Molecular and Morphological Characteristics of a New Species Collected from an Insect (Cicindela transbaicalica) in Korea

  • To exploit insect-derived fungi, insects were collected from seven different regions in Korea, including Gyeongbuk, Goryeong, and several fungi were isolated from them. A fungal strain designated 21-64-D was isolated from riparian tiger beetle (Cicindela transbaicalica) and morphologically identified as a species belonging to the genus Oidiodendron. Phylogenetic analysis using the nucleotide sequences of internal transcribed spacer (ITS) regions and the partial sequence of the large subunit of the nuclear ribosomal RNA (LSU) gene revealed the distinct phylogenetic position of the isolate among recognized Oidiodendron species including its closest neighbors O. chlamydosporicum, O. citrinum, O. maius, and O. pilicola. The hyphal and conidial morphology of the strain, particularly club-shaped hyphae, clearly differentiated it from its close relatives. Results indicated that 21-64-D is a novel species in the genus Oidiodendron, for which the name Oidiodendron clavatum sp. nov. is proposed.

  • Oidiodendron flavum KNU13-6 was isolated for the first time from field soil in Korea and identified based on the internal transcribed spacer region (ITS) of rDNA and morphological characteristics. Based on phylogenetic analysis of ITS and morphological characteristics, the species has not been previously reported in Korea.

Four Unreported Endophytic Fungi Isolated from Roots of Quercus spp. in Korea

  • We isolated endophytic fungi from the roots of Quercus spp. in Korea. The isolates were identified based on morphological and molecular characteristics from the nucleotide sequences of the internal transcribed spacer and large subunit of rDNA regions. We confirmed that the four species we isolated - Oidiodendron citrinum, Acremonium crotocinigenum, Mammaria echinobotryoides, and Pleotrichocladium opacum - have not been recorded previously in Korea. Herein, we present their morphological characteristics and the result of phylogenetic analysis.

광릉요강꽃과 복주머니란의 뿌리에서 분리한 내생균의 다양성 (Diversity of Endophytic Fungi Isolated from Roots of Cypripedium japonicum and C. macranthum in Korea)

  • 본 연구에서는 경기도 포천과 강원도 춘천에 자생하는 광릉요강꽃과 강원도 삼척에서 자생하는 복주머니란의 뿌리에서 내생균을 순수 분리하였고, 분리된 균주의 ITS 부위의 염기서열을 이용하여 계통 분석하였다. 그 결과 광릉요강꽃에서는 11종의 균주가 분리되었고, 복주머니란에서는 15종의 균주가 분리되었으며, Leptodontidium orchidicola, Humicola fuscoatra var. fuscoatra, Umbelopsis dimorpha 등의 균주는 공통으로 분리되었다. 분리된 균주 중 Oidiodendron echinulatum, Pseudogymnoascus roseus, Geomyces vinaceus, Cryptosporiopsis ericae, Umbelopsis dimorpha, Chaetomium cupreum 은 국내 미기록종으로 확인하였다.

Isolation and Identification of Three Newly Reported Ascomycete Fungal Species Isolated from Soil in Korea

  • In this study, three fungal isolates belonging to the phylum Ascomycota under classes Leotiomycetes, Eurotiomycetes, and Sordariomycetes were isolated from soil in Korea. These species were designated as KNUF-22-003, KNUF-22-005, and KNUF-20-NI016, respectively, and identified based on their phylogenetic relationships and morphological characteristics. The isolates were confirmed through molecular phylogenetic analyses of their internal transcribed spacer (ITS) region, 28S rDNA large subunit (LSU), and actin (ACT1 ) gene sequences. Cultural and morphological characteristics of strains KNUF-22-003, KNUF-22-005, and KNUF-20-NI016 were matched with Chaetomella oblonga CBS110.78T, Oidiodendron chlamydosporicum CBS403.69T, and Sarocladium subulatum CBS217.35T, respectively. To the best of our knowledge, this is the first report on C. oblonga, O. chlamydosporicum, and S. subulatum in Korea.

균근균을 활용한 구상나무(Abies koreana E.H.Wilson) 유묘의 생육 증진 효과 (Growth Enhancement Effect of Korean Fir (Abies koreana E.H.Wilson) Seedlings Using Mycorrhizal Fungi )

  • This study investigated the effects of two mycorrhizal fungi, Pisolithus tinctorius and Oidiodendron maius, on the growth and age-specific responses of Korean fir (Abies koreana E.H.Wilson) seedlings. The experiment was conducted from March 2020 to March 2023 at the National Institute of Forest Science in Suwon, South Korea to evaluate plant height, root length, root collar diameter, and biomass in the treatment and control groups. Significant growth increases were observed in all treatment groups compared to the control, with the OI (O. maius) treatment demonstratingthe highest growth increase (p < 0.05). In one-year-old seedlings, the PT (P. tinctorius) treatment demonstrated overall higher growth than the OI treatment and control, with PT demonstrating an approximately 1.3 times increase than OI. The PT and OI treatments both exhibited higher growth than the control in the three-year-old seedlings, with less difference between the two treatments than in the one-year-olds. Both the treatments showed approximately 1.8 times more growth than the control in the five-year-old seedlings, and the OI treatment showed approximately 0.5 times more growth than PT. These results confirm that the OI treatment is generally more effective for the growth of Korean fir seedlings. Although the PT treatment was more beneficial in the one-year-old seedlings, OI was more effective in three-year-old and older seedlings. This suggests that early mycorrhizal inoculation has a significant long-term effecton growth, emphasizing the importance of age-specific mycorrhizal applications in forest conservation and restoration strategies.

Pyrosequencing and Taxonomic Composition of the Fungal Community from Soil of Tricholoma matsutake in Gyeongju

  • Tricholoma matsutake is an ectomycorrhizal fungus that has a symbiotic relationship with the root of Pinus densiflora. Soil microbial communities greatly affect the growth of T. matsutake, however, few studies have examined the characteristics of these communities. In the present study, we analyzed soil fungal communities from Gyeongju and Yeongdeok using metagenomic pyrosequencing to investigate differences in fungal species diversity, richness, and taxonomic composition between the soil under T. matsutake fruiting bodies (Sample 2) and soil where the fairy ring of T. matsutake was no longer present (Sample 1). The same spot was investigated three times at intervals of four months to observe changes in the community. In the samples from Yeongdeok, the number of valid reads was lower than that at Gyeongju. The operational taxonomic units of most Sample 2 groups were less than those of Sample 1 groups, indicating that fungal diversity was low in the T. matsutake-dominant soil. The soil under the T. matsutake fruiting bodies was dominated by more than 51% T. matsutake. From fall to the following spring, the ratio of T. matsutake decreased. Basidiomycota was the dominant phylum in most samples. G-F1-2, G-F2-2, and Y-F1-2 had the genera Tricholoma, Umbelopsis, Oidiodendron, Sagenomella, Cladophialophora, and Phialocephala in common. G-F1-1, G-F2-1, and Y-F1-1 had 10 genera including Umbelopsis and Sagenomella in common. From fall to the following spring, the amount of phyla Basidiomycota and Mucoromycota gradually decreased but that of phylum Ascomycota increased. We suggest that the genus Umbelopsis is positively related to T. matsutake.
