Effects of a Low Calcium Diet and Oxalate Intake on Calcium Deposits in Soft Tissues and Bone Metabolism in Ovariectomized Rats (저 칼슘 및 수산을 첨가한 식이가 난소절제한 흰쥐에서 조직의 칼슘 침착과 골격대사에 미치는 영향)

  • It is controversial whether low calcium intake, commonly associated with osteoporosis, results in calcium accumulation in soft tissues. This study was conducted to investigate the effects of low calcium (Ca) and oxalate (ox) intake on soft-tissue Ca deposits and bone metabolism in ovariectomized (ovx) rats. Eight week old female Sprague-Dawley rats were ovariectomized and divided into four groups. The rats were fed experimental diets containing low (0.1%, w/w) or normal (0.5%, w/w) Ca with or without sodium oxalate (1%, w/w); Sham/NCa, Ovx/NCa, Ovx/LCa, Ovx/NCa-ox, Ovx/LCa-ox for 6 weeks. All ovx rats showed a remarkable increase in body and tissue weight, glutamic oxaloacetic transaminase, glutamic pyruvic transaminase, blood urea nitrogen, alkaline phosphatase, and decreases in weight, ash, and Ca contents, as well as bone breaking force compared to those in sham rats. Serum Ca concentration was not significantly affected by dietary Ca levels or ox intake. Kidney Ca, ox acid content, and microscopic Ca deposition increased remarkably in the Ovx/LCa-ox group compared to those in the other groups. Ca content in the spleen and aorta also increased significantly, but the weight contents, Ca, bone breaking force, and Ca and oxalic acid in feces decreased significantly in the Ovx/LCa-ox group. Serum parathyroid hormone levels were not significantly different among the groups. These results indicate that low Ca intake decreased bone mineral content and increased Ca deposits in soft tissues, which was aggravated by ox intake in ovx rats. Thus, high ox intake may result in a kidney disorder in patients with osteoporosis who eat a low Ca diet.

Effects of Defatted Safflower Seed Powder on Intestinal Physiology and Fecal Short-Chain Fatty Acids in Overiectomized Female Rats Fed High Cholesterol Diets (고콜레스테롤식에 홍화박의 첨가가 난소절제 흰쥐의 장 생리기능과 분변 Short-Chain Fatty Acid 함량에 미치는 효과)

  • Much attention has been given to the role of safflower (Carthamus tinctorius L.) seed in preventing and treating osteroporosis recently. Although numerous studies were done on effects of safflower oil, no attention was given to the role of dietary fiber of safflower seeds. This study was aimed at investigating physiological significance of defatted safflower seed as a source of dietary fiber. Sprague Dawley female rats were assigned one of 5 groups: ovariectomized control (Ovx-control) group and ovariectomized rats 15% (Ovx-ss15) and 30% defatted safflower seed (Ovx-SS30), sham-operated (Sham) group and a normal group. Cholesterol was supplemented to all diets at 0.5% except the normal diet. Ovariectomized rats were pair-fed isocalorically to the Sham group. Ovariectomy caused heavier body weight, but feeding 30% safflower seed brought back to the level of Sham group. Activities of disaccharidases of jejunal mucosa were significantly lowered in Ovx-control group compared to those of Sham, and supplementation of safflower seed tended to increase the activities. Fecal weight of Ovx-SS15 and Ovx-SS30 were almost twice as those of Ovx-control Sham. Propionic acid and butyric acid concentrations per g of feces of Ovx-SS15 and daily excretion of these fatty acids were significantly increased as compared to those of Sham and Ovx-control. In concentration defatted safflower seed supplementation significantly increased fecal bulk and short chain fatty acid production in large intestine of rats.

Effects of estrogen on the ovariectomized rat (에스트로겐이 난소가 제거된 흰쥐에 미치는 영향)

  • The aim of this study was to determine the effects of body weight, organ weight, hematological values and biochemical parameters by estrogen on the ovariectomized (OVX) rats. The animals were divided into 4 groups. Intact group (n=10) received no treatment and operation. Sham group (n=10) received only sham operation and no treatment. OVX group (n=10) received operation and no treatment. OVX+E2(n=10) received operation and estrogen. The body weight of each group increased, but that of OVX+E2 group decreased. There were very significant differences (P<0.001) of body weights between OVX+E2 group and the other groups. Also, the wight of important organs such as heart, liver, spleen and kidney was measured. The heart weight was significantly lower(P<0.05) in the OVX+E2 group than the Intact group. The liver weight of the OVX+E2 group was significantly different in comparison with those of the other groups. On the other hand, there were no significantly differences in the organ weight of spleen and kidney between the OVX+E2 group and the other remaining groups. The hematological values of WBC, RBC, MeV, MCH and MCHC were not significantly different among the groups. The serum total protein and albumin decreased significantly in the OVX+E2 group as compared to those in the intact group. But, there were no significant differences in Ca, IP and Mg among the groups. We conclude that estrogen was significantly decreased the body weight in the ovariectomized rats. Our findings suggest that estrogen may influence the process of lipid packaging and absorption in the ovariectomized rats.

