Objective: Vanin-1 (VNN1) is a pantetheinase that catalyses the hydrolysis of pantetheine to produce pantothenic acid and cysteamine. Our previous studies have shown that the VNN1 is specifically expressed in chicken liver which negatively regulated by microRNA-122. However, the functions of the VNN1 in lipid metabolism in chicken liver haven't been elucidated. Methods: First, we detected the VNN1 mRNA expression in 4-week chickens which were fasted 24 hours. Next, knocked out VNN1 via CRISPR/Cas9 system in the chicken Leghorn Male Hepatoma cell line. Detected the lipid deposition via oil red staining and analysis the content of triglycerides (TG), low-density lipoprotein-C (LDL-C), and high-density lipoprotein-C (HDL-C) after VNN1 knockout in Leghorn Male Hepatoma cell line. Then we captured various differentially expressed genes (DEGs) between VNN1-modified LMH cells and original LMH cells by RNA-seq. Results: Firstly, fasting-induced expression of VNN1. Meanwhile, we successfully used the CRISPR/Cas9 system to achieve targeted mutations of the VNN1 in the chicken LMH cell line. Moreover, the expression level of VNN1 mRNA in LMH-KO-VNN1 cells decreased compared with that in the wild-type LMH cells (p<0.0001). Compared with control, lipid deposition was decreased after knockout VNN1 via oil red staining, meanwhile, the contents of TG and LDL-C were significantly reduced, and the content of HDL-C was increased in LMH-KO-VNN1 cells. Transcriptome sequencing showed that there were 1,335 DEGs between LMH-KO-VNN1 cells and original LMH cells. Of these DEGs, 431 were upregulated, and 904 were downregulated. Gene ontology analyses of all DEGs showed that the lipid metabolism-related pathways, such as fatty acid biosynthesis and long-chain fatty acid biosynthesis, were enriched. KEGG pathway analyses showed that "lipid metabolism pathway", "energy metabolism", and "carbohydrate metabolism" were enriched. A total of 76 DEGs were involved in these pathways, of which 29 genes were upregulated (such as cytochrome P450 family 7 subfamily A member 1, ELOVL fatty acid elongase 2, and apolipoprotein A4) and 47 genes were downregulated (such as phosphoenolpyruvate carboxykinase 1) by VNN1 knockout in the LMH cells. Conclusion: These results suggest that VNN1 plays an important role in coordinating lipid metabolism in the chicken liver.
The purpose of the study was to identify the degree of social support and burden in mothers of children with cancer, and to determine a strategy of an effective social support utility for burden relief in these mothers. The subjects of this study were consisted of mothers of cancer children, registered at a'C'University affiliated hospital in Taejon. The data was collected from July 1 to August 31, 1994. Three instruments were used to collect the data : a semi-structured interview questionnaire which was developed by the researcher was used to identify the content of the mother's burden, a Visual Analogue Scale(VAS) that had a 10cm horizontal line was used to measure the degree of mother's burden and the PRQ part I & part Ⅱ were used to measure the degree of mother's social support. The content of mother's burden collected through in depth interviews was analyzed using content analysis. Also burden and social support data were analyzed by SAS program. The result of this study were summarized as follows : 1. The data on burden content from the interview were categorized as psychological, physiological, family interaction with maternal role, financial burden and personal interaction with social relational burden. 2. The degree of burden measured by VAS had a mean of 8.04(range from 0.5 to 10). 3. The score for social support measured by PRQ part Ⅱ had a mean of 88.9(range from 71 to 113). The highest scores was for reassurance of worth. The lowest score was for opportunity for nuturance. Most of the 15 mothers received the greatest amount of support from their sisters, spouse, friends, neighbors, relations, priest were perceived as eaningful resource person. 4. The correlation coefficient of burden and social support was somewhat negative correlation but no statistical significance(r=-.072). Therefore, a further study is necessary to repeat the qualitative research for exploring factor to be affected family caregiver's burden according to disease proceeding stage. On the basis of the results from this study, future research will be promoted valid and reliable tool development. Through this study, nurses understand and assess the individual psychologic burden and further it would be recommendated to produce professional education program for pediatric oncologic specialist nurse.
