Quality and fermentative characteristics of yogurt added with hot water extract of Welsh onion root (파뿌리 열수추출물을 이용한 요구르트의 발효 및 품질특성)

  • The objective of this study was to investigate the effect of hot water extract of Welsh onion root (HEWO) on growth of lactic acid bacteria and fermentative characteristics of yogurt. The physiochemical characteristics of HEWO such as pH, soluble solid, reducing sugar, total polyphenol content and DPPH radical scavenging activity were studied. The lactic bacterial count in brain heart infusion (BHI) broth with HEWO was about 1 log cycle higher than in control for 24 h at $37^{\circ}C$. The pH of yogurt prepared with HEWO (WY100) and 50% HEWO (WY50) was gradually decreased significantly but increased the viscosity of yogurt with increasing HEWO concentration during fermentation. The viable cells of lactic acid bacteria after fermentation for 24 h were 8.03 (control), 8.77 (WY50), 8.84 (WY100) log CFU/mL, respectively. The DPPH radical scavenging activity of yogurt increased with increasing HEWO concentration. Sensory quality of yogurt prepared with HEWO was higher than that of control. The pH and lactic acid bacteria of all tested yogurts decreased during storage for 10 days at $4^{\circ}C$ but lactic bacterial count of yogurt prepared with HEWO maintained $10^8CFU/mL$ during storage. These results indicated the potential use of HEWO as a valuable resource to improve fermentation and functionality of yogurt.

Effects of Antioxidative Activities and Antibrowning of Extracts from Onion, Apple and Mandarin Orange Peel as Natural Antibrowning Agents (천연 갈변 억제제 개발을 위한 양파, 사과 및 감귤 과피 추출물의 항산화 및 갈변 저해 효과)

  • This study was carried out to investigate antioxidative properties of various extracts and antibrowning effects of extracts in apple slices were investigated by ${\Delta}E$ value and PPO relative activity. Apples were cut into 1.5 cm thickness and they were dipped in 1% extract solutions(OW: water extracts of onion, OE: 80% EtOH extracts of onion, AW: water extracts of apple, AE: 80% EtOH extracts of apple, MW: water extracts of mandarin orange peel, ME: 80% EtOH extracts of mandarin orange peel) for 1 min. OW showed higher than the other treatments for total phenolic contents(94.35 mg/g), PPO inhibition(74.00%). And the highest DPPH free radical scavenging activity(40.27%) measured in ME. ${\Delta}E$ value of apple slices dipped in MW was 2.37 whereas ${\Delta}E$ value of apple slices dipped in AW was 12.12. These results suggest that onion and mandarin orange peel extracts should be a potential source for controlling browning during storage of apple slices.

Reduction in Residual Pesticides and Quercetin Yields in Onion Peel Extracts by Washing (세척방법에 따른 양파껍질추출물의 Quercetin수율 및 잔류농약 제거효과)

  • This study was conducted to assess the removal of residual pesticides and to obtain high amounts of quercetin in onion peel extracts (OPEs) by 4 washing treatments. Washing is one of the standard processing steps in obtaining functional food ingredients from onion peel. After a first detergent wash (0.2% w/v) (DW) and hot air drying ($80^{\circ}C$, 24 hr) (B), 4 washing treatments were tested, including a second DW (C), ultrasonication ($50^{\circ}C$, 10 min) plus DW (D), 0.3% $H_2O_2$ (v/v) plus DW (E), and blanching ($95-97^{\circ}C$, 2 min) plus DW (F). This was followed by 60% (v/v) ethanol extraction and vacuum freeze drying of the OPE. The E treatment yielded 89.04% OPE and a quercetin content of 96.84% in the OPE compared with the B treatment, and had the highest efficiency of all treatments tested. The OPE was tested for the presence of 177 residual pesticides and three compounds were detected in all treatments: cyhalothirn, fluquinconazole and procymidone. Cyhalothirn and fluquinconazole levels were below the permitted levels for fresh onion, while procymidone was present in the high level range of 128.01~133.46 mg/kg in all samples. The E treatment was a better washing method than the others for removal of residual pesticides. It could reduce the level of residual pesticides without changing the functional properties of the OPE.

