The Scattering and Absorption Effects of Fluorescence Spectroscopy in a Real Time Optical Biopsy (실시간 광학적 생검에서 형광분광법의 산란과 흡수에 대한 영향)

  • The fluorescence emanating from a biological tissue contains information about scattering, absorption and the intrinsic fluorescence (fluorescence only due to fluorophores). Becaue fluorescence spectra of biological tissue are often significantly affected by the presence of tissue absorbers and scatterers, the measured tissue fluorescence cannot be interpreted as a linear combination of intrinsic fluorescence spectra of different tissue biochemical. We conducted experiments to examine the influence of scattering and absorption on the experimentally measured fluorescence of a turbid medium such as biological tissue. Therefore, we acquired fluorescence and reflectance spectra of tissue phantoms with a wide range of scatterer and absorber concentrations. By applying a photon migration model, which uses the scattering and absorption information contained in reflectance spectra to remove their distortion also present in fluorescence spectra, we extract the intrinsic fluorescence of these tissue models. We achieved excellent agreement between modeled and actual intrinsic fluorescence spectra. The motivation for this research is that intrinsic fluorescence spectra are expected to change with progression of disease in human tissue, due to changes in the tissue biochemical composition. It is not possible to distinguish the two tissue types by using only the measured fluorescence, however clear separation can be achieved with the intrinsic fluorescence in real time optical biopsy.

Two-Dimensional Behavior and J-Aggregate Formation of Merocyanine Dye Monolayers in Mutual Mixing (상호혼합에 의한 메로시아닌 색소 단분자막의 2차원 거동 및 J-회합체 형성)

  • J-aggregates in the mutual mixing LB films of [6Me-DS]$_{1-x}$ [DO]$_{x}$,[DS]$_{1-x}$ [DO]$_{x}$ and [DSe]$_{1-x}$ [DO]$_{x}$ have been studied by optical absorption, fluorescence and surface pressure-area isotherms. In [6Me-DS]$_{1-x}$ [DO]$_{x}$ films, sharp J-band absorption and fluorescence of [6Me-DS] are linearly shifted to the longer wavelength for the replacement of [6Me-DS] by [DO]. According to the x, a smooth shift of the limited area has been cleared. In the [DS]-[DO] system, the J-band is enhanced at 1:1 composition and strong fluorescence is also observed. Also, the presence of phase separation was suggested in the [DSe]-[DO] system, because the absorption spectra were decomposed into [DSe] and [DO] spectra. On the other hand, in the pressure-area isothermal study, reduction in the molecular occupying area of monolayers has been clarified. This could be ascribed to the enhancement of molecular ordering in J-aggregates. These facts are also believed to reflect the most closely packed arrangement of chromophores in the merocyanine dye monolayers. Thus, it was confirmed that the interaction between mixed dye molecules and the CdC1$_2$+KHCO$_3$subphases affected the J-aggregates of the LB films. Also. it is thought that the J-aggregates are formed non-dimensionally in LB films, such as solution synthesized [DX:DO] assemblies on mixing.s on mixing.

Seasonal Variation of Attenuation Coefficient Spectra Extracted from Yamato Bank Optical Moored Buoy Data

  • Seasonal variation of attenuation coefficient spectra in Japan sea was extracted from underwater radiance/irradiance spectra observed by a moored buoy system developed by National Space Development Agency of Japan (NASDA). The buoy was deployed 9 months from August 31, 1996 to June 1, 1997. Throughout this period, it was collecting downward irradiance and upward radiance spectra under water at the depth of 1.5m and 6.5m everyday. The dairy averaged diffused attenuation coefficient spectra and underwater reflectance spectra were calculated. The results were compared with the absorption spectra of filtered samples obtained by validation cruises, which carried out 5 times during the moored period. Also, the natural fluorescence of chlorophyll a were extracted from the upward radiance spectra observed at 1.5m depth. The seasonal variation of the calculated attenuation coefficient spectra and the natural fluorescence were examined. The result shows a weak blooming of phytoplankton on November and a large blooming on April.

  • In this paper, we calculate the four coefficients for the quantum theory of multiwave mixing including a local-field correction resulting from dipole-dipole interactions. We make contact with the semiclassical calculations of probe absorption and four-wave-mixing coupling coefficients, and illustrate the effects of local field corrections on resonance-fluorescence and coupled-mode-fluorescence spectra. The method uses the hybrid quantum-Langevin-equation master-equation approach of An and Sargent.

  • A series of $Er^{3+}/Yb^{3+}$ co-doped fluorophosphate glasses with varying $YbF_3$ concentration were prepared by a high temperature melt quenching technique. Absorption and emission cross-sections were determined by using the McCumber theory. The larger emission cross-section ($9.86{\times}10^{-21}cm^2$) and longer fluorescence lifetime (12.37 ms) were obtained for the $^4I_{13/2}{\rightarrow}^4I_{15/2}$ transition of ABS3Er4Yb glass. The sensitivity and temperature of the maximum sensitivity were evaluated by the fluorescence intensity ratio method from the measured upconversion spectra. The results were discussed and compared to the other reported glasses.

