Control Efficacy of Several Fungicides against Downy Mildew of Onion at Nursery Seedling Stage (육묘상 양파 노균병에 대한 몇 가지 살균제의 방제 효과)

  • During the nursery seedlings stage at Muan in 2013, moisture and temperature conditions were favorable for the occurrence of onion downy mildew. Several fungicides were evaluated for controlling onion downy mildew at the stage of seedling. The disease incidence of the untreated control was 51.1%, which was sufficient to evaluate the fungicide efficacy for the control of onion downy mildew. Incidence at the mancozeb treatment (MAN treatment) was 4.4% and it was the highest control value of 91.4%. Incidence at the various fungicides treatment (MIX treatment) consisted of amectotradin plus dimethomorph, floupicolide plus propamocarb HCl, or dimethomorph plus pyraclostrobin was 10.0%, and the control value of the MIX treatment was 80.4%. Among the tested fungicides, amectotradin plus dimethomorph (AME plus DIM treatment) was selected and treated. Incidence of AME plus DIM treatment was 12.2%. There was statistically significant difference among the three different treatments and the untreated control. These results suggest that the application of fungicides on onion nursery seedlings was effective to control the onion downy mildew.

Rediscovery of Seven Long-Forgotten Species of Peronospora and Plasmopara (Oomycota)

  • The family Peronosporaceae, an obligate biotrophic group of Oomycota, causes downy mildew disease on many cultivated and ornamental plants such as beet, cucumber, grape, onion, rose, spinach, and sunflower. To investigate the diversity of Peronosporaceae species in Korea, we performed morphological analysis for dried plant herbariums with downy mildew infections by two largest genera, Peronospora and Plasmopara. As a result, it was confirmed that there are five species of Peronospora and two species of Plasmopara, which have been so far unrecorded in Korea, as well as rarely known in the world; Pl. angustiterminalis (ex Xanthium strumarium), Pl. siegesbeckiae (ex Siegesbeckia glabrescens), P. chenopodii-ambrosioidis (ex Chenopodium ambrosioides), P. chenopodii-ficifolii (ex Chenopodium ficifolium), P. clinopodii (ex Clinopodium cf. vulgare), P. elsholtziae (ex Elsholtzia ciliata), and P. lathyrina (ex Lathyrus japonicus). In addition, their phylogenetic relationship was inferred by molecular sequence analysis of ITS, LSU rDNA, and cox2 mtDNA. By rediscovering the seven missing species and barcoding their DNA sequences, this study provides valuable insights into the diversity and evolutionary studies of downy mildew pathogens.

Control of Diseases and Insects for Pesticide-free Cultivation of Leafy Vegetables (엽채류 유기재배의 병해충 관리)

  • This study was conducted to establish agricultural practices to control diseases and insects for chemical pesticide-free cultivation of leafy vegetables. Two diseases, gray mold(Botrytis cinerea) and soft rot(Erwinia carotovora), on lettuce were reduced by controlling temperature and humidity using air-circulation fan. The aphidophagous lady beetle(Harmonia axyridis) and primary parasitoids(Aphidius colemani) showed activity to control aphids density on Chinese cabbage. Co-application of cooking oil and yolk mixture (COY) and BT(Bacillus thuringiensis) decreased diseases including soft rot(Erwinia carotovora), downy mildew(Peronospora brassicae Gaumann), and powdery mildew(Eryslphe polygoni), and insects such as diamondback moth(Plutella xylostella) and beet armyworm(Spodoptera exigua Hubner). Neem extract treatment reduced downy mildew(Peronospora destructor) on Welsh onion.

  • Onion downy mildew, caused by Peronospora destructor, is a major disease in onion cultivation areas in Korea. The causal fungi were collected and analyzed based on sequence similarity and molecular phylogenetic relationships of multi-gene sequences, including the internal transcribed spacer (ITS) region. All isolates from Changnyeong-gun, Hamyang-gun, and Hapcheon-gun in Gyeongnam province, and Muan-gun, Haenam-gun, and Sinan-gun in Jeonnam province were identical in the four types of gene sequences, indicating they were genetically the same strains. In this study, a PCR method was developed based on the ITS gene sequences to amplify the specific DNA fragment for P. destructor only. The detection limit of was total genomic DNA of the P. destructor and the plant $0.7ng/{\mu}L$. Therefore, the developed PCR method could be used to detect P. destructor effectively from symptomless onion leaves.