Changes in the Components of Onion Vinegars by Two Stages Fermentation (2단계 발효에 의한 양파식초의 성분변화)

  • Changes in components of onion vinegar during two-stage fermentation were investigated. One sample group (A) was prepared with onion juice by extraction and concentration (15 brix), and the other group (B) was prepared using the same method but supplemented with sucrose. Comparison of glucose (4.1%), fructose (4.2%), and sucrose (0.6%) found in A as major sugars with those (3.2, 3.3, and 4.6%, respectively) of B showed a significant difference in sucrose concentration. During alcohol fermentation, S. kluyvery DJ97 consumed these sugars completely. At 36 hr of alcohol fermentation, concentrations of reducing and total sugars decreased to 0.5 and 0.8%, in A, and 0.4 and 0.6%, in B, respectively. No further changes were detected in the concentrations of these sugars thereafter. From 12 hr of fermentation, alcohol concentrations in groups A and B increased markedly, maximizing at 7.0% at 36 hr A and at 8.2% at 48 hr in B, and finally decreased thereafter in both groups. The detected alcohol components were ethanol, acetaldehyde, methanol, n-propyl alcohol, iso-butanol, and iso-amyl acohol. During acetic acid fermentation, the concentration of acetic acid as the major organic also increased significantly in both groups (4,776.72 and 4,894.93 mg% in A amd B, respectively). Other organic acid contents such as malic and succinic acids were higher in A than B. these results indicate that vinegar is better produced in onion extract with no sucrose supplementation based on its organic acid contents.

Occurrence of the Onion Moth, Acrolepiopsis sapporensis, in the Welsh Onion Farms and its Treatment Using 'BtPlus' (대파 재배지 파좀나방(Acrolepiopsis sapporensis) 발생 현황과 '비티플러스' 처리 효과)

  • The onion moth, Acrolepiopsis sapporensis, was monitored in the farms cultivating the welsh onion, Allium fistulosum, using sex pheromone from transplantation to harvest. Two occurrence peaks were observed at early June and late July after the overwintering population. However, the population sizes were varied among different years and the cultivating environments. To effectively control A. sapporensis with microbial pesticides, different Bacillus thuringiensis strains were screened to select B. thuringiensis kurstaki (BtK). To enhance the insecticidal virulence of BtK, the culture broth of Photorhabdus temperata temperata (Ptt) was added to the BtK. This mixture of two entomopathogenic bacteria was called 'BtPlus', which was superior to BtK alone in the insecticidal virulence. The enhanced virulence was explained by the immunosuppressive activity of the secondary metabolites contained in the Ptt extract. The metabolites inhibited both cellular and humoral immune responses of A. sapporensis, resulting in the enhanced virulence of BtK. These results suggest that A. sapporensis occurs in the welsh onion fields and the resulting economic damage would be effectively prevented by BtPlus application.

The Hematological Effect of Excessive Crude Allium Tuberosum Extract Administrated in Large Dogs (과량의 부추 생즙 투여가 대형견의 혈액상에 미치는 영향)

