Tris and Tianshan dilutions preserved at 0℃ gradually decreased over time. sperm viability was above 50% for both dilutions on d 9, with a flat rate of decline. The decrease in acrosome integrity rate was faster for Tianshan livestock dilutions than for Tris dilutions when stored at 0℃ for 1 to 6 days. There was no significant difference (p>0.05) in sperm viability between semen preserved in Tris diluent after long-distance transportation and semen preserved in resting condition. The conception rates for Tris dilution and Tianshan livestock dilution were 49.15% and 46.15% respectively, with no significant difference (p>0.05). Conclusion: This shows that Tris diluent is a good long-term protectant. It has been observed that fresh semen can be successfully preserved for long-distance transport when stored under 0℃ conditions. Additionally, it is feasible to distribute semen regionally.
This study was to establish in uitro culture system of mouse preantral follicles and to obtain higher in vitro development rates and production of live young. Preantral follicles were obtained from 12-day-old FI mouse (C57BL $\times$ CBA) by enzymatical methods. Oocyte-granulosa cell complexes (OGCs) of preantral follicles were loaded on Transwell-COL insert and cultured in $\alpha$MEM supplemented with 5% FBS, 100 mIU/$m\ell$ FSH and 100 mIU/$m\ell$ hMG for IVG. IVM was performed in $\alpha$MEM supplemented 1.5 IU/$m\ell$ hCG for 18 hrs and IVF was carried out in Ml6 medium. Embryos were cultured in modified Ml6 medium supplemented 10% FBS for 4 days. The effect of the OGCs size on the nuclear/cytoplasmic maturation was significantly higher in 120-150 ${\mu}{\textrm}{m}$ (MII: 33.0%, $\geq$2-cell: 36.7%, $\geq$morula: 20.9%) than in 70-110 ${\mu}{\textrm}{m}$ (MII: 12.2%, $\geq$2-cell: 10.2%, $\geq$morula: 4.8%) (p<0.001). In period of the IVG days, the rate of $\geq$2-cell was significantly higher in 10 days(38.2%) than in 12 days (20.0%) (p<0.01). This experiment was conducted to establish the technology for artificial hay preparation in Korea. Using far-infrared heater, a device that can control temperature, airflow, and far-infrared radiation was produced and conducted on the fourth harvested alfalfa. The drying conditions were carried out by selecting a total of four conditions. For each condition, the radiation rate was set to around 40% (33-42%), and the temperature was set at 58~65℃, and the speed of the airflow was fixed at 60m/s. The overall drying time was set to 30 min in the single and 60 min (30-30 min) and 90 min (30-30-30 min) in the complex condition, and the radiation rate and temperature were changed by time period. In the case of drying condition 1, the final dry matter (DM) content was 46.26%, which did not reach a DM suitable for hay. However, all of the alfalfa corresponding to the remaining drying conditions 2 to 7 showed a DM content of 80% or more, resulting in optimal alfalfa hay production. In power consumption according to the drying conditions, the second drying condition showed the lowest at 4.7 KW, and the remaining drying conditions were as high as 6.5 to 7.1 KW. The crude protein content was found to be high at an average of 25.91% and it showed the highest content in the 5th drying condition (26.93%) and the lowest value in the 6th drying condition (25.16%). The digestibility showed a high value with an average of 84.90%, and there was no significant difference among treatments (p>0.05). Considering the above results, it was judged that drying condition 2 was the most advantageous.
To enhance the value as a feedstuff of distiller's dried grain (DDG) and develop fermented feedstuff, we investigated the effects of the culture conditions affecting glucoamylase activity, such as pH in submerged culture and moisture content in solid-state culture. Also, we investigated the optimal mixing ratio of DDG and wheat bran for the production of fermented feedstuff containing high content of amino acids. In culture conditions for high fermented activity, pH and moisture were optimum at pH 4 and 60%, respectively. In the case of mixing ratio, the glucoamylase activity was decreased with increase of DDG content. On the other hand, the content of crude protein was increased slowly. For the development of fermented feedstuff, the optimal mixing ratio of DDG and wheat bran was 1 to 4. Finally, we could produce approximately 1 ton (dry matter) of trial product in incubator of pilot-scale. The glucoamylase activity and the crude protein content were 1,024 U/g and 33.6%, respectively.
