Damaged Neuronal Cells Induce Inflammatory Gene Expression in Schwann Cells: Implication in the Wallerian Degeneration

  • Schwann cells play an important role in peripheral nerve regeneration. Upon nerve injury, Schwann cells are activated and produce various proinflammatory mediators including IL-6, LIF and MCP-1, which result in the recruitment of macrophages and phagocytosis of myelin debris. However, it is unclear how the nerve injury induces Schwann cell activation. Recently, it was reported that necrotic cells induce immune cell activation via toll-like receptors (TLRs). This suggests that the TLRs expressed on Schwann cells may recognize nerve damage by binding to the endogenous ligands secreted by the damaged nerve, thereby inducing Schwann cell activation. To explore the possibility, we stimulated iSC, a rat Schwann cell line, with damaged neuronal cell extracts (DNCE). The stimulation of iSC with DNCE induced the expression of various inflammatory mediators including IL-6, LIF, MCP-1 and iNOS. Studies on the signaling pathway indicate that $NF-{\kappa}B$, p38 and JNK activation are required for the DNCE-induced inflammatory gene expression. Furthermore, treatment of either anti-TLR3 neutralizing antibody or ribonuclease inhibited the DNCE-induced proinflammatory gene expression in iSC. In summary, these results suggest that damaged neuronal cells induce inflammatory Schwann cell activation via TLR3, which might be involved in the Wallerian degeneration after a peripheral nerve injury.

Characterization and Immunopotentiating Effects of the Glycoprotein Isolated from Dioscorea Batatas

  • We demonstrate that glycoprotein isolated from Dioscorea batatas (GDB) has immunostimulatory effects including macrophage activation. Analysis of infiltration of inflammatory cells into peritoneal cavity showed GDB treatment significantly increased the recruitment of macrophages, lymphocytes, neutrophils, and monocytes into the peritoneal cavity. Treatment of spleen cells isolated from C57BL/6 mice with GDB significantly increased the proliferation of B cells and T cells induced by LPS and ConA, respectively. Treatment with GDB significantly increased the cytolytic capacity of NK cells and macrophages against YAC-1 and B16 cells, respectively. In order to further confirm and investigate the mechanism of GDB on macrophage activation, we analyzed the effects of GDB on the cytokine expression including iNOS, IL-1${\beta}$, and TNF-${\alpha}$ in mouse macrophage cell line, RAW 264.7 cells. RT-PCR and ELISA showed that GDB increased the expression of IL-1${\beta}$, and TNF-${\alpha}$, whereas iNOS was not induced by GDB. Collectively, this series of experiments indicates that GDB stimulates immune system including macrophage activation.

Cytokines Regulate the Expression of the Thymus and Activation-Regulated Chemokine (TARC; CCL17) in Human Skin Fibroblast Cells

  • Allergic inflammation is thought to be a Th2 cell-dominant immune response during which tissue-resident fibroblasts produce chemokines which contribute to the recruitment of migratory leukocytes to sites of tissue injury. Thymus and activation-regulated chemokine (TARC; CCL17) is a potent member of the CC chemokine family and a selective chemoattractant for Th2 cells. In order to study the regulatory profiles of TARC production by $TNF-{\alpha}$, $IFN-{\gamma}$, and Il-4 in human normal skin fibroblast, CCD-986sk cell line was used. The expression of TARC protein was measured using ELISA, and mRNA level was detected by RT-PCR. The combination of $TNF-{\alpha}$ and IL-4 induced a time-and dose-dependent synergistic increase in the expression of TARC at both protein and mRNA levels in the cultured human skin fibroblasts. Exposure of the cells to single cytokine had no effect on TARC expression. The high concentration (100 ng/ml) and long incubation time (72 h) of $IFN-{\gamma}$ further enhanced the TARC production induced by $TNF-{\alpha}$/lL-4 in the skin fibroblast. This synergistic effect of Th1 and Th2 type cytokines on TARC production by skin fibroblasts may contribute to the inflammatory cell infiltration and tissue damage with allergic inflammation.

한국영화의 초국가적 수용: 영화리뷰를 중심으로 (Transnational Reception of Korean Film: Analyses of Film Reviews)

  • This paper is based on the view that film should be conceived as a form of cultural practice whose meaning is always in the process of being produced within diverse socio-cultural contexts and aims to examine the ways in which the meaning of Korean film is (re)mediated or received in diverse cultural contexts outside the country. In this paper, we employ two theoretical grounds. Firstly, it positions itself in line with the audience studies within the field of cultural studies where the audience is conceived as active agents who produce the meaning of a popular culture text. The recruitment of the theoretical propositions from the audience studies enables recognition of the significance of the reception in film practice which recently seems to be oriented on production and distribution. Secondly, we conceive transnationality of film as that which is being produced in the process of transaction between the film and the audience, that is to say, transnationality is a form of discourse that emerges upon cultural interaction. The empirical work involves examination of a set of reviews of four films--Chihwaseon, Oldboy, Thirt, Poety--that have been published in daily newspapers and some popular film magazines in the U. S., the U. K. and France. Through the analysis of the film reviews, we identify four interpretive schemes or rather discourses recruited via which the Korean films are approached and understood: auteurism, formalism, universal themes, emotional response. We propose that these four kinds of discourse provide a common ground for the audience from different cultural backgrounds to understand Korean film. Furthermore, we also suggest that transnationality of Korean cinema needs to be reconsidered in terms of the reception as the audience from different socio-cultural backgrounds should be understood as active agents who are capable of engaging in Korean cultural texts such as film in their own way producing various meanings and these are also constituent of the meaning of the cultural texts.