  • The Allium tuberosum is a perennial herb. From the ancient times, it has been used for food or medical purpose. Allium tuberosum is widely distributed in Korea, China, Japan and other countries. As herbal medicine, the Allium tuberosum has medical effects on lumbago, sthenia, diuresis, nocturia, hematemesis, anemia, haemorrhoid, hiccups, diarrhea, eczema, sunstroke, pollution and so on. Recently, the Allium tuberosum has been studied on its components, anti-microorganism action, anti-fungal action, anti-arteriosclerosis action, anti-cancer action. However, there has not been any study on hematological effect of allium tuberosum extract on the blood in large dogs whereas the effects of excess garlic and onion on the blood of cattle, horses, dogs, cats and sheep has been reported by many researchers. Thus, this study was performed to observe the hematological effect of excessive Allium tuberosum extract administrated in large dogs. 1. The Red Blood Cell(R3C) count was significantly decreased(p<0.05), Packed Cell Volume (PCV) count were significantly decreased(P<0.05), and Reticulocyte count was significantly increased(p<0.05) 2. The White Blood Cell(WBC) count was significantly increased(p<0.05).3. The Hemoglobin concentration was significantly decreased(p<0.05) and Mean Corpuscular Hemoglobin Concentration (MCHC) was increased. and Mean Corpuscular Volume(MCV) was significantly increased.(p<0.05). 4. The Reduced glutathione(GSH) and Met-Hemoglobin were significantly increased (p<0.05).

Extractive Optimization of Functional Components for Processing of Onion Health Promotion Drink (양파 음료 제조를 위한 기능성 성분 추출 최적화)

  • Onion, licorice, abgelia root, Chinese date, pine needle, and mulberry leaf were used to extract functional components far onion complex drink. No differences were observed between water extraction at room temperature and methanol extraction in electron-donating ability (EDA), thiosulfinate content (TSC), and ascorbic acid content (AAC), whereas water extraction resulted in higher nitrite-scavenging ability at pH 1.2 and 3.0, and lower superoxide dismutase-like ability than methanol extraction. Level of water extracts prepared by hard-boiling for 3 hr at above $100^{\circ}C$ was higher in all functional abilities except TSC than those prepared at room temperature. Optimal conditions for water extraction and storage were $100-120^{\circ}C$ for 6 hr and low-temperature storage, respectively.

Onion peel extract and its constituent, quercetin inhibits human Slo3 in a pH and calcium dependent manner

  • Sperm function and male fertility are closely related to pH dependent $K^+$ current (KSper) in human sperm, which is most likely composed of Slo3 and its auxiliary subunit leucine-rich repeat-containing protein 52 (LRRC52). Onion peel extract (OPE) and its major active ingredient quercetin are widely used as fertility enhancers; however, the effect of OPE and quercetin on Slo3 has not been elucidated. The purpose of this study is to investigate the effect of quercetin on human Slo3 channels. Human Slo3 and LRRC52 were co-transfected into HEK293 cells and pharmacological properties were studied with the whole cell patch clamp technique. We successfully expressed and measured pH sensitive and calcium insensitive Slo3 currents in HEK293 cells. We found that OPE and its key ingredient quercetin inhibit Slo3 currents. Inhibition by quercetin is dose dependent and this degree of inhibition decreases with elevating internal alkalization and internal free calcium concentrations. Functional moieties in the quercetin polyphenolic ring govern the degree of inhibition of Slo3 by quercetin, and the composition of such functional moieties are sensitive to the pH of the medium. These results suggest that quercetin inhibits Slo3 in a pH and calcium dependent manner. Therefore, we surmise that quercetin induced depolarization in spermatozoa may enhance the voltage gated proton channel (Hv1), and activate non-selective cation channels of sperm (CatSper) dependent calcium influx to trigger sperm capacitation and acrosome reaction.

The Effects of Vegetable Extracts on the Growth of Listeria monocytogenes (채소추출물이 Listeria monocytogenes의 생육에 미치는 영향)

  • The inhibitory effects of vegetables, collected from Jinju district, on the growth of Listeria monocytogenes were investigated. Among 12 vegerables, garlic, leek, onion and cabbage were remarkably effective to inhibit the growth of the strain. Especially with the addition of extract equivalent 0.25g of raw garlic per ml broth, the population of the strain was decreased wignificantly, and completely inhibited with the addition of 0.625g or more. During the incubation with the addition of extract equivalent 0.25g of raw garlic per ml broth, the degree of inhibition was gradually increased with the lapse of time, and completely inhibited the growth of the strain after 48 hrs. While the pH value of treated broth remained unchanged almost, those of control were slightly reduced.
