Effect of Sodium Amylosulfate on the Growth of Salmonella typhi (Sodium Amylosulfate의 Salmonella typhi 증식에 대한 영향)

  • Sodium amylosulfate(SAS) has been reported to be an effective substance to inactivate the anti-bacterial activity of blood in blood culture media. The advantage of the use of SAS over sodium polyanethol sulfonate(SPS) is that it does not inhibit the growth of some bacteria! species which are known to be inhibited by SPS. As to S. typhi, SPS is reported to enhance the growth, however the effect of SAS on this organism is not known as yet. Using 43 strains of S. typhi, isolated from clinical materials, the authors tried to determine the effect of SAS on this organism. The methods used for this study were : the SPS and SAS paper disk I sensitivity test, tests on the growth in trypticase soy broth(TSB) with SPS and with SAS, and experimental blood culture in SPS and SAS incorporated TSB. The following results were obtined. 1). S. typhi strains with the turbidity of No. 0.5 tube of MacFarland nepherometer were inoculated onto Mueller-Hinton plate and 1mg disk of SPS and SAS were applied. After 24-hour incubation, none of the 43 strains showed inhibition zone by SPS disk, but all of them showed zones by SAS disk with a mean zone diameter of 9.5mm(Table 1). 2) Inocula consisting of one to 54 viable counts of 37 strains were inoculated into three different media; TSB with 0.05% SPS, TSB with 0.05% SAS and TSB alone. After 24-hour incubation the mean of the optical densities of each medium were 0.483, 0.482 and 0.459 respectively, showing that SAS does not inhibit the growth of S. typhi. Moreover it was shown that there was no correlation between the amount of inocula and growth(Table 2 and Fig. 1). 3). Each set of media in 5 ml amounts consisting of one tube of TSB with 0.05% SPS, one tube of TSB with 0.05% SAS and two tubes of TSB were inoculated with 8, 64. 640 and 6400 viable counts of bacteria. Then 0.5 ml of fresh normal blood was added to all tubes except for one tube of TSB. Macroscopic observation after 24 hour incubation showed a heavy growth in all tubes except for the tube of TSB plus blood, which showed only a light growth in the tube of the heaviest inoculum. This result clearly demonstrates that the growth of S. typhi is inhibited by some antibacterial activities of fresh blood, which are counter acted by SPS and SAS(Table 3). Between SPS and SAS, there was no significant difference found(Table 4 and Fig. 2). With all these results it can be postulated that the addition of SAS into a rountine blood culture media may raise the positivity of S. typhi isolation and shorten the incubation period.

The Thickness Effect on the Determination of Fracture Toughness Jic (파괴인성치 Jic 결정에 대한 시험편 두께의 영향에 관한 연구)

  • Recent Experimental results show that the J integral can be effectively used to obtain a valid parameter for predicting plane strain and plane stress fracture. However, only a few research results have been reported for the effect of thickness where the plane strain state can not be assumed. A purpose of this study is to find the behavior of fracture touhness and tearing modulus varing the specimen thickness. The type of specimen in the present study is compact tension (CT). The thicknesses of the low carbon steel specimens that are used in the experiments are 5, 10, 15, 20 and 25mm. The measurement of crack length is taken by optical measurements method. From the study, the followings are found; 1) The fracture toughness and the tearing modulus which are obtained by using Yoon's and Simpson's formula show more conservative than that by using Rice's and Merkel's. 2) The fracture toughness is increase in specimen thickness which is reached 15mm. Beyond this thickness the fracture toughness is decreased in specimen thickness. 3) In the case of CT specimen with the thickness ranging from 5 to 25mm, the tearing modulus which is applied the same J integral equation is almost constant. 4) By using Yoon's formula, the correlation of the plane slress fracture toughness J sub(C) with specimen thickness B is expressed as the following formula. High Resolution Ocean Color Products Estimation in Fjord of Svalbard, Arctic Sea using Landsat-8 OLI (Landsat-8 OLI를 이용한 북극해 스발바드 피요르드의 고해상도 Ocean Color Product 산출)

  • Ocean Color products have been used to understand marine ecosystem. In high latitude region, ice melting optically influences the ocean color products. In this study, we assessed optical properties in fjord around Svalbard Arctic sea, and estimated distribution of chlorophyll-a and suspended sediment by using high resolution satellite data, Landsat-8 Operational Land Imager (OLI). To estimate chlorophyll-a and suspended sediment concentrations, various regression models were tested with different band ratio. The regression models were not shown high correlation because of temporal difference between satellite data and in-situ data. However, model-derived distribution of ocean color products from OLI showed a possibility that fjord and coastal areas around Arctic Sea can be monitored with high resolution satellite data. To understand climate change pattern around Arctic Sea, we need to understand ice meting influences on marine ecosystem change. Results of this study will be used to high resolution monitoring of ice melting and its influences on the marine ecosystem change at high latitude. KOPRI (Korea Polar Research Institute) has been operated the Dasan station on Svalbard since 2002, and study was conducted using Arctic station.

