Effects of Plant Extracts on Microbial Population, Methane Emission and Ruminal Fermentation Characteristics in In vitro

  • This study was conducted to evaluate effects of plant extracts on methanogenesis and rumen microbial diversity in in vitro. Plant extracts (Artemisia princeps var. Orientalis; Wormwood, Allium sativum for. Pekinense; Garlic, Allium cepa; Onion, Zingiber officinale; Ginger, Citrus unshiu; Mandarin orange, Lonicera japonica; Honeysuckle) were obtained from the Plant Extract Bank at Korea Research Institute of Bioscience and Biotechnology. The rumen fluid was collected before morning feeding from a fistulated Holstein cow fed timothy and commercial concentrate (TDN; 73.5%, crude protein; 19%, crude fat; 3%, crude fiber; 12%, crude ash; 10%, Ca; 0.8%, P; 1.2%) in the ratio of 3 to 2. The 30 ml of mixture, comprising McDougall buffer and rumen liquor in the ratio of 4 to 1, was dispensed anaerobically into serum bottles containing 0.3 g of timothy substrate and plant extracts (1% of total volume, respectively) filled with $O_2$-free $N_2$ gas and capped with a rubber stopper. The serum bottles were held in a shaking incubator at $39^{\circ}C$ for 24 h. Total gas production in all plant extracts was higher (p<0.05) than that of the control, and total gas production of ginger extract was highest (p<0.05). The methane emission was highest (p<0.05) at control, but lowest (p<0.05) at garlic extract which was reduced to about 20% of methane emission (40.2 vs 32.5 ml/g DM). Other plant extracts also resulted in a decrease in methane emissions (wormwood; 8%, onion; 16%, ginger; 16.7%, mandarin orange; 12%, honeysuckle; 12.2%). Total VFAs concentration and pH were not influenced by the addition of plant extracts. Acetate to propionate ratios from garlic and ginger extracts addition samples were lower (p<0.05, 3.36 and 3.38 vs 3.53) than that of the control. Real-time PCR indicted that the ciliate-associated methanogen population in all added plant extracts decreased more than that of the control, while the fibrolytic bacteria population increased. In particular, the F. succinogens community in added wormwood, garlic, mandarin orange and honeysuckle extracts increased more than that of the others. The addition of onion extract increased R. albus diversity, while other extracts did not influence the R. albus community. The R. flavefaciens population in added wormwood and garlic extracts decreased, while other extracts increased its abundance compared to the control. In conclusion, the results indicated that the plant extracts used in the experiment could be promising feed additives to decrease methane gas emission from ruminant animals while improving ruminal fermentation.

Screening of Biological Activity for Phenolic Fraction from Onion (양파 페놀성분획물의 생리활성기능탐색)

  • To develop a material for functional food, the fraction Ⅰ,II and Ⅲ of onion extract were separated by Sephadex LH-20 column. The revealed purification levels of each fraction were 5.2%-6.5%, 12.2%-14.5% and 82.6%-86.7%, respectively. A functional activity test was performed with fraction Ⅲ. The ACE inhibition activity was about 50% at 5mg and about 80% at 10mg. The XOase inhibition activity of Korean Changnyong Daego was 84%, which was the highest. At the experiment of tyrosinase inhibition was detected 21-36% at 10mg. The cholesterol dose in mice blood was as low as low as 79$\pm$5(mg/100 mL) after the intake of fraction Ⅲ. Cholestrol dose in liver was decreased to below 50%. The results show that Sephadex LH-20 column method was very useful as a fraction method for the development of functional food material nsing onions.

  • The desmutagenic effects of seaweed and vegetable extracts were investigated on the mutagenicity of Maillard reaction products (MRP) obtained from equimolar amounts of glucose and amino acid (arginine and lysine${\cdot}$HCl) for Salmonella typhimurium TA 100 without S9 mix. The mutagenicities were inhibited by water-soluble extracts of seaweeds(laver, sea-straghorn, sea-mustard and tangle) and vegetables(ginger, garlic, onion, chinese-pepper, green-onion and cabbage). Cabbage, chinese-pepper, green-onion and sea-straghorn exhibited especially high desmutagenic effects. The desmutagenicities of these extracts(cabbage, green-onion and sea-straghorn) except for sea-straghorn were decreased by heat treatment at $100^{\circ}C$ for 10 min. It is assumed that the desmutagenic effect of seaweed and vegetable extract is due to the reducing power and action of enzyme such as peroxidase and catalase.