  • Different papers such as toilet and towel tissue, newsprint and copy paper made from reclaimed fibres were characterized using UV-VIS reflectance and fluorescence spectroscopy. The emission properties of the extracts of the papers were also evaluated. Photochemical irradiation at ambient temperatures, and thermal aging of the copy papers at 105$^{\circ}C$ and 150$^{\circ}C$ were performed. Post-colour numbers (PC) were calculated from the reflectance values measured from the corresponding UV-VIS reflectance spectra. Light absorption coefficient spectra were calculated before and after irradiation, and the changes in absorption coefficient were related to the content of chromophores in the papers. Photochemical and thermal discolouration of the copy papers, similar to that of virgin fibres, was readily observed. The influence of changes in the concentrations of fluorescent brightening agents and dyes on the spectral properties was also assessed. Photostabilization studies of the copy papers were performed with the use of polymer additives polyethylene glycol (PEG) and polytetrahydrofuran (PTHF).

  • A new laser material, Nd:LSB $(Nd${3}:LaSc_3(BO_3)_4$, lanthanum scandium borate), of microchip type was grown by the Czochralski pulling method, and tested for optical and lasing properties. Nd:LSB, able to be highly doped with $Nd^{3+}$ ions while maintaining good optical, chemical, mechanical properties, was compared to another Nd-type laser material. The absorption and fluorescence spectra, and fluorescence lifetime were measured, and the crystal structure was analyzed. The lasing characteristics were investigated by using Ti:sapphire laser as a pumping light source.

  • The energy relaxation dynamics of the lowest excited singlet and triplet states of the Zn(II)porphyrin monomer and its directly linked arrays were comparatively investigated with increasing the number of porphyrin moieties. While the fluorescence decay rates and quantum yields of the porphyrin arrays increased with the increase of porphyrin units, their triplet-triplet (T-T) absorption spectra and decay times remained almost the same. The difference in the trends of energy relaxation dynamics between the excited singlet and triplet states has been discussed in view of the electronic orbital configurations.

  • The influences of fluorescence, scattering, and flocculation in turbid material by light scattering of N-propyl-N,N-dimethylethanolamine, fluorescence agent and absorption agent were interpreted for the scattered fluorescence intensity and wavelength. They have been studied the molecular properties by the spectroscopy of laser induced fluorescence (LIF) and flocculation. The effects of optical properties in scattering media have been found by the optical parameters(${\mu}_s$, ${\mu}_a$, ${\mu}_t$). Flocculation is an important step in many solid-liquid separation processes and is widely used. When two particles approach each other, interactions of several colloid particles can come into play which may have major effect on the flocculation and LIF process. The values of scattering coefficient ${\mu}_s$ are large by means of the increasing scattering of scatterer, The values have been found that the slope decays exponentially as a function of concentration from laser source to detector by our experimental result. It may also aid in designing the best model for oil chemistry, bio-pharmaceutical, laser medicine and application of medical engineering on LIF and coagulation in particle transport mode.

The Inactivation Effects of UV Light on Bacteriophage f2 (박테리오파지 f2에 대한 자외광선의 살균효과)

  • The effects of ultraviolet light on bacteriophage f2 were investigated to determine the inactivation kinetics and its mechanism. The 260nm light showed a little higher inactivation rate than the one of 300 nm. In this work, our main concern was whether structural and/or conformational changes in the protein capsid could occur by UV irradiation. The inactivation for the first 20 minutes irradiation was rapid with a loss of about 4 logs and followed by a slower rate during the next 40 minutes with no survival noted in the samples irradiated for 90 minutes or longer. The structural change of the protein capsid was examined by optical spectroscopic techniques and electron microscopy. The absorption spectra of the UV irradiated phages showed no detectable differences in terms of the spectral shape and intensity from the control phage. However, the fluorescence emission spectroscopic data, i.e. 1) fluorescence quenching of tryptophan residues upon irradiation of 300 nm light, 2) enhancement of fluorescence emission of ANS (8-aniline-1-naphthalene sulfonate) bound to the intact phages compared to the one in the UV-treated phages, and 3) decrease of energy transfer efficiency from tryptophan to ANS in the UV-treated samples, presented remarkable differences between the intact and UV-treated phages. Such a structural alteration was also observed by electron microscopy The UV-treated phages appeared to be broken and empty capsids. Therefore, the inactivation of the bacteriophage f2 by UV irradiation is thought to be attributed to the structural change in the protein capsid as well as damage in the viral RNA by UV irradiation.