In the present work, novel orange peel was extracted with 100%EtOH (ethanol) and fractionated into four fractions namely F1, F2, F3, F4 which were eluted from paper chromatographs using 100%EtOH, 80%EtOH, 50%EtOH and pure water respectively. The crude extract and its four fractions were evaluated for their total polyphenol content (TPC), total flavonoid content (TFC) and radical scavenging activity using DPPH (1,1-diphenyl-2-picrylhydrazyl) assay. Their cytotoxic activity using WST assay and DNA damage by agarose gel electrophoresis were also evaluated in a human leukemia HL-60 cell line. The findings revealed that F4 had the highest TPC followed by crude extract, F2, F3 and F1. However, the crude extract had the highest TFC followed by F4, F3, F2, and F1. Depending on the values of $EC_{50}$ and trolox equivalent antioxidant capacity, F4 possessed the strongest antioxidant activity while F1 and F2 displayed weak antioxidant activity. Further, incubation HL-60 cells with extract/fractions for 24h caused an inhibition of cell viability in a concentration-dependent manner. F3 and F4 exhibited a high antiproliferative activity with a narrow range of $IC_{50}$ values ($45.9-48.9{\mu}g/ml$). Crude extract exhibited the weakest antiproliferative activity with an $IC_{50}$ value of $314.89{\mu}g/ml$. Analysis of DNA fragmentation displayed DNA degradation in the form of a smear-type pattern upon agarose gel after incubation of HL-60 cells with F3 and F4 for 6 h. Overall, F3 and F4 appear to be good sources of phytochemicals with antioxidant and potential anticancer activities.
The current study was undertaken to determine the effects of the ethanol extracts of loquat (Eriobotrya japonica Lindl.) seeds, flesh or leaves on the differentiation of 3T3-L1 cells and male pig preadipocytes. The cell number was measured with the MTT assay after trypsin digestion. The cell differentiation was determined by measuring the glycerol-3-phosphate dehydrogenase (GPDH) activity and triglyceride(TG) content. No cytotoxicity was observed from the loquat flesh and leaf ethanol extracts at concentrations of 5, 10, 25, 50, 100 or $200{\mu}g/mL$ in 3T3-L1 cells and pig preadipocytes. However, the cell viability of neither cell line were affected by up $50{\mu}g/mL$ of loquat seed ethanol extract. Treatment with the loquat seed and leaf ethanol extracts significantly suppressed the terminal differentiation of both cell lines in a dose-dependent manner, as confirmed by the decrease in the glycerol-3-phosphate dehydrogenase(GPDH) activity and TG content. Treatment with the loquat seed and leaf ethanol extracts inhibited the GPDH activity and reduced the TG content of both cell types more effectively than that with the loquat flesh ethanol extract. The most potent anti-adipogenic effect was obtained in the case of the ethanol extract of loquat seeds.
The objective of this research was to evaluate the effects of dietary supplementation of blood meal (BM) as a source of histidine, and magnesium oxide (MgO) as a catalyst of carnosine synthetase, on carnosine (L-Car) content in the chicken breast muscle (CBM), laying performance, and egg quality of spent old hens. Four hundred eighty laying hens (Hy-Line$^{(R)}$ Brown), 95wk old, were allotted randomly into five replicates of six dietary treatments: T1; 100% basal diet, T2; 100% basal diet+MgO, T3; 97.5% basal diet+2.5% BM, T4; 97.5% basal diet+2.5% BM+MgO, T5; 95% basal diet+5% BM, T6; 95% basal diet+5% BM+MgO. Magnesium oxide was added at 0.3% of diets. The layers were fed experimental diets for 5wk. There were no significant differences in the weekly L-Car content in CBM among all treatments during the total experimental period, but some of the contrast comparisions showed higher L-Car in CBM of T6. The L-Car contents linearly decreased (p<0.01 or p<0.05) as the layers got older except in T4 (p>0.05). There were significant differences in egg weight (p<0.01) and soft and broken egg ratio (p<0.05). The control (T1) was highest in egg weight and T6 was lowest in soft and broken egg ratio. Among the parameters of egg quality, there were significant differences in eggshell strength (p<0.01) and egg yolk color (p<0.05). Magnesium oxide supplementation increased the eggshell strength and BM tended to decrease egg yolk color. Eggshell color, eggshell thickness, and Haugh unit were not influenced by BM and MgO. In conclusion, BM and MgO did not significantly influence the L-Car in CBM of spent layers. The L-Car content rapidly decreased as the layers became senescent. Eggshell strength was increased by MgO supplementation.