Isolation and Identification of Antioxidative Compounds 3,4-Dihydroxybenzoic acid from Black Onion (흑양파로부터 항산화 활성 물질인 3,4-Dihydroxybenzoic acid의 분리 및 동정)

  • The antioxidant substance in black onion was identified. The assays that used 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzthiazo line-6-sulphonic acid) ($ABTS^+$) radicals showed that the ethyl acetate fraction of black onion methanol extract had a higher level of radical-scavenging activity than the other fractions. Two antioxidative compounds were purified and isolated from the ethyl acetate fraction via column chromatographies of silica gel and Sephadex LH-20 using the guided DPPH radical-scavenging assay. The isolated compounds were identified as 3,4-dihydroxybenzoic acid (1) and quercetin (2) based on mass spectrometry and nuclear magnetic resonance. The isolated compounds showed a high level of DPPH and ABTS+ radical-scavenging activity. Compound 2 had a higher level of radical-scavenging activity than 1.

Effects of Dietary Supplements of processed Onion on the Physiochemical Characteristics by Store Period in Duck Meat (양파함유사료를 급여한 오리 고기의 저장기간중 이화학적 특성 변화)

  • The objectives or this study were to improve duck meat qualify during storage period in onion-red ducks which contained antioxidants. Experimental animals (1 day old, Cherry Valley $F_{1}$, 360 chicks) were randomly assigned in eight treatment group. Treatments were control, 3%, 6% of chopped onion-fed, 5%, 10% of onion extract fed, 6% of onion meal, 3%, 6% of fermented onion-fed. pH was significantly high (P<0.05) in treatment group after 3 day storage in pectoralis muscle. Meat color was low in r* and a* value. TBA value was not different in day 1, was significantly low (P<0.05) 6% chopped and 6% fermented group after day 3 storage. Also, TBA value was significantly high (P<0.05) in control group and was significantly low in 3% chopped, 5%, 10% extract after day 6 storage. VBN value was lower in all treatment groups except 5% ferment group than control group. Meat order was high score in 3% and 6% chopped group and duck specific smell was reduced in all treatment groups. Meat appearance was higher score in treatment groups than in control group during 9 day storage. In result, onion feeding was reduce duck specific smell and improve meat quality during long period storage in ducks.

  • Antitumor activities of methanol extract from onion Kochujang (MEOK) were investigated by using aflatoxin $B_{1}-mediated$ Salmonella typhimurium mutagenicity and the model of cytotoxicity on the cancer cell lines. Their immune activities were also investigated using mouse spleen cells and macrophage cell lines, respectively. MEOK showed the enhanced antimutagenicity in a dose-dependent manner. relative to the control group. MEOK decreased over $20\%$ of the proliferation of the A549(lung cancer cell) and MCF(breast cancer cell) cell lines at $1,000\;{\mu}g/mL$ concentration compared with the control cells. The proliferation of mouse spleen cells and the NO production in marcrophage cell lines treated MEOK were increased in a dose-dependent manner, respectively. The activities were higher than that of the control(no added onion) Kochujang.

Selective antibacterial activity of quercetin against Xanthomonas campestris (Quercetin의 식물병원성 세균인 Xanthomonas campestris에 대한 선택적 살균활성)

  • Quercetin is a major flavonoid in onion and known to antimicrobial activity against several pathogenic bacteria. However, the antibacterial activity of quercetin had not been evaluated against plant pathogenic Xanthomonas campestris and Erwinia carotovora. In here, quercetin and the solvent extracts of onion were investigated their antibacterial activity. Quercetin showed a selective inhibitory activity against X. campestris, and the minimal inhibition concentration (MIC) and minimal bactericidal concentration of quercetin were 15.6 and 20.0 ㎍ mL-1 respectively. Otherwise, it showed no inhibitory activity against E. carotovora, and no the additive and the synergistic effects with streptomycin on X. campestris. And the EtOAc extract from the peel of onion that contained quercetin showed 2-fold lower MIC (500 ㎍ mL-1) than the EtOH extract, thus EtOAc was suggested as the extraction solvent for quercetin from onion peel.