Yuklinzu Aqueous Extracts Ameliorate Experimental Climacterium Symptoms Induced by Ovariectomy in Mouse (난소적출로 유발된 갱년기장애 마우스 모델에서 육인주(毓麟珠) 열수 추출물의 증상 개선 효과)

  • Objectives: The object of this study was to observe the complex anti-climacterium potentials of Yuklinzu aqueous extracts (YLZ), using bilateral ovariectomy (OVX) female ddY mice similar to women postmenopausal symptoms, as including cardiovascular diseases, obesity, hyperlipidemia, osteoporosis and hepatic steatosis. Methods: In order to evaluate anti-climacterium effects of YLZ, six groups of mice were used; sham control, OVX control, 17β-estradiol, YLZ 500, 250 and 125 mg/kg treated groups. Since 28 days after bilateral OVX surgery, YLZ were administered orally for 84 days, once a day. And then we evaluated anti-climacterium effects divided into five categories; estrogenic effects, anti-obesity, hypolipidemic effects, hepatoprotective effects and anti-osteoporotic effects. The results of YLZ were compared with 17β-estradiol 0.03 ㎍/head/day subcutaneous treated OVX mice. Results: As a result of OVX, obvious changes related to the estrogen-deficient menopausal symptoms - obesity, hyperlipidemia, hepatic steatosis and osteoporosis were displayed in mice. However, these menopausal symptoms induced by OVX were significantly inhibited by 84 days of consecutive treatment of 17β-estradiol, YLZ 500, 250 and 125 mg/kg, respectively. Especially, YLZ showed obvious dose-dependent inhibitory activities on the OVX-induced climacterium changes in mice, and YLZ 500 mg/kg showed comparable inhibitory effects against menopausal symptoms in comparison with those of 17β-estradiol 0.03 ㎍/head/day subcutaneous treatment. Conclusions: The results suggest that oral administration of YLZ 500, 250 and 125 mg/kg has obvious dose-dependent favorable anti-climacterium effects in OVX mice. Especially, YLZ 500 mg/kg showed comparable inhibitory effects against menopausal symptoms in comparison with those of 17β-estradiol 0.03 ㎍/head/day subcutaneous treatment.

The Biomechanical Characteristics of Isoflavone-Treated Ovariectomized Rat's Femur (이소플라본을 투여한 난소절제 흰쥐 대퇴골의 생체역학적 분석)

  • This study was to investigate the biomechanical characteristics of isoflavone-treated ovariectomized growing rat's femur. 4-week-old Wistar (female rats were randomly divided into 5 groups and assigned to sham(SH) group, and four ovariectomized groups consisting of one ovariectomized(OVX) group and genisetin(10 mg/kg b.w. )(OVX+G), daidzein(10 mg/kg b.w.)(OVX+D), and 17 $\beta$-estradiol(10 $\mu$g/kg b.w.)(OVX+ES) in AIN-76 modified diet for 8 weeks. The OVX groups gained more body weight than the SH group. Femoral weight was increased by feeding genistein and estradiol, whereas femoral length among groups was not significantly different. The breaking farce, stiffness, deformation, and energy values of the OVX group were lower than those of other groups. The stress, strain, and Young's modulus values of the OVX group were higher than those of other groups.

The Effects of Artemisia Princeps var. Orientalis Extracts on Serum Lipids and Connective Tissues Collagen in Ovariectomized Rats (쑥이 갱년기 장애 유도 흰쥐의 혈중 지질 및 결합조직 중 Collagen 함량에 미치는 영향)

  • The aim of this study was to evaluate the effects of Artemisia Princeps Var. Orientalis (AP) on serum lipids and the collagen content of the connective tissues in ovariectomized rats. From day 2 until day 37 after the ovariectomy, Sprague-Dawley female rats were randomly assigned to the following groups: sham-operated rats (Sham), ovariectomized control rats (OVX-control), and ovariectomized rats supplemented with the AP 50 mg/kg bw/day (OVX-AP). The AP ethanol extracts were orally administrated 1 mL per day. The OVX rats were significantly heavier than the sham-operated rats at all time points, but supplementation with the AP extracts tended to gain weight less than OVX-control. Although total-cholesterol was increased at OVX-control, supplementation with the AP extracts tended to result in less than OVX-control. Triglyceride was significantly decreased after supplemented with the AP extracts (p<0.05). HDL-cholesterol is appeared higher AP extracts group than OVX-control. According to the results, we could know the fact that AP extracts were effective on serum lipids content throughout decreasing total-cholesterol, triglyceride and increasing HDL-cholesterol in ovariectomized rats. Supplementation with the AP extracts prevented a decrease in the collagen level in bone and cartilage tissues. These results are consistent with the conclusions based on the estrogenic activities of AP. Therefore, it may be used to possibly improve the quality of life in menopausal women.