  • 최근 고용시장의 불안과 맞물려서 교사 직업에 대한 선호도가 높아짐에 따라 사범대학 및 교육대학의 선호도가 높아지고 있다. 그러나 점차 인구수 감소와 맞물려서 교육부에서 발표되는 임용T.O는 매년 줄어들고 있고 그에 비해 입학학생수 및 재수생숫자는 늘어나 경쟁률은 가파르게 오르고 있는 사정이다. 이로 인하여 사범대학에서는 입학과 동시에 임용시험에 매진하고자하여 대학교가 교원 임용후보자 경쟁시험을 위한 하나의 거대한 학원이 되어가는 분위기이다. 이에 본 연구에서는 학점 및 학생들의 전반적인 학교생활 및 스터디그룹 활용 여부 등을 종합하여 딥 러닝을 이용한 교원임용 후보자 경쟁시험 합격여부의 연관성을 파악하여 사범대학 재학생들에게 학교생활의 청사진을 제시하고자 한다.

사람 Mast cell에서의 케모카인에 대한 백연치효탕(白蓮治哮湯)의 효과(效果) (Effect of Baekryunchihyo-tang(白蓮治哮湯) on Expression of Chemokines in Human Mast Cell Line (HMC-I))

  • Background : Although the pathophysiology of asthma has been reported, its mechanism has not been fully elucidated. The mast cell is an effector cells in allergic inflammation and secretes a number of chemokines. Chemokines are important for the recruitment of leukocytes to sites of infection, which is essential in host defense. Chemokines also contribute to the pathogenesis of several disorders such as asthma, chronic bronchitis, atopic dermatitis, allergic rhinitis, and rheumatoid arthritis. Objective : In this study, the aim was to identify the effect of Baekryunchihyo-tang(白蓮治哮湯) on expression of chemokines. This was examined by RT-PCR using the human mast cell line (HMC-l) Materials and Methods : HMC-l cells were used, which is known to secrete and express chemokines. In order to investigate the protective effect of Baekryunchihyo-tang(白蓮治哮湯), HMC-l cells were incubated with pretreatment of Baekryunchihyo-tang(白蓮治哮湯) for 24 hrs. RT-PCR analyses of chemokine genes of cells pretreated with Baekryunchihyo-tang(白蓮治哮湯) showed that expressions of IL-8, $MIP-l{\beta}$, and RANTES genes in these cells were lower and $MIP-l{\alpha}$ showed a similar pattern compared to the calcium ionophore-treated group. In addition, cell cytotoxicity concentration measurements were performed by MTT assay method. Results : After stimulation with 1 uM calcium ionophore A23178 for 2 hrs, IL-8, major one of CXC chemokines, was highly expressed, and expression of $MIP-l{\beta}$ and RANTES (CC chemokines) increased, while expression of $MIP-l{\alpha}$ did not change. The cell cytotoxicity of Baekryunchihyo-tang(白蓮治哮湯) with treatments at various concentrations and times was not observed, respectively. Conclusion : This study suggests that Baekryunchihyo-tang(白蓮治哮湯) has dose-dependent effects on mRNA expression of IL-8(CXC chemokines), $MIP-l{\beta}$ and RANTES(CC chemokines) in human mast cellline(HMC-l). So these herbal medicines may inhibit the inflammatory process of asthma. Advanced studies are required to investigate the mechanism of inhibition by herbal medicine in the asthma model. This study provides basic data on the possibility of the clinical treatment of Baekryunchihyo-tang(白蓮治哮湯) for allergic disorders.

  • Blood monocytes are the precursors for the lipid-laden foam cells of early atherosclerotic lesions. Monocyte chemoattractant protein-1(MCP-1), a CC chemokine, and chemokine receptor 2 (CCR2) play a crucial role in the recruitment of monocytes to the vascular lesion. Using the human monocyte THP-1 cell line, we investigate the inhibitory effects of methanol extracts of 127 medicinal herbs on MCP-1 induced chemotaxis. Seven kinds of methanol extracts of medicinal herbs showed above 40% inhibitory effect with the concentration of $25{\mu}g/ml$. They were divide three fractions of $CHCI_3$, BuOH, $H_2O$ to use solvent partition. Among them, butanol extract of Junci Medulla and $CHCI_3$ extract of Clematidis Radix are showed significant inhibitory activities (above 50% inhibition) at the same concentration.