Downscaling GPM Precipitation Using Finer-scale MODIS Based Optical Image in Korean Peninsula (MODIS 광학 영상 자료를 통한 한반도 GPM 강우 자료의 상세화 기법)

  • Precipitation is closely related to various hydrometeorological phenomena, such as runoff and evapotranspiration. In Korean Peninsula, observing rainfall intensity using weather radar and rain gauge network is dominating due to their accurate, intuitive and precise detecting power. However,since these methods are not suitable at ungauged regions, rainfall detection using satellite is required. Satellite-based rainfall data has coarse spatial resolution (10 km, 25 km), and has a limited range of usage due to its reliability of data. The aim of this study is to obtain finer scale precipitation. Especially, to make the applicability of satellite higher at ungauged regions, 10 km satellite-based rainfall data was downscaled to 1 km data using MODerate Resolution Imaging Spectroradiometer (MODIS) based cloud property. Downscaled precipitation was verified in urban region, which has complex topographical and environmental characteristics. Correlation coefficient was similar in summer (+0), decreased in spring (-0.08) and autumn (-0.01), and increased in winter (+0.04) season compared to Global Precipitation Measurement (GPM) based precipitation. Downscaling without calibration using in situ data could be useful in areas where rain gauge system is not sufficient or ground observations are rarely available.

Static Behavior of Hollow Cantilever Beam Using Multiplexed FBG Sensors (다중화된 FBG센서를 이용한 중공 내민보의 정적 거동 분석)

  • This paper presents a preliminary study to monitor the lateral behavior of pile foundation using multiplexed fiber Bragg grating(FBG) sensors. In the Preliminary study, an 1.7 meter long cantilever beam with the shape of square hollow box was fabricated and tested under the static loading. Four FBG sensors were multiplexed in a single optical fiber and installed into the top and bottom of the cantilever beam. The strains are directly measured from FBG sensors followed by curvature calculations based on the plane section assumption. Vertical deflections are then estimated using the regression analyses based on the geometric relationships. It has been found that excellent correlation with conventional sensing system was observed. The success of the test encourages the use of the FBG sensing system as a monitoring system for pile foundations. However, further consideration should be given in the case of the sensor malfunction for the practical purpose.

Transition Temperature of Mitochondrial Respiratory Activity as an Index for Chilling Susceptivity of Varieties of Rice Plant (미토콘드리아 호흡활성 전이온도의 측정에 의한 벼품종별 상대적 내냉성판정)

  • The transition temperature of respiratory activity of mitochondria isolated from panicles of rice plant was measured with 41 varieties bred in Korea, which had been agronomically classified into five groups based on their relative cold-tolerance; S (strong cold-tolerance), S-M (moderate to strong cold-tolerance), M (moderate cold-tolerance), M-W (moderate to weak cold-tolerance) and W (weak cold-tolerance). The temperature was found to be $11.5{\sim}13.5^{\circ}C$ for group S, $13.5{\sim}16^{\circ}C$ for group S-M, $16{\sim}17^{\circ}C$ for group M, $17{\sim}18^{\circ}C$ for group M-W, and ${\geq}18^{\circ}C$ for group W respectively with a few exceptions of minor deviation, demonstrating a close correlation between the transition temperature and the chilling susceptivity of a variety. This observation supports the suggestion that the transition temperature of respiratory activity of panicles be adopted as a reliable index for grading a rice variety as to its overall cold tolerance. Also studied is on the methodology for measurement of the transition temperature in the aspect of analytical biochemistry. In addition to the usual polarographic method, a spectrophotometric method which is based on the enzymatic assay of membrane-bound cytochrome C oxidase was examined. This enzyme was found previousely and confirmed again in the present work to be a key factor governing the respiratory trantion at least in plant mitochondria. The optical method was turned out to be very successful, although it was a little more complicate and time-consuming than the electrochemical method.