Bone Mineral Density Changes after Ovariectomy in Rats as an Osteopenic Model : Stepwise Description of Double Dorso-Lateral Approach

  • Objective : This study describes a method for inducing osteopenia using bilateral ovariectomy (OVX), which causes significant changes in bone mineral density (BMD) in rats. Methods : Twenty-five 10-week-old female Sprague Dawley rats were used. Five rats were euthanized after two weeks, and BMD was measured in their femora. The other 20 rats were assigned to one of two groups : a sham group (n = 10), which underwent a sham operation, and an OVX group (n = 10), which underwent bilateral OVX at 12 weeks of age. After six weeks, five rats from each group were euthanized, and BMD was measured in their femora. The same procedures were performed in the remaining rats form each group eight weeks later. Results : The femur BMD was significantly lower in the six-week OVX group than in the six-week sham group, and in the eight-week OVX group than in the eight-week sham group. Conclusion : Bilateral OVX is a safe method for creating an osteopenic rat model. The significant decrease in BMD appears six weeks after bilateral OVX.

The Effects of Dietary Lipids and Ovariectomy on Bone Mass in Rats (식이지방과 종류와 난소절제가 흰쥐가 골질량에 미치는 양향)

  • This study examined the effect of dietary lipids and ovariectomy(OVX) on bone mass in rats. The experimental animals were divided into sham operation(S) or OVX(O) groups and then each group was divided into soybean oil(SS, OS), beef tallow(SB, OB), and fish oil(SF, OF) diet group. Experimental diets were fed for 16 weeks. Weight gain tended to be higher in OVX groups than in sham groups. Weigth gain was the highest in beef tallow group (SB) and the lowest in fish oil group(SF). There were no difference in weight, length, breaking force, strength and circumference of femur mong groups. However, volume of femur tended to be higher in OVX groups than in sham groups. Ash content of femur was significantly lower in OVX than sham groups but there were no difference between SB and OB group. Ca content in the soybean oil and beef tallow group were not influenced by OVX but Ca content in fish oil group was significantly higher in OF than SF group. The SB group showed the hyighest Ca content and the SF group showed the lowest Ca content. P content of femur was significantly lower in OVX groups than in sham groups but there were no difference between SB and OB group. The SS group showed the highest P content and the OS and OF group showed lower P content compare to other groups. From this result, it suggested that beef tallow positively influenced maintenance of bone mass by means of accumulation of body fat in ovariectomy.

Effects of the Herbal-Acupuncture with Acanthopanacis cortex extract at KI10 (Eumgok) on osteoporosis in ovariectomized DDY mice (음곡(陰谷)(KI10)에 시술한 오가피(五加皮) 약침(藥鍼)이 난소적출 생쥐의 골다공증(骨多孔症)에 미치는 영향)

  • Objective : The purpose of this study is to observe the effects of herbal-acupuncture with Acanthopanacis cortex extract (AC-HA) at KI10 (Eumgok) on osteoporosis in ovariectomized (OVX) ddy mice. Methods : We carried out several experimental items to analyze the changes in body weight, uterine weight, uterus index, tibial length, the ash bone weight, tibial BMD, serum ALP, serum creatinine, serum osteocalcin, serum Ca, and the levels of Ca, P, Ca/P ratio in tibia, and we performed histological and histomorphological analysis as well. Results : Tibial BMD was significantly decreased by OVX, while significantly increased by AC-HA at KI10. Serum creatinine level was significantly increased by OVX, while significantly decreased by AC-HA at KI10. Serum osteocalcin level was significantly increased by OVX, while significantly decreased by AC-HA at KI10. Tibial TBV(trabecular bone volume) was significantly decreased by OVX, while significantly increased by AC-HA at KI10. AC-HA at KI10 significantly suppressed the OVX-induced increase of GPL(growth plate length) of tibia in mice. Conclusion : These results suggest that AC-HA at KI10 has a therapeutic effect on osteoporosis in OVX mice.

Study of moxibustion stimulation in the ovariectomized rat model

  • In order to examine the effectiveness of moxibustion stimulation for climacteric disturbance, we administered moxibustion stimulation to ovariectomized (OVX) rats, and compared the tail surface temperature, laboratory parameters, and the level of locomotor activity with those of untreated rats. Twenty-four female Wistar rats (8 weeks old, 160 - 180 g) were divided into three groups. The rats in the OVX-M group underwent moxibustion stimulation after ovariectomy. The rats in the OVX-C group underwent ovariectomy but did not receive moxibustion stimulation. The rats in the Normal group received neither ovariectomy nor moxibustion stimulation. The level of locomotor activity was determined by a metabolism measuring system. The tail surface temperature was significantly lower in the OVX rats before moxibustion stimulation than in the Normal group. In the OVX rats before moxibustion stimulation, there was no clear difference in the level of locomotor activity between the active and resting phases, and the pattern of locomotor activity was irregular. After moxibustion stimulation, the tail surface temperature of the OVX-M group did not significantly differ from that of the Normal group, and the pattern of locomotor activity of the OVX-M group became diphasic with clear active and resting phases, similar to that observed in the Normal group. These results demonstrate that moxibustion stimulation is effective for the treatment of climacteric disturbance.