Producing organic farm products is one of the high-payoff farming practices considering the rapid increase of consumer's purchasing power. The purpose of this study is to identify the optimal organic farming system in relation to farm income. To cope with the above objectives, present farming conditions and profitability of 15 farmers producing organic products including rice and leaf vegetables as lettuse and cabbage were surveyed in Hongsung and Ahsan Gun, Chungnam Province. Based on the surveyed data, maximization of organic farm income were analyzed under the constrained conditions such as limited monthly labour inputs and competitive use of land by Linear Programming Model. The results of this study can be summarized as follows. 1. In the profitability analysis of 15 farmers producing organic products, rice farmers could earn their farm income more than that of conventional farmers by 50%. On the other hand, the controlled lettuce farmers could get more about 100% than that of it. But the controlled cabbage farm could get more about 40% of it. These organic farm products were saled at high prices comparing with the prices of conventional farm products by 170% ~ 230% even though the crop yields produced by organic materials had decreased. 2. According to the labor requirement of the organically produced crops, rice cultivation was needed more labour inputs than the conventional farming method by 130%. On the other hand lettuce and cabbage could saved labour requirement by 40% and 80% of those respectively. Especially for the rice cultivation concern, higher labor requirement was due to the activities as organic fertilization, soil preparation and pest and weed controll, etc. 3. With the surveyed data from farmers who produced farm commodities, L.P. analysis was implemented to find out the optimul farming system and the maximum income. According to the results of L.P. analysis, 58% of total farm income could get more than the conventional farming system In the case of rice cultivation, one ha of paddy was recommendable to save more labour inputs than the controlled leaf vegetables such as lettuse and cabbage. However, in the controlled leaf vegatables, only 73% of total upland equivalent to 0.3 ha should be cultivated under the condition of labor shortage as the restricted 4 workers. And increasing the size of hogs raising should be recommened to achieve self-sufficiency of organic fertilizer. As pointed out the possibility of organic farming, present farmers producing organic farm products could be able to maximize their income by expanding organic farm size with regard to all conditions of our organic industry. Of course, there are many difficulties in the course of developing organic agriculture. So Government should effort to support the development of our organic agriculture considering the various aspects of production, marketing, Quality certification of organic produces.

This study was designed to determine the effect of electric field strength, duration and fusion buffer in fusion parameters on the rate of membrane fusion between the somatic cell and cytoplast for Korean cattle (HanWoo) somatic cell nuclear transfer (SCNT) procedure. Following electrofusion, effect of 5 or $10\;{\mu}M$$Ca^{2+}$-ionophore of activation treatment on subsequent development was also evaluated. Cell fusion rates were significantly increased from 23.1% at 20 V/mm to 59.7% at 26 V/mm and 52.9% at 27 V/mm (p<0.05). Due to higher cytoplasmic membrane rupture or cellular lysis, overall efficiency was decreased when the strength was increased to 30 V/mm (18.5%) and 40 V/mm (6.3%) and the fusion rate was also decreased when the strength was at 25 V/mm or below. The optimal duration of electric stimulation was significantly higher in $25\;{\mu}s$ than 20 and $30\;{\mu}s$ (18.5% versus 9.3% and 6.3%, respectively, p<0.05). Two nonelectrolyte fusion buffers, Zimmermann's (0.28 M sucrose) and 0.28 M mannitol solution for cell fusion, were used for donor cell and ooplast fusion and the fusion rate was significantly higher in Zimmermann's cell fusion buffer than in 0.28 M mannitol (91.1% versus 48.4%, respectively, p<0.05). The cleavage and blastocyst formation rates of SCNT bovine embryos activated by $5\;{\mu}M$$Ca^{2+}$-ionophore was significantly higher than the rates of the embryos activated with $10\;{\mu}M$ of $Ca^{2+}$-ionophore (70.0% versus 42.9% and 22.5% versus 14.3%, respectively; p<0.05). This result is the reverse to that of parthenotes which shows significantly higher cleavage and blastocyst rates in $10\;{\mu}M$$Ca^{2+}$-ionophore than $5\;{\mu}M$ counterpart (65.6% versus 40.3% and 19.5% versus 9.7%, respectively; p<0.05). In conclusion, SCNT couplet fusion by single pulse of 26 V/mm for $25\;{\mu}s$ in Zimmermann's fusion buffer followed by artificial activation with $5\;{\mu}M$$Ca^{2+}$-ionophore are suggested as optimal fusion and activation methods in Korean cattle SCNT protocol.