  • This review summarizes advantage and limitation in infrared spectroscopy and computational chemistry to understand rhizospheric interaction among organic acids, oxyanions and metal oxides. Since organic acids and metal oxides determine dynamics of oxyanions in the soil environment, knowledge of fundamental mechanisms is a prerequisite for understanding the interactions at soil-water interface. Attenuated total reflectance-fourier transform infrared spectroscopy (ATR-FTIR) is a powerful tool to measure the interfacial reactions. However, the ATR-FTIR measurements are abstruse, because the optical characteristics for measurements are variable depending on the experimental setup. In addition, spectral overlapping is a primary obstacle to the analysis of the interfacial reaction; thus, it is essential to detect and to deconvolute bands for signal interpretation. In this review, we expained the fundamental principle for spectrum processing, and four band identification methods, such as derivative spectroscopy, two-dimension correlation spectroscopy, multivariate curve resolution, and computational chemistry with example of aqueous phosphate speciation. As a result, spectrum processing and computational chemistry improved interpretation and spectral deconvolution of overlapped spectra in relatively simple systems, but it was still unsatisfactory for the problems in more complexed system like nature. Nevertheless, we believed that our challenge would contribute practically to develop adequate analytical procedure, signal processing and protocols that could help to improve interpretation and to understand the interfacial interactions of oxyanions in natural systems.

The Accommodative Lag and Refractive Error in Early Adults (초년 성인의 굴절이상과 조절래그 분포의 연관성)

  • Purpose: The purpose of this study was to determine the distribution and correlation of accommodative lag with refractive error. Method: We had tested the clinical refraction and the accommodative lag in clinically normal 49 young adults (total 98 eyes) aged 18 to 25 years without abnormal binocular function. Monocular and binocular accommodative lag were tested with 0.50 D cross-cylinder lens and near vision test chart which had cross-hairs after full correction of LogMAR visual acuity over 0.05. Results: There was no statistical differences in monocular accommodative lag between right ($0.64{\pm}0.64$ D) and left eye ($0.63{\pm}0.64$)(p=0.858). The accommodative lag of male was higher than female and the range of the value was broader than female in binocular accommodative lag (p=0.015). The wider the inter-pupillary distance was, the higher the accommodative lag was (p=0.003). However, there were no differences with age (p=0.800) and dominant eye (p=0.402). The ranges of accommodative lag of low, middle, and high myopia were 0.75 ~ -0.25 D, 1.25 ~ -0.50 D, and 1.50 ~ -0.75 D, respectively, and the regression was 'y = -0.03953x+0.09205'. Conclusions: These data suggest that clinically normal young adults with high amounts of refractive error have more variable accommodative lag and increased spherical equivalent refraction.

A Quantitative Method for Human Face Volume Using 3D Optical Measurement (3D 측정 기기를 이용한 얼굴의 부피 정량법 연구)

  • The evaluation for the visage volume was performed with human and dummy face using PRIMOS $body^{(R)}$. Three researchers measured the dummy face attached different volume of clay (6.6 mL, 13.2 mL, and 19.8 mL). It is identified that the high correlation between measured values and real volume ($R^2=0.99$). The percent (%) of relative standard deviation (RSD) of the data was shown below 10%. In Gage R&R analysis, the percent of contribution, the percent of study variation and distinct categories values were 0.78%, 8.85%, and 15% respectively. Therefore this method was regarded as the good standard condition in aspect of reproducibility and repetitiveness of data. The face volume variation was measured by PRIMOS $body^{(R)}$ on the face in participated 10 healthy Korean women volunteers. Subjects used the product once a day for two weeks as night sleeping pack. After application of the test products, the 3D image data of face were taken 1 and 2 weeks later. We found the face volume was significantly decreased after using products. In conclusion, this novel non-invasive technique was useful in measuring and visualizing alterations in face volume as a consequence of certain treatments. This system will provide a rapid and precise analysis of 3D topographical information for face.

Single mode yield analysis of complex-coupled DFB lasers above threshold for various coupling coefficient ratios and facet reflectivity combinations (문턱 전류 이상에서 Complex-Coupled DFB 레이저 다이오드의 여러 가지 결합 계수 비와 양 단면 반사율 조합에 따른 단일 모드 수율 해석)

  • For complex-coupled (CC) DFB lasers, we found that there might be little correlation between the single mode yields at threshold and above threshold. At threshold, the single mode yield considering f number of in-phase (IP) CC DFB lasers is the same as that of anti-phase (AP) CC DFB lasers. However, the single mode yield as a function of injection current above threshold of IP CC DFB lasers is much different from that of AP CC DFB lasers. In the case of IP CC DFB lasers, the single mode yield increases as the coupling coefficient ratio (CR) increases, while, in the case of AP CC DFB lasers, the single mode yield decreases rapidly regardless of CR as the injection current increases. In the case of AR-HR combinations, the effect of AR ref1ectivity on the single mode yield increases as the coupling strength decreases. As the coupling strength decreases, the CR at which the increase rate of the single mode yield starts to decrease, increases, and the maximum single mode yield increases. Single mode yields of AR-HR and AR-AR combinations are larger than those of AR-CL and CL-CL combinations.