Arsenic Trioxide Promotes Paclitaxel Cytotoxicity in Resistant Breast Cancer Cells

  • A partial response or resistance to chemotherapeutic agents is considered as a main obstacle in treatment of patients with cancer, including breast cancer. Refining taxane-based treatment procedures using adjuvant or combination treatment is a novel strategy to increase the efficiency of chemotherapy. PPM1D is a molecule activated by reactive oxygen species. whose expression is reported to modulate the recruitment of DNA repair molecules. In this study we examined the impact of arsenic trioxide on efficacy of paclitaxel-induced apoptosis in paclitaxel-resistant MCF-7 cells. We also investigated the expression of PPM1D and TP53 genes in response to this combination treatment. Resistant cells were developed from the parent MCF-7 cell line by applying increasing concentrations of paclitaxel. MTT assays were applied to determine the rate of cell survival. DAPI staining using fluorescent microscopy was employed to study apoptotic bodies. Real-time RT-PCR analysis was also applied to determine PPM1D mRNA levels. Our results revealed that combination of arsenic trioxide and paclitaxel elevates the efficacy of the latter in induction of apoptosis in MCF-7/PAC resistant cells. Applying arsenic trioxide also caused significant decreases in PPM1D mRNA levels (p<0.05). Our findings suggest that arsenic trioxide increases paclitaxel-induced apoptosis by down regulation of PPM1D expression. PPM1D dependent signaling can be considered as a novel target to improve the efficacy of chemotherapeutic agents in resistant breast cancer cells.

Membrane Topology of Helix 0 of the Epsin N-terminal Homology Domain

  • Specific interaction of the epsin N-terminal homology(ENTH) domain with the plasma membrane appears to bridge other related proteins to the specific regions of the membrane that are invaginated to form endocytic vesicles. An additional $\alpha$-helix, referred to as helix 0 (H0), is formed in the presence of the soluble ligand inositol-1,4,5-trisphosphate [$Ins(1,4,5)P_3$] at the N terminus of the ENTH domain (amino acid residues 3-15). The ENTH domain alone and full-length epsin cause tubulation of liposomes made of brain lipids. Thus, it is believed that H0 is membrane-inserted when it is coordinated with the phospholipid phosphatidylinositol-4,5-bisphosphate [$PtdIns(4,5)P_2$], resulting in membrane deformation as well as recruitment of accessory factors to the membrane. However, formation of H0 in a real biological membrane has not been demonstrated. In the present study, the membrane structure of H0 was determined by measurement of electron paramagnetic resonance (EPR) nitroxide accessibility. H0 was located at the phosphate head-group region of the membrane. Moreover, EPR line-shape analysis indicated that no pre-formed H0-like structure were present on normal acidic membranes. $PtdIns(4,5)P_2$ was necessary and sufficient for interaction of the H0 region with the membrane. H0 was stable only in the membrane. In conclusion, the H0 region of the ENTH domain has an intrinsic ability to form H0 in a $PtdIns(4,5)P_2$-containing membrane, perhaps functioning as a sensor of membrane patches enriched with $PtdIns(4,5)P_2$ that will initiate curvature to form endocytic vesicles.

서비스직 근로자의 개인-직무 적합성과 직무만족 관계에서 조직후원인식의 매개효과 분석 (The Effect of Person-Job Fit on Job Satisfaction for the Employees of the Service Sector: Focused on the Mediating Effects of Perceived Organizational Support)

  • 본 연구는 서비스직 근로자의 개인-직무 적합성과 직무만족 간의 관계에서 조직후원인식의 매개효과를 규명하고자 하였다. 이를 위하여 개인-직무 적합성, 직무만족, 조직후원인식에 대한 선행연구를 기반으로 연구모형을 설정하고 가설을 도출하였다. 특히, 조직후원인식은 정서적 조직후원인식과 수단적 조직후원인식으로 세분화하였다. 본 연구를 위하여 국내 기업에서 서비스 직무를 담당하고 있는 근로자를 대상으로 설문 조사를 실시하였으며, 온라인으로 총 350명에게 배포되었으며, 최종 281명의 응답 결과를 분석하여, 유효 회수율은 80.3%를 기록하였다. 그 결과 첫째, 개인-직무적합성은 직무만족과 조직후원인식에 정(+)의 영향을 미치는 것으로 나타났다. 둘째, 개인-직무 적합성과 직무만족간의 관계에서 조직후원인식의 하위요인 중 정서적 조직후원인식은 매개효과가 있는 나타났으나, 수단적 조직후원인식은 매개효과가 없는 것으로 나타났다. 본 연구결과를 바탕으로, 고객 점점에서 감정노동을 수행하는 서비스직 근로자의 개인-직무 적합성을 판단해 볼 수 있는 채용 제도의 변화, 조직 구성원의 정서적 지원체계 구축을 위한 조직 문화 및 인사제도 마련, 서비스직 근로자들의 직무만족을 높이기 위한 보상 및 인력개발 체계 구축 등 인사교육 전반에 걸친 이론적 및 실천적 시사점을 제시하였다.
