A novel proteolytic enzyme with fibrinolytic activity, FSP3, was purified from the recently isolated Streptomyces sp. P3, which is a novel bacterial strain isolated from soil. FSP3 was purified to electrophoretic homogeneity by ammonium sulfate precipitation, anion exchange, and gel filtration. FSP3 is considered to be a single peptide chain with a molecular mass of 44 kDa. The maximum activity of the enzyme was observed at 50℃ and pH 6.5, and the enzyme was stable between pH 6 and 8 and below 40℃. In a fibrin plate assay, FSP3 showed more potent fibrinolytic activity than urokinase, which is a clinical thrombolytic agent acting as a plasminogen activitor. The activity was strongly inhibited by the serine protease inhibitor PMSF, indicating that it is a serine protease. Additionally, metal ions showed different effects on the activity. It was significantly suppressed by Mg2+ and Ca2+ and completely inhibited by Cu2+, but slightly enhanced by Fe2+. According to LC-MS/MS results, its partial amino acid sequences are significantly dissimilar from those of previously reported fibrinolytic enzymes. The sequence of a DNA fragment encoding FSP3 contained an open reading frame of 1287 base pairs encoding 428 amino acids. FSP3 is a bifunctional enzyme in nature. It hydrolyzes the fibrin directly and activates plasminogen, which may reduce the occurrence of side effects. These results suggest that FSP3 is a novel serine protease with potential applications in thrombolytic therapy.

The occurrence of THMs, the characteristics of THMs formation and removal of THMs were investigated. The treatment train of M plant consists of prechlorination, flocculation, sedimentation, filtration, ozonation, activated carbon and postchlorination. The study of THM formation indicated that about 92% of the THMs were formed in the flocculation/sedimentation/filtration process which affected by prechlorination. The formation of THMs was highly correlated to $KMnO_4$ consumption and water temperature in raw water. The regression model had showed 0.72~0.80 of determination coefficient so it could be used to predict the amount of THMs formation in finished water. Compared to the prechlorination process, the THMs formation was reduced in interchlorination process. With the addition of PAC, fewer THMs were formed in PAC-chlorination process than in chlorination-PAC process. Our results showed that air stripping could be used to remove the existing THMs.
A total of 260 dogs were randomly selected from two different treed kennels that brucellosis has occurred (group 1, 126 dogs), and random selected breed kennel (group 2, 134 dogs), and monitored for Brucella canis (B. canis) by 2-mercaptoethanol rapid slide agglutination test (2ME-RSAT) and bacterial culture method. For the differentiation, PCR-RFLP using omp-31, wbkA and per genes used for 52 of B canis strains (strain I) isolated in this study and 3 of B. canis strains (strain II) isolated in 1994 in Korea. 2ME-RSAT revealed that 63/126 dogs (50.0%) and 12/134 dogs (9.0%) were positive in group I and group II, respectively. Bacterial culture revealed that 47/126 dogs (37.3%) and 5/134 dogs (3.7%) were positive in group I and group II, respectively. As the results of PCR-RFLP, $\underline{omp}-31$ was amplified from all Brucella spp, except B. abortus. All B. canis isolates showed unique PCR-RFLP pattern following digestion with Bmel8I. However, all Brucella spp. showed the same PCR-RFLP pattern following digestion with SalI. PCR-RFLP analysis of wbkA revealed that all Brucella spp. showed the same pattern following digestion with HindIII. PCR-RFLP analysis of per revealed that B. abortus 544 and B. melitensis 63/9 showed the same pattern, but different from B. suis and B. canis following digestion with HindIII.