The optimal conditions for high yields of mycelial protoplasts from P. cornucopiae were established. The concentraion of enzyme system containing Novozym 234, ${\beta}-D-glucanase$ and ${\beta}-glucuronidase$ was $5mg\;ml^{-1}$ each. The osmotic stabilizer most effective for protoplast isolation was O.6 M sucrose. The optimal reaction time of mycelium with the lytic mixture was 90 min in a shaking condition at 120 strokes $min^{-1}$. When the myelium of P. cornucopiae was cultured for 4 days on mushroom complete medium at $28^{\circ}C$, the formation of protoplast was effective. When the pH of the digestion mixture with O.6 M sucrose as stabilizer varied between pH 4.0 and 7.0, the production of protoplasts was effective in phosphate buffer (pH 6.2) and Na-maleate buffer (pH 5.0). Generally, phosphate buffer was more effective for protoplast isolation than Na-maleate buffer, but 0.6 M sucrose osmotic stabilizer without adjusting pH was most effective. Using these conditions, protoplasts from P. cornucopiae were obtained at a ratio $1{\times}10^7\;ml^{-1}$.
For the practical using of liquid spawn we carried out selection test of nutrient sources, cultural methods and cultural apparatus for liquid spawn production of oyster mushroom(ASTI 2001, ASTI 2018, ASTI 2072, ASTI 2016, ASTI 2070, ASTI 2180, ASTI 2183, ASTI 2042). The optimal temperature and pH range for mycelial growth of Pleurotus species were $25^{\circ}C\;to\;30^{\circ}C$ and 5.5 to 6.5, respectively. The effect of carbon sources, nitrogen sources and mineral salts on the mycelial growth was studied using petridish culture. Generally, the disaccharides and polysaccharides showed good effect for mycelial growth of Pleurotus species, and the polysaccharides were superior to the other classes of carbon sources for mycelial growth. For the mycelial growth of the 8 oyster mushroom stains, soybean flour was superior to the other kinds of nitrogen sources. On the other hand, addition of mineral salts did not affect, and even poor under certain mineral salts, the mycelial growth of the 8 oyster mushroom stains. The brown sugar selected out the carbon source of the agricultural medium. Also the soybean flour selected out the nitrogen source of agricultural medium. In the medium selection, we selected out agricultural optimum medium composed of brown sugar 3%, soybean flour 0.3%, potassium phosphate 0.05%, magnesium sulfate 0.05%. Under the 250 ml erlenmeyer culture, the effects of such factors as the inoculum rate, the working volume, cultural method and flask shapes on the mycelial growth were examined. The optimal inoculum rate and working volume on the mycelial growth of oyster mushroom was 2 mycelial disk (diameter 6 mm) and 100 ml, respectively. The shake flask culture was enhanced the mycelial growth than at the erlenmeyer flask. Pulp form growth of mycelium in the erlenmeyer flask culture was obtained in the culture with glass rod of length 50 mm, diameter 10 mm.
In order to know the effects of rearing conditions such as water temperature, salinity, larval stocking density, kinds and amounts of food organism and seedling collection method on seedling production of the brackish water clam, Corbicula japonica, the growth and survival rate of the larvae were investigated at each rearing condition. The shell length of larvae showed faster growth at $24^{\circ}C$ and $27^{\circ}C$, and survival rate showed good results at $18^{\circ}C$ and $21^{\circ}C$. Based on growth and survival rate according to water temperature, the optimal water temperature for larvae rearing was $24^{\circ}C$. At the salinity of more than 3 psu, the growth and survival rate were higher than 0 psu. The optimal salinities for larvae were 6-9 psu. Growth and survival rate of larvae were high at low stocking density and the optimum stocking density of larvae was 10 inds./ml. Daily feeding concentrations of 10,000-20,000 cell/ml of food organisms mixed Pavolva lutheri, Isochrysis galbana and Chaetoceros calcitrans were very effective. Full grown larvae collection from various substratum was significantly higt in sand, especially sand size was less than 0.25 mm.

④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)

제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