ADL SCORM(Sharable Content Object Reference Model) has been widely accepted as a global reference model for standardizing e-learning technology, and SCORM 2004 4th Edition, a stable version of SCORM, gives content developers the efficient way to build interoperable and reusable e-learning contents. Recently, a number of research efforts have been taken to build on-line SCORM contents based on some traditional training or learning styles. However, they have lacked for supporting more sophisticated learning style such as game based learning, and especially they do not consider employing the specific components of SCORM model for developing game based learning contents in practice. In this work, we elicit some SCORM data elements that is useful for representing game run-time data, and apply those elements to SCORM sequencing of game based learning SCOs(Sharable Content Objects). We thus present the whole procedure of developing SCORM game based learning contents with a sample contents.
Recently, the importance of self-directed learning has emerged in the fields of public education, private education, lifelong education, and vocational training education, in which learners can actively cope with knowledge in an infusion-oriented way. However, there are various theoretical knowledge such as concepts and strategies for self-directed learning, but the situation is insufficient for a system where learners can easily receive content in the academic field they want, depending on the actual self-directed learning operation plan or learning area. Therefore, since it is important to provide various learning content in this paper, we utilize text mining techniques to obtain appropriate information and refine and categorize the meaning. On-line, they want to study a system that provides a variety of content in the academic field that learners are trying to acquire.

New method and device for the on-line measurement of soot concentration in a diesel engine oil are proposed, where the measurement principle is based mainly on attenuated internal total reflection. Various laboratory tests of the detector were performed mainly with carbon black particles of different contamination levels. It was found that the proposed detector could be well used to monitor oil deterioration due to soot contamination. Operational range of the detector was found from 0 to 5 mass percentage of soot content. Test results with water and fuel dilution showed that these effects were not remarkable. However, adsorption of carbon black particles to a measurement surface was considered to be a critical problem in the new detector. Effects of particle deposition onto the interface was experimentally evaluated with the oil temperature and turbulence and discussed throughout this work.
This study investigated the effect of moisture content and loading direction on dowel bearing strength of two types of SCL. Dowel bearing tests of LVL and PSL were conducted with two different MC level, 7.5% and 19%, and two different oriention, L-direction(loading parallel to grain) and X-direction(loading perpendicular to grain). Most of specimens showed typical load-deformation curves and intersected 5% offset line. Failure modes were classified into two categories; spliting(for L-direction specimens) and peeling(for X-direction specimens). Dowel bearing strength generally decreased with increasing MC. The decreasing rate was more significant in X-directon. ESG also decreased with increasing MC, and the ratio of ESG of 7.5% versus 19% was about 1.47. Dowel bearing strength of LVL and PSL in L-direction was higher than that of X-direction. This results indicated that MC and loading orientation had a significant effect on dowel bearing strength of SCL. The average dowel bearing strength of LVL were higher than that of PSL in each loading direction. Two types of probability distribution model were chosen to quantify strength distribution, normal and 2-parameter weibull distribution. The two models showed good agreement with the data, especially in lower tail of the cumulative distribution. Normal and 2-parameter weibull distribution seemed to proper model of the dowel bearing strength for each MC levels.

In support of remote sensing applications for monitoring processes of the Earth system, research was conducted to analyze the basic spectral response related to background soil and vegetation cover characteristics in the visible and reflective infrared wavelengths. Surface samples of seven stations were examined. Five soils were from land-field and two soils from tideland areas. The vegetation cover experiment was conducted on seven soil samples with known natural moisture content (%) by weight. To study the effect of vegetation cover, spectral measurements were taken on five or six vegetation cover treatments of the seven soils with 3 replications in air dry conditions. For collecting RS base data, used spectro-radiometer that measures reflection characteristics between 300~1,100nm was used and measured the reflection of vegetation from bean leaves. The relationships were evaluated for both a general soil line and for the individual lines of five soils, under air-dried condition as well as different vegetation cover ratio, through the determination of the line parameters. As vegetation cover ratio in bean leaves increases, features of soil reflectance decrease and those of plant reflectance become more and more apparent. In proportion to vegetation cover rate, near-infrared reflectance increased and visible reflectance decreased. Analysis results are compared to commonly used vegetation indices(RVI and NDVI ).