Aims: To delineate the histopathological pattern of prostate diseases and to highlight age variations in prostate specific antigen (PSA) values and histopathological features. Materials and Methods: A retrospective review was made of all prostate biopsy reports seen between January 2006 and December 2013 at the King Fahad Hospital, Madinah, Saudi Arabia. Prostate lesions were tabulated and classified into benign and malignant groups. Histological scoring of adenocarcinomas was accomplished using the Gleason system. PSA values were correlated with Gleason scores. Results: Of 417 prostate lesions reviewed, 343 (82.3%) were benign and 74 (17.7%) were malignant, giving a benign to malignant ratio of 4.6:1. Benign prostatic hyperplasia (both with and without inflammation) was the commonest prostatic lesion and accounted for 80.3% of all cases and 97.6% of all benign cases. The age range was 20 to 97 years with a mean of 69.2 years and a peak age group at 70-79 years. Seventy one cases of adenocarcinoma accounted for 95.9% of the total of 74 malignant tumors. It showed an age range of 44 to 95 years, a mean age of 70.9 years and peak prevalence in the 80-89 year age group. Gleason score seven was the most frequent (39.4%) in occurrence. Most adenocarcinomas, 41 cases (57.7%), were moderately differentiated (Gleason score of 5-7). PSA values ranged widely between 16-1,865ng/ml with a mean of 363.4ng/ml. Elevated PSA (>100ng/ml) levels were found in 53 (81.6%) patients. There was a statistically significant positive correlation between serum PSA level and Gleason score (p=0.0304). Conclusions: Prostatic lesions constitute a significant source of morbidity among adult males in Madinah. Benign prostatic hyperplasia was the commonest benign prostatic lesion and adenocarcinoma was the commonest histological subtype of prostatic cancer.
A bacterial disease of tea plants(Camellia sinensis L.) was found in the graftage nursery grown under vinyl house conditions in Suncheon city, Korea, in spring of 2002. The primary symptoms of the disease include small, water-soaked and dark brown spot development on the young leaves. This spot gradually increases in size, especially taking on elongate shape along the midrib or vein of the leaf, and then turns black. The diseased leaves were defoliated easily. Ten strains were isolated from the infected leaf. Inoculation on tea leaf with these isolates produced the same symptoms of naturally infected plants. On the basis of stain reactions, morphological characterization, colony pattern, physiological and biochemical reactions, the bacterium was identified as Pseudomonas syringae pv. theae. This is the first report of brown blight of tea plant in Korea.

Together with the Fusarium graminearum species complex, F. culmorum is a major member of the causal agents of Fusarium head blight on cereals such as wheat, barley and corn. It causes significant yield and quality losses and results in the contamination of grain with mycotoxins that are harmful to humans and animals. In Korea, F. culmorum is listed as a quarantine fungal species since it has yet to be found in the country. In this paper, we report that two isolates (J1 and J2) of F. culmorum were collected from the air at a rice paddy field in Korea. Species identification was confirmed by phylogenetic analysis using multilocus sequence data derived from five genes encoding translation elongation factor, histone H3, phosphate permease, a reductase, and an ammonia ligase and by morphological comparison with reference strains. Both diagnostic PCR and chemical analysis confirmed that these F. culmorum isolates had the capacity to produce nivalenol, the trichothecene mycotoxin, in rice substrate. In addition, both isolates were pathogenic on wheat heads and corn stalks. This is the first report on the occurrence of F. culmorum in Korea.
The stem rot of Chrysanthemum zawadskii var. latilobum occurred in the landscape field of Sanggwan-myeon, Wanju-gun, Jeonbuk, Korea in September 2009. The typical symptoms of the disease were water-soaking and dark brown rot on the stem, wilting and blighting. Most infected plants died of the disease. White mycelial mats were spread over lesions and brown sclerotia were formed on stems and near soil surface. The sclerotia were spherical or irregular, 0.8-3.5 (mean 1.8) mm in size on potato dextrose agar, brown in color. The optimal temperature range of hyphal growth was $25-30^{\circ}C$ and the hyphal diameter was 4-9 ${\mu}m$. Clamp connections were observed in the hyphae of the fungus grown on potato dextrose agar. Pathogenicity of the casual organism was provided on C. zawadskii according to Koch's postulate. The fungus was identified as Sclerotium rolfsii on the basis of mycologiclal characteristics and pathogenicity test to host plants. This is the first report on the stem rot of C. zawadskii caused by S. rolfsii in Korea.