  • This experiment was compared Bordeaux mixture, which used to as good fungicide up to date, with new 6 fungicides for control of powdery mildew on onion from 1963 to 1864 in Pusan. In resulting treated blocks was better effect than untreated block, and specially Difolatan and Dithane M 45 was best. Dithane M 22, Dithane Z 78 and Orthocide was moderate but Sankinon and Bordeaux mixture was lower effect than formers.

Effects of a Slow-Release Fertilizer on Onion (Allium cepa L.) Cultivation in Southwestern Area (서남부 지역 양파 재배 시 완효성 비료 시용효과)

  • This experiment was conducted to evaluate the effect of slow-release fertilizer application on quality elevation of onion (Allium cepa L.) in southwestern sea. The slow-release fertilizers used were NIAST (National Institute of Agricultural Science and Technology) I, II, CDU (Crotonylidene Diurea), UF (Urea-Formaldehydes), and IBDU (Isobutylidene Diurea) 30, 50. Growth of the onion plant treated with slow-release fertilizer showed better than the control. However, a rate of infected plant to downy mildew was low as compared with control. Chlorophyll and total nitrogen contents were high in control, whereas firmness, soluble solid content, number and thickness of scaly leaves were high in slow-release fertilizer plot. In early maturing onion, total marketable bulb yield was higher in slow-release fertilizer plot than all other fertilizer treatments. Total marketable bulb yield in late maturing onion treated with slow-release fertilizer was slightly decreased. Decaying loss and the rate of sprouting during storage period decreased in slow-release fertilizer application. In summary, application of slow-release fertilizer increased marketable yield, storability after harvest and also reduced labor needs for top dressing.

Mild taste and early maturing red onion cultivar 'Eumjinara' (매운맛이 적고 숙기가 빠른 자색양파 '엄지나라')

  • Red onion cultivar 'Eumjinara' with mild taste and early maturation was developed in 2013 by the Allium vegetable crop research team, National Institute Crop Science, Rural development Administration (RDA). 'Eumjinara' was originally selected from red onion breeding line, 'IS1115', It was preliminarily selected and designated 'Mon21-31-3' and advanced yield trial was performed from 2008 to 2009. Regional yield trial test was conducted in six regions from 2010 to 2012. Foliage morphology and leaf color were similar to those of a check cultivar, 'Cheonjujeok', but its shape was comparatively broad ovate. Foliage of 'Eumjinara' lodged seven days earlier than check cultivar on May 31 and the average bulb weight ($234{\pm}49.1g/bulb$) was also higher than that of check cultivar. The new cultivar was relatively resistant to clod hardness and downy mildew infection in the field condition. Average yield of 'Eumjinara' in three-year the regional yield trials was 6,999 kg/10a. The total soluble solid contents and quercetin concentrations were $41.43{\mu}g/g$ FW and $556.11{\mu}g/g$ FW, respectively.

Detection of the Fungicide Iprovalicarb Residues Using a Surface Plasmon Resonance Biosensor (표면플라즈몬공명 바이오센서를 이용한 살균제 Iprovalicarb 잔류물의 검출)

  • Surface plasmon resonance (SPR) biosensor has been used to detect many biochemical reactions, because this label-free sensor has high sensitivity and rapid response. The reactions are monitored by refractive index changes of the SPR biosensor. Iprovalicarb is protective, curative, and eradicative systemic fungicide introduced by Bayer AG in 1999. It has potential far control of downy mildew infesting onion, cucumber, grape and melon, late blight infesting tomato and potato, and anthracnose infesting watermelon and pepper. It is strictly limited to the maximum residue limit. In this study, the applicability of a portable SPR biosensor (Spreeta, Texas instrument, TX, USA) to detect the iprovalicarb residue was examined. The sensor chip was adopted to detect the reaction of iprovalicarb to immobilized iprovalicarb-antibody. The binding of the iprovalicarb onto the biosensor surface was measured by change of the refractive index (RI). Characteristics of the sensor chip including specificity, sensitivity, stability, and reusability were analyzed. In calibration test for seven levels of iprovalicarb concentration (0.32 to 5,000 mg/L) with three replications, a Sigmoidal model with Hill function was obtained between relative RI value and the iprovalicarb concentration with R-square of 0.998. It took 30 minutes to complete a set of detecting assay with the SPR biosensor.