  • Flavonoids are bioactive plant metabolites that have a range of beneficial effects on human health. Quercetin 4'-glycoside (Q4'G), quercetin 3,4'-diglycoside (Q3,4'G), and quercetin aglycone (QA) are the main flavonoids found in onions. QA, in particular, is likely to have a greater biological effect than glycosides. To develop an onion extract with high quercetin content, the optimal extraction conditions for red onion powder containing the outer layer of the onion were determined. The effects of acid treatment on the concentration of quercetin glycosides and QA were evaluated. The flavonoids of red onion powder were optimally extracted under 60-70% ethanol at 70℃ for 2 h. The deglycosylation of Q3,4'G and an increase in Q4'G content occurred within 6 h of 0.2% acetic acid treatment. The QA content and deglycosylation of Q4'G eventually peaked at 24 h. In addition, QA content and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity were highly correlated, with a correlation coefficient of 0.90.

Development of functional soy-based stew sauce including hot water extract of Cornus officinalis S. et Z (산수유 열수추출물을 첨가한 찜류용 기능성 간장소스 개발)

  • We developed a kind of soy-based sauce for stews using three recipes and sensory evaluation processes, and examined the biological activities such as antioxidative, fibrinolytic and ${\alpha}$-glucosidase inhibitory activities of hot water extract of Cornus officinalis S. et Z in order to use as functional ingredient. The developed stew was composed of soy sauce $1\frac{1}{2}$ Tbsp, sugar $1\frac{1}{4}$ Tbsp, garlic, small green onion, powdered sesame and sesame seed oil $\frac{1}{2}$ tsp, respectively, ginger extract $\frac{1}{4}$ tsp, black pepper $\frac{1}{8}$ and water $\frac{2}{3}$C per 300 g of main ingredients(pork rib, chicken and dried pollack). It was evaluated more highly acceptable than the original recipes by sensory test. Replacement of water in the recipe of the developed stew sauce with hot water extract of Cornus officinalis S. et Z did' nt affect the sensory quality. From the above results. we concluded that hot water extract of Cornus officinalis S. et Z might be a excellent source to provide health functionality.

Processing of Oleoresin Onion (양파 Oleoresin의 가공)

  • The purpose of this study was to investigate the extraction yield and quality stability as to the oleoresin process with large amount of onion at one time. The first mixed-product is raw onion juice which was reduced the compression and concentrated by Brix 70% mixed together wit the residue which was extracted and concentrated by ethanol, the second product manufactured by the same method above after the autoclaving with onion, and the other product is made by grinding by 50mesh to freeze-dried onion. Each of yields were 7.3, 9.1 and 0.8% and each of total sugar content was 616.4, 712.3 and 150.3mg/g. Therefore the product extracted by ethanol from freeze-dried onion was very low in yield and total sugar content. By the index of the overall odor intensity, contents of total pyruvate were 1,733.7, 520.6, and 2,716.5$\mu\textrm{g}$/g for each product. As a result, oleoresin onion processing that desired to use raw onion was remarkable for odor recovery. For the homogenous mixture with concentrate of onion juice and ethanol extract were emulsified by the addition of 2% of PGDR(polyglycerol condensed ricinoleate) and agitation(10,000rpm, 30 minutes). At this time, interfacial tension was 1.9 dyne/cm and the formation of emulsion was for 96.2% when left over 24hours in 6$0^{\circ}C$. When it was to be centrifuged(2,000$\times$G, 80 minutes) after emulsification, the volume of emulsion level without seperation was 92.6%, and very high in emulsification stability. The induced heating-oxidize with soy bean oil and sesame oil added to 1% of onion oleoresin, induction-time extension effect appeared with antioxidant activity that was applicable for 80.8~82.2% as to the effect of addition of 0.02% BHA.