A total of 80 cows, including 40 top mastitis resistant and 40 top mastitis susceptible animals as Group I and Group II, were selected from a population of 520 cows based on clinical mastitis occurrence. PCR-SSCP analysis on four fragments within the 5'region and two fragments of Exons 4,15 of bovine lactoferrin (bLF) revealed that four fragments-P1,P4,E4,E15-had polymorphisms which totally included six base mutations, and only two of them had significant differences in allele frequencies between resistant and susceptible groups, P1 (53.7% vs. 70.0%, p<0.05) and P4 (55.0% vs. 68.8%, p<0.05). Further study on these two promising markers combined with the milk performance traits of cows demonstrated that their selection would result in higher fat percentage (p<0.05), lower Somatic Cell Score (SCS) (p<0.05) and Clinical Mastitis Residuals (CMR) (p<0.01) indicating higher mastitis resistance and lower milk yield (p<0.05). The putative transcription factor binding sites in the 5'region were also studied by using MatInspector 7.2.2 software, and two signal pathways regulating the expression of bLF including the NF-${\kappa}B$ pathway and nuclear hormone receptor pathway were predicted.
Four of nine exoplanets found by microlensing were detected by the resonant caustic, which represents the merging of the planetary and central caustics at the position when the projected separation of a host star and a bounded planet is s~1. One of the resonant caustic lensing events, OGLE-2005-BLG-169, was a caustic-crossing high-magnification event with A_max ~800 and the source star was much smaller than the caustic, nevertheless the perturbation was not obviously apparent on the light curve of the event. In this paper, we investigate the perturbation pattern of the resonant caustic to understand why the perturbations induced by the caustic do not leave strong traces on the light curves of high-magnification events despite a small source/caustic size ratio. From this study, we find that the regions with small magnification excess around the center of the resonant caustic are rather widely formed, and the event passing the small-excess region produces a high-magnification event with a weak perturbation that is small relative to the amplification caused by the star and thus does not noticeably appear on the light curve of the event. We also find that the positive excess of the inside edge of the resonant caustic and the negative excess inside the caustic become stronger and wider as q increases, and thus the resonant caustic-crossing high-magnification events with the weak perturbation occur in the range of q $\leq$ 10-4. We determine the probability of the occurrence of events with the small excess $|\varepsilon|{\leq}3%$ in high-magnification events induced by a resonant caustic. As a result, we find that for the Earth-mass planets with a separation of ~2.5 AU the resonant caustic high-magnification events with the weak perturbation can occur with a significant frequen.
The adrenergic receptor beta 2 (ADRB2) plays a role in various physiological responses of the muscle to exercise, such as contraction and relaxation. Given its important role in muscle function, we investigated the structure of the horse ADRB2 gene and its expression pattern after exercise to determine if it can serve as a putative biomarker for recovery. Evolutionary analyses using synonymous and non-synonymous mutation ratios, were compared with other species (human, chimpanzee, mouse, rat, cow, pig, chicken, dog, and cat), and revealed the occurrence of positive selection in the horse ADRB2 gene. In addition, expression analyses by quantitative polymerase chain reaction exhibited ubiquitous distribution of horse ADRB2 in various tissues including lung, skeletal muscle, kidney, thyroid, appendix, colon, spinal cord and heart, with the highest expression observed in the lung. The expression of ADRB2 in skeletal muscle was significantly up-regulated about four folds 30 minutes post-exercise compared to pre-exercise. The expression level of ADRB2 in leukocytes, which could be collected with convenience compared with other tissues in horse, increased until 60 min after exercise but decreased afterward until 120 min, suggesting the ADRB2 expression levels in leukocytes could be a useful biomarker to check the early recovery status of horse after exercise. In conclusion, we identified horse ADRB2 gene and analyzed expression profiles in various tissues. Additionally, analysis of ADBR2 gene expression in leukocytes could be a useful biomarker useful for evaluation of early recovery status after exercise in racing horses.
